Abstract:In this paper,the effects of 6-BA and NAA combinations of different organs and different concentrations on the rapid culture of Caryopteris clandonensis are lucubrated.The results show that the germination and rootability of the middle section of newly-growing twigs are the highest;the inducing ratio is higher than 85%;MS + 6-BA 0.5 mg/L + NAA 0.5 mg/L is the optimal successive transfer culture medium,and the germination and rootability of the twigs of Caryopteris clandonensis are high.The rootability of stem apex seedlings is high during rooting,and the rooting ratio of stem apex seedlings can be up to 85% in short time.Short but strong seedlings can be cultured in PP333,thus,the survival ratio of the seedlings of Caryopteris clandonensis can be increased.

Abstract:The segments from various part of \%Liriope platyphylla \%Wang et Tang var. \%variegata \%Hort. were cultured on MS medium supplemented with variable ratio of hormones. The results showed that the highest induction frequency (100%) of adventitious buds was obtained by culturing the buds of rootstocks on MS medium supplemented with BA 1.5 mg/L+NAA 0.5 mg/L, but the mosaic of the plant can't be kept by this way. In order to solve this problem, the liquid culture with MS medium supplemented with BA 3.0 mg/L+NAA 0.5 mg/L can avoid dedifferentiation and keep the mosaic of the proliferated buds. The adventitious buds could be transplanted once a month, while MS medium supplemented with IBA 0.25 mg/L is suitable for rooting. After a period of training, the rooted plants could be planted on soil.

Abstract:This study involved three investigations: (1) The effect of relative moisture on frequency of anther-derived callus induction in cold pretreatment; (2) The effect of cold treatment to the callus on plant regeneration; and (3) highly frequent and rapid establishment of barley embryogenic cell suspensions. Research results showed that the frequency of anther-derived callus induction was promoted obviously by higher relative moisture in cold pretreatment; The frequency of green plantlets from anther-derived callus was improved by cold treatment; and fast-growing, well-dispersed, and homogeneous cell suspensions were established from 7-week-old anther embryogenic callus. The frequency of establishment was 25%-50%. It was very difficult to establish embryogenic cell suspensions from 8-9-week-old anther-derived embryogenic callus.

MS+pH 5.8 on rooting culture better,1/2MS+pH 5.8 clumps on the induction of good. This test is white and build a plant breeding system provides a scientific reference.

Abstract:Platycerium wallichii Hook is a rare and endangered tropical epiphytic fern with ornamental and medicinal value. It is usually propagated by spore germination to produce sporophytes. This study established an in vitro protocols for spore germination, leading to filament formation and development of prothallia, and finally development of sporophytes following fertilization. The prothallia could be proliferated by 8.7 time within 30 days. The development of archegonia and antheridia on prothallia was asynchronous and prothallia were mainly dioecious (over 90%) and to a lesser extent hermaphroditic (5%). Prothallia were subcultured and propagated on plant growth regulator (PGR)-free Murashige and Skoog (MS) medium before fertilization. After fertilization, the frond lobes of in vitro sporophytes, which were used as explants, induced green globular bodies (GGBs) and adventitious sporophyte buds (ASBs). Among the PGRs tested, 1.0 mg L − 1 6-benzyladenine (BA) induced the most ASBs (16.7 per frond) and 1.0 mg L − 1 thidiazuron (TDZ) induced the largest number of GGBs (17.7 per frond), while 2,4-dichlorophenoxyacetic acid (2,4-D) induced the fewest ASBs and GGBs (12.8 per frond) and α -naphthaleneacetic acid (NAA) induced only a few ASBs (12.6 per frond), with mostly formation of rhizoids. When root sections were used as explants, optimal ASB induction required MS medium with 0.3 mg L − 1 BA and 0.1 mg L − 1 NAA and SPC could reach 5.7 within 60 d. Sporophytes could also be proliferated from rhizomes on the optimal MS medium with 0.3 mg L − 1 BA and its sporophyte proliferation coefficient (SPC) could reach 6.3 within 45 d. When 1⁄2MS medium was supplemented with 0.3–1.0 mg L − 1 NAA or indole-3-butyric acid (IBA), rooting of sporophytes was essentially 100%. After wrapping rhizomes in moss and transplanting them to a humus and river sand (1: 1, v/v) substrate, 100% of the plantlets survived after one month (98.7% after three months). The regeneration protocols presented in this work have a potential to assist in for the conservation and propagation of this endangered horticultural and medicinal fern.

Abstract:Extreme environmental conditions and limited understanding of ecosystem processes and community dynamics pose significant challenges for arctic ecosystem restoration activities. As pioneer species, bryophytes are critical to the function and structure of northern ecosystems and play an important role in facilitating soil and microhabitat development. A total of 11 bryophytes species were collected from a mixed community near Lac de Gras in the Northwest Territories, Canada and a 12 week laboratory study was conducted. Three propagule types (large (2.1-40 mm), medium (1.0-2 mm), small (<1 mm) phyllids) and three slurries (distilled water, beer, buttermilk) were used to determine effective bryophyte propagation techniques and slurries for species introduction in arctic restoration sites. Medium size fragments were more effective than large or small for propagating bryophytes as they led to greater bryophyte plant count and cover. Water and beer were more effective than buttermilk, although their effects were lessening after 12 weeks. Bryum pseudotriquetrum was the most abundant species propagated, followed by Aulacomium turgidum and Ceratodon purpureus. This study suggests propagation with medium size fragments and distilled water would be most efficient for field application in arctic ecosystem restoration if bryophyte revegetation is a focus.

Abstract:The present study was conducted to study the tissue culture and rapid propagation technique for Bletilla striata.The lateral buds and tubers of Bletilla striata were used as explants and sterilized with 0.2% HgCl2 at different times to select suitable explants and sterilization time.The basic MS medium or half MS medium was used with different concentrations of 6-BA or NAA to conduct the induction and multiplication of cluster buds,and rooting culture of Bletilla striata.It was found that the lateral bud of Bletilla striata was suitable for explants and had the best sterilization effect,when sterilized with 0.2% mercury bichloride for 8 minutes.The MS medium+1.5 mg / L 6-BA + 0.1mg / L NAA showed better effects on the induction of cluster buds for lateral buds.The best suitable multiplication medium for cluster buds was MS+2.0mg / L 6-BA + 0.2mg / L NAA with the multiplication coefficient 5.0,every thirty days.The better medium for the induction of roots was found to be 1 / 2MS + 1.0mg / L NAA + 0.1mg / L 6-BA with 83.3% of rooting rate.

Abstract:Tissue culture and rapid propagation of Salvia prionitis were reported in this paper. Effects of several factors such as different explants,collecting periods on the influence of sterilization, different phytohormones concentration,medium on the induction of buds and roots,and transplantation of seedlings were studied. Results showed that stem was the best explant for sterilization,the next one was the tip of stem and the third one was the low tip on the ground before the stem outgrowth. The best collecting periods of explants was from April to June. The most suitable medium for bud induction was MS+BA 0.8～1.6 mg/L+NAA 0.2 mg/L,MS+BA 0.2 mg/L+NAA 0.02 mg/L was optimum for proliferation, and MS+NAA1.0 mg/L was best for root induction. The survival rate of the seedlings was about 90%.

Abstract:This article mainly studied on the fast propagation technology system of tissue according to Canna glauca L. with buds as explant. The result show that: MS+BA1.0~4.0mg/L+NAA0.2mg/L culture medium can induce movable bud better than general methods, MS+BA6.0~8.0mg/L+NAA0.2mg/L culture medium has the best effect in reproducing aspect, there both have different degree root detach when using culture medium MS (not include hormone) and MS+NAA0.2mg/L culture medium culture root. Young plant can come out bettle when its height reaches to 6~8cm. Moss can be used to exercise young plant. The survive ratio comes to 95%.

Abstract:Gentiana rhodantha is a perennial herb of the genus Gentiana (Tourn.) L. This study was novel in establishing a regeneration system of G. rhodantha using young leaves as explants on the Murashige and Skoog (MS) medium supplemented with different plant growth regulators (PGRs). The roots, stems, and leaves of G. rhodantha were used as explants. The effects of the optimal explant disinfection method, type of explant used, concentrations of PGRs added to the culture media on tissue culture, and rapid propagation of G. rhodantha were studied. The results showed that the optimal disinfection method for stems and roots consisted of disinfection using 75% ethanol for 50 s, followed by 4% sodium hypochlorite (NaClO) for 10 min. The optimal disinfection technique for leaves consisted of disinfection using 75% ethanol for 50 s, followed by 4% NaClO for 8 min. Root explant was the most suitable for inducing the callus of G. rhodantha on the MS medium supplemented with different PGRs. The optimal conditions for callus induction included 1.0 mg/L 6-benzylaminopurine (6-BA) and 0.5 mg/L α-naphthalene acetic acid (NAA). The callus induction rate using the root explant reached 94.28%. MS supplemented with 2.0 mg/L 6-BA and 0.1 mg/L NAA was the optimal medium for inducing adventitious shoots from the callus of G. rhodantha . The best medium for propagation and plantlets strengthening was MS supplemented with 0.8 mg/L 6-BA and 0.3 mg/L NAA, and the propagation index was 8.62. MS supplemented with 0.3 mg/L 3-indolebutyric acid was the best culture medium for inducing the rooting of adventitious buds, with the maximum rooting rate reaching up to 100%.

Abstract:Genus Zephyranthes consists of economically important plant species due to their high ornamental value and presence of valuable bioactive compounds. However, this genus propagates by asexual division only which gives slow propagation rate. Plant tissue culture has the potential to provide efficient techniques for rapid multiplication and genetic improvement of the genus. In this work, a dual in vitro regeneration system through callus mediated shoot regeneration and direct shoot regeneration in species Zephyranthes candida, Zephyranthes grandiflora and Zephyranthes citrina was investigated. Bulb, leaf and root explants were cultured on Murashige and Skoog (MS) medium amended with different plant growth regulators (PGR's) viz. 2,4-dichlorophenoxyacetic acid (2,4-D), 1-Naphthalene acetic acid (NAA), 6-benzyl amino purine (BAP), N-phenyl-N'-1,2,3 -thiadiazol-5-ylurea (TDZ), 6-Furfuryl- aminopurine (KIN) alone or in combinations for callus induction and regeneration. Only bulb explants showed callus induction and regeneration response on different PGR combinations with a varied response in callus induction percentage, callus color and callus texture. Creamish compact callus (CC) was induced on 2 mg L[Formula: see text] 2,4-D, brown friable callus (BF) on 2 mg L[Formula: see text] NAA + 1 mg L[Formula: see text] BAP and green friable callus (GF) callus on 1 mg L[Formula: see text] KIN + 3 mg L[Formula: see text] NAA. The maximum shoot multiplication from different callus types (indirect organogenesis) was achieved on 2 mg L[Formula: see text] BAP alone without combinations. Bulb explants of Z. grandiflora induced maximum callus induction percentage (86.4%) and shoot regeneration percentage (83.5%) with the maximum 08 shoots per 150 mg callus mass. The induction and regeneration response was followed in the order of Z. grandiflora > Z. candida > Z. citrina. Similarly, maximum direct organogenesis from bulb explants was obtained in Z. grandiflora (93.3%) followed by Z. candida (91.5%) and Z. citrina (90.4%) on 3 mg L[Formula: see text] TDZ amended MS media. Adventitious root induction was achieved on 2 mg L[Formula: see text] IBA with a maximum of 8 roots per shoot. The in vitro raised plantlets were successfully acclimatized in the field with 85% survival efficiency. The genome size (2C DNA content) of the field-grown plants and in vitro regenerated plants, evaluated through flow cytometry technique, were similar and showed no ploidy changes. An efficient mass propagation protocol was established for obtaining plants with unaltered genome size in the three species of Zephyranthes.

Abstract:Abstract As a traditional Chinese medicinal material, Cynanchum atratum Bunge has been widely used in traditional Chinese medicine for its treatment of abscesses, acute urinary infection and hectic fever.Thus, wild resources of it have been endangered by overharvesting. Plant tissue culture technology is an important measure to protect wild resources of medicinal plants, including C. atratum . Therefore, a fast and efficient propagation system of C. atratum through axillary bud proliferation pathwayhas been established. Through axillary bud proliferation, the medium [MS+sucrose 30 g/L+Agar 7 g/L+NAA 0.2 mg/l+IBA 1.5mg/l+KT 0.5 mg/l] can effectively proliferate adventitious buds, and the induction rate was 100 %, proliferation coefficient could reach 8.56. MS medium was used to induce adventitious bud rooting, with rooting rate of 98% and no callus. The highest survival rate was 90% when the ratio of grass mud pond and orchard red soil was 1:1. To our knowledge this is the first report of rapid propagation system in C. atratum , it achieve rapid reproduction of C. atratum .

Abstract:Axillary node segment from test-tube plant provided by CIP was used as explants to establish the regeneration system of Ullucus tuberousus.Propagation was selected according to experimental design using orthogonal table using Mean multiple per generation and Mean length of shoot as index. Result showed that MS medium supplemented with 0.08 mg·L-1BA+0.03 mg·L-1NAA+0.5 mg·L-1GA3 was the most suitable one for proliferation of axillary node segment taken from growing shoot,and axillary bud could proliferation above 20 times per 25 days in 5 clones provided by CIP,except clone U2,mean length of total axillary node increasing above 7cm.In addition,1/2 primary medium supplement with 0.03 mg/L NAA was efficient for rooting of node,rhizogenesis start after 7 to 10 d,15 d later 3 to 6 fibres root could extend 1.0~2.0cm long,radication rate was 100%.

Abstract:[Objective] The aim was to carry out study on tissue culture of Lespedeza cyrtobotrya. [Method] The seeds of L. cyrtobotrya were used as materials to study on its tissue culture. [Result] The best sterilization time to L. cyrtobotrya seeds was 8 min with 2.1% NaClO, in which shooting percent reached 37.8% and no polluted situations occurred. In the primary culture with the MS as basal medium, the concentration of 6-BA showed a significant effect on the index of buds differentiation, the optimum differentiation culture medium was MS + BA 1.0 mg/L +NAA 0.1 mg/L +2, 4-D 0.01 mg/L, on which the index of generation could reach 6.69. The optimum subculture medium was MS +6-BA 1.0mg/L +2, 4-D 0.05 mg/L. The plants can generate the highest roots and rooting percent with IBA 0.50 mg/L. [Conclusion] This study had provided theoretical basis for genetic improvement of L. cyrtobotrya.

Abstract:Abstract. Isnaini Y, Praptosuwiryo TNg. 2020. In vitro spore germination and early gametophyte development of Cibotium barometz (L.) J. Sm. in different media. Biodiversitas 21: 5373-5381. Cibotium barometz (L.) J. Sm. is known as the golden chicken fern and included in Appendix II of CITES. It is an important export commodity for traditional and modern medicine. Globally, populations of this species are under significant pressure due to overexploitation in the wild. In vitro culture is one of the technologies used for ex-situ propagation and conservation of rare and endangered ferns and lycophytes. This study’s objectives were: (i) to observe in vitro spore germination and early gametophyte development of C. barometz, and (ii) to determine the best culture medium for rapid spore germination and early development of the gametophytes. The sterilized spores were sown in half-strength Murashige & Skoog (1⁄2MS) basal medium supplemented with combinations of 6-Benzylaminopurine (BAP) and α-Naphthalene acetic acid (NAA). A factorial combination of four BAP concentrations (0, 2, 4, and 6 mg L-1) with four concentrations of NAA (0; 0.01; 0.03 and 0.05 mg L-1) created 16 treatments replicated in a Completely Randomized Design. Spore germination of C. barometz was observed to be Vittaria-type, and its prothallial development was Drynaria-type. Spore germination started 7-14 days after sowing. Young heart-shape gametophytes consisting of 110-240 cells were formed in 45-61 days after sowing. The two best spore culture media for rapid spore germination and development of C. barometz gametophytes were 1⁄2 MS with or without 2 mg L-1 BAP.

Abstract:The primarily technology of tissue culture and propagation of Siraitia grosvenorii was reported,contains the condition of tissue culture,confection of medium,selection and sterilization of explant,inoculation and culture,preservation of superior provenance,hardening-seedling and transplanting,plant pack and transportation. And 5 kinds of referenced medium were reported too. The medium of induced stem was MS+BA0.5～1.0 mg/L+IAA(NAA)0.05～0.1 mg/L+sugar 3%+agar 4.5 g/L,pH5.8. The medium of induced stem apex was MS+BA0.5～1.0 mg/L+NAA0.05～0.1 mg/L+coconut milk 100 mL+sugar 3%+agar 4.5 mg/L,pH5.8. The medium of subculture of fasciculate bud was MS+BA0.3～0.7 mg/L+NAA0.05/IAA0.1 mg/L+sugar 3%+agar 4.5 mg/L,pH5.8. The medium of subculture of cutting was MS+BA0.1 mg/L+IAA0.3 mg/L+active carbon 0.07 g/L+sugar 3%+agar 4.5 mg/L,pH5.8. The rooting medium was MS+BA0.07 mg/L+IBA0.15 mg/L+IAA0.1 mg/L active carbon 0.1 g/L+sugar 3%+agar 4.5 mg/L,pH5.8. On the other hand,the cause of ill phenomenon in the process of tissue culture was analyzed,preventive measure was put forward. And feasible condition of hardening-seedling and transplanting was clear,the relevant manage method was formed. The integrated technology procedure of tissue cultured plantlets of S.grosvenorii was established.

Abstract:With young branches as explants,the effects of plant growth regulators on proliferation and rooting of Paraserianthes falcataria(L.) Nielsen were studied.The result showed: the best proliferating medium was MS+6-BA at 1.0 mg/L+NAA at 0.075 mg/L,with the proliferation rate up to 2.65;The optimum rooting medium was 1/3 MS+IBA at 0.1 mg/L+NAA at 0.01 mg/L,and the rooting rate reached 54.6%;The survival rate after transplantation was 95.8%.

Abstract:Tissue culture and rapid propagation of a Yunyan 97 giant variation plant was carried out. Result indicated that the most suitable medium for callus induction and proliferation of bud clusters was MS+6-BA2mg?L-1+NAA0.2mg?L-1, while the suitable medium for buds amplification was 1/2MS+6-BA 0.10 mg?L-1+IBA 0.5mg?L-1. It took about 80 days from explants inoculated to small seedling. Attention was also paid to slicing time of explants with bud clusters and how phytohormone was added. Result suggested that, to slice explants 15 to 20 days after bud spots were formed could promote healthy bud clusters which were also easy to slice. Phytohormone added before and after medium sterilization resulted little influence to phytohormone activity.

Abstract:For setting up the technological system of tissue culture of Pinellia ternate(Thunb.)Breit.,took corms as explants,MS as basic medium and added different compounds of NAA / N6-BA,to culture the cuttings of leafstalk. Results showed that: 1.The suitable compounds of media were MS +NAA 0.1-0.2/N6-BA 2.0-3.0,normally on such media explants dedifferentiation forming callus needed 7 d,forming embryoids 15 d,set up regenerated plantlets with shoot and adventitious root 30 d. multiple of proliferation reached to 6-10.2.If added only NAA or N6-BA to MS,the cuttings may forming callus,but it was difficult to re-differentiating. 3.If MS added low concentration of NAA(0.1)and paired up with high concentration of N6-BA (4.0) would induce the embriods and regenerated plantlets de-differentiating and formed callus again. But raised the concentration of NAA,such appearance would be put off.

Abstract:In vitro culture of Amorphophallus konjac C.Koch with Rhizomes as explants.The optimum media for callus induction was screened out by using orthogonal design.Different results can get when cultivate on different media.The results show that the media of NN+1.0mg/L BA+1.0mg/L NAA is the best for callus induction.Media of MS+2.0mg/L BA+1.0mg/L IAA,can get the highest ratio of adventitious buds differentiation,and IAA is better than NAA in this phase.MS+0.5mg/L IBA+0.5mg/L GA_3 can induce roots occurrence preferably,up to 100%.Antioxidants of 100mg/L PVP,100mg/L Vc and sorbent of 500mg/L AC can control browning efficiently.PVP and Vc restrain callus growth,whenas AC can stimulate it.
