Abstract:Objective To investigate the effect of combined treatment with zoledronic acid( ZA) and parathyroid hormone( PTH) on fracture healing in OVX rats. Methods Rats in this study received bilateral ovariectomy. Twelve weeks later,the unilateral transverse osteotomy on the tibia was performed in rats. Then,the tibia was fixed with intramedullary nail. After the establishement of fracture model,the rats were then randomly treated with vehicle,ZA( 1. 5μg / kg weekly),PTH( 60 μg / kg, three times a week),or ZA plus PTH. After 4-or 8-week treatment,the fractured tibia was collected. And micro-CT,histology, and biomechanical test were performed. Results Compared with that in control group,the formation and strength of the callus increased in all treatment groups. The best promotion of relative bone volume( BV / TV), the trabecular thickness, and biomechanical strength was observed in ZA + PTH group. Conclusion The combination of ZA and PTH has a cumulative effect on fracture healing in OVX rats.
