Abstract:We report a colorimetric sensor based on the supramolecular complex between a conformation-sensitive cationic polythiophene derivative, poly(3-(4-methyl-3-thienyloxy)propyltrimethylammonium) (PMTPA) and 2-naphthalenesulfonate (NS). It could be applied to discern 19 primary amino acids by the wavelength of PMTPA/NS. The sensing performance and mechanism was investigated by spectroscopic studies such as absorption and emission spectra. This approach relies on the disassembly of PMTPA/NS aggregates induced by the amphiphilic isoindole derivatives of amino acids formed in situ. 100% confidence limits for this discrimination could be achieved by using linear discriminant analysis (LDA).

Abstract:An innovative approach based on a surface functional monomer-directing strategy for the construction of a sensitive and selective molecularly imprinted electrochemical sensor for patulin recognition is described. A patulin imprinted platinum nanoparticle (PtNP)-coated poly(thionine) film was grown on a preformed thionine tailed surface of PtNP-nitrogen-doped graphene (NGE) by electro-polymerization, which provided high capacity and fast kinetics to uptake patulin molecules. Thionine acted not only as a functional monomer for molecularly imprinted polymer (MIP), but also as a signal indicator. Enhanced sensitivity was obtained by combining the excellent electric conductivity of PtNPs, NGE, and thionine with multisignal amplification. The designed sensor displayed excellent performance for patulin detection over the range of 0.002-2 ng mL(-1) (R-2 = 0.995) with a detection limit of 0.001 ng mL(-1) for patulin. In addition, the resulting sensor showed good stability and high repeatability and selectivity. Furthermore, the feasibility of its applications has also been demonstrated in the analysis of real samples, providing novel tactics for the rational design of MIP-based electrochemical sensors to detect a growing number of deleterious substances.

Abstract:Organic semiconductors have enough molecular versatility to feature chemo-specific electrical sensitivity to large families of chemical substituents via different intermolecular bonding modes. This study demonstrates that one single conducting polymer can be tuned to either discriminate water-, ethanol- or acetone-vapors, on demand, by changing the nature of its dopant. Seven triflate salts differ from mild to strong p-dopant on poly(3-hexylthiophene) sensing micro-arrays. Each material shows a pattern of conductance modulation for the polymer which is reversible, reproducible, and distinctive of other gas exposures. Based on principal component analysis, an array doped with only two different triflates can be trained to reliably discriminate gases, which re-motivates using conducting polymers as a class of materials for integrated electronic noses. More importantly, this method points out the existence of tripartite donor-acceptor charge-transfer complexes responsible for chemospecific molecular sensing. By showing that molecular acceptors can have duality to p-dope semiconductors and to coordinate donor gases, such behavior can be used to understand the role of frontier orbital overlapping in organic semiconductors, the formation of charge-transfer complexes via Lewis acid-base adducts in molecular semiconductors.

Abstract:Poly(4-vinylpyridine) was crosslinked and quaternized with 1,4-bromobutane to form a polyelectrolyte humidity sensitive film on an interdigitated gold electrode, which was further coated with a layer of polypyrrole by a facile method of vapor phase polymerization. The composite so prepared was characterized by UV–vis spectroscopy and scanning electron microscopy. The investigations on the humidity sensitive properties of the composite revealed that it exhibited an impedance as low as 105Ω even at 0%RH due to the existence of intrinsic conducting polypyrrole, thus conquering the difficulties in measuring low humidity with resistive-type humidity sensors. The impedance of the composite changed linearly with humidity in the range of 0–60%RH with good sensitivity. In addition, its response time (t90%) for adsorption and desorption between 33% and 97%RH was estimated to be 33s and 110s, respectively, and a hysteresis of 5%RH was observed. All these suggest it is promising as a sensitive material for low humidity detection. The effect of concentration and ratio of oxidizing agent to doping agent, polymerization temperature of pyrrole on the humidity sensitive properties of the composite have been investigated. A sensitive mechanism of the composite was proposed by taking into account the contribution of both the intrinsic electronic conduction and ionic conduction.

Abstract:The identification of phosphates holds significant importance in many physiological processes and disease diagnosis, and traditional detection techniques struggle to simultaneously detect and distinguish phosphates. The complexity of synthesizing sensing units restricts the construction of sensor arrays as well. In this study, a bifunctional dicopper chloride trihydroxide (Cu2Cl(OH)3) nanozyme with conspicuous laccase- and peroxidase-like activities has been synthesized in basic deep eutectic solvents (DES). Exploiting the various regulatory impacts of multiple phosphates on the dual-enzyme mimicking activities, the sensor array based on the laccase mimic and peroxidase mimic properties of Cu2Cl(OH)3 was designed, which has been successfully harnessed for the identification of eight phosphates (ATP, ADP, AMP, PPi, Pi, GTP, GDP, and GMP). This approach streamlines the creation of sensor arrays. Besides, the three simulated actual samples (healthy individuals, moderately ill patients, and severely ill patients) have been accurately distinguished. This work makes a substantial contribution to enhancing the highly effective construction of array channels and promoting discrimination of phosphates in intricate samples.
Oligonucleotide Discrimination Enabled by Tannic Acid-Coordinated Film-Coated Solid-State Nanopores

Abstract:Discrimination of nucleotides serves as the basis for DNA sequencing using solid-state nanopores. However, the translocation of DNA is usually too fast to be detected, not to mention nucleotide discrimination. Here, we utilized polyphenolic TA and Fe3+, an attractive metal-organic thin film, and achieved a fast and robust surface coating for silicon nitride nanopores. The hydrophilic coating layer can greatly reduce the low-frequency noise of an original unstable nanopore, and the nanopore size can be finely tuned in situ at the nanoscale by simply adjusting the relative ratio of Fe3+ and TA monomers. Moreover, the hydrogen bonding interaction formed between the hydroxyl groups provided by TA and the phosphate groups of DNAs significantly increases the residence time of a short double-strand (100 bp) DNA. More importantly, we take advantage of the different strengths of hydrogen bonding interactions between the hydroxyl groups provided by TA and the analytes to discriminate between two oligonucleotide samples (oligodeoxycytidine and oligodeoxyadenosine) with similar sizes and lengths, of which the current signal patterns are significantly different using the coated nanopore. The results shed light on expanding the biochemical functionality of surface coatings on solid-state nanopores for future biomedical applications.

Abstract:Functionalization of π-conjugated polymers is dispensable for solubilization of the rigid and hydrophobic backbones in water. However, polymer aggregation is always present and leads to issues with complication and reproducibility in spectral properties. Herein, we reported a simple and robust method to make a series of conjugated polymer nanostructures by a crosslinking strategy. In favor of multivalency effect, polythionenes with various chain lengths were functionalized with branched polyethylenimine (PEI) via ester aminolysis reaction under mild conditions. Photophysical studies revealed the conjugated backbones could be well stabilized and dispersed in water. By taking advantage of intermolecular recognition interaction between copper ions and cationic PEI, we applied crosslinked polythiophenes as a nano probe at very low concentration (0.01 mg/mL) to fluorescently detect copper ions with high sensitivity up to 10 nM and selectivity over other metal ions in aqueous solutions, without occurrence of detectable aggregates. The overall performance of our nano probes outperforms reported water-soluble polymers-based probes, particularly in probe availability and manipulation as well as selective copper detection capability.

Abstract:Abnormal photoluminescent properties of polyphenylphenyl compounds 6,7-dicyano-2,3-di-[4-(2,3,4,5-tetraphenylphenyl)phenyl]quinoxaline (CPQ) and 6,7-dimethyl-2,3-di-(4-(2,3,4,5-tetraphenylphenyl)phenyl)quinoxaline (MPQ) in aqueous solutions have been investigated. Photoluminescent emissions of CPQ and MPQ in tetrahydrofuran (THF)–water mixtures do not change monotonously with increasing their effective concentration in solution. This phenomenon is different from both aggregation-induced emission quenching and aggregation-induced emission enhancement, and can be ascribed to the results of combinational effects of intramolecular rotation, intermolecular hydrogen bonds, and solvent viscosity and hydration. We also observed that organic nanoparticles of CPQ and MPQ are formed in the aqueous solutions. Based on detection of the fluorescence of CPQ nanoparticles in aqueous solutions when introducing nucleobases, we report for the first time the application of organic nanoparticles for nucleobase sensing and found that CPQ nanoparticles can recognize the nucleobases with a sensitivity of guanine > adenine > thymine ≥ cytosine. These findings elucidate the photoluminescent behavior of polyphenylphenyl compounds in aqueous solutions and provide an approach to apply fluorescent organic nanoparticles for biosensing instead of metal and inorganic nanoparticles.

Abstract:ThT-E (ethyl-substitutive Thioflavin T), a derivative of Thioflavin T (ThT), is capable of inducing a guanine -rich nucleic acid sequence that forms a parallel G-quadruplex under certain conditions and exhibits significant fluorescence enhancement. In this work, we used Pb2+ to induce parallel G-quadruplex into anti-parallel G-quadruplex to reduce fluorescence of ThT-E and found a simple Pb2+ detection method. Through this approach, Pb2+ in aqueous solutions can be detected at 0. 76 nM with fluorescence spectrometry in a facile way, with high selectivity against other metal ions. These results indicate the method for detecting Pb2+ is simple, sensitive, specific and label-free.

Abstract:Simple, rapid, and sensitive technologies to detect nucleic acid modifications have important applications in genetic analysis, clinical diagnosis, and molecular biology. Because genetic modifications such as single nucleotide polymorphisms (SNP), DNA methylation, and other lesions can serve as hallmarks of human disease, interest in such methods has increased in recent years. This Account describes a new strategy for the optical detection of these DNA targets using cationic conjugated polymers (CCPs).Because of their unique signal amplification properties, researchers have extensively investigated conjugated polymers as optical transducers in highly sensitive biosensors. Recently, we have shown that cationic polyfluorene can detect SNPs within the DNA of clinical samples. When we incorporated deoxyguanosine triphosphate (dGTP-FI) into the DNA chain at an SNP site where the target/probe pair is complementary, we observed higher fluorescence resonance energy transfer (FRET) efficiency between cationic polyfluorene and fluorescein label on the dGTP. By monitoring the change in emission intensity of cationic polyfluorene or fluorescein, we identified the homozygous or heterozygous SNP. The high sensitivity of this assay results from the 10-fold enhancement of fluorescein emission intensity by the FRET from polyfluorene. This method can detect allele frequencies (the proportion of all copies of a gene that is made up of a particular gene variant) as low as 2%. Using this novel method, we clearly discriminated among the SNP genotypes of 76 individuals of Chinese ancestry.Improving on this initial system, we designed a method for multicolor and one-tube SNP genotyping assays based on cationic polyfluorene using fluorescein-labeled deoxyuridine triphosphate (dUTP-FI) and Cy3-labeled deoxycytidine triphosphate (dCTF-Cy3) in extension reactions. We also developed a one-step method for direct detection of SNP genotypes from genomic DNA by combining allele-specific PCR with CCPs. In 2008, we developed a new method for DNA methylation detection based on single base extension reaction and CCPs. Treatment of DNA with bisulfite followed by PCR amplification converts unmethylated DNA into a C/T polymorphism, which allows us to characterize the methylation status of the target DNA. Furthermore, we used CCPs to detect DNA lesions caused by ultraviolet light irradiation for the first time. By monitoring the color change of cationic polythiophene before and after DNA cleavage, we also detected oxidative damage to DNA by hydroxyl radical.These CCP-based new assays avoid primer labeling, cumbersome workups, and sophisticated instruments, leading to simpler procedures and improved sensitivity. We expect that these features could lead to major advances in human disease diagnostics and genomic study in the near future.

Abstract:Nucleotide hybridization technology is an effective method for achieving optical-based selective recognition of nucleotides. Unfortunately, the optimal work performance is always restricted by the lack of the coupling method used to bind sensors with nucleotide sequences. In this work, a series of nucleotide hybridization detection experiments based on optical fiber surface plasmon resonance (SPR) are conducted, aimed at comparing the ability to bind nucleotide of Polyethylenimine (PEI) and Poly-L-Lysine (PLL), which are common coupling agents in optical-based nucleotide hybridization study. In our work, qualitative nucleotide measurements using PEI and PLL are carried out in positive order, negative order, and temperature fluctuation, and confirm PEI has a higher work performance in nucleotide-binding and nucleotide-detection. Quantitative determination in the concentration range of 0–100 nmol/L demonstrates the DNA sensitivity reaches 0.13 nm/(nmol/L) and the detection limit (DL) lies in 4.9 nmol/L, which is outstanding compared with similar optical sensors. In this paper, two kinds of nucleotide molecule detection methods based on optical fiber SPR sensing technology with PEI and PLL are proposed and experimentally verified, and the nucleotide-binding capability of PLL and PEI is contrast analyzed according to experimental results for the first time. This study is of great significance for improving the measurement scheme of DNA sensors and the selection of gene carriers for manufacturing DNA complexes.

Abstract:It is of great significance to propose simple methods to detect DNA bases sensitively for biological analysis and medical diagnosis. Herein, a highly crystalline polyimide covalent organic framework (TAPM-COF) has been successfully synthesized via a solvothermal route using pyromellitic dianhydride (PMDA) and tris(4-aminophenyl) amine (TAPA), which possessed large specific surface area (2286 m 2 g −1 ) and excellent thermal stability. Intriguingly, the crystallinity of the TAPM-COF improved significantly with the increase of water content in the reaction medium. To verify this phenomenon, we synthesized TPPM-COF with two pores by pyromellitic dianhydride (PMDA) and N,N,N′,N'-tetrakis(4-aminophenyl)-1,4-benzenediamine (TPDA), which bonding was similar to TAPM-COF. Furthermore, the prepared TAPM–COF–0.3 was used to construct a novel and independent electrochemical biosensor on glassy carbon electrode for simultaneously determination of adenine (A) and guanine (G) without other additives. However, to further improve signal of TAPM-COF in the field of electrochemical sensors, the crystalline TAPM–COF–0.3 can be readily integrated with amino-functionalized multiwalled carbon nanotubes (NH 2 -MWCNT) to form core-shell TAPM–COF–0.3@NH 2 -MWCNT driven by a π–π stacking interaction for more sensitive electrochemical sensing toward purine bases. In comparison to TAPM-COF/GCE, the TAPM-COF@NH 2 -MWCNT/GCE exhibited more favorable linear range and lower limit of detection. The work provided a new strategy for amplifying signal of COF in the field of electrochemical sensors.

Abstract:Polythiophene (PT) derivatives are in the spotlight as functional materials with their conductivity and optical properties, which depend on the distortion of coplanarity of the flexible backbone. The PT derivatives can be applied to chemical sensors owing to their tunability of chemical and physical properties, while on-site sensing in aqueous media using PT-based solid-state devices is still challenging. In this Account, we describe a strategy for on-site supramolecular analytical devices. To maximize the applicability of PTs, we designed two types of PT-based chemical sensors. In the optical sensors, microarrays made of a hydrogel containing PTs on portable glass chips can simultaneously analyze multi-analytes combined with pattern recognition techniques. The various optical responses by adding analytes on the microarray chips can be recorded by easy-to-handle methods such as a digital camera, achieving on-site and high-throughput analysis without large spectrophotometers. Next, water-gated polythiophene transistors can offer highly sensitive detection due to a synergy effect of field-effect doping and interfacial charge modulation. Moreover, the electrical device with a microfluidic chamber allows real-time monitoring. We believe that our approaches would be a guideline to pave the way for the realization of supramolecular analytical devices.Polythiophene is one of the promising materials for chemical sensors owing to their tunability of chemical and physical properties, while on-site sensing in aqueous media using polythiophene-based solid-state devices is still challenging. We herein propose several strategies to realize supramolecular analytical devices in accordance with the philosophy of "lateral thinking of established technology".

Abstract:Electronic tongue capable of identifying thiophene compounds can promote early-stage environmental pollution monitoring and assist in environmental governance. Herein, a pH-modulated colorimetric sensor array based on PtNi nanowires (NWs) with peroxidase-like activity is designed for discrimination of thiophene compounds. The simplicity of the construction of this sensor array lies in the fact that it is composed of only one nanomaterial-3,3′,5,5′-tetramethylbenzidine (TMB)-H 2 O 2 system at three different pH values, which largely simplifies the programmatic design of the sensor array. Besides, in terms of sensing principle, the adsorption of sulfur from thiophene compounds on PtNi NWs alters the electronic state of their active centers, resulting in different peroxidase-like activities at different pH values. The color variances brought by the changes serve as "fingerprints" to successfully recognize six thiophene compounds as low as 1 μM level. Notably, PtNi NWs show splendid peroxidase-like activity (12.66 U/mg), far outweighing that of Pt NWs (2.62 U/mg), thanks to synergistic catalytic effect of Pt and Ni.

In this sensing design (schematic representation of the sensing procedure for adenosine triphosphate detection, Scheme 1 in manuscript text), which contains an enhanced three-step magnification process, SMSs with "clean" surface were first used to separate the undesirable aptamer and aptamer-adenosine triphosphate (ATP) complex attached on SMSs surface after aptamer-ATP interaction, which lead to the detachment of blocker DNA into the solution phase. Then, under the assistance of blocker DNA, an amplified method based on the inherent signal-transduction mechanism of the hairpin probe and strand-displacement property of DNA polymerase was introduced. The obtained duplex DNA was used to hybridize with an acceptor DNA assembled on electrode to form triplex DNA, which could bring a more obvious detection signal compared with the duplex DNA without the amplification. The electrochemical signal came from the GSGH-based enhanced sensing interface containing positively charged ferrocene-appended poly(ethyleneimine) (Fc-PEI). Using the above multiple effects, we could achieve the sensitive analysis of a model small molecule-ATP (an important "molecular currency" of intracellular energy transfer) in a wide detection range from 0.05 nM to 56.5 nM with the detection limit of 0.023 nM.

Abstract:An ambipolar organic field-effect transistor (OFET) based on poly(diketopyrrolopyrrole-terthiophene) (PDPPHD-T3) is shown by P. Sonar, H. Haick, and co-workers on page 4012 to sensitively detect xylene isomers at low to 40 ppm level in multiple sensing features. Combined with pattern-recognition algorithms, a sole ambipolar FET sensor, rather than arrays of sensors, is able to discriminate highly similar xylene structural isomers from each other.

Abstract:An extensible multidimensional colorimetric sensor array for the detection of protein is developed based on DNA functionalized gold nanoparticles (DNA-AuNPs) as receptors. In the presence of different proteins, the aggregation behavior of DNA-AuNPs was regulated by the high concentrations of salt and caused different color change; while DNA-AuNPs grew induced by the reduction of HAuCl4 and NH2OH as a reductant on the surface of nanoparticles exhibited different morphologies and color appearance for different proteins. The transducers based on AuNPs modified by specific and nonspecific DNA enables naked-eye discrimination of the target analytes. This extensible sensing platform with only two receptors could simultaneously discriminate ten native proteins and their thermally denatured conformations using hierarchical cluster analysis (HCA) at the concentration of 50nM with 100% accuracy. This opens up the possibility of the sensor array to investigate the different conformational changes of biomacromolecules, and it gives a new direction of developing multidimensional transduction principles based on plasmonic nanoparticle conjugates. Furthermore, the sensing system could discriminate proteins at the concentration of 500nM in the presence of 50% human urine, which indicated this sensor array has great potential ability in analyzing real biological fluids. In addition, the multidimensional colorimetric sensor array is suitable for analysis of target analytes in the resource-restricted regions because of rapid, simple, low cost, and in-field detection with the naked eye.

Abstract:Water-soluble, regioregular polythiophene (PT) derivatives are conformational sensitive to external stimuli, and their spectral responses are determined by conformation changes, π-π interactions and scattering of visible lights originated from PT aggregates. In this paper, we review the synthesis of water-soluble PTs and their photophysical properties, and then highlight our efforts toward the development of PT-based optical sensors for small molecules. Mechanism studies indicate that the interplay between side-chain steric/electrostatic repulsive and interchain attractive interactions is responsible for the assembly and disassembly of water-soluble PTs in aqueous media. Consequently, the planarization/deplanarization of PT backbones provides the possibility for colorimetric and fluorescent dual detection of small bioanalytes and chemical species.
