Abstract:A new practical method for the synthesis of a non-cross-linked polystyrene supported 2-phenylimino-2-oxazolidine chiral auxiliary is reported. This method started from the same material as in a previous synthesis, N-Boc-L-tyrosine ethyl ester, but the overall yield was 42.5%, which was higher than the copolymerisation method.

Abstract:Herein, we report the first intramolecular C-N bond formation of phenylpropanamide derivatives via organocatalytic oxidative reactions, affording 3,3-disubstituted oxindole derivatives with up to 99% yield. The high efficiency of this reaction is exemplified by the transition metal-free mild conditions and the ability to perform the reaction on a gram scale. Meanwhile, the DFT calculation of the catalytic oxidative transformation pathway has also been studied.

Abstract:A wide range of isoquinoline ring-modified Quinap oxides with different steric and electronic variations have been constructed through a palladium-catalyzed imidoylative cyclization of arylethenyl isocyanides with 2-diphenylphosphinyl-1-naphthyl bromides. The Pd-catalysis plays dual roles in the formation of both axial C–C bond and isoquinoline ring. This de novo synthetic strategy features good functional group tolerance, high yields and easy scale-up, providing Quinap derivatives with substitution patterns that could not be obtained using coupling reactions. Chiral ligands 7 and 12 can be readily prepared by transformation of the resulting Quinap oxide to their BINOL esters, and have been proven to be superb chiral ligands for the copper-catalyzed enantioselective A3-coupling and alkynylation of quinoline reactions. In general, the enantioselectivies obtained using ligands 7 and 12 are excellent, and the ee values are higher than those using Quinap as ligand, even three times higher in some cases. Mechanism studies revealed that a monomeric copper(I) complex bearing a single chiral ligand was formed and served as the catalytically active species.

Abstract:An attractive and novel methodology involving Pd/Cu-catalyzed tandem head-to-tail dimerization/cycloisomerization of terminal ynamides for the synthesis of 3,5-disubstituted oxazolones was developed. Under Pd(PPh3)2Cl2/CuI cooperative catalyzed reaction conditions, it provided efficient access to 5-vinyloxazolones with exceptional functional group tolerance and good chemoselectivity. The control experiments demonstrated that Pd(PPh3)2Cl2 serves a key role in the dimerization of terminal ynamides and shows low catalytic activity in the intramolecular cyclization. Moreover, the hetero-Diels-Alder reaction of product 5-vinyloxazolones was also described, which provided polycyclic oxazolones in good yield.

Abstract:An efficient synthesis toward 2-alkyl substituted oxazoles has been achieved through Suzuki cross-coupling reaction. Treatment of various alkyl iodides with 9-MeO-BBN, followed by in situ palladium-catalyzed carbon–carbon formation with 2-iodooxazoles obtained the 2-alkyl substituted oxazoles in good to excellent yields. Regioselective C2-alkylation on 2,4-di-iodooxazole under the same reaction conditions was firstly disclosed.

Abstract:A Pd(II)-catalyzed aerobic intermolecular 1,2-diamination of conjugated dienes was developed for the regio- and chemoselective preparation of a variety of functionalized tetrahydroquinoxalines, using simple sulfonyl protected o-phenylendiamines as a nitrogen source. This methodology provides a direct and efficient synthesis of tetrahydroquinoxalines. O2 was used as the stoichiometric oxidant, and reaction conditions were applied to a series of o-phenylendiamines and conjugated dienes. 35 examples are described, and good yields and selectivities are obtained for the majority of the products.

Abstract:An efficient palladium(0)-catalyzed, Cu(I)-mediated synthetic route to trisubstituted olefins and conjugate dienes has been developed via oxo directing Liebeskind-Srogl cross-coupling reactions of gem-dihaloolefin-type alpha-oxo ketene dithioacetals with aryl and alkenylboronic acids. The synthetic protocol has demonstrated rare examples of transition-metal-promoted transformations of ketene dithioacetals, providing a novel route to highly functionalized conjugate dienes.

Abstract:Poly(2-oxazoline)s have received more and more attention in biomedical fields for their hydrophilicity and excellent biocompatibility. Poly(2-oxazoline)s can act as functional peptide mimics and promising substitutes for polyethylene glycol. Therefore, it is of great importance to develop new polymerization methods for the synthesis of functional poly(2-oxazoline)s with diverse structures. Herein, we report a controllable and facile synthesis of poly(2-oxazoline)s using trimethylsilyl trifluoromethanesulfonate (TMSOTf) as the initiator. This polymerization presents temperature-dependent characteristics, showing a decrease in polymerization time from a week to hours by increasing the reaction temperature. Based on TMSOTf-initiated polymerization, poly(2-oxazoline)s were obtained in a controllable way with expected chain length, variable side chain functional groups and narrow dispersities. This living polymerization on 2-oxazolines also enables facile synthesis of statistical and block copolymers of poly(2-oxazoline) and will contribute to the functional study and application of poly(2-oxazoline).
