Abstract:The alpha(2B)-adrenergic receptor (ADRA2B) plays an important role in vasoconstriction and blood pressure regulation. One common variant in the ADRA2B gene (del301-303) has been identified, and results in markedly decreased receptor desensitization in vitro but does not alter vascular sensitivity in vivo. Therefore, we fully characterized genetic variations in ADRA2B and related them to phenotype in vivo. We examined 5812 bp of contiguous sequence of ADRA2B (promoter, exonic, and 3'-untranslated region; 3'-UTR) using the polymerase chain reaction to amplify the genomic target followed by bidirectional sequencing (n = 68). Haplotypes were inferred using an expectation maximization algorithm. Vasoconstriction in response to increasing doses of the highly selective alpha(2)-adrenergic receptor agonist, dexmedetomidine (0.01-1000 ng/min) was measured in the dorsal hand vein using a linear variable differential transformer. The dose that produced 50% (ED50) of maximum venoconstriction (E-max) was determined for each subject from the individual dose-response curves. ED50 and Emax were compared in subjects with and without variant alleles and haplotypes of interest. We identified 24 variable sites, 12 in the promoter region, five in the coding region (including two previously described as non-synonymous variants) and seven in the 3'-UTR region. Four haplotypes were inferred, representing approximately 95% of the cohort. One haplotype, characterized by two single nucleotide polymorphisms in the promoter region, and one in the 3'-UTR, occurred in seven of 38 African-Americans, and was associated with a lower E-max, 61.3% [95% confidence interval (CI) 39.5-83.0, n = 7] compared to 78.1% (CI 73.8-82.5) in wild-types (n = 61) (P = 0.02). There was no association between the nine common variants and dexmedetomidine ED50. We have described novel variants and haplotypes of the ADRA2B gene. These do not alter sensitivity to a selective alpha(2)-adrenergic receptor agonist but some may decrease maximal venoconstriction in vivo. (c) 2005 Lippincott Williams & Wilkins.

Abstract:There are marked interethnic differences in beta (1)-adrenclceptor-mediated responsiveness, with sensitivity decreased in African-Americans and increased in Chinese compared with Caucasians, Therefore, the frequency of a common naturally occurring polymorphism of the human beta (1)-adrenoceptor gene (Arg(389)Gly), which has functional importance in vitro, was determined in 194 African-Americans, 316 Caucasian-Americans, 221 Hispanic-Americans and 142 Chinese. African-Americans were found to hare a significantly lower frequency of the Arg(389) allele than the other three ethnic groups (all P < 0.01), In the populations studied, the order of the distribution of the Arg(389) allele was: Chinese (74%) > Caucasians (72%) > Hispanics (67%) > African-Americans (58%). To determine the functional significance of the Arg(389)Gly beta (1)-adrenoceptsr polymorphism in-vivo heart rate responses to exercise were compared in healthy subjects homozygous for the Arg (n = 9) and Gly (n = 8) alleles, Heart rate response to exercise was not affected by genotype (P = 0.4). Although ethnic differences in the frequency of the beta (1)-adrenoceptor Arg(389)Gly polymorphism exist, the polymorphism does Hot appear to have functional significance in healthy subjects and therefore may not contribute to ethnic differences in response to drugs acting through the beta (1)-adrenoceptor, Pharmacogenetics 11:191-197 (C); 2001 Lippincott Williams & Wilkins.

Abstract:BACKGROUND:The alpha2C-adrenergic receptor plays an important role in the regulation of the sympathetic nervous system and, therefore, blood pressure and heart rate. A deletion polymorphism in its gene (ADRA2C del322-325), ten times more common in black than white Americans, has been associated with a loss of function in vitro and, under controlled study conditions, raised blood pressure and catecholamine secretion. We therefore examined the hypothesis that the ADRA2C deletion variant would alter sympathetic activity and contribute to ethnic differences in blood pressure. METHODS:We measured resting plasma norepinephrine and epinephrine concentrations, blood pressure and heart rate in 224 healthy subjects (127 whites), and determined their ADRA2C del322-325 genotype. Additionally, we analyzed heart rate variability (HRV) in a subgroup of 50 black subjects. RESULTS:Systolic (SBP) and diastolic blood pressure (DBP) were higher in blacks than whites [difference (95% confidence interval), 4.4 (1.5-7.4) mmHg, P = 0.003; and 2.7 (0.7-4.6) mmHg, P = 0.01, respectively]. Norepinephrine concentrations did not differ among subjects with 0, 1 and 2 copies of the deletion variant [median (interquartile range), 185.0 (147.5-269.8), 200.0 (154.9-257.0) and 173.8 (158.5-235.8) pg/ml, respectively; P = 0.54]. Similarly, none of the HRV parameters differed among the genotype groups. In multiple linear regression analyses adjusting for multiple covariates, the deletion genotype was not associated with SBP or DBP. In contrast, black ethnicity was associated with higher SBP (P = 0.001) and DBP (P = 0.005). CONCLUSION:The ADRA2C deletion polymorphism had no effect on markers of resting sympathetic activity and cardiovascular measures, and did not account for ethnic differences in blood pressure.

Abstract:Objective: The alpha(1A)-adrenergic receptor is highly expressed in human vasculature including resistance arteries and veins, and its stimulation is primarily responsible for adrenergically mediated smooth muscle contraction. Variability in sensitivity to phenylephrine, an alpha(1A) adrenergic agonist, has a large genetic component. We examined the hypothesis that a common polymorphism of alpha(1A)-adrenergic receptor (Arg347Cys) affects in vivo response.Methods. We measured vascular sensitivity to phenylephrine using the dorsal hand vein linear variable differential transformer technique and determined alpha(1A)-adrenergic receptor genotype in 74 healthy, nonsmoking adults (28 Arg/Arg, 30 Arg/Cys, and 16 Cys/Cys).Results. Sensitivity to phenylephrine, expressed as the dose of phenylephrine resulting in 50% venoconstriction (Phe(50)), was not significantly different in subjects with the 3 alpha(1A) adrenergic receptor genotypes: Phe(50) geometric mean (95% confidence interval) was 513 ng/min (287-918 ng/min) for Arg/Arg, 431 ng/min (274-680 ng/min) for Arg/Cys, and 471 ng/min (197-1124 ng/min) for Cys/Cys (P = .90).Conclusion: We conclude that the Arg347Cys receptor polymorphism does not alter agonist-mediated venoconstriction in vivo.
Frequency of Functionally Important Beta-2 Adrenoceptor Polymorphisms Varies Markedly among African-American, Caucasian and Chinese Individuals.

Abstract:There are ethnic differences in the prevalence and severity of hypertension and asthma and in beta-2 adrenergic receptor (BAR2)-mediated vascular responses. Two common polymorphisms of the human BAR2, Arg16 to Gly and Gln27 to Glu, are associated with alterations in BAR2 response, both in vitro and in vivo. Ethnic differences in disease manifestations and responses to treatment may be explained by the altered frequency of BAR2 polymorphisms. To determine the relative frequencies of the Arg16 to Gly and Gln27 to Glu BAR2 polymorphisms in different ethnic groups we studied 415 (123 African-American, 188 Caucasian-American and 104 Chinese) healthy individuals. There was a marked interethnic difference in the frequency of the BAR2 polymorphisms among the ethnic groups. The Glu27 allele was more frequent in Caucasian-American (34.8%) than in African-American individuals (20.7%) (P = 0.0001) and much less frequent in Chinese individuals (7.2%) (P = 0.0001 versus African-American or Caucasian-American). The homozygous Glu27 genotype was more frequent in Caucasian-American (15.4%) than African-American individuals (4.9%) (P = 0.003) and was not observed in Chinese. The Gly16 allele (54.3% versus 41.3%) and homozygous genotype (35.1% versus 18.3%) were more common in Caucasian-American than Chinese individuals (P = 0.003 for both). There is a marked ethnic difference in the frequency of these two common BAR2 polymorphisms among African-American, Caucasian-American and Chinese individuals, with a markedly reduced frequency of the Glu27 polymorphism, the polymorphism associated with resistance to desensitization and increased BAR2 responses, in African-American and Chinese individuals. Such ethnic genotypic differences may explain previously observed alterations in the response to the BAR agonists in different ethnic groups.

The relationship between six ADRA2B variants (one promoter, three coding, and two in the 3' untranslated region) with an allele frequency of more than 5% and desensitization was determined. Results Forty-one individuals (22 men, 21 whites, age 18-45 years) were studied. The ADRA2B 301-303 deletion allele (ins/del and del/del, n = 18) was associated with resistance to desensitization [1.01 (interquartile range 0.90-1.06)] as compared with ins/ins homozygous individuals (n = 23) [0.91 (interquartile range 0.73-0.99)], P = 0.026. In addition, the -98 GG, 1182 CC, and 1776 CC genotypes were associated with significantly less desensitization than GC or CC, and CA or AA genotypes, respectively. Conclusion Common ADRA2B variants contribute to the interindividual variability in vascular desensitization to an alpha(2)-AR agonist in vivo. Abstract:Response inhibition refers to the suppression of inappropriate or irrelevant responses. It has a central role in executive functions, and has been linked to a wide spectrum of prevalent neuropsychiatric disorders. Increasing evidence from neuropharmacological studies has suggested that gene variants in the norepinephrine neurotransmission system make specific contributions to response inhibition. This study genotyped five tag single-nucleotide polymorphisms covering the whole alpha-2B-adrenergic receptor (ADRA2B) gene and investigated their associations with response inhibition in a relatively large healthy Chinese sample (N=421). The results revealed significant genetic effects of the ADRA2B conserved haplotype polymorphisms on response inhibition as measured by stop-signal reaction time (SSRT) (F(2, 418)=5.938, p=0.003). Individuals with the AAGG/AAGG genotype (n=89; mean SSRT=170.2 ms) had significantly shorter SSRTs than did those with either the CCAC/AAGG genotype (n=216; mean SSRT=182.4 ms; uncorrected p=0.03; corrected p=0.09) or the CCAC/CCAC genotype (n=116; mean SSRT=195.8 ms; corrected p<0.002, Cohen's d=0.51). This finding provides the first evidence from association research in support of a critical role of the norepinephrine neurotransmission system in response inhibition. A better understanding of the genetic basis of response inhibition would allow us to develop more effective diagnosis, treatment, and prevention of deficient or underdeveloped response inhibition as well as its related prevalent neuropsychiatric disorders.

Abstract:Background and Objectives: The most common polymorphisms of the human beta(2)-adrenergic receptor - Arg16 --> Gly and Gln27 --> Glu - are associated with alterations in beta(2)-adrenergic receptor responses, both in vitro and in vivo. beta(2)-Adrenergic receptor-mediated vascular responses are affected by ethnicity, blood pressure, and genotype. We tested the hypothesis that these two common beta(2)-adrenergic receptor genetic variants are associated with essential hypertension in black or white Americans.Subjects and Methods: In a population-based case-control association study, the relationship between beta(2)-adrenergic receptor genotypes and hypertension was examined in 307 normotensive subjects (128 black and 179 white) and 356 hypertensive subjects (155 black and 201 white). A polymerase chain reaction-based single-stranded conformational polymorphism method with direct sequencing of the bands of interest was used to detect the two frequently occurring beta(2)-adrenergic receptor variants (Arg16-->Gly, Gln27-->Glu).Results: No significant differences in the distributions of alleles and genotypes of the tested beta(2)-adrenergic receptor variants were found between normotensive and hypertensive groups from either black or white Americans (all P >.05). There was a marked interethnic difference in the frequency of the Gln27-->Glu beta(2)-adrenergic receptor polymorphism in both normotensive and hypertensive subjects. Ln normotensive white: subjects, the variant Glu27 allele (35.2% versus 18.0%; P <.0001) and Glu27 homozygous genotype (14.0% versus 4.7%; P <.01) mere more common than in black subjects. Similarly, in hypertensive white subjects, the variant Glu27 allele (35.8% versus 18.4%; P <.0001) and the Glu27 homozygous genotype (15.9% ret sus 2.6%; P <.0001) were more common than in black subjects.Conclusions: These data suggest that although there are marked ethnic differences in their distribution, the common genetic polymorphisms of the human beta(2)-adrenergic receptor gene do not cosegregate with the presence of hypertension in either black or white Americans.

Abstract:OBJECTIVE:The study was conducted to investigate single nucleotide polymorphism(SNP) in beta2-adrenoceptor(beta2-AR) gene and the distribution of these identified SNPs in Chinese Han ethnic group.METHODS:beta2-AR gene was sequenced to detect SNPs by fluorescent labeling automatic sequencing method in 80 unrelated samples from territory of Dabie Mountain in Anhui province.RESULTS:A total of 8 SNPs were identified in length of 3.8 kb, including 5 SNPs in code region, 3 SNPs in regulatory region. Although the variations, -468C to G, -367T to C, -47C to T,-20T to C, +79C to G, +100G to A, +491C to T, +1098T to C have been identified in other ethnic groups, they have not been found in our study. The allele distribution of SNPs is in good unity with the Hardy-Weinberg equilibrium.CONCLUSION:The distribution of SNPs in beta2-AR gene is not equable and the SNPs in different ethnic groups differ greatly. Abstract:A dysfunction of the central noradrenergic system has long been suggested to be involved in attention-deficit/hyperactivity disorder (ADHD). Pharmacological evidence from animal studies and clinical practice has identified the alpha-2A adrenergic receptor gene (ADRA2A) as a candidate gene in ADHD. Some findings from Caucasian populations seem to support a role for this gene in ADHD. The current study first examined the association of the ADRA2A MspI and DraI polymorphisms with ADHD in the Han Chinese population, which differs quite substantially from the Caucasian population in the frequencies of alleles at these polymorphisms. No biased transmission of alleles of either polymorphism was observed using transmission disequilibrium. test (TDT) analysis in a sample of 268 nuclear families with an ADHD proband; however, haplotype analysis only identified a trend toward over-transmission of the M/C haplotype to probands with the combined subtype of ADHD (chi(2) = 3.233, P = 0.072). The mm genotype of the MspI polymorphism was also marginally related (P = 0.051) to lower ADHD symptom scores in a sample of 559 Chinese children with ADHD, which is inconsistent with data from Caucasian samples. Our results provide weak evidence for a possible role of ADRA2A in ADHD symptom expression. (C) 2006 Wiley-Liss, Inc.
SNP Identification in Α_(2a)-Adrenergic Receptor Gene in Chinese and the Effect on Gene Expression

Abstract:Objective : To scan single nucleotide polymorphism ( SNP ) in Chinese alpha-2A adrcnergic receptor(α2A-AR] gene and study the cffects of the SNP on the gene expres-sion. Methods: The complete sequence of α2A-AR gene was analyzed with automated DNA se-quencer to scan SNPs . Genomic DNA was extracted from whole blood and a 239 bp fragment containing the G/C polymorphism was amplified with PCR using a pair of specific primers. PCR-RFLP was used to perform the genotyping of the SNP at the site-1 296 bp of the people in the North of China . Electrophoresis mobility shift assay( EMSA ) was used to study the binding of the 390 bp fragments ( -1414-1025 bp) with G or C at the site-1296 bp and nuclear ex-tracts . Results : In our study, two SNPs were found in α2A-AR gene . Allele frequencies of the SNP at the site-1296 bp were 0.61 and 0.39 for G and C, and the genotype frequencies were 0.34,0.54 and 0.13 for GG, GC and CC respectively from the people in the North of China . In the EMSA, a specific binding appeared in the complex of nuclear extracts and DNA with C at-1296 bp . Conclusion : Two SNPs exist in α2A-AR gene from the people in the North of China , and DNA fragment with allele C of the SNP at the site-1296 bp could bind with a specific protein , which could influence the gene expression .

Abstract:beta-adrenoceptors are the common pharmacological targets for the treatment of cardiovascular diseases and asthma. Genetic modifications of beta-adrenergic system in engineered mice affect their lifespan. Here, we tested whether genes encoding for key components of the beta-adrenergic signaling pathway are associated with human longevity. We performed a 10-year follow-up study of the Chinese longitudinal healthy longevity survey. The Han Chinese population in this study consisted of 963 long-lived and 1028 geography-matched young individuals. Sixteen SNPs from ADRB1, ADRB2, ADCY5, ADCY6, and MAPK1 were selected and genotyped. Two SNPs, rs1042718 (C/A) and rs1042719 (G/C), of ADRB2 in linkage disequilibrium (D' = 1.0; r2 = 0.67) were found to be associated with enhanced longevity in men in two geographically isolated populations. Bonferroni-corrected P-values in a combined analysis were 0.000530.010. Men with haplotype A-C showed an increased probability to become centenarians (the frequency of A-C in long-lived and young individuals are 0.332 and 0.250, respectively, OR = 1.49, CI 95% = 1.171.88, P = 0.0007), in contrast to those with haplotype C-G (the frequency of C-G in long-lived and young individuals are 0.523 and 0.635, respectively, OR = 0.63, CI 95% = 0.510.78, P = 0.000018). The permuted P-values were 0.00005 and 0.0009, respectively. ADRB2 encodes the beta 2-adrenergic receptor; the haplotype A-C markedly reduced its translational efficiency compared with C-G (P = 0.002) in transfected HEK293 cells. Thus, our data indicate that enhanced production of beta 2-adrenergic receptors caused by genetic variants is inversely associated with human lifespan.

Abstract:Aim: Genetic polymorphisms causing Ser49Gly and Gly389Arg mutants of beta(1)-adrenoceptor may result in significant changes in the function of this receptor. The aim of the present study was to investigate the frequencies of the Ser49Gly and Gly389Arg mutant alleles in healthy Chinese populations and to investigate the differences between 2 Chinese ethnic groups (Han and Dai populations) with respect to the frequencies of these alleles. Methods: A total of 225 Han Chinese and 175 Dai Chinese unrelated healthy volunteers were recruited for this study. Genomic DNA was extracted from peripheral blood leukocytes by using a standard manual chloroform-phenol extraction. Fragments spanning the 2 polymorphisms were amplified by using polymerase chain reaction with template genomic DNA and relevant primers. The DNA products including the polymorphic loci were subjected to restriction endonuclease digestion with Eco0109I and BcgI. Digested fragments were detected with an ultraviolet detector after electrophoresis (100 V for approximately 1.5 h). Results: The frequencies of the Gly49 and Arg389 alleles were, respectively, 16.2% and 76.4% in the Han population and 14.6% and 75.7% in the Dai population. Conclusion: The polymorphisms causing the Ser49Gly and Gly389Arg mutations of the beta(1)-adrenoceptor existed in both healthy Han and Dai Chinese populations. The frequencies of the Ser49Gly and Gly389Arg mutant alleles were not significantly different in the Han and Dai populations. However, the frequency of the Gly389 variant seems to be significantly lower in these 2 populations than in an African-American population.
