Abstract:2-Amino-3-methyl pyridine was prepared with 2-cyano-methylpyridine as raw material via hydrolysis in dilute basic solution using H_2O_2 as oxidant and Hofmann degradation with NaBrO.The feasible reaction conditions were as follows:hydrolysis,(50°C),5% NaOH,3.5 h,10% of H_2O_2;Hofmann degradation,(60°C),12% NaOH,0.5 h.The overall yield was 74%(mol).The product obtained was identified by GC-MS、IR、()~1H NMR and elemental analysis.

Abstract:A phosgene- and metal-free synthesis of unsymmetrical arylurea derivatives utilizing 3-substituted dioxazolones and commercially available amines has been developed. The 3-substituted dioxazolones serve as the precursors of thein situgenerated isocyanate intermediates in the presence of non-toxic sodium acetate as a base and methanol as a solvent under mild heating conditions. A series of unsymmetrical phenylureas includingN, N'- mono-, di-, and trisubstituted derivatives are chemoselectively obtained in moderate to excellent yields from a broad scope of both 3-substituted dioxazolones and amines without a significant amount of the symmetrical byproducts. In many cases, the resulting unsymmetrical phenylureas are easily separated by filtration allowing no chromatographic purification. These investigations provide an opportunity for facile, practical, and eco-friendly synthesis of unsymmetrical diarylureas. Application of this method is demonstrated toward the gram-scale preparation of anti-cancer drugs, sorafenib and the 3-cycle continuous synthesis in conjugation with the recycle of sodium acetate and methanol process of known herbicide, daimuron.

Abstract:A new practical approach to prepare 1,3-(diethoxymethyl)-5-N,N-dimethylamino-6-methyluracil (5) has been realized from 6-methyluracil (1). Compound 5 is a key intermediate material in the synthesis of HIV-1 RT and methionine synthase inhibitors. 6-Methyluracil (1) reacted with HNO3-P2O5 giving 5-nitro-6-methyluracil (2), which alkylated at N-1,N-3 through reaction with ethoxymethyl chloride to give 3. Compound 3 was then reduced through treatment with HCOONH4-Pd/C and the derived C-5 amino group reacted with CH3I to convert into compound 5 in good yield.

Abstract:1-[Hexahydrocyclopenta[c]pyrrol-2(1H)-yl]-3-(4-methylbenzenesulfonyl)urea (Gliclazide) was synthesised over three steps from the starting material of cyclopentane-1,2-dicarboxylic anhydride. N-aminocyclopentane-1, 2-dicarboximide was obtained from the condensation reaction between the material and 80% hydrazine hydrate. The reduction of N-aminocyclopentane-1,2-dicarboximide and followed by the condensation with p-toluenesulfonyl urea generated Gliclazide. Every step was modified to be more simple and gave a 60% overall yield. The procedure is suitable for large-scale production.

Abstract:OBJECTIVE To find a commercially useful route for the preparation of 1-(2′-bromo-2′-deoxy-3′,5′-di-O-propionyl-β-D-ribosyl)thymine. METHOD To uses thymine and tetra-O-acylribofuranose as starting materials. The process involved the following steps: silylanization, condensation, deacetylation and halogenation. RESULTS The procedure was simple and the yield of this route was higher than other synthetic routes by using cheaper reagents.CONCLUSION The improved synthetic process is practical.

Abstract:Without the use of chemical oxidants and metal catalysts, an aqueous electrochemical synthesis of 5-trifluoromethyluracil in batch and continuous flow was reported. The green and economically friendly approach employing commercially available CF3SO2Na as a CF3 source and cheap (NH4)2SO4 as a supporting electrolyte afforded the 5-trifluoromethyluracil in 10 g scale in batch electrolysis. The continuous flow-electrolysis technique was employed to further facilitate the scale-up synthesis of 5-trifluoromethyluracil.

Abstract:Fluoroalkylated enaminones, such as trifluridine and 5-trifluoromethyluracil, have widespread applications in pharmaceuticals and agrochemicals. Although these kinds of pharmaceutical agent often bear CF3 and perfluoroalkyl motifs in the core structure, access to such analogues typically requires multi-step synthesis. Here, we report a mild, metal-free and operationally simple strategy for the direct perfluoroalkylation of uracils, cytosines and pyridinones through a visible-light induced pathway from perfluoroalkyl iodides. This photochemical transformation features synthetic simplicity, mild reaction conditions without any photoredox catalyst, and high functional group tolerance, providing a facile route for applications in medicinal chemistry.

Abstract:Abstract A variety of 2,3‐unsaturated‐O‐glycosides have been prepared by the Ferrier rearrangement of acetyl protected glycals under microwave irradiation using silica gel as an acid catalyst. Environmental friendliness, high yields, and short reaction times are the key features of this method. Furthermore, the method was applicable not only to the Ferrier reaction of 3,4,6‐tri‐O‐acetyl glucal and 3,4,6‐tri‐O‐acetyl galactal but also to the Ferrier reaction of 3,4‐di‐O‐acetyl arabinal.

Abstract:A new and efficient method has been reported for the synthesis of 2-amino-9-[4-acetoxy-3-(acetoxymethyl)butyl-1-yl]purine (famciclovir) starting from guanine. The route involves chlorination of guanine, optimized Mitsunobu reaction, coupling with diethyl malonate, hydrogenation, reduction and esterification, and the overall yield is about 29%. This method does not require any form of chromatographic purification to give pure famciclovir, and it is an industrially viable manufacturing process for this drug.

Abstract:A stereoselective radical-mediated hydrogermy- lation of the protected 5-ethynyluracil nucleosides with trialkyl-, triaryl,- or tris(trimethylsilyl)germanes gave (Z)-5-(2- germylvinyl)uridine, 2'-deoxyuridine, or ara-uridine as major products. Reaction of the β-triphenylgermyl vinyl radical intermediate with oxygen and fragmentation of the resulting peroxyradical provided also 5-(2-(triphenylgermyl)acetyl)- pyrimidine nucleosides in low to moderate yields. Thermal isomerization of the latter in MeOH occurred via a four-centered activated complex, and subsequent hydrolysis of the resulting O-germyl substituted enol yielded 5-acetyluracil nucleosides in quantitative yield.

Abstract:An efficient, diversity-orientated synthesis of 5-sulfone-substituted uracils was established. The use of protecting groups to synthesize sulfones from N-heterocycles was avoided. Various heterocycles were synthesized for the first time from favorable, easily accessible starting materials. Diversity-orientated syntheses are important for the medicinal chemistry of virostatics and chemotherapeutics. This approach provides a broad substrate tolerance and excellent yields of up to 98%.
