Abstract:Abstract Elongation factor 1&agr; (EF-1&agr;) is an essential factor for protein synthesis in eukaryotes. Here, we demonstrated that Tetrahymena EF-1&agr; induced bundles of rabbit skeletal muscle F-actin as well as Tetrahymena F-actin in vitro, although Tetrahymena and skeletal muscle actins are different in some parts of their primary structures and in the binding abilities to some actin-binding proteins. Co-sedimentation experiments showed that the binding ratio of Tetrahymena EF-1&agr; to skeletal muscle F-actin in the bundles was 1:1 . Electron microscopic observation showed that alkaline pH or high ionic strength reduced the bundling activity of Tetrahymena EF-1&agr; to some extent, although the EF-1&agr; seemed to be able to induce bundling of the F-actin within the range of physiological condition.

Abstract:Ethyl carbamate (EC), widely found in alcoholic beverages, has been revealed to be a probable carcinogen in humans. Urethanase (EC 3.5.1.75) is an effective enzyme for the degradation of EC; however, the previously identified urethanases exhibited insufficient acid and alcohol resistance. In this study, an enantioselective amidase (AmdA) screened from Agrobacterium tumefaciens d3 exhibited urethanase activity with excellent alcohol resistance. AmdA was first overexpressed in Escherichia coli; however, the recombinant protein was primarily located in inclusion bodies, and thus, co-expression of molecular chaperones was used. The activity of AmdA increased 3.1 fold to 307 U/L, and the specific activity of urethanase with C-terminal His-tags reached 0.62 U/mg after purification through a Ni-NTA column. Subsequently, the enzymatic properties and kinetic constants of AmdA were investigated. The optimum temperature for AmdA was 55 °C, it showed the highest activity at pH 7.5, and the Km was 0.964 mM. Moreover, after 1 h of heat treatment at 37 °C in a 5-20% (v/v) ethanol solution, the residual urethanase activity was higher than 91%, considerably more than that reported thus far.
Biological Degradation of Aflatoxin B1 by Cell-Free Extracts of Bacillus velezensis DY3108 with Broad PH Stability and Excellent Thermostability

Abstract:(1) Background: Aflatoxin contamination in food and grain poses serious problems both for economic development and public health protection, thus leading to a focus on an effective approach to control it; (2) Methods: Aflatoxin B1 (AFB1) degrading bacteria were isolated using a medium containing coumarin as the sole carbon source, and the biodegradation of AFB1 by the isolate was examined by high performance liquid chromatography, and liquid chromatography mass spectrometry; (3) Results: a bacterial strain exhibiting strong AFB1 degradation activity (91.5%) was isolated and identified as Bacillus velezensis DY3108. The AFB1 degrading activity was predominantly attributed to the cell-free supernatant of strain DY3108. Besides, it was heat-stable and resistant to proteinase K treatment but sensitive to sodium dodecyl sulfate treatment. The optimal temperature for the maximal degradation of AFB1 was 80 °C. Even more notable, the supernatant showed a high level of activity over a broad pH (4.0 to 11.0) and exhibited the highest degradation (94.70%) at pH 8.0. Cytotoxicity assays indicated that the degradation products displayed significantly (p < 0.05) lower cytotoxic effects than the parent AFB1; (4) Conclusions: B. velezensis DY3108 might be a promising candidate for exploitation in AFB1 detoxification and bioremediation in food and feed matrices.

Some differentials in infant and child mortality risks in Pakistan 1962 - 1986.

Abstract: This paper draws upon a group of demographic surveys to provide direct and indirect estimates of infant and child mortality risks in Pakistan for the period 1962-1986. Differentials and trends in the estimates are subsequently explored. The 1962-65 Population Growth Estimation (PGE), the 1968-71 and 1976-79 Population Growth Surveys (PGS), the 1984-86 Pakistan Demographic Surveys, the 1975 Pakistan Fertility Surveys (PFS), the 1979-80 Population, Labor Force and Migration Survey (PLM), and the 1984-85 Pakistan Contraceptive Prevalence Survey were the sources used for the study. Results indicate that infant and child mortality rates are declining, with male-female differentials in child mortality risks also growing less marked. Both of these observations were more pronounced in urban areas. The importance of both mothers' education and improved rural living conditions in reducing the risk of child mortality is stressed in the paper.
Detoxification of aflatoxin B1 by Stenotrophomonas sp. CW117 and characterization the thermophilic degradation process

Abstract:Mycotoxins are high toxic, widely distributed contaminants in foodstuff. In this study, a aflatoxin B1 (AFB1) degrading strain S. acidoaminiphila CW117 was screened, and its detoxification characteristics were investigated. Substrate AFB1 at 45 μg/L was degraded by CW117 within 24 h; meanwhile, 4.1 mg/L AFB1 was almost degraded within 48 h. After 24 h degradation, the biotoxicity of the detoxified culture was eliminated. Strain CW117 efficient degradation to AFB1 (especially to low AFB1 concentrations) suggested its potential significance to detoxification development on food and feedstuff. The active degradation components present in the cell-free supernatant. The degradation ratio increased constantly with increasing incubation temperature raised (0-90 °C) and was even stable at 90 °C. Degradation was optimal at pH 6-7, and was only partially inhibited by metal-chelators (EDTA and EGTA), proteinase K, and a protein denaturant (sodium dodecyl sulfate, SDS). The recombinant laccase rLC1 (0.5 mg/mL) from CW117 degraded 29.3% of AFB1 within 24 h; however, the cell-free supernatant degraded 76.7% of the toxin in same time, with much lower protein content. The results indicated the CW117 degrades AFB1 via a combination of enzymes and micro-molecule oxides.

Abstract:Aflatoxin B1 (AFB1) is one of the most harmful mycotoxins, raising serious global health and economic problems. Searching for biological approaches for effective and safe AFB1 degradation is imminent. In our study, Microbacterium proteolyticum B204 isolated from bovine faeces degraded 77% of AFB1 after 24 hour, becoming the first reported bacteria from the Microbacterium family to possess AFB1 degradation characteristics. Temperature variation showed little effect on its degradation ratio, demonstrating high thermostability of 75% and 79% after boiling and sterilization, respectively. We suppose that the components playing a key role during this process were proteins, considering the decreased degradation rate caused by Proteinase K. Cell viability detection on HepG2 cells indicated that the degradation products were much less toxic than pure AFB1. Furthermore, B204 cell-free culture supernatant also degraded AFB1-contaminated food, such as peanuts, corn and cheese. These results suggested that this strain with AFB1 degradation properties could be a prospective candidate for application in the food and feed industries.

Abstract:The aim of this study is to determine the best aflatoxin B1 degradation conditions which was optimized using a combination of the Plackett-Burman and Box-Behnken methods with Panus neostrigosus culture filtrate. Panus neostrigosus was grown in a modified Kirk Broth medium to determine optimal degradation conditions. As a result, aflatoxin B1 was degraded under varying culture conditions. The Plackett-Burman method was designed after sixteen different experiments with fifteen variables. The three most effective variables (Sucrose, yeast extract, wheat bran) were chosen for the Box-Behnken methodology. The aflatoxin B1 degradation rate was 49% in just 1 h exposure to culture filtrate which was obtained under optimal growth conditions; (g-ml/L) sucrose 10, yeast extract 3, wheat bran 3, soytone 5, KH2PO4 2, MgSO4.7H2O 0.5, CaCl2.H2O 0.1, ammonium tartrate 2, trace element solution 10; 28 °C of incubation temperature, medium pH 5, 7.5% inoculum rate, 125 rpm of agitation speed, and a twelve-day incubation period. The SDS-PAGE studies show that the enzyme responsible for AFB1 degradation has 38 kDa molecular weight and has no laccase or MnP activity. To the best of our knowledge, this is the first report for AFB1 degradation by Panus neostrigosus.
Degradation and Detoxification of Aflatoxin B1 by Tea-Derived Aspergillus niger RAF106

Abstract:Microbial degradation is an effective and attractive method for eliminating aflatoxin B1 (AFB1), which is severely toxic to humans and animals. In this study, Aspergillus niger RAF106 could effectively degrade AFB1 when cultivated in Sabouraud dextrose broth (SDB) with contents of AFB1 ranging from 0.1 to 4 μg/mL. Treatment with yeast extract as a nitrogen source stimulated the degradation, but treatment with NaNO3 and NaNO2 as nitrogen sources and lactose and sucrose as carbon sources suppressed the degradation. Moreover, A. niger RAF106 still degraded AFB1 at initial pH values that ranged from 4 to 10 and at cultivation temperatures that ranged from 25 to 45 °C. In addition, intracellular enzymes or proteins with excellent thermotolerance were verified as being able to degrade AFB1 into metabolites with low or no mutagenicity. Furthermore, genomic sequence analysis indicated that the fungus was considered to be safe owing to the absence of virulence genes and the gene clusters for the synthesis of mycotoxins. These results indicate that A. niger RAF106 and its intracellular enzymes or proteins have a promising potential to be applied commercially in the processing and industry of food and feed to detoxify AFB1.

Abstract:Aflatoxins seriously threaten the health of humans and animals due to their potential carcinogenic properties. Enzymatic degradation approach is an effective and environmentally friendly alternative that involves changing the structure of aflatoxins. In this study, Trametes versicolor aflatoxin B1-degrading enzyme gene (TV-AFB1D) was integrated into the genome of Pichia pastoris GS115 by homologous recombination approach. The recombinant TV-AFB1D was expressed in engineering P. pastoris with a size of approximately 77 kDa under the induction of methanol. The maximum activity of TV-AFB1D reached 17.5 U/mL after the induction of 0.8% ethanol (v/v) for 84 h at 28 °C. The AFB1 proportion of 75.9% was degraded using AFB1 standard sample after catalysis for 12 h. In addition, the AFB1 proportion was 48.5% using AFB1-contaminated peanuts after the catalysis for 18 h at 34 °C. The recombinant TV-AFB1D would have good practical application value in AFB1 degradation in food crops. This study provides an alternative degrading enzyme for the degradation of AFB1 in aflatoxin-contaminated grain and feed via enzymatic degradation approach.

Abstract:Aflatoxin B1 (AFB1) contamination is a food safety issue threatening human health globally. Biodegradation is an effective method for overcoming this problem, and many microorganisms have been identified as AFB1-degrading strains. However, the response mechanisms of these microbes to AFB1 remain unclear. More degrading enzymes, especially of new types, need to be discovered. In this study, a novel AFB1-degrading strain, DDC-4, was isolated using coumarin as the sole carbon source. This strain was identified as Bacillus halotolerans through physiological, biochemical, and molecular methods. The strain's degradation activity was predominantly attributable to thermostable extracellular proteins (degradation rate remained approximately 80% at 90 °C) and was augmented by Cu2+ (95.45% AFB1 was degraded at 48 h). Alpha/beta hydrolase (arylesterase) was selected as candidate AFB1-degrading enzymes for the first time as a gene encoding this enzyme was highly expressed in the presence of AFB1. Moreover, AFB1 inhibited many genes involved in the nucleotide synthesis of strain DDC-4, which is possibly the partial molecular mechanism of AFB1's toxicity to microorganisms. To survive under this stress, sporulation-related genes were induced in the strain. Altogether, our study identified a novel AFB1-degrading strain and explained its response mechanisms to AFB1, thereby providing new insights for AFB1 biodegradation.

Screening and identification of an aflatoxin B1-degrading strain from the Qinghai-Tibet Plateau and biodegradation products analysis

Abstract:This research aimed to address the issue of aflatoxin B 1 (AFB 1 ) contamination, which posed severe health and economic consequences. This study involved exploring unique species resources in the Qinghai-Tibet Plateau, screening strains capable of degrading AFB 1 . UPLC-Q-Orbitrap HRMS and NMR were employed to examine the degradation process and identify the structure of the degradation products. Results showed that Bacillus amyloliquefaciens YUAD7, isolated from yak dung in the Qinghai-Tibet Plateau, removed 91.7% of AFB 1 from TSB-AFB 1 medium with an AFB 1 concentration of 10 μg/mL (72 h, 37°C, pH 6.8) and over 85% of AFB 1 from real food samples at 10 μg/g (72 h, 37°C), exhibiting strong AFB 1 degradation activity. Bacillus amyloliquefaciens YUAD7's extracellular secretions played a major role in AFB 1 degradation mediated and could still degrade AFB 1 by 43.16% after boiling for 20 min. Moreover, B. amyloliquefaciens YUAD7 demonstrated the capability to decompose AFB 1 through processes such as hydrogenation, enzyme modification, and the elimination of the -CO group, resulting in the formation of smaller non-toxic molecules. Identified products include C 12 H 14 O 4 , C 5 H 12 N 2 O 2 , C 10 H 14 O 2 , C 4 H 12 N 2 O, with a structure consisting of dimethoxyphenyl and enoic acid, dimethyl-amino and ethyl carbamate, polyunsaturated fatty acid, and aminomethyl. The results indicated that B. amyloliquefaciens YUAD7 could be a potentially valuable strain for industrial-scale biodegradation of AFB 1 and providing technical support and new perspectives for research on biodegradation products.

Effective degradation of aflatoxin B1 using a novel thermophilic microbial consortium TADC7

Abstract:We constructed a novel thermophilic microbial consortium, TADC7, with stable and efficient aflatoxin B1 (AFB1) degradation activity. The microbial consortium degraded more than 95% of the toxin within 72h when cultured with AFB1, and the optimum temperature was 55-60°C. TADC7 tolerated high doses of AFB1, with no inhibitory effects up to 5000μgL-1 AFB1; moreover, the degradation kinetics fit well with the Monod model. The proteins or enzymes in the TADC7 cell-free supernatant played a major role in AFB1 degradation. AFB1 degradation by the cell-free supernatant was stable up to 90°C, with an optimal pH of 8-10. We performed 16S rRNA sequencing to determine TADC7 community structure dynamics; the results indicated that Geobacillus and Tepidimicrobium played major roles in AFB1 degradation.
Detoxification of Aflatoxin B1 by a Potential Probiotic Bacillus amyloliquefaciens WF2020

Abstract:Microbial degradation is considered as an attractive method to eliminate exposure to aflatoxin B1 (AFB1), the most toxic mycotoxin that causes great economic losses and brings a serious threat to human and animal health, in food and feed. In this study, Bacillus amyloliquefaciens WF2020, isolated from naturally fermented pickles, could effectively degrade AFB1 ranging from 1 to 8 μg/ml, and the optimum temperature and pH value were 37–45°C and 8.0, respectively. Moreover, B. amyloliquefaciens WF2020 was considered to be a potential probiotic due to the synthesis of active compounds, absence of virulence genes, susceptibility to various antibiotics, and enhanced lifespan of Caenorhabditis elegans . Extracellular enzymes or proteins played a major role in AFB1 degradation mediated by B. amyloliquefaciens WF2020 into metabolites with low or no mutagenicity and toxicity to C. elegans . AFB1 degradation by the cell-free supernatant was stable up to 70°C, with an optimal pH of 8.0, and the cell-free supernatant could still degrade AFB1 by 37.16% after boiling for 20 min. Furthermore, B. amyloliquefaciens WF2020 caused a slight defect in fungal growth and completely inhibited AFB1 production when co-incubated with Aspergillus flavus . Additionally, B. amyloliquefaciens WF2020 suppressed the expression of 10 aflatoxin pathway genes and 2 transcription factors ( alfR and alfS ), suggesting that B. amyloliquefaciens WF2020 might inhibit AFB1 synthesis in A. flavus . These results indicate that B. amyloliquefaciens WF2020 and/or its extracellular enzymes or proteins have a promising potential to be applied in protecting food and feed from AFB1 contamination.

Abstract:Aflatoxin B1 (AFB1), a mycotoxin and natural carcinogen, commonly contaminates cereals and animal feeds, posing serious health risks to human and animal. In this study, Bacillus amyloliquefaciens ZG08 isolated from kimchi could effectively remove 80.93% of AFB1 within 72 h at 37 °C and pH 7.0. Metabolome and transcriptome analysis showed that metabolic processes including glycerophospholipid metabolism and amino acid metabolism were most affected in B. amyloliquefaciens ZG08 exposed to AFB1. The adaptation mechanism likely involved activation of the thioredoxin system to restore intracellular redox equilibrium. The key genes, tpx and gldA, overexpressed in Escherichia coli BL21, achieved degradation rates of 60.15% and 47.16% for 100 μg/kg AFB1 under optimal conditions of 37 °C and pH 8.0 and 45 °C and pH 7.0, respectively. The degradation products, identified as AFD1, were less cytotoxic than AFB1 in HepG2 cells. These findings suggest potential strategies for utilizing probiotics and engineered enzymes in AFB1 detoxification.

Abstract:High-T c A-15 Nb 3 Ge superconductors have been prepared by chemical vapour deposition. Superconducting transition temperatures, crystal structures, and growth morphology for different samples are reported here. Recombinant Oxidase from Armillaria tabescens as a Potential Tool for Aflatoxin B1 Degradation in Contaminated Cereal Grain

Abstract:Forage grain contamination with aflatoxin B1 (AFB1) is a global problem, so its detoxification with the aim of providing feed safety and cost-efficiency is still a relevant issue. AFB1 degradation by microbial enzymes is considered to be a promising detoxification approach. In this study, we modified an previously developed Pichia pastoris GS115 expression system using a chimeric signal peptide to obtain a new recombinant producer of extracellular AFB1 oxidase (AFO) from Armillaria tabescens (the yield of 0.3 g/L), purified AFO, and selected optimal conditions for AFO-induced AFB1 removal from model solutions. After a 72 h exposure of the AFB1 solution to AFO at pH 6.0 and 30 °C, 80% of the AFB1 was degraded. Treatments with AFO also significantly reduced the AFB1 content in wheat and corn grain inoculated with Aspergillus flavus. In grain samples contaminated with several dozen micrograms of AFB1 per kg, a 48 h exposure to AFO resulted in at least double the reduction in grain contamination compared to the control, while the same treatment of more significantly (~mg/kg) AFB1-polluted samples reduced their contamination by ~40%. These findings prove the potential of the tested AFO for cereal grain decontamination and suggest that additional studies to stabilize AFO and improve its AFB1-degrading efficacy are required.
Characterization and mechanism of aflatoxin degradation by a novel strain of Trichoderma reesei CGMCC3.5218

Abstract:Aflatoxins, which are produced mainly by Aspergillus flavus and A. parasiticus , are recognized as the most toxic mycotoxins, which are strongly carcinogenic and pose a serious threat to human and animal health. Therefore, strategies to degrade or eliminate aflatoxins in agro-products are urgently needed. We investigated 65 Trichoderma isolates belonging to 23 species for their aflatoxin B 1 (AFB 1 )-degrading capabilities. Trichoderma reesei CGMCC3.5218 had the best performance, and degraded 100% of 50 ng/kg AFB 1 within 3 days and 87.6% of 10 μg/kg AFB 1 within 5 days in a liquid-medium system. CGMCC3.5218 degraded more than 85.0% of total aflatoxins (aflatoxin B 1 , B 2 , G 1 , and G 2 ) at 108.2–2323.5 ng/kg in artificially and naturally contaminated peanut, maize, and feed within 7 days. Box–Behnken design and response surface methodology showed that the optimal degradation conditions for CGMCC3.5218 were pH 6.7 and 31.3°C for 5.1 days in liquid medium. Possible functional detoxification components were analyzed, indicating that the culture supernatant of CGMCC3.5218 could efficiently degrade AFB 1 (500 ng/kg) with a ratio of 91.8%, compared with 19.5 and 8.9% by intracellular components and mycelial adsorption, respectively. The aflatoxin-degrading activity of the fermentation supernatant was sensitive to proteinase K and proteinase K plus sodium dodecyl sulfonate, but was stable at high temperatures, suggesting that thermostable enzymes or proteins in the fermentation supernatant played a major role in AFB 1 degradation. Furthermore, toxicological experiments by a micronucleus assay in mouse bone marrow erythrocytes and by intraperitoneal injection and skin irritation tests in mice proved that the degradation products by CGMCC3.5218 were nontoxic. To the best of our knowledge, this is the first comprehensive study on Trichoderma aflatoxin detoxification, and the candidate strain T. reesei CGMCC3.5218 has high efficient and environment-friendly characteristics, and qualifies as a potential biological detoxifier for application in aflatoxin removal from contaminated feeds.

Abstract:Aflatoxin B1 (AFB1) contamination is an important issue for the safety of edible oils. Enzymatic degradation is a promising approach for removing mycotoxins in a specific, efficient, and green manner. However, enzymatic degradation of mycotoxins in edible oil is challenging as a result of the low activity and stability of the enzyme. Herein, a novel strategy was proposed to degrade AFB1 in peanut oil using an amphipathic laccase-inorganic hybrid nanoflower (Lac NF-P) as a biocatalyst. Owing to the improved microenvironment of the enzymatic reaction and the enhanced stability of the enzyme structure, the proposed amphipathic Lac NF-P showed 134- and 3.2-fold increases in the degradation efficiency of AFB1 in comparison to laccase and Lac NF, respectively. AFB1 was removed to less than 0.96 μg/kg within 3 h when using Lac NF-P as a catalyst in the peanut oil, with the AFB1 concentration ranging from 50 to 150 μg/kg. Moreover, the quality of the peanut oil had no obvious change, and no leakage of catalyst was observed after the treatment of Lac NF-P. In other words, our study may open an avenue for the development of a novel biocatalyst for the detoxification of mycotoxins in edible oils.

Abstract:Significant progress has been made in the biological degradation of aflatoxin B1 (AFB1), which is the most toxic and carcinogenic mycotoxin commonly found in food and feed products; however, research in this area has currently focused on the effective methods for detoxifying AFB1 and the mechanisms underlying the degradation process. The relationship between AFB1 biotransformation and the metabolic feedback by the AFB1-degrading strain has rarely been reported. This study investigated the biotransformation pathway of AFB1 in a highly efficient degrading strain, Aspergillus niger FS10, and the metabolic regulation of FS10 during AFB1 degradation. We analyzed the AFB1 metabolic degradation products and evaluated its toxicity based on its structures by triple quadrupole-linear ion trap-mass spectrometry (Q-Trap-MS) coupled with LightSight™ Software. Four metabolic degradation products were obtained: Product 1 (C17H14O6), Product 2 (C17H14O7), Product 3 (C16H12O5), and Product 4 (C27H31N3O13, AFB2-GOH). Two biotransformation pathways were proposed based on the structures of these degradation products. Metabolomics data revealed that FS10 exhibited different degrees of bioremediation under low-, medium- and high- dose AFB1. The results also suggested the significant involvement of glutathione, a metabolite, in the AFB1 biotransformation pathway and its formation of AFB2-GOH with AFB1. The four products could probably be less toxic than AFB1, as reflected by changes in the AFB1 toxicity site. Therefore, the AFB1-degrading strain FS10 exhibits great potential for application in the detoxification of AFB1 in food and feed products.
