The effects of glucose with different concentration on MCP-1 expression and synthesis in rats mesangial cells

Abstract:Objective To investigate the significance of monocyte chemoattractant protein-1(MCP-1)in diabetic nephropathy by comparing the levels of MCP-1 which expressed and synthesized in rats mesangial cells(RMCs)with the different concentration of glucose in vitro.Methods RMCs,used as target cells,were divided into control group(5.6 mmol/L,group A),intermediate glucose group(17.5 mmol/L,group B)and high glucose group(30 mmol/L,group C).The concentration of MCP-1 in supernatants was determined by ELISA and the expression of MCP-1 mRNA in RMCs was detected by RT-PCR.Results The expressions of MCP-1 mRNA were poor in group A,which in group B and C were obviously higher than those in group A(all P<0.01),and in group C were further than those in group B at the same time(all P<0.01),except for 48 h;The expressions of MCP-1 protein in the supernatant persistently increased in group B and C by comparing with group A(all P<0.01),except for 12 h at group B.and which in group C were obviously higher than those in group B(all P<0.01).Conclusion The expression of MCP-1mRNA and protein synthesis in RMCs can be induced by the glucose in dose-and time-dependent manners and in the certain range.Elevated expressions of MCP-1mRNA and protein synthesis may be taken an important role in the progression of diabetic nephropathy.
