Abstract:A rapid and efficient micropropagation protocol was developed for Curcuma zedoaria , using shoot buds as explants. Explants established on basal MS medium were transferred to 12 different medium compositions with different combinations and concentrations of plant growth hormones 6-benzylaminopurine (BAP), kinetin (Kn), and thidiazuron (TDZ). The best shoot multiplication was recorded in MS medium supplemented with BAP (2.0 mg L −1 ) and TDZ (1.5 mg L −1 ), resulting in 5.3 ± 0.24 shoots of per explant and shoot length of 6.12 ± 0.19 cm. For the best in vitro root induction, the aseptic shoots excised from the culture flasks and inoculated individually on MS medium supplemented with 1.5 mg L −1 indole-3-butyric acid (IBA) resulted in 74.2% root growth and 8.82 ± 0.31 roots per explant. Although callus induction was observed in MS containing 2,4-dichlorophenoxyacetic acid (2, 4-D) and 1-naphthaleneacetic acid (NAA), the highest callus proliferation was observed with MS and NAA (2.0 mg L −1 ). Micropropagated plants established on soil showed 83% survival on acclimatization to the field conditions.

Abstract:The present study aimed to determine the foliar micro-morpho-anatomical features of in vitro cultured Gardenia jasminoides J. Ellis (Rubiaceae) to compare the effect of exogenous supplementation of growth regulators (cytokinins; 6-benzylaminopurine and Kinetin), in order to attenuate heterotrophic nutrition (in vitro) induced structural disorders in the proliferating shoots. Murashige and Skoog’s (MS) medium supplemented with 2.0 mg L−1 6-benzylaminopurine (BAP) in combination with 0.15 mg L−1 indole-3-acetic acid (IAA) was detected optimal for axillary bud induction. Nutrient medium containing 0.5 mg L−1 BAP and 0.25 mg L−1 IAA was found appropriate combination for proliferation of multiple shoots, and yielded 37.2 shoots (per explant) with 7.45 cm average length after 2nd subculture (8 weeks). Supplementation of NAA with cytokinins resulted in callus formation. The proliferation of shoots on kinetin (Kn) and IAA combination resulted in the formation of fragile shoots and the leaves with increased structural impairments like underdeveloped photosynthetic, vascular, and ground tissue systems, non-functional stomata, and reduced vein density. Comparatively, BAP and IAA treatment favoured healthy shoot proliferation and development of stable tissue systems with reduced structural abnormalities. Half strength MS medium augmented with 3.0 mg L−1 indole-3-butytric acid (IBA) was the optimal medium for root induction (32.0 roots per shoot with 3.5 cm in length). The regenerated plantlets showed 97% survival success during acclimatization and exhibited normal growth characteristics and morphology. The light microscopic evaluation of foliages provided technical support to in vitro regeneration techniques to understand the structural adaptational mechanism of in vitro raised plantlets, thereby substantially contributed in the reduction of the rate of mortality of regenerated plants of G. jasminoides.
Efficient in vitro regeneration of Zingiber zerumbet Smith (a valuable medicinal plant) plantlets from rhizome bud explants

Abstract:This study was conducted to develop an efficient protocol for mass propagation ofZingiber zerumbet Smith. Explants from rhizome buds were cultured on Murashige and Skoog (MS) medium supplemented with 6-Benzylaminopurine (BAP) alone (0 to 5 mg/l) or a combination of BAP (0 to 5 mg/l) and indole 3-acetic acid (IAA) (0 to 2 mg/l). MS medium supplemented with a combination of 5.0 mg/l BAP and 2.0 mg/l IAA or 3.0 mg/l BAP and 0.5 mg/l IAA produced the highest mean number of shoots (5.6) per explant as compared to other concentrations. The best shoots length (9.44 cm) was obtained on the medium containing 1.0 mg/l of BAP and 2.0 mg/l IAA. Thus, combined effects of BAP and IAA improved significantly the shoot growth and proliferation. MS medium supplemented with a combination of 5.0 mg/l BAP and 2 mg/l IAA gave the highest number of roots (17). However, longest roots per explant were obtained with 1.0 mg/l BAP alone. The proliferated shoots were green and healthy in appearance. Finally, healthy and complete plants with well developed roots were hardened, acclimatized and planted in the field successfully with a survival rate of 80%. Auxin and cytokinin synergism in micropropagation for mass production of Aloe vera

Abstract:Aloe vera [Aloe vera (L.) Burm. f.] is considered a valuable medicinal plant worldwide due to its remarkable beneficial effects on human health. However, challenges in A. vera propagation hinder meeting the increasing demand in the health and beauty sectors. As an alternative method, in vitro propagation is crucial for the mass production of Aloe plants, which is a rapid method as well. Therefore, the present study aimed to establish an efficient micropropagation protocol for A. vera by in vitro optimization of the effect of different plant growth regulators (PGRs). For shoot proliferation, sterilized explants were inoculated on the Murashige and Skoog (MS) medium supplemented with 6-benzylaminopurine (BAP) and thidiazuron (0.5, 1.0, 2.0, and 4.0 mg/l) in combination with 0.5 mg/l naphthaleneacetic acid (NAA). Subsequently, indole-3-butyric acid (IBA) (1.0, 2.0, and 3.0 mg/l) was used for root induction. It was found that the explants cultured on the MS medium supplemented with 4.0 mg/l BAP + 0.5 mg/l NAA showed the highest percentage of response (90 ± 1.29) for shoot induction within the minimum number of days (5 ± 0.33). The highest number of shoots (2.7 ± 0.36) and length of shoots (4.7 ± 0.42 cm) per explant were also observed with the same concentration of PGRs. However, the highest number of roots (3.2 ± 0.57), length of roots (5.67 ± 0.21 cm), and root induction (80 ± 1.97 %) were noticed within the minimum number of days (11 ± 0.79) on the MS medium supplemented with 1.0 mg/l IBA. Thus, the proposed method is a quick and effective approach for the mass propagation of A. vera with appropriate dosages of auxins and cytokinins, which may allow meeting the increasing commercial demand.
Micropropagation of Curcuma zedoaria Roscoe and Zingiber zerumbet Smith

Abstract:The aim of study is to establish a micropropagation protocol that can be applied to both C. zedoaria and Z. zerumbet for the germblasm conservation of these two species. Buds from rhizomes were used to establish the in vitro plantlets of C. zedoaria and Z. zerumbet. The shoot explants of the in vitro plantlets of both species that were cultured in liquid medium produced twice the number of shoots than those cultured in solid medium of the same composition. The aseptic shoots of C. zedoaria plantlets that were cultured on MS solid medium supplemented with 0.5 mg L-1 BA and 0.5 mg L-1 IBA (shoot proliferation medium) produced 2.3 shoots per explant while those cultured in the liquid MS medium with same combination of plant growth regulators produced 6.1 shoots per explant. For Z. zerumbet, each shoot explant produced 3.9 shoots when they were cultured in the solid proliferation medium while an average of 6.4 shoots could be induced form each shoot explant when cultured in liquid medium. Half-shoot of C. zedoaria produced significantly higher number of shoots than undivided or whole shoots after culturing in the liquid shoot proliferation medium for 4 weeks. An average of 5.9 shoots could be produced from each half shoot which was equivalent to 12 shoots obtained from a single shoot. All the in vitro plantlets of both species survived after acclimatization and transferred to the soil.

Abstract:A rapid and efficient plant regeneration protocol for a wide range of alfalfa genotypes was developed via direct organogenesis. Through a successive excision of the newly developed apical and axillary shoots, a lot of adventitious buds were directly induced from the cotyledonary nodes when hypocotyl of explants were vertically inserted into modified Murashige and Skoog (MS) medium supplemented with 0.025 mg dm −3 thidiazuron (TDZ) and 3 mg dm −3 AgNO 3 . When the lower part of shoots excised from explants were immersed into the liquid medium with 1.0 mg dm −3 α-naphthaleneacetic acid (NAA) for 2 min, and then transferred to hormone free half-strength MS medium, over 83.3 % of the shoots developed roots, and all plantlets could acclimatize and establish in soil. The protocol has been successfully applied to eight genotypes, with regeneration frequencies ranging from 63.8 to 82.5 %.
Influence of plant growth regulators on micropropagation and in vitro flowering of Trichodesma indicum (Linn) R. Br

Abstract:An efficient protocol for micropropagation and in vitro flowering of Trichodesma indicum (Linn) R. Br. was developed using shoot tip explants. The physiological role of cytokinin and its combination with auxins on micropropagation and in vitro flowering was investigated. The highest number of shoots (9.94 ± 0.10) and the maximum average shoot length (5.56 ± 0.35 cm) were recorded on Murashige and Skoog (MS) medium supplemented with benzylaminopurine (BAP) (4.44 μM) and naphthaleneacetic acid (NAA) (2.69 μM). The effect of sucrose concentration on in vitro floral development was studied in plantlets cultured on MS medium supplemented with gibberellic acid (GA3) and BAP. The highest percentage of flowering (93.2%) was obtained on MS medium supplemented with GA3 (1.44 μM), BAP (1.33 μM) and sucrose (30 g l−1). Root formation from the adventitious shoots was easily achieved on MS medium containing indole-3-butyric acid (IBA) (2.46 μM). The regenerated plantlets showed 86% survival rate and were phenotypically normal. The described method can be successfully employed for large-scale multiplication and in vitro flowering of T. indicum.
An Improved Micropropagation Protocol by Ex Vitro Rooting of Passiflora edulis Sims. f. flavicarpa Deg. through Nodal Segment Culture

Abstract:A procedure for rapid clonal propagation of Passiflora edulis Sims. f. flavicarpa Deg. (Passifloraceae) has been developed in this study. Nodal explants were sterilized with 0.1% HgCl2 and inoculated on Murashige and Skoog (MS) basal medium. The addition of 2.0 mgL(-1) 6-benzylaminopurine (BAP) to MS medium caused an extensive proliferation of multiple shoots (8.21 ± 1.13) primordial from the nodal meristems. Subculturing of these multiple shoots on the MS medium augmented with 1.0 mgL(-1) of each BAP and Kinetin (Kin) was successful for the multiplication of the shoots in vitro with maximum numbers of shoots (25.73 ± 0.06) within four weeks of incubation. Shoots were rooted best (7.13 ± 0.56 roots/shoots) on half strength MS medium supplemented with 2.0 mgL(-1) indole-3 butyric acid (IBA). All in vitro regenerated shoots were rooted by ex vitro method, and this has achieved 6-7 roots per shoot by pulsing of cut ends of the shoots using 200 as well as 300 mgL(-1) IBA. The plantlets were hardened in the greenhouse for 4-5 weeks. The hardened plantlets were shifted to manure containing nursery polybags after five weeks and then transferred to a sand bed for another four weeks for acclimatization before field planting with 88% survival rate.
Micropropagation of Tolumnia Orchids through Induction of Protocorm-like Bodies from Leaf Segments

Abstract:A protocol for plant regeneration via direct induction of protocorm-like bodies (PLBs) from leaf segments of Tolumnia Snow Fairy was developed as a basis for mass production. Ten-month-old, in vitro–grown donor plantlets were obtained by inducing shoots from buds on the flower stalk. Leaf segments harvested from plantlets of different heights and from expanding leaves at different positions were compared, as were two BA concentrations with 0.5 mg·L −1 NAA. The greatest rate of PLB induction (16.7%) was observed when leaf segments taken from 1- to 2-cm height plants were cultured in Murashige and Skoog (MS) basal medium supplemented with 2 mg·L −1 BA and 0.5 mg·L −1 NAA after 16 weeks of culture. When using leaf explants, only inner, expanding leaves cultured on MS basal medium supplemented with 4 mg·L −1 BA and 0.5 mg·L −1 NAA resulted in PLB induction, at an average rate of 25.5 PLBs per explant. After 16 weeks of culture, histological and scanning electron microscopy (SEM) observations revealed that PLBs originated from epidermal cells of leaf explants. PLBs of 1 to ≤2 mm in diameter continued to proliferate after 4 weeks of culture. These secondary PLBs could be produced from either whole PLBs or the upper side of PLBs. Finally, PLBs were regenerated into plantlets. After ≈14 months of culture, fully developed plants exhibiting well-developed roots and shoots were acclimatized. These plants grew well, with 1-year survival rates of nearly 73%, for plants originating as explants taken from 1- to 2-cm tall plants, and of 79%, for plants originating as explants taken from inner leaves. Some mature plants flowered 1 year after transplantation. This study presents a simple system that can provide a large number of PLBs for mass propagation in a short time that can be converted into plants and also used for the new cultivars of Tolumnia orchids.

Ploidy Status, Nuclear DNA Content and Start Codon Targeted (SCoT) Genetic Homogeneity Assessment in Digitalis purpurea L., Regenerated In Vitro

Abstract:Digitalis purpurea L. is a therapeutically important plant that synthesizes important cardiotonics such as digitoxin and digoxin. The present work reports a detailed and efficient propagation protocol for D. purpurea by optimizing various PGR concentrations in Murashige and Skoog (MS) medium. The genetic homogeneity of in vitro regenerants was assessed by the flow cytometric method (FCM) and Start Codon Targeted (SCoT) marker technique. Firstly, the seeds inoculated in full MS medium added with 0.5 mg/L GA3 produced seedlings. Different parts such as hypocotyl, nodes, leaves and apical shoots were used as explants. The compact calli were obtained on BAP alone or in combinations with 2, 4-D/NAA. The hypocotyl-derived callus induced somatic embryos which proliferated and germinated best in 0.75 mg/L BAP-fortified MS medium. Scanning electron microscopic (SEM) images confirmed the presence of various developmental stages of somatic embryos. Shoot regeneration was obtained in which BAP at 1.0 mg/L and 2.0 mg/L BAP + 0.5 mg/L 2,4-D proved to be the best treatments of PGRs in inducing direct and indirect shoot buds. The regenerated shoots showed the highest rooting percentage (87.5%) with 24.7 ± 1.9 numbers of roots/shoot in 1.0 mg/L IBA augmented medium. The rooted plantlets were acclimatized in a greenhouse at a survival rate of 85-90%. The genome size and the 2C nuclear DNA content of field-grown, somatic embryo-regenerated and organogenic-derived plants were estimated and noted to be 3.1, 3.2 and 3.0 picogram (pg), respectively; there is no alteration in ploidy status and the DNA content, validating genetic uniformity. Six SCoT primers unveiled 94.3%-95.13% monomorphic bands across all the plant samples analyzed, further indicating genetic stability among in vitro clones and mother plants. This study describes for the first time successful induction of somatic embryos from hypocotyl callus; and flow cytometry and SCoT marker confirmed the genetic homogeneity of regenerated plants.
Micropropagation and reintroduction of the endemic Tripleurospermum ziganaense (Asteraceae) to its natural habitat

Abstract:In this study, a rapid and efficient biotechnological method was developed for the critically endangered (CR) endemic species Tripleurospermum ziganaense ( Asteraceae ) in Turkey to reintroduce it to its natural habitat. Murashige and Skoog basal medium (MS) supplemented with 1.0 mg L −1 gibberellic acid (GA 3 ) was highly effective with 80% germination percentage. MS medium containing 1.0 mg L −1 6-BA and 0.1 mg L −1 IBA was superior for the highest average shoot number of 11.73-fold per explant. The same basal medium strengthened with 1.0 mg.L −1 2iP plus 0.1 mg L −1 IBA was highly favorable for the highest average shoot length of 47.77 mm. In vitro micropropagated plants had similar DNA ploidy levels ( x ) and chromosome number (2 n = 18) compared to mother plants. Rooting success was achieved between 93.33 and 100% in all rooting media. MS medium strengthened with 1.0 mg L −1 IBA was remarkably effective in terms of the highest average root number with 11.71-fold per explant. On the other hand, MS medium fortified with 0.25 mg L −1 NAA was quite conspicuous with an average root length of 124.03 mm per explant. All rooted plantlets were initially acclimatized in climate room conditions and then transferred from optimal greenhouse conditions into a botanical garden with 100% and 90% survival rates, respectively. Afterward, plantlets were reintroduced into experimental plot near their wild population of T. ziganaense , with 90% survival. Flow cytometry and cytology-assisted cytogenetic fidelity assessments of the micropropagated plantlets exhibited ploidy as well as chromosomal stability within the plantlets and with coherence to their mother plant. The preliminary results obtained from the combination of micropropagation with reintroduction efforts offered an excellent opportunity for in situ and ex situ conservation efforts to bolster populations of threatened endemic T. ziganaense . This approach consisting of the components of conservation, propagation, and reintroduction (CPR) may potentially serve as a model for saving and enriching other species at risk.

Abstract:Plumbago zeylanica is a unique model for studying flowering plant gametogenesis, heterospermy, and preferential fertilization, yet understanding the control of related molecular mechanisms is impossible without efficient and reproducible regeneration and stable genetic transformation. We found three key factors for enhancing successful regeneration: (1) tissue source of explants, (2) combination and concentration of growth regulators, and (3) culture conditions. The highest frequency of shoot regeneration was achieved using hypocotyl segments cultured on MS basal medium supplemented with BA 2.0 mg/l, NAA 0.75 mg/l, adenine 50 mg/l and 10% (v/v) coconut milk under subdued light at 25±2°C; under these conditions, each hypocotyl segment produced over 30 shoots, arising primarily through direct organogenesis after 3 weeks of culture. Regenerated shoots rooted easily on half-strength basal MS medium and were successfully established in the greenhouse. Using this tissue culture protocol, reporter gene GUS under the constitutive CaMV 35S promoter was introduced into P. zeylanica cells of petiole, cotyledon and hypocotyl with A. tumefaciens strains AGL1 and LBA4404. Transient expression was observed in all recipient tissues. Stable transgenic calli originating from petiole were obtained.

Effects of culture methods and plant growth regulators on in vitro propagation of Brassavola nodosa (L.) Lindl. hybrid

Abstract:Brassavola nodosa (L.) Lindl. is an epiphytic orchid with great potential for the ornamental plant industry. The lack of information on propagation and production techniques limits the development of large-scale commercial production. Furthermore, this species is experiencing a reduction in population due to habitat destruction, the impact of climate change, and over-collection from native habitats. This study aimed at developing an efficient protocol for micropropagation of B. nodosa , which could be valuable towards the large-scale commercial production as well as for conservation of this species. Six different concentrations of plant growth regulators (BA or IBA, with or without adenine sulfate) were evaluated in modified Murashige and Skoog (MS) medium for shoot multiplication. In addition, two concentrations of either NAA or IBA were evaluated for rooting. Explants were cultured under three different culture media conditions: semi-solid medium, liquid medium (partial immersion), and liquid medium (complete immersion). Results indicate that B. nodosa could be successfully micropropagated in liquid culture with partial immersion. The modified MS medium supplemented with 2.0 mg L −1 BA and 30.0 mg L −1 adenine sulfate resulted in higher multiplication rates. Rooting was obtained using either 0.5 mg L −1 NAA or 1.0 mg L −1 IBA with no significant differences between both rooting treatments. Plantlets achieved 100% ex vitro survival after 30-d acclimatization.
Effects of plant growth regulators on efficient plant regeneration efficiency and genetic stability analysis from two Ocimum tenuiflorum L. morphotypes

Abstract:An effective, rapid and improved protocol was established for in vitro plant regeneration from nodal explants of two Ocimum tenuiflorum L. morphotypes, an medicinally important species of immense industrial value. The treatment medium used was Murashige and Skoog (MS) medium supplemented with different concentrations and combination of cytokinins (BA/KN/2-iP) and auxins (NAA/IBA/IAA). The adventitious shoot buds formation and plant regeneration capacity were significantly different (p < 0.05) between two morphotypes. The highest number of shoots (5.01 ± 0.11) in O. tenuiflorum purple leaves was obtained in the medium containing 0.5 mg l−1 BA, while in O. tenuiflorum green leaves the highest number of shoots was 7.03 ± 0.10 on a medium containing 1.0 mg l−1 BA. Regenerated shoots were rooted in vitro on low-salt MS derived media. Media were supplemented with different auxins that affected roots formation of particular morphotypes. The acclimatization of micropropagated plantlets of two morphotypes showed the best results with the sand:soil:compost (2:2:1). Genetic stability of regenerated plants as well as mother plants of two morphotypes of O. tenuiflorum L. was assessed using random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR) markers. Both RAPD and ISSR displayed a homogenous amplification profile for all the micropropagated plants of O. tenuiflorum L. when compared to their mother plants. Gas chromatography results showed that the essential oils extracted from in vitro-derived plants and their mother stock plants have similar profiles. Rapid and high multiplication frequency, molecular genetic and essential oil content stability ensures the efficacy of the protocol developed for the production of these industrially important medicinal plants.

Abstract:An accelerated, unique and efficient alginate-encapsulation (synthetic seed)-based genetically true-to-type mass propagation system in stevia ( Stevia rebaudiana Bert.) was successfully developed. Multiple shoot cultures in stevia were initially established from shoot tip explants cultured on Murashige and Skoog (MS) semi-solid medium supplemented with 0.5 mg/l N 6 -benzyladenine. Synthetic seeds were developed from individual shoot tips (obtained from in vitro multiple shoots) using 3% ( w/v ) sodium alginate and 75 mM calcium chloride solutions. In comparison with non-encapsulated in vitro shoot tips, alginate-encapsulated ones (i.e. synthetic seeds) exhibited accelerated germination (in the form of fresh shoot emergence) (~ 2 days), higher shoot proliferation rate and elongation (9.47 shoots with 10.94 mm length, and 64.33 leaves) as well as quicker root initiation (~ 9 days), multiplication and elongation (~ 13 roots with 38 mm length) after 30 days of inoculation in filter paper (M-bridge)-immersed half-strength MS liquid medium without plant growth regulator. Non-encapsulated in vitro shoot tips failed to initiate roots and showed reduced shoot growth. Synthetic seed-regenerated complete plantlets were successfully acclimatized in cocopeat. Clonal fidelity analysis of synthetic seed-regenerated plantlets was performed using ten inter-simple sequence repeats primers that exhibited monomorphic banding pattern, ensuring no genetic variations among the plantlets as well as with their mother plant. The present protocol will be beneficial not only for the smooth exchange of germplasms but also as an alternative approach for accelerated-cum-enhanced in vitro mass propagation of stevia.

Abstract:An efficient mass propagation method for Feronia limonia was developed from excised shoot tips and nodal explants of in vitro grown seedlings. Explants were cultured on MS medium with different conc. of NAA, Kn, IAA and BAP singly or in combinations. Highest number of micro shoots and better plant growth were obtained from these two explants on MS medium supplemented with 0.2 mg/l BAP alone. The regenerated shoots were successfully rooted on MS medium supplemented with 0.5 mg/l NAA. The in vitro raised plantlets were successfully established in soil following the formation of roots with 100% survivability under ex vitro condition. Abstract:An efficient in vitro propagation protocol for a medicinal plant, Persicaria glabra (Willd.) M. Gomez, has been established. Nodal explants were cultured in vitro on agar-gelled (0.8%) Murashige and Skoog (MS) medium containing different concentrations of cytokinins, namely N 6 -benzyladenine (BA), 6-(γ,γ-dimethylallylamino)purine (2-iP), or kinetin (KIN). BA (2.5 μM) was found to be the most effective cytokinin for axillary bud proliferation. A growth retardant ancymidol (ANC) was combined with 2.5 μM BA to enhance the multiple shoot induction and ensure the overall health of micro shoots. The nodal explants taken from micro shoots developed in 2.5 μM BA were subcultured on agar-gelled MS medium supplemented with different concentrations of ANC along with 2.5 μM BA. Among the different concentrations tested, 4.0 μM ANC combined with 2.5 μM BA produced a maximum number of multiple shoots (13 shoots per explant). A progressive decline in the shoot height was observed with increasing concentrations of ANC and significantly reduced shoot length (0.5 cm) at a higher concentration (12.0 μM ANC). Reduced leaf area and more chlorophyll were noticed in ANC-treated plants than in BA-alone-treated plants. This facilitated better growth and development of robust micro shoots. Well-developed micro shoots were excised from 4.0 μM ANC along with 2.5 μM BA and subjected to ex vitro rooting experiments using different concentrations of auxins, including α-naphthaleneacetic acid (NAA) or indole-3-butyric acid (IBA). The maximum number of roots (46.33 roots per shoot) and root length (14.53 cm) were observed in 0.5 mM IBA with 89.11% of the response. The rooted plants were successfully acclimatized in field conditions with 100% survival. The genetic fidelity analysis of in vitro –raised plants was tested using inter simple sequence repeat (ISSR) markers, and they showed genetic monomorphism with the mother plant. The present study suggests that adding ANC and BA would enhance the multiple shoots induction and high rate of acclimatization of P. glabra cultures. The addition of ANC resulted in a 2.6-fold increase in shoot number and avoided undesirable elongation of shoots. Healthy shoots, thus, produced in ANC combined with BA-added medium doubled the survival of plants during acclimatization over a BA-alone medium and can be used in many other plant species that have the problem of developing shoots of an unhealthy nature coupled with abnormal internodal elongations.

Abstract:Platycerium wallichii Hook is a rare and endangered tropical epiphytic fern with ornamental and medicinal value. It is usually propagated by spore germination to produce sporophytes. This study established an in vitro protocols for spore germination, leading to filament formation and development of prothallia, and finally development of sporophytes following fertilization. The prothallia could be proliferated by 8.7 time within 30 days. The development of archegonia and antheridia on prothallia was asynchronous and prothallia were mainly dioecious (over 90%) and to a lesser extent hermaphroditic (5%). Prothallia were subcultured and propagated on plant growth regulator (PGR)-free Murashige and Skoog (MS) medium before fertilization. After fertilization, the frond lobes of in vitro sporophytes, which were used as explants, induced green globular bodies (GGBs) and adventitious sporophyte buds (ASBs). Among the PGRs tested, 1.0 mg L − 1 6-benzyladenine (BA) induced the most ASBs (16.7 per frond) and 1.0 mg L − 1 thidiazuron (TDZ) induced the largest number of GGBs (17.7 per frond), while 2,4-dichlorophenoxyacetic acid (2,4-D) induced the fewest ASBs and GGBs (12.8 per frond) and α -naphthaleneacetic acid (NAA) induced only a few ASBs (12.6 per frond), with mostly formation of rhizoids. When root sections were used as explants, optimal ASB induction required MS medium with 0.3 mg L − 1 BA and 0.1 mg L − 1 NAA and SPC could reach 5.7 within 60 d. Sporophytes could also be proliferated from rhizomes on the optimal MS medium with 0.3 mg L − 1 BA and its sporophyte proliferation coefficient (SPC) could reach 6.3 within 45 d. When 1⁄2MS medium was supplemented with 0.3–1.0 mg L − 1 NAA or indole-3-butyric acid (IBA), rooting of sporophytes was essentially 100%. After wrapping rhizomes in moss and transplanting them to a humus and river sand (1: 1, v/v) substrate, 100% of the plantlets survived after one month (98.7% after three months). The regeneration protocols presented in this work have a potential to assist in for the conservation and propagation of this endangered horticultural and medicinal fern.

Abstract:This work aims to develop a rapid protocol for the in vitro establishment of Polygonatum verticillatum (L.) All., an astounding medicinal herb of Astavarga group of plants. Successful in vitro establishment was achieved utilizing rhizome bud segments of approx. 0.5 to 1.0 cm size as explants. Sterilized explants were cultured on MS medium containing graded concentration of different plant growth regulators used either singly or in various combinations. Maximum (85±10.0%) per cent shoot induction with maximum shoot length (6.50 ± 0.11cm) were recorded in TDZ (2.0 mg/l) + NAA(1.0 mg/l) + GA3 (2.0 mg/l) supplemented MS medium. NAA (1.0 mg/l) + Kn (0.5 mg/l) and IBA(1.0 mg/l) supplemented full strength MS medium with 3% sugar showed highest 70 ± 5.0% rooting response.1⁄2 MS+1.5 % sucrose medium augmented with IBA (1.0mg/l) showed the maximum average number of roots per explants (12.8 ± 1.07) with highest root length(6.12±0.27 cm).
