The distribution of somatostatin precursor 1 and preproenkephalin mRNA in chicken intestinal nerve of Remak

Abstract:The intestinal nerve of Remak (INR) is unique to birds. Commencing at the jejunal-duodenal border, it traverses the mesentery and mesorectum and runs parallel to the gut until it terminates at the cloaca. In order to study the distributions of somatostatin precursor 1 (PSS1) mRNA and preproenkephalin (PPE) mRNA in chicken INR, sense and anti-sense digoxigenin (DIG) labeled RNA probes were prepared. Fragments of PSS1 and PPE genes were obtained by RT-PCR from the total RNA of chicken brains. Amplified cDNA fragments were subcloned into pGM-T easy vectors, and then the plasmids were transformed into E.coli DH5α and chosen by “white-blue plaque selection”. Recombinant SS1/pGM-T and PPE/pGM-T easy vectors which have been identified by sequencing were linearized with the restriction enzyme of Nco Ⅰ and Sal Ⅰ respectively. According to the protocol of “DIG RNA Labeling Kit (SP6/T7)”, sense and anti-sense DIG labeled RNA probes were producted by SP6 and T7 RNA polymerase respectively. Certificated by dot blot hybridization, both the sense and anti-sense RNA probes were prepared successfully. The distributions of PSS1 mRNA and PPE mRNA in the INR were examined by means of in situ hybridization histochemistry (ISHH). Many PPE mRNA and PSS1 mRNA positive neurons were found in the INR. Of these labeled neurons, 83.79±7.96 and 96.04±4.53 PPE mRNA positive neurons were distributed in juxta-jejunoileum and juxta-rectal portion of INR respectively; 86.98±7.93 and 86.07±6.11 PSS1 mRNA positive neurons were distributed in juxta-jejunoileum and juxta-rectal portion of INR respectively. These findings show that the PSS1 mRNA and PPE mRNA are co-expressed in the INR. Most of the fusiform or oval labeled neurons are distributed in the ganglia in small groups, some of the labeled neurons were also found in the nerve trunk between ganglia. PSS1 mRNA and PPE mRNA were transcribed in most of the INR neurons. The possible contribution of these positive neurons to the innervation of intestinal and oviductal wall is also discussed.
