Abstract:Two novel angular-type furanocoumarin glycosides, peucedanoside A (1) and peucedanoside B (2), along with a known compound apterin (3), were isolated from the roots of Peucedanum praeruptorum Dunn. Their chemical structures were determined by MS, NMR spectroscopy and chemical analysis. Complete assignments of the H-1 and C-13 NMR spectroscopic data were achieved by 1D and 2D NMR experiments including DEPT, HSQC, HMBC and ROESY. Copyright 2007 John Wiley & Sons, Ltd.

Abstract:The root of Peucedanum harry-smithii var. subglabrum was extracted with methanol, then separated with solvents at different polarity into four fractions: aqueous (H2O), ethyl acetate (AcOEt), chloroform (CHCl3) and petroleum ether (DAB-6). From AcOEt psoralen, bargapten, xanthotoxin, marmesin, umbelliferone, scopoletin, (+/-) peuformosin, Pd-I b, (+/-) selinidin, praeruptorin D were isolated by column chromatography on silica gel, using petroleum ether/ethyl acetate as eluent. The structures of the coumarins were identified by 1H-NMR and 13C-NMR.

Abstract:AIM:To study the chemical constituents of Peucedanum decursivum.METHODS:Various column chromatographies with silica gel and HPLC were employed for the isolation and purification. The structures of the compounds were elucidated on basis of spectral analyses and chemical methods.RESULTS:Seven compounds were isolated from the roots of Peucedanum decursivum (Miq.) Maxim. 1 is 3'(S)-hydroxy-4' (R)-angeloyloxy-3',4'-dihydroxanthyletin, named Decursitin D; 2 is 3'(S)-acetoxy-4'(R)-hydroxy-3',4'-dihydroxanthyletin, named decurstin F; the others are 3'(S)-acetoxy-4'(R)-angeloyloxy-3', 4'-dihydroxanthyletin (3), Pd-C-IV (4), Pd-C-II (5), (+)-3'S-decursinol (6) and (+)-trans-decursidinol (7).CONCLUSION:Compounds 1 and 2 are new compounds, while compounds 6 and 7 were isolated from Peucedanum decursivum (Miq.) Maxim for the first time.

Abstract:The phytochemical study of Peucedanum praeruptorum led to the isolation of twenty‐five coumarins (1‐25). Of which, (±) praeruptol A (±1), one pair of previous undescribed seco‐coumarin enantiomers were obtained. Their structures were established according to HR‐ESI‐MS, NMR, X‐ray single crystal diffraction analysis, as well as ECD calculation. All compounds were tested for anti‐inflammatory activity in the RAW264.7 macrophage model, and eight compounds (7‐10, and 13‐16) exhibited significant inhibitory effects with IC50 values ranging from 9.48 to 34.66 μM. Among them, compound 7 showed the strongest inhibitory effect, which significantly suppressed the production of IL‐6, IL‐1β, and TNF‐α, as well as iNOS and COX‐2 in a concentration‐dependent manner. Further investigated results showed that compound 7 exerted an anti‐inflammatory effect via the NF‐κB signaling pathway.

Abstract:Two minor coumarins, including a new compound qianhucoumarin J, were isolated from Peucedanum praeruptorum by a multi-step separation procedure using high-speed counter-current chromatography (HSCCC) and preparative high performance liquid chromatography (prep-HPLC) monitored by high performance liquid chromatography coupled with mass spectrometry (HPLC/MS) analysis. The crude extract was analyzed first by HPLC/MS to detect unknown compounds. The target compounds were enriched by HSCCC, using a two-phase solvent system of petroleum ether–ethyl acetate–methanol–water (5:5:6:4, v/v) and then isolated and purified by preparative HPLC monitored by HPLC/MS. The structure of the new compound was elucidated mainly by analysis of its 1D and 2D NMR spectral data. Its absolute configuration was determined by chemical degradation followed by chiral HPLC analysis.

Abstract:Eight compounds were isolated from the root of Peucedanum wawrii (Wolff) Su By means of physico-chemical data and spectra analysis (UV, IR, ~1HNMR, ~13CNMR), their structures were elucidated as Pd-1b (V), peucedanocumarin II (VI), d-laserpitin (VII), bergapten (IV), d-mannitol (I), hexacosanoic acid (III), beta-sitosterol (II) and daucosterol (VIIII). These compounds were found in this species for the first time.

Abstract:The accumulation of pyranocoumarins, the main active ingredient of Peucedanum praeruptorum Dunn, is strictly regulated by key enzyme genes in the coumarin biosynthesis pathway. However, little is understood about the correlation between the expression of key enzyme genes and the accumulation of coumarin content at various growth and development phases. In this work, a high-performance liquid chromatographic method and the real-time quantitative PCR system were prepared to establish the pyranocoumarins contents and the key enzyme expression in the roots, stems, and leaves of Peucedanum praeruptorum Dunn. The results showed that the coumarin content in the roots was the highest before blotting and was the lowest after flowering. Particularly, the content of Praeruptorin A, Praeruptorin B and Praeruptorin E was reduced to the lowest in roots after flowering. Consistent with the variation trend of the coumarin content in the stem, the relative expression of most key enzyme genes in the coumarin synthesis pathway was also significantly decreased. Especially for PpBMT gene, the relative expression in roots were extremely significantly decreased up to 90%. However, the variation trend of the coumarin content and the key enzyme genes expression was opposite in the roots and leaves. These results contribute to our knowledge of the process by which important enzymes control the production and accumulation of coumarin.
