Abstract:From the fruits of Celastrus orbiculatus Thunb. a new beta-dihydroagarofuran sesquiterpene ester named 1beta,2beta,13-triacetoxy-9alpha-cinnamoyloxy-beta-dihydroagarofuran (1) has been isolated along with two known compounds, 6alpha-acetoxy-1beta, 9beta-dibenzoyloxy-8beta-hydroxy-beta-dihydroagarofuran (2), and 1beta, 6alpha-diacetoxy-9beta-benzoyloxy-8beta-hydroxy-beta-dihydroagarofuran ( 3). Their structures were elucidated on the basis of spectroscopic data.

Abstract:A new sesquiterpene polyol ester, 1-beta-acetoxy-2-beta, 8-beta, 9-alpha-tribenzoyloxy-6-alpha-hydroxy-beta-dihydroagarofuran, was isolated from the root bark of Celastrus rosthornianus. Its structure was elucidated mainly on the basis of 1D and 2D NMR spectrometry. The complete assignment of the C-13 NMR chemical shifts, and the determination of H-1-H-1 weak couplings observed in the H-1 NMR spectrum were also carried out on the basis of the 2D NMR spectra.

Its structure was elucidated, on the basis of H-1 NMR, C-13 NMR, NOE, IR and high resolution mass spectrometry, as 1-beta-acetoxy-8-beta,9-alpha-dibenzoyloxy-4-alpha,6-alpha-dihydroxy-2-beta (alpha-methylbutanoyloxy)-beta-dihydroagarofuran. The long-range H-1-H-1 COSY spectrum of this compound was also investigated.

Abstract:Two new sesquiterpenes, 1 and 2, as well as the five known compounds 37, were isolated from the stems of Euonymus alatus. Compounds 27 have a beta-dihydroagarofuran skeleton. The structures of these compounds were elucidated mainly by spectroscopic methods (1D-, 2D-NMR, ESI-MS, and HR-ESI-MS). We also report the X-ray crystal structure of evonine (3) for the first time.

Abstract:A new norditerpenoid 1, as well as 15 known terpenoids 2-16, was isolated from the ethanol extract of the branches and leaves of Euonymus grandiflorus Wall. The structures of these compounds were elucidated mainly due to spectroscopic methods (1D NMR, 2D NMR, ESI-MS and HR-ESI-MS). Compound 11, betulinic acid, displayed moderate cytotoxicity against four human tumor cell lines A549, HCT116, SK-BR-3 and HepG2, with IC50 values of 10.3, 12.2, 3.6 and 4.5 μg/mL, respectively.

Abstract:Ten new dihydroagarofuran sesquiterpene polyol esters, tripterdines A-J (1−10) were isolated from the stem and branch of Tripterygium wilfordii. The structures of new compounds were elucidated on the basis of extensive spectroscopic analyses, including UV, IR, HRESIMS, NMR, and CD exciton chirality method. The structures of compounds 1, 3, and 6 were confirmed by X-ray crystallographic analyses. The anti-inflammatory and cytotoxic activities were assessed for all the compounds (1–10). Compounds 3, 5 and 10 exhibited potent anti-inflammatory activities with the secretion level of TNF-α ranging from 43.86% to 51.27%, and the IL-6 ranging from 32.44% to 50.64%. In addition, compounds 1, 3, 7 and 9 showed weak cytotoxicities against three human tumor cell lines (Huh7, MCF-7 and HCT-116).

Abstract:Novel heteroatom-incorporated antofine and cryptopleurine analogues were designed, synthesized, and tested against a panel of five cancer cell lines. Two new S-13-oxo analogues (11 and 16) exhibited potent cell growth inhibition in vitro (GI(50): 9 nM and 20 nM). Interestingly, both compounds displayed improved selectivity among different cancer cell lines, in contrast to the natural products antofine and cryptopleurine. Mechanism of action (MOA) studies suggested that R-antofine promotes dysregulation of DNA replication during early S phase, while no similar effects were observed for 11 and 15 on corresponding replication initiation complexes. Compound 11 also showed greatly reduced cytotoxicity against normal cells and moderate antitumor activity against HT-29 human colorectal adenocarcinoma xenograft in mice without overt toxicity.

Abstract:Two new beta-dihydroagarofuran sesquiterpene polyesters were isolated from Celastrus paniculatus. The structures were deduced using various 2D NMR techniques (H-H COSY, C-H COSY, COLOC, NOESY), NOE difference spectra and mass spectra. The positions of the ester moieties were located by 2D heteronuclear correlated spectra via long-range coupling, or combined with NOE experiments. Their stereochemical structures were studied and the complete assignments of the H-1 and C-13 chemical shifts were made.

Abstract:Euonymus hederaceus is distributed widely in the south of China; its stems and leaves have been used as folk medicines to treat many diseases such as renal deficiency and chronic diarrhea, traumatic injury, and abnormal menstruation. Chemical investigation of the leaves and stems of Euonymus hederaceus resulted in the isolation forthe first time and characterization of a new friedelane type triterpene with a molecular mass of 472 and molecular formula of C30H48O4 by high resolution mass spectrometry. The 1H-NMR 13C-NMR and DEPT1350 spectra matched the characteristic data of the proposed triterpene skeleton.The compound was finally identified as 28-hydroxyfriedelan-3-one-29-oic acid on the basis of spectroscopic evidence, including two dimensional nuclear magnetic resonance as well as its IR spectrum.

Abstract:A new phenylpropanoid derivative ( 1 ), along with five phenylpropanoids ( 2–6 ), two monoepoxy lignans ( 8–9 ), one bisepoxy lignan ( 10 ), two cyclolignans ( 11–12 ), six neolignans ( 7 , 13–17 ), two mixed lignan-neolignans ( 18–19 ), two lignan glycosides ( 20–21 ), and four flavonolignans ( 22–25 ), were isolated from the stems and twigs of Euonymus acanthocarpus . Compounds 2–3 , 6–8 , 12 , and 14–25 were obtained from Celastraceae family for the first time, and compounds 5 and 9 were isolated from Euonymus genus for the first time. All the compounds were tested for cytotoxicity against SK-OV-3 and MCG-803 human tumor cell lines. Compounds 3 , 10 , 12 , and 18 showed weak cytotoxicity against SK-OV-3 cell line, and compounds 3–4 , 10–13 , and 19 showed weak cytotoxicity against MCG-803 cell line.

Abstract:1 A hitherto unidentified endophytic strain of the genus Chaetomium, isolated from the medicinal plant Otanthus maritimus, yielded a new tetrahydrofuran derivative, aureonitolic acid (), along with 5 known natural products, 2 - 6. The structure of 1 was determined by extensive spectroscopic analysis and comparison with reported data. Extracts of the fungus, grown either in liquid culture or on solid rice media, exhibited considerable cytotoxic activity when tested in vitro against L5178Y mouse lymphoma cells. Compounds 2 and 6 showed significant growth inhibition against L5178Y cells with EC50 values of 7.0 and 2.7 μg/mL, respectively, whereas 1 was inactive

Abstract:Four new and five known eudesmane-type sesquiterpenes were isolated from the aerial parts of Erigeron annus. The structures of these new compounds were elucidated by spectral means, including 2D-NMR analysis, to be 1 beta,4 beta-dihydroxyeudesman-11-ene; 1 beta,7 alpha-dihydroxyeudesman-4(15)-ene; (1 R,5 S,6 S,7 S,10 R)-1 beta, 6 alpha-dihydroxyeudesman-4-one and 1 beta- D-glucopyranosyloxy-6 alpha-hydroxyeudesman-4(15)-ene. In addition, a chemical transformation of a known compound 1 beta,6 alpha-dihydroxy-eudesman-4 (15)-ene was carried out and the derivative 1beta,6alpha-dihydroxy-4beta(15)-epoxyeudesmane was obtained. Some of the compounds were evaluated for their in vitro antitumor activities.

Abstract:Thirteen undescribed dihydroagarofuran sesquiterpenes, hypogricins A-M, were isolated from the twigs of Tripterygium hypoglaucum. Extensive spectroscopic techniques were used for the structural elucidation of these compounds. All the compounds showed no significant cytotoxicity against adriamycin-sensitive HepG-2 and adriamycin-resistant HepG-2/Adr cell lines. When HepG-2/Adr cell line was co-treated with those compounds plus adriamycin, hypogricins A and B showed potent activity to reverse multidrug resistance (MDR) of HepG2/Adr cells to adriamycin (reversal fold: RF = 102 and 200, respectively). In addition, hypogricins E, H, and L showed moderate activity to inhibit NO production in LPS-stimulated RAW264.7 cells with the IC50 of 22.3, 17.8, and 19.8 μM, respectively.
