Abstract:Abstract Mycotoxins are secondary metabolites of different species of fungi. Aflatoxin B 1 (AFB 1 ), deoxynivalenol (DON), zearalenone (ZEN) and fumonisin B 1 (FB 1 ) are the main mycotoxins contaminating animal feedstuffs. These mycotoxins can primarily induce hepatotoxicity, immunotoxicity, neurotoxicity and nephrotoxicity, consequently cause adverse effects on the health and performance of animals. Therefore, physical, chemical, biological and nutritional regulation approaches have been developed as primary strategies for the decontamination and detoxification of these mycotoxins in the feed industry. Meanwhile, each of these techniques has its drawbacks, including inefficient, costly, or impractically applied on large scale. This review summarized the advantages and disadvantages of the different remediation strategies, as well as updates of the research progress of these strategies for AFB 1 , DON, ZEN and FB 1 control in the feed industry.

Abstract:Mycotoxins are toxic secondary metabolites produced by different fungal species under specific environmental conditions. The common and regulated mycotoxins are such as deoxynivalenol (DON), zearalenone (ZEN), ochratoxin (OTA), aflatoxin B1 (AFB1), and fumonisins (FB). These mycotoxins are highly regulated in feed and food because their effects start to exert from their lowest exposures and are abundant in our common environment. However, there are other emerging mycotoxins such as apicidin, beauvericin, aurofusarin, and enniatins which are also harmful. Thus, making a total of around 500 forms of mycotoxins. The existence of mycotoxins in feed and food has a significant impact on animal and human health, which ultimately, slows down economic growth globally. According to this review, different approaches to removing multi-mycotoxin separately or simultaneously have been stated. Mostly, the review focused on the simultaneous removal of different multiple mycotoxins. This is because the current studies show a growing trend in reporting the co-existence of multiple mycotoxins in feed and food materials, however, most detoxifying approaches are for singular mycotoxins. Therefore, the physical, chemical, and biological approaches to remove multi-mycotoxin have been elucidated as well as their advantages and limitations. Furthermore, the authors give suggestions on the way forward to reduce exposure to mycotoxins and diminish their health effects in society. Lastly, the authors emphasized introducing more stringent limits for co-existing mycotoxins, especially those that have the same health effects by acting synergistically, such as AFB1 and OTA, which both act as carcinogenic agents.

Abstract:Mycotoxins are known worldwide as fungus-produced toxins that adulterate a wide heterogeneity of raw feed ingredients and final products. Consumption of mycotoxins-contaminated feed causes a plethora of harmful responses from acute toxicity to many persistent health disorders with lethal outcomes; such as mycotoxicosis when ingested by animals. Therefore, the main task for feed producers is to minimize the concentration of mycotoxin by applying different strategies aimed at minimizing the risk of mycotoxin effects on animals and human health. Once mycotoxins enter the production chain it is hard to eliminate or inactivate them. This paper examines the most recent findings on different processes and strategies for the reduction of toxicity of mycotoxins in animals. The review gives detailed information about the decontamination approaches to mitigate mycotoxin contamination of feedstuffs and compound feed, which could be implemented in practice.

Abstract:Mycotoxins are highly toxic metabolites produced by fungi that pose a huge threat to human and animal health. Contamination of food and feed with mycotoxins is a worldwide issue, which leads to huge financial losses, annually. Decades of research have developed various approaches to degrade mycotoxins, among which the biological methods have been proved to have great potential and advantages. This review provides an overview on the important advances in the biological removal of mycotoxins over the last decade. Here, we provided further insight into the chemical structures and the toxicity of the main mycotoxins. The innovative strategies including mycotoxin degradation by novel probiotics are summarized in an in-depth discussion on potentialities and limitations. We prospected the promising future for the development of multifunctional approaches using recombinant enzymes and microbial consortia for the simultaneous removal of multiple mycotoxins.

Abstract:Mycotoxins are secondary metabolites produced by fungi in food and feed, which can cause serious health problems. Bioenzymatic degradation is gaining increasing popularity due to its high specificity, gentle degradation conditions, and environmental friendliness. We reviewed recently reported biosynthetic mycotoxin-degrading enzymes, traditional and novel expression systems, enzyme optimization strategies, food and feed applications, safety evaluation of both degrading enzymes and degradation products, and commercialization potentials. Special emphasis is given to the novel expression systems, advanced optimization strategies, and safety considerations for industrial use. Over ten types of recombinases such as oxidoreductase and hydrolase have been studied in the enzymatic hydrolysis of mycotoxins. Besides traditional expression system of Escherichia coli and yeasts, these enzymes can also be expressed in novel systems such as Bacillus subtilis and lactic acid bacteria. To meet the requirements of industrial applications in terms of degradation efficacy and stability, genetic engineering and computational tools are used to optimize enzymatic expression. Currently, registration and technical difficulties have restricted commercial application of mycotoxin-degrading enzymes. To overcome these obstacles, systematic safety evaluation of both biosynthetic enzymes and their degradation products, in-depth understanding of degradation mechanisms and a comprehensive evaluation of their impact on food and feed quality are urgently needed.

Abstract:Mycotoxins have substantial impacts on agricultural production and food preservation. Some have high similarities in bioactivity but subtle differences on structures from various fungal producers. Understanding of their complex cross-biosynthesis will provide new insights into enzyme functions and food safety. Here, based on structurally related mycotoxins, such as aurovertins, asteltoxin, and citreoviridin, we showed that methyltransferase (MT)-catalyzed methylation is required for efficient oxidation and polyketide stability. MTs have broad interactions with polyketide synthases and flavin-containing monooxygenases (FMOs), while MT AstB is required for FMO AstC functionality in vivo. FMOs have common catalysis on pyrone-polyene intermediates but different catalytic specificity and efficiency on oxidative intermediates for the selective production of more toxic and complex mycotoxins. Thus, the subtle protein interaction and elaborate versatile catalysis of biosynthetic enzymes contribute to the efficient and selective biosynthesis of these structure-related mycotoxins and provide the basis to re-evaluate and control mycotoxins for agricultural and food safety.

Abstract:Abstract Mycotoxins are secondary metabolites produced by fungus. Many mycotoxin species are highly toxic and are frequently found in cereals and feedstuffs. So, powerful detection methods are vital and effective ways to prevent feed contamination. Traditional detection methods can no longer meet the needs of massive, real-time, simple, and fast mycotoxin monitoring. Rapid detection methods based on advanced material and sensor technology are the future trend. In this review, we highlight recent progress of mycotoxin rapid detection strategies in feedstuffs and foods, especially for simultaneous multiplex mycotoxin determination. Immunoassays, biosensors, and the prominent roles of nanomaterials are introduced. The principles of different types of recognition and signal transduction are explained, and the merits and pitfalls of these methods are compared. Furthermore, limitations and challenges of existing rapid sensing strategies and perspectives of future research are discussed.

Abstract:Mycotoxins are secondary metabolites produced during the growth, storage, and transportation of crops contaminated by fungi and are physiologically toxic to humans and animals. Aflatoxin, zearalenone, deoxynivalenol, ochratoxin, patulin, and fumonisin are the most common mycotoxins and can cause liver and nervous system damage, immune system suppression, and produce carcinogenic effects in humans and animals that have consumed contaminated food. Physical, chemical, and biological methods are generally used to detoxify mycotoxins. Although physical methods, such as heat treatment, irradiation, and adsorption, are fast and simple, they have associated problems including incomplete detoxification, limited applicability, and cause changes in food characteristics (e.g., nutritive value, organoleptic properties, and palatability). Chemical detoxification methods, such as ammonification, ozonation, and peroxidation, pollute the environment and produce food safety risks. In contrast, bioenzymatic methods are advantageous as they achieve selective detoxification and are environmentally friendly and reusable; thus, these methods are the most promising options for the detoxification of mycotoxins. This paper reviews recent research progress on common mycotoxins and the enzymatic principles and mechanisms for their detoxification, analyzes the toxicity of the degradation products and describes the challenges faced by researchers in carrying out enzymatic detoxification. In addition, the application of enzymatic detoxification in food and feed is discussed and future directions for the development of enzymatic detoxification methods are proposed for future in-depth study of enzymatic detoxification methods.

Abstract:Mycotoxins secreted by fungi or molds that infest agricultural products pose a serious threat to the safety and quality of plant-derived foods. Conventional food processing methods frequently face challenges in achieving ideal mycotoxin detoxification. Therefore, novel strategies inspired by non-thermal technologies and biological resources are emerging as promising alternatives. This review summarizes the recent contamination and toxicity manifestations of common mycotoxins in plant-derived foods. Furthermore, the underpinning mechanisms involved in emerging non-thermal technologies and current biological means for mycotoxin detoxification are comprehensively reviewed. It also elaborates on the research progress made with several mycotoxin removal strains (such as lactic acid bacteria, yeast, and other microorganisms), as well as specific enzymes like laccase. Drawing inspiration from practical experience, application limitations, and regulatory status with non-thermal treatments and biological resources for mycotoxin detoxification, priority should be given to integrating these two methods. Simultaneously, the review discusses the fundamentals, benefits, and drawbacks of this innovative strategy while identifying key directions for future research. This approach kills two birds with one stone in detoxifying mycotoxins from plant-derived foods. It offers valuable insights into nontoxic, eco-friendly, cost-effective, and targeted detoxification of mycotoxins in agricultural production, thereby laying the foundation for advancing mycotoxin detoxification techniques.

Abstract:Food contamination with aflatoxins is the focus of current concern. The i ntermittent incidence of these toxins in agricultural commodities has negative effects on the economy of the affected regions where harvest and postharvest techniques for the prevention of mold growth are seldom practiced. Aflatoxins are highly hepatotoxic, hepatocarcinogenic,teratogenic, and mutagenic in nature and they contaminate a wide variety of important agricultural commodities, such as peanuts, maize, rice and cottonseed, before, during and after harvest in various environmental conditions/settings. The biosynthesis of aflatoxins in nature depends upon the various factors such as carbon, nitrogen, temperature, water activity,pH, developmental stages, oxidative stress, and plant metabolites. These toxins can cause several ailments and can be controlled by blocking and interfering with the absorption and disturbing the metabolic enzymes. The present article overviews the biosynthesis, infection, transport, metabolism and toxicity of aflatoxins. Detailed studies on their biosynthesis,mechanism of toxicity, structure-function relationship, metabolism and transport will provide deep insight into the technology involved in the development of an effective control measures for aflatoxins.

Abstract:Mycotoxin contamination in food poses health hazards to humans. Current methods of controlling mycotoxins still have limitations and more effective approaches are needed. During the past decades of years, variable environmental factors have been tested for their influence on mycotoxin production leading to elucidation of a complex regulatory network involved in mycotoxin biosynthesis. These regulators are putative targets for screening molecules that could inhibit mycotoxin synthesis. Here, we summarize the regulatory mechanisms of hierarchical regulators, including pathway-specific regulators, global regulators and epigenetic regulators, on the production of the most critical mycotoxins (aflatoxins, patulin, citrinin, trichothecenes and fumonisins). Future studies on regulation of mycotoxins will provide valuable knowledge for exploring novel methods to inhibit mycotoxin biosynthesis in a more efficient way.

Abstract:Mycotoxin contamination is a globally concerned problem for food and agricultural products since it may directly or indirectly induce severe threats to human health. Sensitive and selective screening is an efficient strategy to prevent or reduce human and animal exposure to mycotoxins. However, enormous challenges exist in the determination of mycotoxins, arising from complex sample matrices, trace‐level analytes, and the co‐occurrence of diverse mycotoxins. Appropriate sample preparation is essential to isolate, purify, and enrich mycotoxins from complicated matrices, thus decreasing sample matrix effects and lowering detection limits. With the cross‐disciplinary development, new solid‐phase extraction strategies have been exploited and integrated with nanotechnology to meet the challenges of mycotoxin analysis. This review summarizes the advance and progress of solid‐phase extraction techniques as the methodological solutions for mycotoxin analysis. Emphases are paid on nanomaterials fabricated as trapping media of SPE techniques, including carbonaceous nanoparticles, metal/metal oxide‐based nanoparticles, and nanoporous materials. Detection of 13 mycotoxins in feed using modified QuEChERS with dispersive magnetic materials and UHPLC-MS/MS.

Abstract:An analytical method for the simultaneous determination of 13 mycotoxins in feed by magnetic dispersive solid-phase extraction combined with ultra-high performance liquid chromatography and tandem mass spectrometry was developed. The samples were extracted with acetonitrile/water (80:20, v/v, containing 3% acetic acid), and separated by centrifugation after salting-out, and then treated with magnetic adsorbents to remove interferences. The separation of target mycotoxins was performed on an ACQUITY UPLC HSS T3 column using a mobile phase consisting of 1 mmol/L ammonium acetate with 0.1% formic acid and methanol by gradient elution. Good linearities for the 13 mycotoxins were achieved with correlation coefficients over 0.99, and the recoveries of mycotoxins were in the range of 89.3-112.6% at spiking at levels of 5, 20, and 100 mu g/kg, with relative standard deviations of 0.9-10.4%. Based on the functional magnetic materials (MDN@Fe3O4, PSA@Fe3O4, ZrO2@Fe3O4) applied in dispersive solid-phase extraction, the pretreatment process is more convenient and it is beneficial to reduce the experimental cost by reusing the recycled magnetic materials. It is a simple, rapid, and environmentally friendly analytical method for the determination of mycotoxins in feed.

Abstract:Mycotoxins are a major threat to animal and human health, as well as to the global feed supply chain. Among them, aflatoxins, fumonisins, zearalenone, T-2 toxins, deoxynivalenol, and Alternaria toxins are the most common mycotoxins found in animal feed, with genotoxic, cytotoxic, carcinogenic, and mutagenic effects that concern the animal industry. The chronic negative effects of mycotoxins on animal health and production and the negative economic impact on the livestock industry make it crucial to develop and implement solutions to mitigate mycotoxins. In this review, we summarize the current knowledge of the mycotoxicosis effect in livestock animals as a result of their contaminated diet. In addition, we discuss the potential of five promising phytogenics (curcumin, silymarin, grape pomace, olive pomace, and orange peel extracts) with demonstrated positive effects on animal performance and health, to present them as potential anti-mycotoxin solutions. We describe the composition and the main promising characteristics of these bioactive compounds that can exert beneficial effects on animal health and performance, and how these phytogenic feed additives can help to alleviate mycotoxins' deleterious effects.

Abstract:Among microorganisms found in food, fungi stand out because they are adaptable and competitive in a large range of water activities, temperatures, pHs, humidities and substrate types. Besides sporulating, some species are toxigenic and produce toxic metabolites, mycotoxins, under adverse biotic and abiotic variables. Microorganisms are inactivated along the food chain, but mycotoxins have stable structures and remain in ready-to-eat food. The most prevalent mycotoxins in food, which are aflatoxins, fumonisins, ochratoxin A, patulin, tenuazonic acid, trichothecenes and zearalenone, have maximum tolerable limits (MTLs) defined as ppb and ppt by official organizations. The chronic and acute toxicities of mycotoxins and their stability are different in a chemical family. This critical review aims to discuss promising scientific research that successfully mitigated levels of mycotoxins and focus the results of our research group on this issue. It highlights the application of natural antifungal compounds, combinations of management, processing parameters and emergent technologies, and their role in reducing the levels and bioaccessibility. Despite good crop management and processing practices, total decontamination is almost impossible. Experimental evidence has shown that exposure to mycotoxins may be mitigated. However, multidisciplinary efforts need to be made to improve the applicability of successful techniques in the food supply chain to avoid mycotoxins' impact on global food insecurity.

Abstract:Ruminant diets include cereals, protein feeds, their by-products as well as hay and grass, grass/legume, whole-crop maize, small grain or sorghum silages. Furthermore, ruminants are annually or seasonally fed with grazed forage in many parts of the World. All these forages could be contaminated by several exometabolites of mycotoxigenic fungi that increase and diversify the risk of mycotoxin exposure in ruminants compared to swine and poultry that have less varied diets. Evidence suggests the greatest exposure for ruminants to some regulated mycotoxins (aflatoxins, trichothecenes, ochratoxin A, fumonisins and zearalenone) and to many other secondary metabolites produced by different species of Alternaria spp. (e.g., AAL toxins, alternariols, tenuazonic acid or 4Z-infectopyrone), Aspergillus flavus (e.g., kojic acid, cyclopiazonic acid or β-nitropropionic acid), Aspergillus fuminatus (e.g., gliotoxin, agroclavine, festuclavines or fumagillin), Penicillium roqueforti and P. paneum (e.g., mycophenolic acid, roquefortines, PR toxin or marcfortines) or Monascus ruber (citrinin and monacolins) could be mainly related to forage contamination. This review includes the knowledge of mycotoxin occurrence reported in the last 15 years, with special emphasis on mycotoxins detected in forages, and animal toxicological issues due to their ingestion. Strategies for preventing the problem of mycotoxin feed contamination under farm conditions are discussed.

Abstract:The global occurrence of mycotoxins is considered to be a major risk factor for human and animal health. Contamination of different agricultural commodities with mycotoxins still occurs despite the most strenuous prevention efforts. As a result, mycotoxin contaminated feed can cause serious disorders and diseases in farm animals. A number of approaches, such as physical and chemical detoxification procedures, have been used to counteract mycotoxins. However, only a few of them have practical application. A recent and promising approach to protect animals against the harmful effects of mycotoxin contaminated feed is the use of substances for reduction of the contamination of feed by mycotoxins. These substances, so-called mycotoxin binders (MB), are added to the diet in order to reduce the absorption of mycotoxins from the gastrointestinal tract and their distribution to blood and target organs, thus preventing or reducing mycotoxicosis in livestock. Recently, the use of such substances as technological feed additives has been officially allowed in the European Union. The efficacy of MB appears to depend on the properties of both the binder and the mycotoxin. Depending on their mode of action, these feed additives may act either by binding mycotoxins to their surface (adsorption), or by degrading or transforming them into less toxic metabolites (biotransformation). Biotransformation can be achieved by mycotoxin-degrading enzymes or by microorganisms producing such enzymes. Various inorganic adsorbents, such as hydrated sodium calcium aluminosilicate, zeolites, bentonites, clays, and activated carbons, have been tested and used as MB. An interesting alternative to inorganic adsorbents for the detoxification of mycotoxins is the use of organic binders, such as yeast cell wall components, synthetic polymers (cholestyramine, polyvinylpyrrolidone), humic substances and dietary fibres. This paper gives an overview of the current knowledge and situation in the field of MB. The most important types of MB, mechanism of their action, and their application as a part of general strategy to counteract mycotoxins are described in this review. Recent advances in the use and study of MB, as well as data of their in vitro and in vivo effectiveness are given. Problems, potential, current trends and perspectives associated with the use of MB are discussed as well in the review.

Abstract:Mycotoxins are toxic secondary metabolites generated from toxigenic fungi in the contaminated food and agro-food, which have been regarded as a serious threat to the food safety and human health. Therefore, the control of mycotoxins and toxigenic fungi contamination is of great significance and has attracted the increasing attention of researchers. As we know, nano-semiconductors have many unique properties such as large surface area, structural stability, good biocompatibility, excellent photoelectrical properties, and low cost, which have been developed and applied in many research fields. Recently, nano-semiconductors have also been promisingly applied in mitigating or controlling mycotoxins and toxigenic fungi contaminations in food and agro-food. In this review, the type, occurrence, and toxicity of main mycotoxins in food and agro-food were introduced. Then, a variety of strategies to mitigate the mycotoxin contamination based on nano-semiconductors involving mycotoxins detection, inhibition of toxigenic fungi, and mycotoxins degradation were summarized. Finally, the outlook, opportunities, and challenges have prospected in the future for the mitigation of mycotoxins and toxigenic fungi based on nano-semiconductors.
