Abstract:Adenosine has received great attentions acting as a potential biomarker for monitoring lung cancer. Most of the reported studies for adenosine detection require large instruments and complicated procedures. Herein, a sensitive, rapid and in-situ colorimetric aptasensor was developed for adenosine detection. Moreover, a homemade biomimetic electronic-eye (E-eye) was established and utilized as a portable in-time detection equipment. The entire measurement can be completed within 20 min, including the combination of aptamer with adenosine or AuNPs and the detection of adenosine. Four different kinds of aptamer were compared and the results showed that the AuNPs-aptamer-biotin system was the most stable and with the widest detection range of 5.0 mu M-60.0 mu M and the lowest LOD of 0.17 mu M. Moreover, the artificial urine samples were also tested with a linear range from 5.0 to 50.0 mu M and a LOD of 0.48 mu M. The results validated that the aptasensor together with the E-eye can be a promising platform for adenosine detection. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Adenosine can be a possible biomarker for monitoring the progress of lung cancer. Here, a sensitive and easily operated colorimetric methods based on aptasensor for the detection of adenosine has been developed. Gold nanoparticles (AuNPs) will aggregate in high-salt solutions, which causes color change, and single-stranded DNA will prevent this from happening. Based on changes in solution color, adenosine can be quantitatively determined. Besides a home-made bionic electronic-eye (E-eye) system was developed as a portable in-situ detection platform. Two different aptamers were used for comparison for adenosine detection. The linear correlation existed between 5.0-60.0μM with a detection limit of 0.17μM in water with modified aptamer. Moreover, the performance of the method was validated by testing the artificial urine samples with the linear range from 5.0 to 50.0μM and a detection limit of 0.48μM in artificial urine. This simple and cost-effective approach indicating its great potential for point-of-care applications.

Abstract:We report herein an exonuclease-assisted aptamer-based target recycling amplification strategy for sensitive and selective chemiluminescence (CL) determination of adenosine. This aptasensor is based on target-induced release of aptamers from capture probes immobilized on the 96-well plate surface, and thus leading to a decreased hybridization with gold nanoparticle-functionalized reporter sequences followed by a CL signal. The introduction of exonuclease III catalyzes the stepwise removal of mononucleotides from 3′-hydroxyl termini of duplex DNAs of aptamers, liberating the adenosine. Therefore, a single copy of target adenosine can lead to the release and digestion of numerous aptamer strands from the 96-well plates and ultimately an enhanced sensitivity is achieved. Experimental results revealed that the exonuclease-assisted recycling strategy enabled the monitoring of adenosine with wide working ranges and low detection limits (LOD: 0.5nM). This new CL strategy might create a novel technology for the detection of various targets and could find wide applications in the environmental and biomedical fields.

Abstract:We report a novel multimode detection aptasensor with three signal responses (i.e., fluorescence recovery, enhanced Raman signal, and color change). The presence of adenosine induces the conformational switch of the adenosine aptamer (Apt), forming adenosine-aptamer complex and releasing quantum dots (QDs) from AuNPs, resulting in the recovered fluorescence, the enhanced Raman signal, and color change of the solution. The multimode signal recognition is potentially advantageous in improving the precision and reliability of the detection in complex environments compared to the conventional single-mode sensing system. The multimode detection strategy opens up a new possibility in sensing and quantifying more other target molecules.

Abstract:Adenosine, as an endogenous nucleoside modulator, plays an important role in heart rate regulation, neurotransmission, and control of the respiratory system and thus it is significantly important to realize its sensitive detection. Herein, a highly sensitive electrochemical aptasensor for adenosine detection was proposed by using multi-walled carbon nanotubes (MWCNTs) as support matrix loading PtCu nanoparticles (PtCu-MWCNTs) to amplify signal. On one hand, disposable screen-printing gold electrodes (SPGEs) were used as superb sensing base to ensure the stable connection of aptamers 1 (ssDNA1). On the other hand, the PtCu-MWCNTs complex was synthesized through a one-pot method, which not only can precisely control the proportion of metal mass in the product but also exhibited superior electrocatalytic activity towards H2O2. The recognition reactions were achieved by stepwise incubation of ssDNA1, ssDNA2-PtCu-MWCNTs (denoted as ssDNA2-label), and adenosine on the SPGEs. As a result, the constructed electrochemical aptasensor exhibited a wide linear range from 10 nM to 1.0 μM with a low detection limit of 1.0 nM (S/N = 3) for adenosine detection. The aptasensor also successfully realized the adenosine detection in human serum samples, which means that the proposed aptasensor holds a potential application in point-of-care detection.

Abstract:Adenosine is an endogenous molecule that plays a vital role in biological processes. Research indicates that abnormal adenosine levels are associated with a range of diseases. The development of sensors capable of detecting adenosine is pivotal for early diagnosis of disease. For example, elevated adenosine levels are closely associated with the onset and progression of cancer. In this study, we designed a novel DNA biosensor utilizing chaperone copolymer-assisted catalytic hairpin assembly for highly sensitive detection of adenosine. The functional probe comprises streptavidin magnetic beads, an aptamer, and a catalytic chain. In the presence of adenosine, it selectively binds to the aptamer, displacing the catalytic chain into the solution. The cyclic portion of H1 hybridizes with the catalytic strand, while H2 hybridizes with the exposed H1 fragment to form an H1/H2 complex containing a G-quadruplex. Thioflavin T binds specifically to the G-quadruplex, generating a fluorescent signal. As a nucleic acid chaperone, PLL-g-Dex expedites the strand exchange reaction, enhancing the efficiency of catalytic hairpin assembly, thus amplifying the signal and reducing detection time. The optimal detection conditions were determined to be a temperature of 25 °C and a reaction time of 10 min. Demonstrating remarkable sensitivity and selectivity, the sensor achieved a lowest limit of detection of 9.82 nM. Furthermore, it exhibited resilience to interference in complex environments such as serum, presenting an effective approach for rapid and sensitive adenosine detection.

Abstract:In this paper, a simple, label-free and regenerative method was proposed to study the interaction between aptamer and small molecule by using methylene blue (MB+) as an electrochemical indicator. A thiolated capture probe containing twelve bases was firstly self-assembled on gold electrode by gold–sulfur affinity. Aptamer probe containing thirty two bases, which was designed to hybridize with capture DNA sequence and specifically recognize adenosine, was then immobilized on the electrode surface by hybridization reaction. MB+ was abundantly adsorbed on the aptamer probe by the specific interaction between MB+ and guanine base in aptamer probe. MB+-anchored aptamer probe can be forced to dissociate from the sensing interface after adenosine triggered structure switching of the aptamer. The peak current of MB+ linearly decreased with the concentration of adenosine over a range of 2×10−8–2×10−6M with a detection limit of 1×10−8M. In addition, we examined the selectivity of this electrochemical biosensor for cytidine, uridine and guanosine that belonged to the nucleosides family and possessed a similar structure with adenosine. The peak current of MB+ slightly decreased even then 1mM cytidine, uridine or guanosine was added to reaction cell for 2h, which confirmed this biosensor had a good selectivity for adenosine. After each of detection, the electrode was immersed into aptamer solution for regeneration. The recovery of the electrochemical signal of MB+ indicated the sensor surface of electrode was successfully regenerated.

Abstract:A simple and sensitive colorimetric aptasensor for rapid and facile detection of adenosine triphosphate (ATP) has been demonstrated here based on aptamer-based hairpin probes and unmodified gold nanoparticles (AuNPs). The hairpin probe is constructed by adding another five nucleotides to the 5′-end of an anti-ATP aptamer which can hybridize to nucleotides at the 3′-end of the aptamer, forming a hairpin-shaped structure. In the absence of ATP, the hairpin probes are rigid, and the AuNPs are susceptible to salt-induced aggregation. Conversely, upon binding with target ATP, the hairpin probes undergo conformational changes, forming aptamer-ATP complexes and exposing flexible ends which coat the surface of AuNPs to inhibit their aggregation in the high salt solution. Subsequently, a blue-to-red color change can be recognized by the naked eye. The aptasensor achieved selective responses toward ATP with a detection limit of 0.1μM, and exhibited high-quality detection performance in biological samples. In addition, this detection method is simple, rapid and cost-effective, holding great potential for further applications in point-of-care research.

Abstract:Adenosine triphosphate (ATP) is a small but significant biological molecule that plays an important role in regulating cellular metabolism processes. Accurate and sensitive analytical techniques for detecting ATP are urgently needed. Herein, a new electrochemical aptasensor was designed in light of DNA-templated copolymers that parallel to the electrode for sensitive gauging of ATP. The ATP aptamer decorated by the electronic medium ferrocene can be regarded as a.bridge. connecting two DNA-templated copolymers. When ATP exists, because of the extraordinary binding selectivity of DNA-templated copolymers for target ATP, the rapid electron transfer of ferrocene was beneficial to the sensitive detection of target ATP. Specifically, our parallel DNA copolymers are more stable than upright DNA copolymers and have a faster signal transmission because of the close distance to the electrode; meanwhile, the nonspecific pollution is less. Consequently, the developed novel aptasensor exhibited a wide range of linear response toward ATP that was from 0.1 to 10 000 pM and high selectivity with a detection limit down to 11.5 fM. The electrochemical aptasensor has a broad application prospect in the detection of small biological molecules.

Abstract:Aptamers, which are in vitro selected functional oligonucleotides, have been employed to design novel biosensors (i.e., aptasensors) due to their inherent selectivity, affinity, and their multifarious advantages over traditional recognition elements. In this work, we reported a multifunctional reusable label-free electrochemical biosensor based on an integrated aptamer for parallel detection of adenosine triphosphate (ATP) and alpha-thrombin, by using electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). A An electrode as the sensing surface was modified with a part DNA duplex which contained a 5'-thiolated partly complementary strand (PCS) and a mixed aptamer (MBA). The unimolecular MBA contained small-molecule ATP binding aptamer (ABA) and also protein alpha-thrombin binding aptamer (TBA). Thus, the aptasensor could be used for detection of ATP and alpha-thrombin both. The detection limit of ATP was 1 x 10(-8) M, and its detection range could extend up to 10(-4) M, whereas the detection limit of alpha-thrombin was 1 x 10(-11) M, and its detection range was from 1 x 10(-11) to 1 x 10(-7) M. Meanwhile, after detecting alpha-thrombin, the sensing interface could be used for ATP recognition as well. The aptasensor regeneration could be realized by rehybridizing of the MBA strand with the partly complementary strand immobilized on the Au surface after ATP detection or by treating with a large amount of ATP and then rehybridizing the MBA strand with the partly complementary strand immobilized on the An surface after alpha-thrombin detection. The aptasensor fabricated exhibited several advantages such as label-free detection, high sensitivity, regeneration, and multifunctional recognition. It also showed the detectability in biological fluid. Therein it held promising potential for integration of the sensing ability such as the simultaneous detection for multianalysis in the future.

Abstract:We designed a novel aptamer based biosensor (aptasensor) for ultrasensitive detection of adenosine triphosphate (ATP) through resonance energy transfer (RET). The ATP aptamer was modified with Cy3 at the 3' end, and a green quantumdot (525) was attached to the 5' end of its complementary sequence respectively. The ATP aptamer and its complementary sequence could assemble into a duplex structure in the absence of target ATP, and then decrease the distance between the quantum dot and Cy3 which could produce significant RET signal. Upon ATP binding, the ATP aptamer could dissociate with its complementary sequence and then increase the distance between the quantumdot and Cy3 which would significantly decrease the RET signal. Therefore, the ATP detection could be easily achieved through detection of the fluorescence intensity ratio between 525 nm and 560 nm. The results show that the emission fluorescence intensity ratio of 525/560 is linearly related to the logarithmic concentration of ATP. The linear range of this aptasensor is from 0.1 nM to 1 mu M, and the detection limit is lower down to 0.01 nM. Excellent selectivity of this aptasensor for ATP has been demonstrated through the detection of thymidine triphosphate (TTP), cytidine triphosphate (CTP), guanosine triphosphate (GTP) and adenosine diphosphate (ADP) respectively as control. The method we described here could easily detect ATP with excellent selectivity, linearity and sensitivity down to the nanomolar range, as well as avoid photobleaching.

Abstract:In this study, we fabricated adenosine imprinted poly(hydroxyethyl methacrylate-N-methacryloyl-L-cysteine methyl ester-silver nanoparticles (AgNPs)-N-methacryloyl-L-phenylalanine methyl ester) (MIP-Ag) sensor. It is a sensitive, simple, low-cost, and label-free adenosine determination in an aqueous solution without the need for any complicated coupling processes. The sensor without the addition of AgNPs (MIP) was prepared for control experiments to examine the effect of incorporated AgNPs to increase the signal response of surface plasmon resonance. Also, non-imprinted (NIP-Ag) sensor were designed using the same polymerization recipe without adding adenosine to evaluate the imprinting efficiency. The characterization studies of MIP-Ag, NIP-Ag and MIP sensors were carried out by atomic force microscopy and contact angle measurements. It was determined that the MIP-Ag sensor detected the target adenosine molecule 5.10 times more selectively than the competitor guanosine and 3.92 times more than thymidine. Imprinting efficiency was determined by comparing the signal response of MIP-Ag and NIP-Ag sensors. Repeatability studies of the MIP-Ag sensor were statistically analyzed for the 1.0 ppm adenosine, and the percent relative standard deviation of the intraday assays of less than 1.0% showed an insignificant decrease in the detection of adenosine after four adsorption-desorption cycles.

Abstract:We have developed a novel sensitive chemiluminescence (CL) aptasensor for the target assay as exemplified by using adenosine as a model target. In this work, we have demonstrated the signaling mechanism to make detection based on magnetic separation and 3,4,5-trimethoxyl-phenylglyoxal (TMPG), a special CL reagent as the signaling molecule, which reacts instantaneously with guanine nucleobases (G) of adenosine-binding aptamer strands. Briefly, amino-functioned capture DNA sequences are immobilized on the surface of carboxyl-modified magnetic beads, and then hybridized with label-free G-rich (including 15 guanine nucleobases) adenosine-binding aptamer strands to form our CL aptasensor. Upon the introduction of adenosine, the aptamer on the surface of magnetic beads is triggered to make structure switching to the formation of the adenosine/aptamer complex. Consequently, G-rich aptamer strands are forced to dissociate from magnetic beads sensing interface, resulting in a decrease of CL signal. The decrement of peak signal is proportional to the amount of adenosine. The effects of the amounts of capture DNA, aptamer, magnetic beads are investigated and optimized. It was found that the CL intensity had a linear dependency on the concentration of adenosine in the range of 4×10−7 to 1×10−5M. With a low detection limit of 8×10−8M and simplicity in CL detection, this novel technique will offer a great promise for future target/aptamer analysis.
Adenosine detection by using gold nanoparticles and designed aptamer sequences.

Abstract:Based on gold nanoparticles (AuNPs) and engineered DNA aptamers, we designed a novel bioassay strategy for the detection of adenosine as a small target molecule. In this design, an aptamer is engineered to consist of two pieces of random-coil like ssDNA which are respectively attached to AuNPs through their 50-thiol-modified end. They can reassemble into the intact aptamer tertiary structure and induce nanoparticle aggregation in the presence of the specific target. Results have demonstrated that gold nanoparticles can effectively differentiate these two different DNA structures via their characteristic surface plasmon resonance-based color change. With this method, adenosine can be selectively detected in the low micromolar range, which means that the strategy reported here can be applicable to the detection of several other small target molecules.

Abstract:A novel strategy for the fabrication of electrochemical aptasensor is proposed in this work, and the strategy has been employed to develop an aptasensor for the assay of adenosine deaminase activity. While a well-designed oligonucleotide containing three functional regions (an adenosine aptamer region, a G-quadruplex halves region, and a linker region) is adopted in our strategy as the core element, the enzymatic reaction of adenosine catalyzed by adenosine deaminase plays a key role as well in the regulation of the binding of the G-quadruplex halves with hemin, the electroactive probe, which is to reflect the activity of the enzyme indirectly but accurately. The detection limit of the fabrication biosensor can be lowered to 0.2 U mL(-1) of adenosine deaminase, and 1 nM of the inhibitor erythro-9-(2-hydroxy-3-nonyl) adenine hydrochloride is enough to present distinguishable electrochemical response. Moreover, since the electroactive probe is not required to be bound with the oligonucleotide, this strategy may integrate the advantages of both the labeled and label-free strategies.

Abstract:Graphene (GR) can strongly interact with aptamers due to hydrophobic and π–π stacking interactions between nucleotide bases and GR, but hardly interact with aptamer–target complexes or rigid double-stranded DNA. The unique GR/aptamers interaction has been fascinatingly developed in fluorescent assay, colorimetric biosensing and mass spectrometric analysis. However, exploration of the unique GR/aptamers interaction in electrochemical biosensing is still at an early stage. In this regard, we reported herein a novel, sensitive and relatively cushy approach to construct electrochemical aptasensors by immobilizing GR/aptamers complex on the surface of poly(o-phenylenediamine) (PoPD) modified glassy carbon electrodes (GCE). Adenosine triphosphate (ATP) was chosen as a model analyte and a well-studied 27-mer single-stranded DNA aptamer was tailor-made to have methylene-blue (Mb) at both ends for use as an electrochemical probe. The Mb-dual-labeled aptamers/GR/PoPD/GCE exhibited a strong electrochemical signal due to the high electrons transfer and large surface area of GR and using double Mb labeled at two ends of aptamers. The presence of ATP triggers the structure switching of aptamer to form aptamer–ATP complex, causing the destruction of the interaction between aptamer and GR and thus the release of Mb-dual-labeled aptamer from the GR, resulting in a low electrochemical signal. Under optimal conditions, we have successfully detected ATP down to a detection limit of 0.3nM with a wide detection range of 10nM–2mM. And from the experiment of cancer cellular ATP assay, it is proved that the method may have great prospects in the field of biological analysis.

Abstract:Aptamers, single-stranded nucleic acids that can selectively bind to various target molecules, have been widely used for constructing biosensors. A major challenge in this field, however, is direct sensing of analytes in complex biological media such as undiluted serum. While progress has been made in developing an inhomogeneous assay by using a preseparation step to wash away the interferences within serum, a facile strategy for direct detection of targets in homogeneous unprocessed serum is highly desired. We herein report a turn-on luminescent aptamer biosensor for the direct detection of adenosine in undiluted and unprocessed serum, by taking advantage of a terbium chelate complex with long luminescence lifetime to achieve time-resolved detection. The sensor exhibits a detection limit of 60 μM adenosine while marinating excellent selectivity that is comparable to those in buffer. The approach demonstrated here can be applied for direct detection and quantification of a broad range of analytes in biological media by using other aptamers.
A Novel Aptasensor Based on Silver Nanoparticle Enhanced Fluorescence

Abstract:In the present study, we report a novel aptasensor based on silver nanoparticle enhanced fluorescence for the detection of adenosine. First, the distance dependence nature of silver nanoparticle enhanced fluorescence was investigated through fluorescent dyes modified oligonucleotides to control the spacing distance between dyes and AgNP. The results showed that the fluorescence intensity reached the maximum value with the spacing distance of dyes about 8 nm from AgNP surface. The fluorescence intensity decreases when the spacing distance is either above or below this value. Based on this result, a fluorescence switch is constructed. In the "OFF" state, without the target molecules, there is a greater spacing distance between the Cy3 dyes and the AgNP giving comparatively lower fluorescence intensity. While in the "ON" state, in the presence of target molecules, the fluorescence signals increased for the conformation structure change of the aptamer which shorten the spacing distance between the Cy3 dyes and the AgNP to 8 nm. Using adenosine as target, the aptasensor produced a linear range from 200 nM to 200 μM with a correlation coefficient of 0.9949 and the detection limit was 48 nM estimated using 3σ. The aptasensor was also found to be specific in targeting adenosine. The presented method shows a new strategy of combining aptamer recognition and silver nanoparticle for fluorescence signal enhancement and increasing sensitivity.
Resonance Light Scattering Detection of Adenosine Based on Aptamer Functionalized Nanogold Probe Recognition

Abstract:Coupling nano-probe technology,aptamer recognition with resonance light scattering analysis,through recognizing reaction between adenosine and its specific aptamer in homogeneous matrix,causing the disaggregation of the aggregated network structure of adenosine specific aptamer functionalized nanogold structure,a sharply change on the resonance light scattering signal can be obtained.Based on it,a novel homogeneous aptamer recognizing based method for adenosine detection was established.The linear concentration range was 0.1～10μmol/L for adenosine with a detection limit of 0.04μmol/L(3S/N).The proposed method also shows good selectivity for adenosine detection and no interference from guanosine,thymidine,cytidine and uridine.
