Abstract:Numerous advances have been made in X-ray technology in recent years. X-ray imaging plays an important role in the nondestructive exploration of the internal structures of objects. However, the contrast of X-ray absorption images remains low, especially for materials with low atomic numbers, such as biological samples. X-ray phase-contrast images have an intrinsically higher contrast than absorption images. In this review, the principles, milestones, and recent progress of X-ray phase-contrast imaging methods are demonstrated. In addition, prospective applications are presented.

Abstract:We report a novel approach to diffraction phase microscopy (DPM) with automatic pinhole alignment. The pinhole, which serves as a spatial low-pass filter to generate a uniform reference beam, is made out of a liquid crystal display (LCD) device that allows for electrical control. We have made DPM more accessible to users, while maintaining high phase measurement sensitivity and accuracy, through exploring low cost optical components and replacing the tedious pinhole alignment process with an automatic pinhole optical alignment procedure. Due to its flexibility in modifying the size and shape, this LCD device serves as a universal filter, requiring no future replacement. Moreover, a graphic user interface for real-time phase imaging has been also developed by using a USB CMOS camera. Experimental results of height maps of beads sample and live red blood cells (RBCs) dynamics are also presented, making this system ready for broad adaption to biological imaging and material metrology.

Abstract:Singlet microscopy is very attractive for the development of cost-effective and portable microscopes. In contrast to conventional microscope objectives, which consist of multiple lenses, the manufacturing process for singlet lenses is done without extensive assembling and aligning. In this manuscript, we report a novel singlet virtual Zernike phase contrast microscopy setup for unstained pathological tumor tissue slides. In this setup, the objective consists of only one lens. There is no need for the inset Zernike phase plate, which is even more expensive than a whole brightfield microscopy setup. The Zernike phase contrast is virtually achieved by the deep learning computational imaging method. For the practical virtual Zernike phase contrast microscopy setup, the computational time is less than 100 ms, which is far less than that of other computational quantitative phase imaging algorithms. With a conceptual demo experimental setup, we proved our proposed method to be competitive with a research-level conventional Zernike phase contrast microscope and effective for the unstained transparent pathological tumor tissue slides. It is believed that our deep learning singlet virtual phase contrast microscopy is potential for the development of low-cost and portable microscopes and benefits resource-limited areas.

Abstract:Diffraction Phase Microscopy (DPM) is a widely used quantitative phase imaging method, whose common-path nature endows it with low noise and high sensitivity. Current applications of DPM include biological topography as well as biological dynamics for its nondestructive feature. Many different forms of DPM based on the original idea have appeared according to the different demands. In this paper, both the principle and the DPM classification will be given a comprehensive description. Furthermore, the future trend of DPM development is also discussed.

Abstract:An axial adjustable common-path interferometric phase imaging system with simple structure is proposed based on the theory of interference imaging. In this design, making full use of the reversibility of the optical path and the reflection and refraction characteristics of the splitter prism, the incident beam is split into two parallel beams by a prism. One beam is incident on the sample and reflected by the reflecting stage as object light, and the other beam is directly reflected as reference light. The interferograms can be captured by combining object beam with reference beam through the same beam splitter. The interferometer can achieve different types of interference by adjusting the angle between the layer of the beam splitter prism and the optic axis. Taking Fourier transform and three steps phase shift norm method as examples, the phase of the interference fringe is recovered, and the performance of the system is evaluated by the analysis of the results. The feasibility of the proposed design for biological cells phase imaging is demonstrated. The system has the characteristics of simple structure, easy operation and low system error, which can provide an easy and simple method for label-free morphological detection of homogeneous cell.

Abstract:Two-photon microscopy normally suffers from the scattering of the tissue in biological imaging. Multidither coherent optical adaptive technique (COAT) can correct the scattered wavefront in parallel. However, the determination of the corrective phases may not be completely accurate using conventional method, which undermines the performance of this technique. In this paper, we theoretically demonstrate a method that can obtain more accurate corrective phases by determining the phase values from the square root of the fluorescence signal. A numerical simulation model is established to study the performance of adaptive optics in two-photon microscopy by combining scalar diffraction theory with vector diffraction theory. The results show that the distortion of the wavefront can be corrected more thoroughly with our method in two-photon imaging. In our simulation, with the scattering from a 450-[Formula: see text]m-thick mouse brain tissue, excitation focal spots with higher peak-to-background ratio (PBR) and images with higher contrast can be obtained. Hence, further enhancement of the multidither COAT correction performance in two-photon imaging can be expected.

Abstract:We develop a confocal system equipped with optimal elliptical apertures to improve axial point spread function and signal-to-background ratio (SBR) for different detector sizes. By adjusting the parameters of the elliptical apertures, the axial half width at half-maximum can be reduced to 4.986 (described in optical coordinates) and SBR can be improved to 0.176. We evaluate this system with the 1951 USAF resolution test chart and the primary cultured neuron from SD rat stained by Map-2, and observe better imaging performance, which indicates the potential applications in biological science.

Abstract:The strong need in materials and biological science has prompted the development of high-speed quantitative phase imaging. However, for phase retrieval applying digital micromirror devices (DMDs), the accuracy of the retrieved phase will be disturbed by the DMD-induced aberrations. Here, we propose a phase retrieval method based on measuring and correcting errors caused by phase non-uniformity of the device. Using only four binary amplitude masks and corresponding diffraction intensities, the proposed method achieves rapid convergence and high-quality reconstruction. The experiments prove the practical feasibility for general samples and the effective improvement of the retrieved phase accuracy.
Tri-directional X-Ray Phase Contrast Multimodal Imaging Using One Hexagonal Mesh Modulator.

Abstract:Objective. X-ray phase contrast imaging is a promising technique for future clinical diagnostic as it can provide enhanced contrast in soft tissues compared to traditional x-ray attenuation-contrast imaging. However, the strict requirements on the x-ray coherence and the precise alignment of optical elements limit its applications towards clinical use. To solve this problem, mesh-based x-ray phase contrast imaging method with one hexagonal mesh is proposed for easy alignment and better image visualization.Approach. The mesh produces structured illuminations and the detector captures its distortions to reconstruct the absorption, differential phase contrast (DPC) and dark-field (DF) images of the sample. In this work, we fabricated a hexagonal mesh to simultaneously retrieve DPC and DF signals in three different directions with single shot. A phase retrieval algorithm to obtain artifacts-free phase from DPC images with three different directions is put forward and false color dark-field image is also reconstructed with tri-directional images. Mesh-shifting method based on this hexagonal mesh modulator is also proposed to reconstruct images with better image quality at the expense of increased dose.Main results. In numerical simulations, the proposed hexagonal mesh outperforms the traditional square mesh in image evaluation metrics performance and false color visualization with the same radiation dose. The experimental results demonstrate its feasiblity in real imaging systems and its advantages in quantitive imaging and better visualization. The proposed hexagonal mesh is easy to fabricate and can be successfully applied to x-ray source with it spot size up to 300μm.Significance. This work opens new possibilities for quantitative x-ray non-destructive imaging and may also be instructive for research fields such as x-ray structured illumination microscopy (SIM), x-ray spectral imaging and x-ray phase contrast and dark-field computed tomography (CT).

Abstract:Recent development of the medical imaging is the changing from macro-imaging to the micro-imaging by improving the spatial resolution and the contrast resolution of the soft tissues. Cells and microstructure of μm magnitude can be visualized in vivo non-invasively and dynamically by medical micro-imaging technique. Medical micro-imaging is capable of making the pathological diagnosis more early and the treatment of the leision more precisely by γ knife. Because the phase contrast resolution of X ray in soft tissues is about one thousand times higher than that of the absorption X ray method, so the phase contrast imaging is becoming the hot spot of the medical imaging. In this paper, the synchrotron X ray source at BSRF was used to investigate the microstructure of lung, liver and kidney by means of the DEI phase contrast micro-imaging method. Results indicated that the microstructures of lung alveoli, renal tubulous, liver lobules etc, which could not be seen by conventional absorption X ray method but could be revealed clearly by phase contrast imaging method. The spatial resolution reached about 20 μm. It is concluded that the medical phase contrast micro-CT is a new developing technique of the medical imaging in the near future.

Abstract:In-line Phase Contrast Imaging using a micro-focus X-ray source enables weakly absorbing specimens to be imaged successfully with X-rays and enhances the visibility of fine scale structure in more strongly absorbing specimens greatly. Getting the intensity on the image plane, we can get the phase contrast of an object by phase retrieval method. We got phase contrast of a black ant in different position and obtained the phase retrieval result by Transport of Intensity Equation method. On the base of edge visibility formula and entropy method, we quantitatively compare different edge visibility results of the images in different position and the differences between original phase contrast image and phase retrieval image.

Abstract:Propagation-based phase-contrast imaging was simulated based on paraxial Fresnel-Kirchoff diffraction integral and spherical wave illumination. Under a developed micro-CT system parameters, the effects of focal-spot size and imaging geometry on phase-contrast imaging have been investigated using a 2 mm thickness polystyrene edge phantom. An equivalent mono-energy was used to substitute the polychromatic spectrum of the micro-focus x-ray source. To consider effects of focal-spot size and detector resolution, the obtained phase-contrast image with an ideal point source was convolved with source intensity distribution and point spread function of detector. Simulations show reasonable influences of the two parameters and are in good agreement with experimental results.

Abstract:Hard X-ray phase-contrast imaging has extremely high sensitivity of weakly absorbing low-Z samples in medical and biological fields. Phase-contrast radiographic images have superposition phenomenon, while phase-contrast computed tomographic images reveal quantitative distribution of refractive index of the samples. But computed tomography requires long exposure time and lots of doses which are likely to damage biological specimens or live samples. We propose differential phase-contrast tomosynthesis based on diffraction enhanced imaging (DEI) to rapidly inspect the samples with a few projections. An experiment on biological samples is performed to prove it can highly enhance the information of detailed structures and mostly remove superposition phenomenon.

Abstract:Micro-CT is a new three dimensional imaging tool based on x-ray imaging mechanism and with ultrahigh spatial resolution. It can be used to image all kinds of samples or live small animalsunder non-destructive condition. In this paper, a micro-CT prototype system with high spatial resolution was developed. From the projective images and reconstructed cross-sectional images of a small insect sample, the phase-contrast effect with the edge-enhancement trait can be observed clearly. The cross-sectional images also show that the developed micro-CT system has the detail detectability down to 12 micrometers.

Abstract:Hard X-ray phase-contrast imaging can provide high sensitivity of weakly absorbing low-Z objects in medical and biological fields. Grating-based differential phase-contrast (DPC) method which relies on Talbot self-imaging effect have been paid more attention because it can work with conventional X-ray tube and show great potential for clinic use. Meanwhile tomosynthesis as a 3D reconstruction method is especially useful in mammography where low does is necessary. In this paper, an phase-contrast tomosynthetic experimental system is implemented based on the differential phase contrast imaging method according to moiré deflectometry (named MD-DPC method) with weakly coherent hard X-rays. The source grating of former grating-based method can perhaps be removed and the energy utilization efficiency of the X-ray tube is increased. The effectiveness of MD-DPC method is proved by actual experiments. In our phase-contrast tomosynthetic experimental system, the x-ray tube and the detector is stationary while the sample is rotated on a rotation stage, which is equivalent to conventional linear tomosynthesis. However, for phase contrast image, the refraction-angle is the line integral of differentials of refractive index decrement in the sample along the beam path whose vector direction is vary as the tomosynthetic projection direction changes, so refraction-angle projections can’t be simply added up like scalar quantities as in conventional tomosynthesis. A mapping method is adopted here to map each refraction-angle projection to the same direction and then refraction-angle tomosynthetic images can be implemented by adopting the shift-and-add approach. Our experimental system is validated and phase-contrast tomosynthesis based MD-DPC would be considered as a promising imaging method for mammography to enhance the contrast of early cancer tissues in the near future.

Abstract:Objective: To report on a micro computed tomography (micro-CT) system capable of x-ray phase contrast imaging and of increasing spatial resolution at constant magnification. Approach: The micro-CT system implements the edge illumination (EI) method, which relies on two absorbing masks with periodically spaced transmitting apertures in the beam path; these split the beam into an array of beamlets and provide sensitivity to the beamlets' directionality, i.e., refraction. In EI, spatial resolution depends on the width of the beamlets rather than on the source/detector point spread function (PSF), meaning that resolution can be increased by decreasing the mask apertures, without changing the source/detector PSF or the magnification. Main results: We have designed a dedicated mask featuring multiple bands with differently sized apertures and used this to demonstrate that resolution is a tuneable parameter in our system, by showing that increasingly small apertures deliver increasingly detailed images. Phase contrast images of bar pattern-based resolution phantom and a biological sample (a mouse embryo) were obtained at multiple resolutions. Significance: The new micro-CT system could find application in areas where phase contrast is already known to provide superior image quality, while the added tuneable resolution functionality could enable more sophisticated analyses in these applications, e.g., by scanning samples at multiple scales.

Abstract:Objective To study the methodology of X-ray phase contrast imaging using synchrotron radiation, and evaluate the quality of phase contrast images. Methods Several experiments to obtain phase contrast images and absorption contrast images of various biological samples were conducted in Beijing Synchrotron Radiation Facility (BSRF), and then these images were interpreted to find out the difference between the two kinds of imaging methods. Results Satisfactory phase contrast images of these various samples were obtained, and the quality of these images was superior to that obtained with absorption contrast imaging. The phase contrast formation is based on the phenomenon of fresnel diffraction which transforms phase shifts into intensity variations upon a simple act of free-space propagation, so it requires highly coherent X-rays and appropriate distance between sample and detector. This method of imaging is very useful in imaging of low-absorption objects or objects with little absorption variation, and its resolution is far higher than that of the conventional X-ray imaging. The photographs obtained showed very fine inner microstructure of the biological samples, and the smallest microstructure to be distinguished is within 30-40 μm. There is no doubt that phase contrast imaging has a practical applicability in medicine. Moreover, it improves greatly the efficiency and the resolution of the existing X-ray diagnostic techniques. Conclusions X-ray phase contrast imaging can be performed with synchrotron radiation source and has some advantages over the conventional absorption contrast imaging. ;
