Effect of advanced glycation end products of different concentration on the expression of Fractalkine in human tubular epithelial cells

Abstract:Objective It has been proved that advanced glycosylation end products(AGE) can upregulate the expressions of MCP-1 and RANTES in cultured human renal mesangial cells(HRMC),while the role of fractalkine(FKN) in diabetic nephropathy is not yet clear.The aim of the present study is to observe the effects of AGEs on the expression of FKN in HK-2 cells,a human tubular epithelial cell line.Methods HK-2 cells were treated with AGE-BSA at different doses for different times.FKN mRNA and protein expressions were analyzed by RT-PCR and ELISA.In addition,the HK-2 cells were cultured in AGE-BSA with or without the anti-FKN antibody.The transmigration and adhesion of U937 monocytes to HK-2 cells were detected with the Transwell system and fluorescent staining. Results Compared with the control groups,the levels of FKN mRNA and protein in the HK-2 cells incubated with AGE-BSA increased significantly in a dose-and time-dependent manner(P<0.05).The AGE-BSA group also showed a significantly larger number of U937 monocytes migrated and adhered to HK-2 cells than the control and anti-FKN antibody-blocking groups(P<0.01). Conclusion AGEs may be involved in the development of diabetic nephropathy partly via FKN.
