Abstract:Two new polyene polyketides, namely aurovertins T and U (1 and 2), were isolated from Calcarisporium arbuscula, together with aurovertins B (3), D and E (4 and 5), and M (6). The structures were elucidated by extensive spectroscopic methods (especially 2D‐NMR techniques). The cytotoxic activities of all isolates against human triple‐negative breast cancer cell line (MDA‐MB‐231) were evaluated. As a result, compounds 3, 4, and 6 exhibited more potent cytotoxic activities against MDA‐MB‐231 cell line than the positive control taxol. Also, discussion about the relationships between structure and activity of these aurovertins was presented.

Abstract:A new norditerpenoid 1, as well as 15 known terpenoids 2-16, was isolated from the ethanol extract of the branches and leaves of Euonymus grandiflorus Wall. The structures of these compounds were elucidated mainly due to spectroscopic methods (1D NMR, 2D NMR, ESI-MS and HR-ESI-MS). Compound 11, betulinic acid, displayed moderate cytotoxicity against four human tumor cell lines A549, HCT116, SK-BR-3 and HepG2, with IC50 values of 10.3, 12.2, 3.6 and 4.5 μg/mL, respectively.

Their chemical structures were elucidated on the basis of MS, NMR spectroscopic analyses coupled with chemical degradation. The cytotoxic activities of two isolated compounds were evaluated against two human gastric cancer cell lines BGC-823 and AGS using MTT assay.

Abstract:Novel heteroatom-incorporated antofine and cryptopleurine analogues were designed, synthesized, and tested against a panel of five cancer cell lines. Two new S-13-oxo analogues (11 and 16) exhibited potent cell growth inhibition in vitro (GI(50): 9 nM and 20 nM). Interestingly, both compounds displayed improved selectivity among different cancer cell lines, in contrast to the natural products antofine and cryptopleurine. Mechanism of action (MOA) studies suggested that R-antofine promotes dysregulation of DNA replication during early S phase, while no similar effects were observed for 11 and 15 on corresponding replication initiation complexes. Compound 11 also showed greatly reduced cytotoxicity against normal cells and moderate antitumor activity against HT-29 human colorectal adenocarcinoma xenograft in mice without overt toxicity.

Abstract:Nine undescribed eremophilane sesquiterpenes, one undescribed guaiane sesquiterpene, along with ten known analogues were isolated and identified from fungus Penicillium roqueforti, which was separated from the root soil of Hypericum beanii N. Robson collected from the Shennongjia Forestry District, Hubei Province. Their structures were elucidated on the basis of various spectroscopic analyses, mainly including NMR and HRESIMS data, 13C NMR calculations with DP4+ probability analyses, ECD calculations, and single-crystal X-ray diffraction experiments. Furthermore, all twenty compounds were evaluated for the in vitro cytotoxic activities against seven human tumor cell lines, and the result suggested that 14-hydroxymethylene-1(10)-ene-epi-guaidiol A exhibited considerable cytotoxic activity against the Farage (IC50 < 10 μM, 48 h), SU-DHL-2, and HL-60 cells. Further mechanism study demonstrated that 14-hydroxymethylene-1(10)-ene-epi-guaidiol A could significantly promote apoptosis by inhibiting tumor cell respiration and decreasing intracellular ROS levels, thereby inducing S-phase blockade in tumor cells.

Abstract:Two new tigliane diterpenes, named eupneonoids A (1) and B (2), together with four known analogues (3-6) were isolated from the whole plant of Euphorbia neorubella Bruyns. Their structures were deduced based on the detailed spectroscopic analysis. All the isolates displayed cytotoxic effects against A549 human tumor cell lines with IC50 values ranging from 1.318 to 7.042 mu M.

Abstract:Four new and five known eudesmane-type sesquiterpenes were isolated from the aerial parts of Erigeron annus. The structures of these new compounds were elucidated by spectral means, including 2D-NMR analysis, to be 1 beta,4 beta-dihydroxyeudesman-11-ene; 1 beta,7 alpha-dihydroxyeudesman-4(15)-ene; (1 R,5 S,6 S,7 S,10 R)-1 beta, 6 alpha-dihydroxyeudesman-4-one and 1 beta- D-glucopyranosyloxy-6 alpha-hydroxyeudesman-4(15)-ene. In addition, a chemical transformation of a known compound 1 beta,6 alpha-dihydroxy-eudesman-4 (15)-ene was carried out and the derivative 1beta,6alpha-dihydroxy-4beta(15)-epoxyeudesmane was obtained. Some of the compounds were evaluated for their in vitro antitumor activities.

Abstract:Nine new sesquiterpene alkaloids, eurochevalierines A-I (1-9), were separated from the rice cultures of the endophytic fungus Penicillium sp. HZ-5 originated from the fresh leaf of Hypericum wilsonii N. Robson. The structures' illumination was conducted by single-crystal X-ray diffraction, extensive spectroscopic analysis, alkaline hydrolysis reaction, and Snatzke's method. Importantly, the antitumor activities screen of these isolates indicated that 1 could suppress triple negative breast cancer (TNBC) cell proliferation and induce apoptosis, with an IC50 value of 5.4 μM, which is comparable to the positive control docetaxel (DXT). Flow cytometry experiments mentioned that compound 1 significantly reduced mitochondrial membrane potential (MMP) of TNBC cells. In addition, 1 could activate caspase-3 and elevated the levels of reactive oxygen species (ROS) and expressions of suppressive cytokines and chemokines. Further Western blot analysis showed that 1 could selectively induce mitochondria-dependent apoptosis in TNBC cells via the BAX/BCL-2 pathway. Remarkably, these finding provide a new natural product skeleton for the treatment of TNBC.

Abstract:Celahypodiol 1, an unusual 17-membered carbon diterpenoid with a novel skeleton, and a new triterpenoid 12-oleanene-3beta,6alpha-diol 2, together with four known compounds furreginol 3, suigol 4, 20(30)-lupene-3beta, 29-diol 5, and 20(29)-lupene-1beta,3beta-diol 6, were isolated from the stalks of Celastrus hypoleucus (Oliv.) Warb. Their structures were established by means of spectroscopic analysis, including 2D NMR. The new compounds exhibited anti-tumor activities against a panel of human tumor cell lines.

Abstract:Two previously undescribed coumarins ( 1 and 2 ) were isolated from the root of Hansenia weberbaueriana which have been used to cure inflammatory diseases over thousands of years by Chinese. The structures of new findings were confirmed by comprehensive analyses of spectral evidences in HRESIMS, 1D and 2D NMR combined with chemical calculations. Compounds 1 and 2 exhibited potential anti-inflammatory properties by reducing the mRNA expression levels of TNF-α , IL-6 and IL-1β in lipopolysaccharide (LPS)-induced RAW264.7 macrophages at a concentration of 15 μM.

Abstract:Ten new dihydroagarofuran sesquiterpene polyol esters, tripterdines A-J (1−10) were isolated from the stem and branch of Tripterygium wilfordii. The structures of new compounds were elucidated on the basis of extensive spectroscopic analyses, including UV, IR, HRESIMS, NMR, and CD exciton chirality method. The structures of compounds 1, 3, and 6 were confirmed by X-ray crystallographic analyses. The anti-inflammatory and cytotoxic activities were assessed for all the compounds (1–10). Compounds 3, 5 and 10 exhibited potent anti-inflammatory activities with the secretion level of TNF-α ranging from 43.86% to 51.27%, and the IL-6 ranging from 32.44% to 50.64%. In addition, compounds 1, 3, 7 and 9 showed weak cytotoxicities against three human tumor cell lines (Huh7, MCF-7 and HCT-116).

Abstract:Ten new germacrane-type sesquiterpenoids (1-10) were isolated from a whole plant extract of Eupatorium chinense. The structures were elucidated by analysis of their NMR and MS data as well as by comparison with literature values. The absolute configuration of eupachinsin A (1) was determined by single-crystal X-ray diffraction analysis. Compounds 3 and 4 exhibited cytotoxicity against a human breast cancer cell line (MDA-MB-231), with IC50 values of 0.8 and 3.4 μM, respectively. In addition, compounds 3-5 showed cytotoxicity against the human hepatocellular carcinoma cell line (HepG2), with IC50 values ranging from 3.6 to 7.6 μM.

Abstract:Three undescribed sesquiterpenoids, phaeocaulisguatriol (1) and phaeocaulistriol A-B (19, 20), along with twenty known sesquiterpenoids were isolated from a chloroform-soluble extract of Curcuma phaeocaulis Valeton rhizome. Their structures were determined using comprehensive spectroscopic methods. All isolated compounds were evaluated for their cytotoxicity against five human tumor cell lines Hela, HepG2, MCF-7, BGC823, and A549. Among them, phaeocaulisguatriol (1) and phaeocaulistriol B (20) showed potent cytotoxic activity against MCF-7 (IC50 of 40.7 and 58.8 mu M, respectively), by inducing apoptosis. The effectiveness of compounds 1 and 20 was supported by predictive intermolecular interactions in molecular docking. Molecular dynamics simulation studies further probed the interaction mechanism under simulated physiological conditions between the two most active compounds and the TP53 protein, an apoptotic effector protein. Moreover, Western blot experiments confirmed that compound 1 induces apoptosis by activating the expression of TP53 and caspase 3 proteins. This work provides a deep insight into new sesquiterpenoids isolated from C. phaeocaulis Valeton acting as potential antitumor activity inhibitors.

Abstract:Nine undescribed dihydro‐β‐agarofuran sesquiterpenoid derivatives (1–9), along with a known analogue (10) were obtained from the leaves of Tripterygium wilfordii. Their gross structures were determined via extensive spectroscopic data, and the absolute configurations were elucidated by means of single‐crystal X‐ray diffraction analysis and electron circular dichroism (ECD) techniques which include ECD exciton chirality, octant rule of saturated cyclohexanone and comparison between the experimental and calculated ECD spectra. All the isolated compounds were tested for their neuroprotective activities against H2O2‐induced cell injury in human neuroblastoma SH‐SY5Y cells. Compounds 5 and 6 improved cell viability by 16.15 and 15.12% compared with the H2O2 treated group at 25 μM, respectively.This article is protected by copyright. All rights reserved.

Abstract:A new dimeric guaianolide (8) and a new glaucolide (3), a seco-guaiaretic acid (4), two guaianolides (6 and 7), and five known sesquiterpene lactones (1, 2, 5, 9, and 10) were isolated from the aerial part of Artemisia anomala. Their structures were determined on the basis of chemical and spectroscopic analysis.
