Abstract:According to the helix theory, in almost all structures of organic compounds exist helices. The helix structure is the basic reason of the molecular optical rotation. A left-hand helix will result in left rotation of the plane polarized light and a right-hand helix will result in right rotation of the plane polarized light. The total molecular optical rotatory power (and direction) depends on the algebraic sum of rotations of all helices existed in the molecular structure.Optical active cyclophane derivatives have special structures and can have a variety of changes. In this type of compound, designated P, M helix (plus or mines helix) is not the only helix existing in the molecule, even is not the helix which has the most substantial contribution to the optical rotation, so some particular analysis should be carried out. After carefully analyzing every helix existing in the structure, we can find their optical rotation behavior is similar to the behavior of all other kinds of optical active organic compounds, and comply well with those rules based on the helix theory. Thus, their optical rotation direction from the absolute configuration can be predicted. Conversely, their absolute configuration can be deduced from the optical rotation direction.

Abstract:A series of cyclic esters, which are optically active as a consequence of their helical structures, were synthesized to investigate the relationships between their structures and their optical activities. This paper reports the electron impact fragmentation mechanisms of these six cyclic esters. Accurate mass measurements and mass analyzed ion kinetic energy spectrometry confirmed fragmentation patterns. The stability of the fragment ions has a great influence on the fragmentation pathways, but no correlation with the optical activity was found.

Abstract:Despite the profound implications of chiral self-sorting in various fields such as biology, materials, catalysis, and supramolecular chemistry, the elucidation of intrinsic factors that affect the efficiency of this process remains a formidable challenge. Here, we present a theoretical analysis based on a recent example of a molecular cinquefoil knot. By introducing various amino acids into the chiral ligands, we probe the potential factors contributing to the absence of chiral self-sorting. Energy profile shows that the thermodynamic stability of the diastereoisomeric helicates is considerably influenced by the ratio and arrangement of chiral ligands. From our calculations, the interaction strength linked to chiral ligands exerts a greater influence on determining the chiral self-sorting outcome compared to structural or steric effects. A better understanding of the mechanism of chiral self-sorting formation of circular helicates could aid in the design of topologically chiral molecules with diverse functionalities. [GRAPHICS]

Abstract:The enantioseparation of 1-phenyl-1-ethanol 1-phenyl propanol and 2-phenyl-propionic acid methyl ester was studied on two different kinds of chiral columns-self-prepared cellulose tris(3,5-dimethylphenylcarbamate) (CDMPC), Pirkle's type chiral stationary phase (CSP) (S,S)-Whelk-O 1, (R,R)-DNB-DPEDA chiral columns. The influence of the alcohol modifiers in mobile phase and solute structure on enantioseparation was studied., The difference of chiral recognition mechanism for these compounds on CDMPC and ( S, S) Whelk-O 1, (R,R)-DNB-DPEDA columns was compared. The results indicate that on (S,S)-Whelk-O 1 chiral column and (R,R)-DNB-DPEDA chiral column, the primary interaction is attraction between solute and stationary phase, while on CDMPC the inclusion is the dominant chiral recognition mechanism for the solutes, and the solute molecular volume size and its geometric shape must play a principle role in meeting the requirement for a stereoselective fit into the cellulose chiral cavity, which may be critical to the chiral recognition of these three solutes on CDMPC CSP. The hydrogen-bond interaction is not important for the chiral recognition.

Abstract:Three series of cholesteryl esters with different linkages and terminal groups have been synthesised and characterised. All the esters except cholesteryl 6-bromohexanoate can exhibit thermotropic liquid crystalline phases in certain temperature regions. The insertion of a 4-phenoxyl and a 4-biphenylyloxy group in the ester can increase the molecular length, broaden the whole mesophase width and heighten the clearing temperature. The increasing molecular length can enforce the layered arrangement in series A and B (resulting in a monolayer smectic A phase), while destroying the layer stacking in series C (resulting in a suppressed or absent interdigitated smectic A phase). Meanwhile, the molecular structure played an important role in the molar helical twisting power and its temperature dependence.
Synthesis of a novel helical cyclophane using chiral binaphthyl as structural templates

Abstract:The chiral binaphthyls is one of the most important precursors for constructing optical molecules. In this paper, (R)- and (S)-2,2'-diethynyl-1,1'-binaphthyl with highly stable chiral configurations were employed as structural templates, and a new type of helical cyclophane compounds (R,P)-3 and (SM-3 were synthesized in enantiopure form by the introduction of benzyl sulfone through Sonogashira coupling reaction, followed by the treatment of disulfone with lithium diisopropylamide (LDA). MS, IR, H-1 and C-13 NMR as well as elemental analysis characterized compound 3. The enantiomeric relation of two isomers was reflected unambiguously by their circular dichroism spectra with exact mirror images and specific rotations [alpha](D) data.

Abstract:Based on (S)-ethynylpyrrolidine, propargylamine and propargyl alcohol, five monosubstituted ethynyl monomers, including (S)-N-[(4-chlorophenyl)carbamoyl]-2-ethynyl pyrrolidine (I), (S)-N-(4-chlor robenzoyl)-2-ethynyl pyrrolidine (II), 1-(p-chlorophenyl)-3-(2-propynyl)urea (III), p-chlorophenyl N-propargylcarbamate (IV) and propargyl N-(4'-chlorophenyl)carbamate (V) were synthesized. Optically active helical copolymers, I-50-ran-II50, I-80-ran-II20, I-95-ran-III5, I-95-ran-IV5 and I-95-ran-V-5, were prepared through coordination polymerization catalyzed by Rh(nbd)BPh4 of I with II to V, respectively. The effect of the structure and composition of the copolymers on the optical activity were studied, and the enantioseparation performance of the copolymers as coated chiral stationary phase toward 9 standard racemates were evaluated by high performance liquid chromatography. I-80-ran-II20 has the highest optical activity, showing better chiral recognition performance than other copolymers, and exhibited well chiral resolution ability for benzoin (alpha=1.40) and cobalt acetylacetonate (alpha= 1.88). The introduction of chiral monomer II has little effect on the helicity of the copolymer backbone, but obviously interferes with the asymmetric arrangement of side groups and reduces the enantioseparation performance. The introduction of achiral propargyl monomers significantly reduces the optical activity and enantioseparation of copolymers regardless of the bond orientation. [GRAPHICS] .
Unraveling the amplified chiroptical responses of rylene-bladed quintuple [6]helicenes

Abstract:Unraveling the key structural features to maximize the chiroptical properties is of significance for developing high-performance chiral materials. Here we present our first attempt to elucidate and understand the molecular design of excellent chiroptical properties via the combination of multiplicity and the alignment of subhelicenes. Two stereoisomeric PDI-bladed quintuple [6]helicenes, namely D 5 -CRP and C 2 -CRP , were revealed to show distinct spatial arrangements of subhelicenes. Circular dichroism (CD) spectra showed that the Cotton effects (Δ ε ) are reaching 1,412 mol −1 L cm −1 for D 5 -CRP and 669 mol −1 L cm −1 for C 2 -CRP in the visible spectrum. The greatly amplified Δ ε relative to the smaller analogue NPDH arises from the circular annulation of helicenes and high molecular symmetry that could significantly regulate the transition dipole moments and thereby make them tend to be (anti)parallel, as supported by TDDFT calculations for the rotatory strength ( R ). Consequently, the maximal dissymmetry factors (| g abs | and | g lum |) of this kind of chiral molecular carbon imides were estimated to be up to 0.021 and 0.012, respectively. This study provides a deep insight into the chiroptical properties of complicated chiral systems.

Abstract:The structure and clustering propensity of a chiral derivative of cis-1,2-cyclohexanediol, namely, 1-phenyl-cis-1,2-cyclohexanediol (cis-PCD), has been studied under supersonic expansion conditions by combining laser spectroscopy with quantum chemistry calculations. The presence of the phenyl substituent induces conformational locking relative to cis-1,2-cyclohexanediol (cis-CD), and only one conformer of the bare molecule is observed by both Raman and IR-UV double resonance spectroscopy. The homochiral preference inferred for the dimer formation at low enough temperature is in line with the formation of a conglomerate in the solid state. The change in clustering propensity in cis-PCD relative to trans-1,2-cyclohexanediol (trans-CD), which shows heterochiral preference, is explained by the presence of the phenyl substituent rather than the effect of cis-trans isomerism. Indeed the transiently chiral cis-CD also forms preferentially heterodimers, whose structure is very close to that of the corresponding trans-CD dimer.

Effects of Compositions of Chiral-Achiral N-Propargylamide Copolymers on Their Optical Activities

Abstract:An achiral and a chiral substituted acetylene monomer underwent copolymerizations smoothly,providing copolymers high yields(over 97%).The molecular weights of the copolymers are moderate(15000～26400).The obtained copolymers possessed the same composition with the corresponding monomer feed ratio.All the copolymers adopted stable helices under the investigated conditions.However,the composition of the copolymers not only influenced the helical structure of the polymer main chains and further their optical activity,but also exerted large effects on the screw sense of the helices in the polymer backbones.Accordingly,the variation in copolymer composition led to the change of circular dichroism(CD) signal in both intensity and sign.

Abstract:Background:: α-phenylethanol and its derivatives are important intermediates for the synthesis of a variety of chiral drugs. Method:: The interaction mechanism of the two enantiomers of α-phenylethanol with β-CD was investigated using 1H-NMR and ROESY. The loading of β-cyclodextrin (β-CD) as the host with the chiral drug intermediate α-phenylethanol as the guest was investigated using high-resolution NMR in D2O and quantum chemical calculations. Results:: The results showed that both α-phenylethanol enantiomers were able to enter into the hydrophobic cavity of β-CD and undergo enantiospecific interactions, while the combination of 2D ROESY and quantum chemical calculations showed that the benzene ring of both R and S α- phenylethanol was inserted into the β-CD cavity toward the small-port end. However, the most stable poses of the two enantiomers were different, so the benzene ring of the S-enantiomer was oriented more toward the small-port end of β-CD than that of the R-enantiomer, which was reflected in the signals of 2D ROESY, which were obviously different. Conclusion:: β-CD can enantioselectively recognize the α-phenylethanol enantiomers, and the 2D ROESY method is a very direct and powerful tool in the recognition process of chiral host and guest research.

Abstract:An experimental and theoretical study of structure and properties of chiral amphiphilic chlorin e6 13(N)-methylamide-15-methyl,17-alkyl esters (17-pentadecyl 1 and 17-phytyl 2 ) and their Ni complexes ( Ni-1 and Ni-2 ) and their helical twisting efficiency in liquid crystal were carried out. Quantum chemical simulation of their structure was performed, the adequacy of which was established by comparing the calculated and experimental UV/Vis, CD, 1 H and 13 C NMR spectra as well as optical rotation angles. Low values of g-factors of dissymmetry for macroheterocycle (MHC) with the phytol substituent were experimentally revealed. The reason for this effect, associated with the transfer of the chirality from the stereogenic centers of the substituent to macrocyclic chromophore in the associate, was established by quantum-chemical calculation. Using polarization microscopy, the formation of chiral nematic phases was detected by doping a mixture of cyanobiphenyls with macroheterocycles 1, 2, Ni-1 and Ni-2 . The values of the clearance temperature and helical pitch were measured, the slopes of phase diagrams β and helical twisting power (HTP) were calculated. The orientation order parameters of dopants in liquid crystal (LC) were determined by polarization spectroscopy. The dielectric properties of LC–MHC systems were studied within the chiral nematics and isotropic phases, and the values of the Kirkwood's correlation factors were calculated. Using quantum chemical simulation of MHC solvates, the features of the influence of aliphatic chains structure and complex formation on the physical properties of induced mesophases were established.

Abstract:A series of new optically active aromatic poly(ester amide)s containing a chiral group in the side chain prepared from the p-toluenesulfonic acid salt of o,o'-bis(leucyl)-hexanediol (TS-+LHD+TS-) and p-phthaloyl chloride and styrene-2,5-dicarbonyl chloride styrene have been synthesized by interfacial polymerization. The structure of the monomer is elucidated by FT-IR and elemental analysis. The thermal properties of the polymers were studied by DSC and TGA. The chiroptical properties of the above polymer have also been studied by circular dichroism (CD) spectroscopy. Results indicated that these polymers form helical structures.

Abstract:5 Series of chiral-achiral N-propargylamide copolymers were synthesized, providing copolymers with moderate number-average molecular weights in high Yields. For the copolymers front the achiral monomers with appropriate bulky pendent groups, the moderate steric hindrance between the pendent groups enabled the copolymer main chains to adopt helices preferring the same handed screw sense taken by the chiral units; the corresponding copolymers exhibited higher CD signal intensity even when compared with the pure chiral polymer. However, for the achiral co-monomers with too small or too large pendent groups, they were unfavorable for the resulting copolymers to prefer the helical handedness adopted by the chiral units I leading to low CD signal intensity. These findings would be helpful for us to design and synthesize novel helical N-propargylamide poly- and copolymers with high optical activity

Abstract:The conformations of O-uridine-5'-yl O-alkyl N-phosphoryl serine methyl esters (1) and (2) have been investigated by nuclear magnetic resonance (NMR) and circular dichroism (CD) techniques. Nuclear Overhauser effect (NOE) measurements and CD studies revealed that the glycosyl bonds in both compounds favor the syn orientation. The stereosensitive P-31-C-13 coupling constants were also measured and used for the analysis of the trans and gauche conformations. The three-dimensional structural characteristics of compounds (1) and (2) were discussed.

Abstract:This work explores the influence of helical elongation of symmetric oxa[n]helicenes on their structural, photophysical, and chiroptical features via experimental and computational analysis. A slight variation in the helical scaffold induced around a three‐fold increase in dissymmetry factors from|glum| = 0.0008 for oxa[7]helicene to|glum| = 0.0022 for oxa[9]helicene. The adjustment of the main helical scaffold in helicenes is a fundamental strategy for modulating their optical features, thereby enhancing their potential for diverse applications. This work explores the influence of helical elongation (n = 5–9) on the structural, photophysical, and chiroptical features of symmetric oxa[n]helicenes. Crystal structure analyses revealed structural variations with helical extension, impacting torsion angles, helical pitch, and packing arrangements. Through theoretical investigations using density functional theory (DFT) calculations, the impact of helical extension on aromaticity, planarity distortion, and heightened chiral stability were discussed. Photophysical features were studied through spectrophotometric analysis, with insights gained through time‐dependent DFT (TD‐DFT) calculations. Following optical resolution via chiral high‐performance liquid chromatography (HPLC), the chiroptical properties of both enantiomers of oxa[7]helicene and oxa[9]helicene were investigated. A slight variation in the main helical scaffold of oxa[n]helicenes from [7] to [9] induced an approximately three‐fold increase in dissymmetry factors with the biggest values of|glum| of oxa[9]helicene (2.2 × 10−3) compared to|glum|of oxa[7]helicene (0.8 × 10−3), findings discussed and supported by TD‐DFT calculations.

Abstract:The association between molecular chirality and helical characteristics known as the chirality-helicity equivalence is determined for the first time by quantifying a chirality-helicity measure consistent with photoexcitation circular dichroism experiments. This is demonstrated using a formally achiral SN 2 reaction and a chiral SN 2 reaction. Both the achiral and chiral SN 2 reactions possess significant values of the chirality-helicity measure and display a Walden inversion, i. e. an inversion of the chirality between the reactant and product. We also track the chirality-helicity measure along the reaction path and discover the presence of chirality at the transition state and two intermediate structures for both reactions. We demonstrate the need for the chirality-helicity measure to differentiate between steric effects due to eclipsed conformations and chiral behaviors in formally achiral species. We explain the significance of this work for asymmetric synthetic reactions including the intermediate structures where the Cahn-Ingold-Prelog (CIP) rules cannot be used.

Abstract:In this study, we explore the synthesis and solid-state characterization of four coumarin-3-carboxylic acid esters, each modified at the C-3 position with different cycloalkyl groups: cyclohexyl, menthyl, and iso-pulegyl. We conducted a detailed analysis of these compounds utilizing a variety of techniques such as a single-crystal X-ray diffraction, nuclear magnetic resonance (NMR), and Fourier-transform infrared (FTIR) spectroscopy. Additionally, we calculated the dipole moments for these molecules. Our findings include a thorough structural assessment, highlighting the role of noncovalent interactions through Full Interaction Maps and Hirshfeld surface analysis. This study reveals the critical influence of the weak C-H...O hydrogen bonds in determining the solid-state architecture of these esters, whereas π-π stacking interactions appear to be negligible among the studied derivatives.

Abstract:Copolymerizations of two novel N-propargylamide monomers [HC CCH2NHCOR, in M1, R: CH = CH2; in M2, R: CH2CH2CH2CH = CH2] with the other two N-propargylamide monomers (M3: achiral monomer; M4: chiral monomer), which were synthesized previously, were carried out in varied monomer feed ratios in the presence of (nbd)Rh+B-(C6H5)(4), with the aim to prepare optically active polymers simultaneously possessing helical polymer backbones and pendent polymerizable vinyl groups. Among the four homopolymers derived from M1-M4, polymer 1 and polymer 2 cannot dissolve completely in any of the usual solvents, while polymer 3 and polymer 4 are known to form stable helical conformations. The expected copolymers were obtained in high yields (>= 95%) and exhibited good solubility in several organic solvents. The obtained copolymers were found to adopt stable helical structures under the investigated conditions. The as-prepared helical copolymers bearing vinyl groups were further employed as macromonomers to copolymerize with N-isopropylacrylamide (NIPAM) in chloroform at 55 degrees C using 2,2'-azobis(isobutyronitrile) as an initiator and N,N'-methylenebisacrylamide as a cross-linking agent. The copolymerizations took place smoothly, quantitatively providing hydrogels containing helical polymer chains preformed in the first copolymerization step. The helical structures remained in the hydrogels according to circular dichroism spectroscopy measurements. The hydrogels preferentially adsorbed D-tryptophan and (R)-(+)-1-phenylethylamine in the corresponding two enantiomer pairs, demonstrating the potential applications of the novel optically active hydrogels in the areas of chiral recognition and chiral resolution.
