Abstract:Objective To analyze the characteristics of intramembranous as well as endochondral bone formation during osteoporotic fracture healing in a rat ovariectomy model. Methods Fifty-seven female SD rats(6 months old) were randomized into OVX group or SHAM group.Osteoporosis was induced in OVX group through bilateral ovariectomy following three months of BMD monitoring. Then transverse fracture was created in mid-shaft of the right femur in both groups using a three-point-bending apparatus.Weekly radiographic follow-up was performed with the external callus measured in lateral view radiography.Samples were collected at 2,4,and 8 weeks and scanned with micro-CT.Histological quantifications were carried out subsequently. The mechanical property of the femur was checked at the end of the study. Results In the first three weeks after fracture,SHAM group showed significantly higher values of external callus areas and volumes than the OVX group in the subperiosteal region.More active chondrogenesis was also observed in the SHAM group than the OVX group without significant differences in quantitative analysis. With endochondral ossification,SHAM group presented significantly larger volume of mineralized callus than OVX group at fracture site.At 8 weeks,SHAM group showed better mechanical property than OVX group. Conclusion Both intramembranous and endochondral bone formation were impaired in an ovariectomy induced rat osteoporotic fracture model.
