Abstract:A novel route was employed to synthesize 8-phthalimido-3,6-dioxaoctan-1-ol(3) from triethylene glycol.The experimental results show that route Ⅱ is an economical one.The total yield of 8-amino-3,6-(dioxaoctanoic) acid(5) by route Ⅱ is 36%.

Abstract:A new, practical and cost-effective route for scalable synthesis of 2-[(4-dodecyloxyphenyl)sulfonyl] butanoic acid, a key intermediate of a new cyan dye-forming coupler containing a sulfone group, was developed by adopting phenol as the starting material. The synthesis was accomplished in five steps with etherification, chlorosulfonation, reduction, nucleophilic reaction by C-S coupling and hydrolyzation. An important objective of the new synthetic route was the synthesis of 2-[4-(dodecyloxyphenyl)sulfonyl]butanoate. Overall yield obtained at optimized conditions increased to 66 %. The synthetic strategy was proven to be a process enabling rapid delivery of the target product with high purity.

Abstract:A new route for the synthesis of 6-bromo-4,4-dimethylthiochroman was proposed and the optimized conditions for each step were obtained.Bromobenzene was selected as starting material and 6-bromo-4,4-dimethylthiochroman was prepared via chlorosulfonation,reduction,thioetherification and cyclization with the yield of 88.5%.Except the reduction,which had to be performed at a higher temperature,the other three steps proceeded at temperatures lower than 50oC.Thus both the energy cost and byproducts were decreased.Compared to previous routes,the proposed one in this work is advanced for its lower cost and less contamination,so it is suitable to be industrially amplified.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:Methyl 3-oxo-2,6,6-trimethylcyclohex-1-ene-1-carboxylate was synthesised over eight steps from the starting material of citral instead of α-ionone. KMnO4, HC(OEt)3 and O2/TEMPO/CuCl were substituted for NaIO4, CH3I and pyridinium chlorochromate, respectively. Every step was simplified and gave a 48% overall yield. The procedure is suitable for large scale production.

Abstract:1-Amino-3,6,9,12-tetraoxapentadecan-15-oic acid 8 was synthesised from tetraethylene glycol through a 7 step sequence including esterification, mesylation, azide substitution with subsequent reduction followed by hydrolysis. The structure of product 8 was identified by 1H and 13C NMR spectroscopy, elemental analysis and electrospray ionisation mass spectrometry (ESI-MS). All reaction conditions were optimised and easy to control. The key advantages of this process are the high yields of products and a new route to synthesise 1-amino-3,6,9,12-tetraoxapentadecan-15-oic acid.

Abstract:An asymmetric aldol strategy has been developed for the synthesis of L-daunosamine and D-ristosamine derivatives starting from noncarbohydrate precursors. Lithium and boron enolate mediated aldol reactions of 12 with O-TBS lactaldehyde gave non-Evans syn and Evans syn aldol products, respectively, with high selectivity. The chemical efficiency of the lithium enolate reactions were higher than the corresponding reactions with the boron enolates. Curtius rearrangement of lactone acids 23 and 26 gave the corresponding N-BOC amino lactones 30 and 32 in 64% and 62%, respectively, with complete retention of configuration. Lactone 30 was converted by a two-step sequence to N-benzoyldaunosamide 40. The overall yield for the amino sugar 40 was 18% over six steps. Similarly, lactone 32 was converted to N-benzoylristosamide 42 with an overall yield of 18% starting from 12.

Abstract:OBJECTIVE To study the synthetic method of 2-aminoxy-1- aminoethane dihydrochloride. METHOD The objective compound was synthesized by steps of ring-opening, condensation, hydrazinolysis and hydrolysis, starting from phthalic anhydride.RESULTS The chemical structure of synthetic product was confirmed by IR and 1HNMR, the total yield was amount to 39.4%. CONCLUSION This synthetic route is practical.

Abstract:Objective To prepare(2E,6E)-octadiene-3,7-dimethyldiol-1acetate.Methods The target compound was synthesized by five steps(acetylating,addition-elimination,replacement,oxidation and rearrangement)with geraniol.Results The desired product was obtained in over all yield of 49.7% and its structure was confirmed by 1H-NMR and IR.Conclusions A new route for the synthesis of (2E,6E)-octadiene-3,7-dimethyldiol-1acetate was explored.

Abstract:3-[(E)-5-(2,3,4,5-Tetr~ethoxy-6-methylphenyl)-3-methylpent-3-enyl]-2,2-dimethyloxirane (8) is a key intermediate for total synthesis of coenzyme QlO (1) and has previously been synthesized from p-cresol via bromination, methylation, Grignard reaction and epoxidation.'S2 However, this procedure is not attractive for the large-scale preparation of 8 due to drawbacks such as low yield (ca. 30% overall yield), low reaction temperature (-78°C). chromatographic separation and the use of several expensive and hazardous reagents. Therefore, a practical method for synthesis of 8 is desirable. Herein, we report a new efficient and convenient method for the preparation of 8 from commercially available starting material 2.

Abstract:To conquer disadvantages in existing synthetic routes of diethyl 4-amino-8-deazatetrafolate,just as high polarity and poor solubility of some intermediates as well as unsatisfying overall yield,a novel synthesis method with protecting groups introduced for 2,4-diamino groups was reported.Starting from 6-acetoxymethyl-2,4-dichloropyridopyrimidine,diethyl 4-amino-8-deazatetrafolate as a key intermediate of anti-folate inhibitors was obtained by a series of successive reactions including protection,condensation,reduction and debenzylation.Relatively shorter reaction time,higher yield and convenient work-up were achieved using this method.The regioselectivity of benzylation and reduction method of pyridine ring were also investigated.This novel synthesis route is of significance for synthesis of tetrahedronfolate analogues.

Abstract:The total synthesis of 3,7-dimethyl-7-hydroxy-2-octen-1,6-olide and (E)-6,7-dihydroxy-3,7-dimethyl-2-octenic acid were carried out though Wittig reaction, hydrolyzation, epoxidation and intramolecular cyclization or ring-opening in overall yields of 6.4% and 23.7% using 6-methyl-5-hepten-2-one as raw materials, and their structures were characterized by H-1 NMR, C-13 NMR and ESI-MS spectral data.

Abstract:6-Hydroxy-8-chlorooctanoate ethyl ester, an intermediate of α-lipoic acid, was synthesized via a chemoenzymatic method. High-throughput screening revealed that keto reductase HGD-1 could effectively catalyze the preparation of 6-hydroxy-8-chlorooctanoate ethyl ester, achieving a substantial conversion rate. The reaction conditions were optimized subsequently, the parameters for the complexation reaction were established as follows: a weight of AlCl 3 1.8 times that of the substrate; a reaction time of 4 h; a temperature range of 20–25 °C. The parameters for the enzyme-catalyzed reaction were as follows: a temperature range of 25–30 °C; a reaction time of 4 h; a solution pH of 6.5–7.5; a substrate concentration of 50 g/L; concentrations of keto reductase HGD-1, coenzyme glucose dehydrogenase, and nicotinamide adenine dinucleotide phosphate of 3 g/L, 4 g/L, and 0.05 g/L, respectively. Under these optimal conditions, the substrate conversion rate exceeded 95%, and 92% yield of 6-hydroxy-8-chlorooctanoate ethyl ester is obtained, suggesting a viable, eco-friendly method for synthesizing α-lipoic acid, and it provides a green process route for the industrial production of α-lipoic acid. Abstract:2,4-Dimethylthiophene-3-amine is an important intermediate of pesticides,2,4-Dimethylthiophene-3-amine was synthesized from thioglycolic acid and acetyl-acetone as starting material via seven steps reactons including nucleophilic addition,esterification,cyclization,hydrolysis,nitration,decarboxylation and reduction.The optimum reaction condition of esterification was studied,such as the kinds of and the quantity of catalyst,reaction time,the ratio of raw material and the time of azeotropic dehydration.structure of synthesized compound was confirmed by IR、1H NMR.compared to the traditional technics,this process can improve the yield and purity of the product.It is simple,warm and economical,which is suitable for industrial scale manufacture.

Abstract:The synthetic route of 6,8-diselenooctanoic acid was developed from ethyl 6,8-dichlorooctanoate via three steps,that is hydrolysis,selenylation and acidification,with water as a solvent and a small amount of ethanol as a co-solvent.The reaction process is carried out in a homogeneous phase,and the final product 6,8-diselenooctanoic acid can be directly precipitated from the aqueous phase,which was ever extracted with a lot of organic solvent according to literature.Using dripping time of sodium diselenide,reaction temperature and time after dripping off sodium diselenide as the influencing factors to carry out the orthogonal experiment,and the best reaction conditions were as follows:the dripping time was 2 h,reaction temperature was 70 ℃,reaction time was 3 h and the total yield was 73%.

Abstract:The total synthetic route of using the ring closure and acylation reactions,to synthesize the N-dichloroacetyl-5-chloro-2,3-dihydro-benzoxazole with 2-amino-4-chlorophenol,dichloromethane and dichloroacetyl chloride as raw materials was investigated.The factors affecting the yields of the intermediate 5-chloro-2,3-dihydro-benzoxazole and the title compound N-dichloroacetyl-5-chloro-2,3-dihydro-benzoxazole,such as the polarity of solvent and the species of attaching acid agent used,reaction temperature,stirring time,and the molar ratio of reactants,were studied by orthogonal experiments,and the results were discussed.The optimum experimental conditions for the preparation of the intermediate 5-chloro-2,3-dihydro-benzoxazole were found as following: N,N-dimethylacetamide is used as solvent,reaction temperature used is 90℃,stirring time is 1 h and NaOH is used as attaching acid agent.Under above conditions,the yield of 5-chloro-2,3-dihydro-benzoxazole is 72.6%.The optimum synthetic conditions found for preparing the title compound are as follows: N,N-dimethylacetamide is used as solvent,reaction temperature used is 20～25℃,stirring time is 2 h and the molar ratio of 5-chloro-2,3-dihydro-benzoxazole and dichloroacetyl chloride is 1:1.2;and the yield of the title compound could be 91.8%.The structures of the obtained compounds were characterized by IR,1H-NMR,13C-NMR and ultimate analysis.The proposed synthetic route is simple,the reaction conditions are mild and the raw materials are cheap and easy to be obtained.

Abstract:To increase the yield of(R)-α-lipoic acid(Ⅱ) and reduce its cost,an one-step synthesis of Ⅱ by using(R)-6,8-dichlorooctanoic acid(Ⅰ) as starting material was described.The synthesis steps involved chiral resolution of 6,8-dichlorooctanoic acid by(R)-α-phenylethylamine and substitution reaction in water phase.The optimized parameters were as follow.In the chiral resolution,n(6,8-dichlorooctanic acid)∶n((R)-α-phenylethylamine)=1∶0.48,and reaction temperature is 35 ℃;in the substitution reaction,n(Ⅰ)∶n(Na2S2)=1∶1.2,the reaction temperature is 75 ℃,and the dropping time keeps 3.0 h.Under the optimal conditions,the total yield of Ⅱ was 30.0%.When starting material 6,8-dichlorooctanic acid was expanded to 600 g and Ⅰ was increased to 222.0 g,the total yield of Ⅱ was up to 34.3%.
