Abstract:Due to the structural similarity with sphinganine,sphinganine analogue mycotoxins（SAMT） fumonisin and AAL-Toxin execute their toxicity through the competitive inhibition of sphinganine N-acyltransferase.In this paper,the structure,biosynthesis,and toxicology of SAMT as well as the detection methods of SAMT and SAMT-producing strains,and the contamination of SAMT in agriculture products in different areas were reviewed.The ways to control the contamination of SAMT in agriculture products were also suggested.

Abstract:Fumonisins are mycotoxins produced by the fungus Fusarium verticillioides, which is a widespread pathogen of corn. The mycotoxins are known to cause fatal diseases in some domestic animals and have been linked to human esophageal cancer in China and South Africa. Here, we describe a simple method for direct and quantitative analysis of the toxins in food products. The method is based on high-performance liquid chromatography coupled with an evaporative laser scattering detector (HPLC–ELSD) without any prior derivatization of the samples. Using this method, we have analyzed corn-based food samples from central markets in eastern China. The results showed that FB1 was the main contaminant in the samples. The overall level of fumonisin contamination was relatively low, with a range of 0.25–1.80μg/g (mean 0.74μg/g) in 66.7% (16 of 24) of corn samples from Middle-eastern Area, 0.21–0.29μg/g (mean 0.24μg/g) in 28.6% (6 of 21) of corn samples from Northeastern Area, and 0.30–3.13μg/g (mean 0.47μg/g) in 30.0% (6 of 20) of corn samples from Southeastern Area.

Abstract:Abstract Mycotoxins are secondary metabolites produced by fungus. Many mycotoxin species are highly toxic and are frequently found in cereals and feedstuffs. So, powerful detection methods are vital and effective ways to prevent feed contamination. Traditional detection methods can no longer meet the needs of massive, real-time, simple, and fast mycotoxin monitoring. Rapid detection methods based on advanced material and sensor technology are the future trend. In this review, we highlight recent progress of mycotoxin rapid detection strategies in feedstuffs and foods, especially for simultaneous multiplex mycotoxin determination. Immunoassays, biosensors, and the prominent roles of nanomaterials are introduced. The principles of different types of recognition and signal transduction are explained, and the merits and pitfalls of these methods are compared. Furthermore, limitations and challenges of existing rapid sensing strategies and perspectives of future research are discussed.

Abstract:Mycotoxins are carcinogenic, teratogenic and mutagenic toxic compounds produced by some filamentous fungi, which are extremely harmful to corn, rice, wheat, peanut, soybean, rapeseed and other grain and oil crops, and seriously threaten environmental safety, food safety and human health. With the rapid increase in the global population and the expansion of the main crop planting area, mycotoxin contamination has increased year by year in agricultural products. The current review aimed to summarize the contamination status and harmful effects of major mycotoxins of grain and oil crops and the environmental factors that impact mycotoxin contamination. Further, control measures of mycotoxin contamination, especially the biological control strategies, were discussed.

Abstract:Food contamination with aflatoxins is the focus of current concern. The i ntermittent incidence of these toxins in agricultural commodities has negative effects on the economy of the affected regions where harvest and postharvest techniques for the prevention of mold growth are seldom practiced. Aflatoxins are highly hepatotoxic, hepatocarcinogenic,teratogenic, and mutagenic in nature and they contaminate a wide variety of important agricultural commodities, such as peanuts, maize, rice and cottonseed, before, during and after harvest in various environmental conditions/settings. The biosynthesis of aflatoxins in nature depends upon the various factors such as carbon, nitrogen, temperature, water activity,pH, developmental stages, oxidative stress, and plant metabolites. These toxins can cause several ailments and can be controlled by blocking and interfering with the absorption and disturbing the metabolic enzymes. The present article overviews the biosynthesis, infection, transport, metabolism and toxicity of aflatoxins. Detailed studies on their biosynthesis,mechanism of toxicity, structure-function relationship, metabolism and transport will provide deep insight into the technology involved in the development of an effective control measures for aflatoxins.

Abstract:Mycotoxins, which are secondary metabolites produced by toxicogenic fungi, are natural food toxins that cause acute and chronic adverse reactions in humans and animals. The genus Fusarium is one of three major genera of mycotoxin-producing fungi. Trichothecenes, fumonisins, and zearalenone are the major Fusarium mycotoxins that occur worldwide. Fusarium mycotoxins have the potential to infiltrate the human food chain via contamination during crop production and food processing, eventually threatening human health. The occurrence and development of Fusarium mycotoxin contamination will change with climate change, especially with variations in temperature, precipitation, and carbon dioxide concentration. To address these challenges, researchers have built a series of effective models to forecast the occurrence of Fusarium mycotoxins and provide guidance for crop production. Fusarium mycotoxins frequently exist in food products at extremely low levels, thus necessitating the development of highly sensitive and reliable detection techniques. Numerous successful detection methods have been developed to meet the requirements of various situations, and an increasing number of methods are moving toward high-throughput features. Although Fusarium mycotoxins cannot be completely eliminated, numerous agronomic, chemical, physical, and biological methods can lower Fusarium mycotoxin contamination to safe levels during the preharvest and postharvest stages. These theoretical innovations and technological advances have the potential to facilitate the development of comprehensive strategies for effectively managing Fusarium mycotoxin contamination in the future.

Abstract:The consumption of agricultural products contaminated with mycotoxins poses a significant threat to the health of both humans and animals. Maize frequently becomes contaminated with toxic fungi while it is still growing in the field. Therefore, more proactive measures should be implemented to reduce mycotoxin levels during the storage and processing of maize after harvest. This article analyzes the prevalent mycotoxins found in maize, specifically aflatoxins, ochratoxins, trichothecenes, fumonisins, and zearalenone. The study provides a comprehensive analysis of the occurrence of mycotoxins in maize during storage, as well as fate analysis of them during processing. It summarizes the impacts of storage time, environmental conditions, storage methods, and agricultural practices on mycotoxin occurrence during storage in the post-harvest period. Furthermore, the different distribution of mycotoxins across various fractions during both dry- and wet-milling processes in the post-harvest processing period is analyzed. Additionally, the strategies to control mycotoxins in maize are also proposed during the post-harvest period. This review offers valuable insights for future research on mycotoxin contamination in maize during the post-harvest period.

Abstract:Mycotoxins have substantial impacts on agricultural production and food preservation. Some have high similarities in bioactivity but subtle differences on structures from various fungal producers. Understanding of their complex cross-biosynthesis will provide new insights into enzyme functions and food safety. Here, based on structurally related mycotoxins, such as aurovertins, asteltoxin, and citreoviridin, we showed that methyltransferase (MT)-catalyzed methylation is required for efficient oxidation and polyketide stability. MTs have broad interactions with polyketide synthases and flavin-containing monooxygenases (FMOs), while MT AstB is required for FMO AstC functionality in vivo. FMOs have common catalysis on pyrone-polyene intermediates but different catalytic specificity and efficiency on oxidative intermediates for the selective production of more toxic and complex mycotoxins. Thus, the subtle protein interaction and elaborate versatile catalysis of biosynthetic enzymes contribute to the efficient and selective biosynthesis of these structure-related mycotoxins and provide the basis to re-evaluate and control mycotoxins for agricultural and food safety.
Occurrence of Mycotoxins in Feed Ingredients and Complete Feeds Obtained from the Beijing Region of China

Abstract:Background:The current study was carried out to provide a reference for the control of mycotoxin contamination in feed ingredients and complete feeds for swine.Methods:A total of 55 feed ingredients,including 14 corn,13 wheat bran,11 soybean meal and 17 dried distillers grains with solubles(DDGS) as well as 76 complete swine feeds including 7 creep feeds,14 starter feeds,14 grower feeds,18 grower-finisher feeds,10 gestating sow feeds,and 13 lactating sow feeds were randomly collected from15 swine farms located in the Beijing region of China from July to August 2011.Immunoaffinity clean-up,using High-Performance Liquid Chromatography(HPLC) in combination with UV or Fluorescence Detection,was used for quantitative analysis of aflatoxin B,(AFB,),deoxynivalenol(DON),zearalenone(ZEA) and ochratoxin A(OTA) in the ingredients and complete feeds.Results:DON and ZEA were the most prevalent mycotoxins found.DON was detected at percentages of 93,92,54,100 and 97%with a mean level of 1.01,0.44,0.05,1.36 and 0.65 ppm in the samples of corn,wheat bran,soybean meal,DDGS and complete feeds,respectively.The detected percentages of ZEA were 100,100,54,100 and 100 with mean levels of 109.1,14.9,9.2,882.7 and 58.9 ppb in the same samples.In the wheat bran and soybean meal samples,the content of all four mycotoxins were below the maximum limits set by Chinese regulations while the percentage of samples that exceeded regulatory limits were 7,57 and 7%for corn,and 7,14 and 3%for the complete feeds for AFB,,DON and OTA respectively.DDGS showed the most serious mycotoxin contamination and the percentage of samples that exceeded regulatory limits were 6,88 and 41%,for AFB,,DON and ZEA,respectively.Conclusions:This paper is the first to present data on the natural occurrence of AFB,,DON,ZEA and OTA in ingredients and complete feeds obtained from swine farms in China’s Beijing region.The data shows that feed ingredients and complete swine feeds obtained from these farms are most often contaminated with DON followed by contamination with A

Abstract:Mycotoxin contamination in food poses health hazards to humans. Current methods of controlling mycotoxins still have limitations and more effective approaches are needed. During the past decades of years, variable environmental factors have been tested for their influence on mycotoxin production leading to elucidation of a complex regulatory network involved in mycotoxin biosynthesis. These regulators are putative targets for screening molecules that could inhibit mycotoxin synthesis. Here, we summarize the regulatory mechanisms of hierarchical regulators, including pathway-specific regulators, global regulators and epigenetic regulators, on the production of the most critical mycotoxins (aflatoxins, patulin, citrinin, trichothecenes and fumonisins). Future studies on regulation of mycotoxins will provide valuable knowledge for exploring novel methods to inhibit mycotoxin biosynthesis in a more efficient way.

Abstract:Mycotoxins have the potential to enter the human food chain through carry-over of contaminants from feed into animal-derived products. The objective of the study was to develop a reliable and sensitive method for the analysis of 30 mycotoxins in animal feed and animal-derived food (meat, edible animal tissues, and milk) using liquid chromatography–tandem mass spectrometry (LC–MS/MS). In the study, three extraction procedures, as well as various cleanup procedures, were evaluated to select the most suitable sample preparation procedure for different sample matrices. In addition, timed and highly selective reaction monitoring on LC–MS/MS was used to filter out isobaric matrix interferences. The performance characteristics (linearity, sensitivity, recovery, precision, and specificity) of the method were determined according to Commission Decision 2002/657/EC and 401/2006/EC. The established method was successfully applied to screening of mycotoxins in animal feed and animal-derived food. Abstract:Mycotoxin contamination has become a challenge in the field of food safety testing, given the increasing emphasis on food safety in recent years. Mycotoxins are widely distributed, in heavily polluted areas. Food contamination with these toxins is difficult to prevent and control. Mycotoxins, as are small-molecule toxic metabolites produced by several species belonging to the genera Aspergillus, Fusarium, and Penicillium growing in food. They are considered teratogenic, carcinogenic, and mutagenic to humans and animals. Food systems are often simultaneously contaminated with multiple mycotoxins. Due to the additive or synergistic toxicological effects caused by the co-existence of multiple mycotoxins, their individual detection requires reliable, accurate, and high-throughput techniques. Currently available, methods for the detection of multiple mycotoxins are mainly based on chromatography, spectroscopy (colorimetry, fluorescence, and surface-enhanced Raman scattering), and electrochemistry. This review provides a comprehensive overview of advances in the multiple detection methods of mycotoxins during the recent 5 years. The principles and features of these techniques are described. The practical applications and challenges associated with assays for multiple detection methods of mycotoxins are summarized. The potential for future development and application is discussed in an effort, to provide standards of references for further research.

Abstract:Investigation and risk monitor were carried out on aflatoxin B1,ochratoxin,vomitoxin in maize for feedstuffs,Piglet feed,Fattening pig feed and sow feed.ELISA rapid detection kit was used to detect the 100batches of samples.The results showed that the detection rate of aflatoxin B1was 72.37%,standard rate was 4%.Ochratoxin did not exceed the standard,but the detection rate was 48.48%.Detection rate of vomitoxin was 96.97%,standard rate was 52%,which was the highest rate of pollution in the involved products.Therefore,vomitoxin is the most serious pollution toxin,aflatoxin B1is the second,ochratoxin is relative low in feed in Southern Shaanxi and Guanzhong area.

Abstract:Agricultural food commodities are highly susceptible to contamination by fungi and mycotoxins, which cause great economic losses and threaten public health. New technologies such as gamma ray irradiation, ultraviolet radiation, electron beam irradiation, microwave irradiation, pulsed light, pulsed electric fields, plasma, ozone, etc. can solve the problem of fungal and mycotoxin contamination which cannot be effectively solved by traditional food processing methods. This paper summarizes recent advancements in emerging food decontamination technologies used to control various fungi and their associated toxin contamination in food. It discusses the problems and challenges faced by the various methods currently used to control mycotoxins, looks forward to the new trends in the development of mycotoxin degradation methods in the future food industry, and proposes new research directions.

Analysis of Potential Fumonisin-Producing Fusarium Species in Corn Products from Three Main Maize-Producing Areas in Eastern China.

Abstract:BACKGROUND: Fusarium species are common fungal contaminants of maize and a number of them can produce mycotoxin fumonisins. China is one of the largest maize producers in the world. This study investigated the contamination of maize samples from three areas in eastern China by Fusarium and fumonisin-producing fungi as well as their fumonisin-producing potential. RESULTS: A total of 22 Fusarium strains were isolated, 19 of which were able to produce fumonisin. Among the 19 strains, 16 belonged to F. verticillioides, two to F. subglutinans and one to F. proliferatum. The majority (17/19) of the fumonisin-forming strains were high FB1 producers, which is a potential health risk for the population in these areas. Fusarium contamination in samples from the mideastern area was the most serious (11 Fusarium strains, with nine producing fumonisin, isolated from 24 samples), followed by the northeastern area (nine Fusarium strains, with all nine producing fumonisin, isolated from 21 samples) and the southeastern area (two Fusarium strains, with one producing fumonisin, isolated from 19 samples). CONCLUSION: Although the overall levels of FBs and contamination by fumonisin-producing fungi in corn samples were not serious, the contaminating Fusarium strains possessed fairly strong toxicogenic ability and potential risk for food safety. Abstract:Mycotoxin contamination is a globally concerned problem for food and agricultural products since it may directly or indirectly induce severe threats to human health. Sensitive and selective screening is an efficient strategy to prevent or reduce human and animal exposure to mycotoxins. However, enormous challenges exist in the determination of mycotoxins, arising from complex sample matrices, trace‐level analytes, and the co‐occurrence of diverse mycotoxins. Appropriate sample preparation is essential to isolate, purify, and enrich mycotoxins from complicated matrices, thus decreasing sample matrix effects and lowering detection limits. With the cross‐disciplinary development, new solid‐phase extraction strategies have been exploited and integrated with nanotechnology to meet the challenges of mycotoxin analysis. This review summarizes the advance and progress of solid‐phase extraction techniques as the methodological solutions for mycotoxin analysis. Emphases are paid on nanomaterials fabricated as trapping media of SPE techniques, including carbonaceous nanoparticles, metal/metal oxide‐based nanoparticles, and nanoporous materials. Abstract:An analytical method for the simultaneous determination of 13 mycotoxins in feed by magnetic dispersive solid-phase extraction combined with ultra-high performance liquid chromatography and tandem mass spectrometry was developed. The samples were extracted with acetonitrile/water (80:20, v/v, containing 3% acetic acid), and separated by centrifugation after salting-out, and then treated with magnetic adsorbents to remove interferences. The separation of target mycotoxins was performed on an ACQUITY UPLC HSS T3 column using a mobile phase consisting of 1 mmol/L ammonium acetate with 0.1% formic acid and methanol by gradient elution. Good linearities for the 13 mycotoxins were achieved with correlation coefficients over 0.99, and the recoveries of mycotoxins were in the range of 89.3-112.6% at spiking at levels of 5, 20, and 100 mu g/kg, with relative standard deviations of 0.9-10.4%. Based on the functional magnetic materials (MDN@Fe3O4, PSA@Fe3O4, ZrO2@Fe3O4) applied in dispersive solid-phase extraction, the pretreatment process is more convenient and it is beneficial to reduce the experimental cost by reusing the recycled magnetic materials. It is a simple, rapid, and environmentally friendly analytical method for the determination of mycotoxins in feed.
