Abstract:It aimed at isolating and bioinformatics analyzing the gene encoding a 10-deacetylbaccatin Ⅲ-10-O-acetyl transferase(DBAT)in Taxus chinensis var.mairei,a medicinal plant of the Chinese endemic genera.By using the primers which were designed according to the cDNA sequence deposited in the GenBank database,an RT-PCR cloning strategy was employed to amplify a full-length cDNA(designated as Tc-DBAT).The template of this amplication was the total RNA isolated from leaves of Taxus chinensis var.mairei.Tc-DBAT has an open reading frame(ORF) of 1 323 bp corresponding to a deduced protein of 440 residues and one intron was found in the genomic sequence.Tc-DBAT contained 5 predicted N-terminal acetylation site and two conserved N-glycosylation site in the N-terminal region,a conserved Big-1(bacterial Ig-like domain 1) domain profile,multi-phosphorylation sites and a Calponin-like repeat profile.Sequence homology comparison,phylogenetic analysis and advanced structures prediction all suggested that Tc-DBAT is a functional enzyme which belonging to the transferase superfamily.It catalyzes the acetylation of the C-10 hydroxyl group of the advanced metabolite 10-deacetylbaccatin Ⅲ(10-DAB) to yield baccatin Ⅲ,the last diterpene intermediate in the Taxol biosynthetic pathway.With successful cloning and characterization of related gene involved in biosynthesis of baccatin III,it may be feasible to augment the semisynthetic approaches in producing the immediate diterpenoid precursor of Taxol and,ultimately,to replace them entirely with the combinational expression system for enhanced Taxol production.
