Abstract:There are marked interethnic differences in beta (1)-adrenclceptor-mediated responsiveness, with sensitivity decreased in African-Americans and increased in Chinese compared with Caucasians, Therefore, the frequency of a common naturally occurring polymorphism of the human beta (1)-adrenoceptor gene (Arg(389)Gly), which has functional importance in vitro, was determined in 194 African-Americans, 316 Caucasian-Americans, 221 Hispanic-Americans and 142 Chinese. African-Americans were found to hare a significantly lower frequency of the Arg(389) allele than the other three ethnic groups (all P < 0.01), In the populations studied, the order of the distribution of the Arg(389) allele was: Chinese (74%) > Caucasians (72%) > Hispanics (67%) > African-Americans (58%). To determine the functional significance of the Arg(389)Gly beta (1)-adrenoceptsr polymorphism in-vivo heart rate responses to exercise were compared in healthy subjects homozygous for the Arg (n = 9) and Gly (n = 8) alleles, Heart rate response to exercise was not affected by genotype (P = 0.4). Although ethnic differences in the frequency of the beta (1)-adrenoceptor Arg(389)Gly polymorphism exist, the polymorphism does Hot appear to have functional significance in healthy subjects and therefore may not contribute to ethnic differences in response to drugs acting through the beta (1)-adrenoceptor, Pharmacogenetics 11:191-197 (C); 2001 Lippincott Williams & Wilkins.
Frequency of Functionally Important Beta-2 Adrenoceptor Polymorphisms Varies Markedly among African-American, Caucasian and Chinese Individuals.

Abstract:There are ethnic differences in the prevalence and severity of hypertension and asthma and in beta-2 adrenergic receptor (BAR2)-mediated vascular responses. Two common polymorphisms of the human BAR2, Arg16 to Gly and Gln27 to Glu, are associated with alterations in BAR2 response, both in vitro and in vivo. Ethnic differences in disease manifestations and responses to treatment may be explained by the altered frequency of BAR2 polymorphisms. To determine the relative frequencies of the Arg16 to Gly and Gln27 to Glu BAR2 polymorphisms in different ethnic groups we studied 415 (123 African-American, 188 Caucasian-American and 104 Chinese) healthy individuals. There was a marked interethnic difference in the frequency of the BAR2 polymorphisms among the ethnic groups. The Glu27 allele was more frequent in Caucasian-American (34.8%) than in African-American individuals (20.7%) (P = 0.0001) and much less frequent in Chinese individuals (7.2%) (P = 0.0001 versus African-American or Caucasian-American). The homozygous Glu27 genotype was more frequent in Caucasian-American (15.4%) than African-American individuals (4.9%) (P = 0.003) and was not observed in Chinese. The Gly16 allele (54.3% versus 41.3%) and homozygous genotype (35.1% versus 18.3%) were more common in Caucasian-American than Chinese individuals (P = 0.003 for both). There is a marked ethnic difference in the frequency of these two common BAR2 polymorphisms among African-American, Caucasian-American and Chinese individuals, with a markedly reduced frequency of the Glu27 polymorphism, the polymorphism associated with resistance to desensitization and increased BAR2 responses, in African-American and Chinese individuals. Such ethnic genotypic differences may explain previously observed alterations in the response to the BAR agonists in different ethnic groups.

Abstract:Background and Objectives: The most common polymorphisms of the human beta(2)-adrenergic receptor - Arg16 --> Gly and Gln27 --> Glu - are associated with alterations in beta(2)-adrenergic receptor responses, both in vitro and in vivo. beta(2)-Adrenergic receptor-mediated vascular responses are affected by ethnicity, blood pressure, and genotype. We tested the hypothesis that these two common beta(2)-adrenergic receptor genetic variants are associated with essential hypertension in black or white Americans.Subjects and Methods: In a population-based case-control association study, the relationship between beta(2)-adrenergic receptor genotypes and hypertension was examined in 307 normotensive subjects (128 black and 179 white) and 356 hypertensive subjects (155 black and 201 white). A polymerase chain reaction-based single-stranded conformational polymorphism method with direct sequencing of the bands of interest was used to detect the two frequently occurring beta(2)-adrenergic receptor variants (Arg16-->Gly, Gln27-->Glu).Results: No significant differences in the distributions of alleles and genotypes of the tested beta(2)-adrenergic receptor variants were found between normotensive and hypertensive groups from either black or white Americans (all P >.05). There was a marked interethnic difference in the frequency of the Gln27-->Glu beta(2)-adrenergic receptor polymorphism in both normotensive and hypertensive subjects. Ln normotensive white: subjects, the variant Glu27 allele (35.2% versus 18.0%; P <.0001) and Glu27 homozygous genotype (14.0% versus 4.7%; P <.01) mere more common than in black subjects. Similarly, in hypertensive white subjects, the variant Glu27 allele (35.8% versus 18.4%; P <.0001) and the Glu27 homozygous genotype (15.9% ret sus 2.6%; P <.0001) were more common than in black subjects.Conclusions: These data suggest that although there are marked ethnic differences in their distribution, the common genetic polymorphisms of the human beta(2)-adrenergic receptor gene do not cosegregate with the presence of hypertension in either black or white Americans.

Abstract:Objective: The alpha(1A)-adrenergic receptor is highly expressed in human vasculature including resistance arteries and veins, and its stimulation is primarily responsible for adrenergically mediated smooth muscle contraction. Variability in sensitivity to phenylephrine, an alpha(1A) adrenergic agonist, has a large genetic component. We examined the hypothesis that a common polymorphism of alpha(1A)-adrenergic receptor (Arg347Cys) affects in vivo response.Methods. We measured vascular sensitivity to phenylephrine using the dorsal hand vein linear variable differential transformer technique and determined alpha(1A)-adrenergic receptor genotype in 74 healthy, nonsmoking adults (28 Arg/Arg, 30 Arg/Cys, and 16 Cys/Cys).Results. Sensitivity to phenylephrine, expressed as the dose of phenylephrine resulting in 50% venoconstriction (Phe(50)), was not significantly different in subjects with the 3 alpha(1A) adrenergic receptor genotypes: Phe(50) geometric mean (95% confidence interval) was 513 ng/min (287-918 ng/min) for Arg/Arg, 431 ng/min (274-680 ng/min) for Arg/Cys, and 471 ng/min (197-1124 ng/min) for Cys/Cys (P = .90).Conclusion: We conclude that the Arg347Cys receptor polymorphism does not alter agonist-mediated venoconstriction in vivo.
Single Nucleotide Polymorphisms of the S CN5A Gene in Han Chinese and Their Relation with Brugada Syndrome

Abstract:BACKGROUND:Mutations in the cardiac sodium channel gene (SCN5A) may lead to a broad spectrum of familial arrhythmias, including long QT syndrome (LQTS), idiopathic ventricular fibrillation (IVF), and isolated cardiac conduction diseases. Recent studies have shown that polymorphisms in the SCN5A gene also play an important role in the manifestation of disorders involving cardiac excitability. In this study, we investigated the polymorphisms of the SCN5A gene in Han Chinese and its relation to Brugada syndrome (BS).METHODS:Genomic DNA was isolated from 120 unrelated healthy volunteers and 48 unrelated Brugada syndrome patients by means of standard procedures. All exons including the putative splicing sites of the SCN5A gene were amplified by PCR and sequenced directly or after subcloning using an ABI Prism 377 DNA sequencer.RESULTS:A total of 5 single nucleotide polymorphisms (SNPs) were identified in the Han Chinese population, including 3 novel ones: G87A(A29A), 4245 + 82A > G, and G6174A. The allele frequencies of each SNP in the Han Chinese population were as follows: G87A (A29A) 27.5%, A1673G (H558R) 10.4%, 4245 + 82A > G 32.8%, C5457T (D1819D) 41.3%, and G6174A 44.9%. S1102Y and 10 other SNPs identified in other ethnic populations were not detected in this study. There was no significant difference in the allele frequency of A1673G (H558R) between different ethnic populations (all P > 0.5). On the other hand, the allele frequency of C5457T (D1819D) among Han Chinese was similar to its frequency among Japanese (P > 0.5), but higher than that among Americans (P < 0.005). The allele G1673 (R558) was over-represented in BS patients compared to controls (P < 0.005), but there was no significant difference in genotype frequencies at this locus. There were also no differences in either the allele or genotype frequencies of the 4 other identified SNPs when comparing BS patients with healthy controls.CONCLUSIONS:The distribution of SCN5A SNPs may vary between different ethnicities. The polymorphism of A1673G might be associated with BS and may contribute to a susceptibility to BS in Han Chinese.

Abstract:Objectives Black patients may be less responsive to beta-blockers than whites. Genetic variants in the beta(1)-adrenergic receptor (beta(1)-AR) associated with lesser response to beta-blockers are more common in blacks than in whites. The purpose of this study was to determine whether ethnic differences in response to beta-blockade can be explained by differing distributions of functional genetic variants in the beta(1)-AR. Methods We measured sensitivity to beta-blockade by the attenuation of exercise-induced tachycardia in 165 patients (92 whites), who performed a graded bicycle exercise test before and 2.5 h after oral atenolol (25 mg). We determined heart rate at rest and at three exercise levels from continuous ECG recordings and calculated the area under the curve. We also measured plasma atenolol concentrations and determined genotypes for variants of the beta(1)-AR (Ser49Gly, Arg389Gly) and alpha(2C)-AR (del322-325). The effects of ethnicity, genotype, and other covariates on the heart rate reduction after atenolol were estimated in multiple regression analyses. Results Atenolol resulted in a greater reduction in exercise heart rate in whites than in blacks (P = 0.006). beta(1)-AR Arg389 (P = 0.003), but not the alpha(2C)-AR 322-325 insertion allele (P = 0.31), was independently associated with a greater reduction in heart rate area under the curve. Ethnic differences in sensitivity to atenolol remained significant (P = 0.006) after adjustment for beta(1)-AR and alpha(2C)-AR genotypes. Conclusion Ethnic differences in sensitivity to the beta(1)-blocker atenolol persist even after accounting for different distributions of functional genetic beta(1)-AR variants, suggesting that additional, as yet unidentified factors contribute to such ethnic differences.

Norepinephrine concentrations did not differ among subjects with 0, 1 and 2 copies of the deletion variant [median (interquartile range), 185.0 (147.5-269.8), 200.0 (154.9-257.0) and 173.8 (158.5-235.8) pg/ml, respectively; P = 0.54]. Similarly, none of the HRV parameters differed among the genotype groups. Abstract:Background. A common polymorphism of the beta(1)-adrenergic receptor Arg389Gly markedly affects function in vitro, but little is known about its in vivo significance.Methods and Results: Resting and exercise hemodynamic responses were measured in subjects homozygous for Arg389 (n = 21) or Gly389 (n = 13) alleles before and 3 hours after administration of a beta-blocker, atenolol. Demographic characteristics and atenolol concentrations were similar in the two genotypic groups. Genotype had a marked effect on resting hemodynamic responses to atenolol, with Arg389-homozygous subjects having a larger decrease in resting systolic blood pressure (8.7 +/- 1.3 mm Hg versus 0.2 +/- 1.7 mm Hg, P < .001) and mean arterial blood pressure (7.2 +/- 1.0 mm Hg versus 2.0 1.7 mm Hg, P = .009). Attenuation of exercise-induced hemodynamic responses by atenolol was not affected by genotype.Conclusions. There is reduced sensitivity of Gly389 homozygotes to a beta-adrenergic receptor antagonist, and this polymorphism may be an important determinant of variability in response to P-blockade.
Meta-analysis of C242T Polymorphism in CYBA Genes: Risk of Acute Coronary Syndrome is Lower in Asians but Not in Caucasians

Abstract:Background A lot of studies have demonstrated that C242T polymorphism in CYBA genes may play an important role in the pathological process of acute coronary syndrome (ACS). However, the results are not consistent. To further evaluate this debate, we performed a meta-analysis to determine the relationship between C242T polymorphism and ACS. Methods and results We screened PubMed/MEDLINE, EBSCIO, and EMBASE research reports until Mar. 2014 and extracted data from 10 studies involving 6102 ACS patients and 8669 controls. Subgroup analysis by ethnicity documented a significant decreased risk of ACS for C242T polymorphism in the Asian population under allelic comparison (odd ratio (OR) 0.73; 95% confidence intervals (CI) 0.64–0.83), dominant model (OR 0.71; 95% CI 0.62–0.82), and homozygote comparison (OR 0.57; 95% CI 0.35–0.92). However, in the overall population and especially with Caucasians, no significant association was uncovered. Further meta-regression analysis revealed that the heterogeneity among studies was largely attributed to ethnicity. No publication bias was detected through a funnel plot and an Egger’s linear regression test. Conclusions Taken together, our results suggest that the C242T polymorphism might be a protective factor against developing ACS in the Asian population. Further researches will be needed to identify the confounding factors which modified the protective effect of T allele among Caucasians.

Abstract:Objective: No clear consensus has been reached on the β2-adrenergic receptor polymorphisms (A46G and C79G) and essential hypertension risk. We performed a meta-analysis to summarize the possible association. Methods From 301 reports, we included 18 articles (20 studies) at the inclusion criteria. The fixed-effects model and the random-effects model were applied for dichotomous outcomes to combine the results of the individual studies. Results: There was no statistical association between A46G and hypertension risk in all subjects, Asians or Caucasians. However, the association was observed in dominant genetic model (AA vs. (AG + GG)) (P = 0.04, OR = 1.38, 95% CI [1.01–1.87], Pheterogeneity = 0.98) in the subgroup of the mixed Africans. No statistical association could be found between C79G and hypertension risk in overall or any ethnic subgroup. In the research conducted on the severe hypertension (SBP ≥160mmHg and/or DBP≥95mmHg hypertensive population), significant association could be found in dominant genetic model (CC vs. (CG + GG)) (P = 0.04, OR = 1.38, 95% CI [1.02 - 1.86], Pheterogeneity = 0.03), and there was also a boardline significance between the C79 allele and severe hypertension (P = 0.05, OR = 1.26, 95% CI [1.00–1.57], Pheterogeneity = 0.04). No association could be found in the study between these two polymorphisms and stage 2 hypertension. Conclusion Significant association between ADRB2 A46G polymorphism and hypertension was found in the population of the mixed African. The ADRB2 C79G polymorphism has significant association with the SBP ≥160 mmHg and/or DBP ≥95 mmHg hypertensive population.
Novel human α1a-adrenoceptor single nucleotide polymorphisms alter receptor pharmacology and biological function

Abstract:We identified nine naturally-occurring human single nucleotide polymorphisms (SNPs) in the α1a-adrenoceptor (α1aAR) coding region, seven of which result in amino acid change. Utilizing rat-1 fibroblasts stably expressing wild type α1aAR or each SNP at both high and low levels, we investigated the effect of these SNPs on receptor function. Compared with wild type, two SNPs (R166K, V311I) cause a decrease in binding affinity for agonists norepinephrine, epinephrine, and phenylephrine, and also shift the dose–response curve for norepinephrine stimulation of inositol phosphate (IP) production to the right (reduced potency) without altering maximal IP activity. In addition, SNP V311I and I200S display altered antagonist binding. Interestingly, a receptor with SNP G247R (located in the third intracellular loop) displays increased maximal receptor IP activity and stimulates cell growth. The increased receptor signaling for α1aAR G247R is not mediated by altered ligand binding or a deficiency in agonist-mediated desensitization, but appears to be related to enhanced receptor–G protein coupling. In conclusion, four naturally-occurring human α1aAR SNPs induce altered receptor pharmacology and/or biological activity. This finding has potentially important implications in many areas of medicine and can be used to guide α1aAR SNP choice for future clinical studies.
Genetic variants in the α2C-adrenoceptor and G-protein contribute to ethnic differences in cardiovascular stress responses:

Abstract:Objectives Cardiovascular responses to stressors are regulated by sympathetic activity, increased in black Americans, and associated with future cardiovascular morbidity. Our aim was to determine whether two functional variants in genes regulating sympathetic activity, a deletion in the alpha(2C)-adrenergic receptor (ADRA2C del322-325) and a G-protein beta 3-subunit variant (GNB3 G825T), affect cardiovascular responses to physiologic stressors and contribute to their ethnic differences. Methods We measured heart rate and blood pressure responses to a cold pressor test (CPT) in 79 healthy participants (40 blacks, 39 whites), aged 25.7 +/- 5.3 years, and determined genotypes for the ADRA2C and GNB3 variants. We examined the response variables (increase in heart rate and blood pressure) in multiple linear regression analyses adjusting first for baseline measures, ethnicity, and other covariates, and then additionally for genotypes. Results Black participants had a greater heart rate response to CPT than whites [mean difference, 9.9 bpm; 95% confidence interval (CI), 4.1 to 15.6; P=0.001]. Both the ADRA2C del/del (115.8 bpm; 95% CI, 8.0-23.7; P<0.001) and GNB3 T/T genotypes (6.8 bpm; 95% CI, 0.9-12.7; P=0.026) were associated with greater heart rate response. After adjusting for genotypes, the ethnic difference was abrogated (1.3 bpm; 95% CI, -5.4-8.0; P=0.70), suggesting that the genetic variants contributed substantially to ethnic differences. Conclusion Variation in genes that regulate sympathetic activity affects hemodynamic stress responses and contributes to their ethnic differences. This study elucidates how genetic factors may in part explain ethnic differences in cardiovascular regulation.

Abstract:The alpha(2B)-adrenergic receptor (ADRA2B) plays an important role in vasoconstriction and blood pressure regulation. One common variant in the ADRA2B gene (del301-303) has been identified, and results in markedly decreased receptor desensitization in vitro but does not alter vascular sensitivity in vivo. Therefore, we fully characterized genetic variations in ADRA2B and related them to phenotype in vivo. We examined 5812 bp of contiguous sequence of ADRA2B (promoter, exonic, and 3'-untranslated region; 3'-UTR) using the polymerase chain reaction to amplify the genomic target followed by bidirectional sequencing (n = 68). Haplotypes were inferred using an expectation maximization algorithm. Vasoconstriction in response to increasing doses of the highly selective alpha(2)-adrenergic receptor agonist, dexmedetomidine (0.01-1000 ng/min) was measured in the dorsal hand vein using a linear variable differential transformer. The dose that produced 50% (ED50) of maximum venoconstriction (E-max) was determined for each subject from the individual dose-response curves. ED50 and Emax were compared in subjects with and without variant alleles and haplotypes of interest. We identified 24 variable sites, 12 in the promoter region, five in the coding region (including two previously described as non-synonymous variants) and seven in the 3'-UTR region. Four haplotypes were inferred, representing approximately 95% of the cohort. One haplotype, characterized by two single nucleotide polymorphisms in the promoter region, and one in the 3'-UTR, occurred in seven of 38 African-Americans, and was associated with a lower E-max, 61.3% [95% confidence interval (CI) 39.5-83.0, n = 7] compared to 78.1% (CI 73.8-82.5) in wild-types (n = 61) (P = 0.02). There was no association between the nine common variants and dexmedetomidine ED50. We have described novel variants and haplotypes of the ADRA2B gene. These do not alter sensitivity to a selective alpha(2)-adrenergic receptor agonist but some may decrease maximal venoconstriction in vivo. (c) 2005 Lippincott Williams & Wilkins.

Abstract:Background Chronic obstructive pulmonary disease (COPD) is a heterogeneous and complex disease of which the pathogenesis remains largely unknown. Many factors could influence COPD development and progression. One of them is the genetic risk factor. A severe hereditary deficiency of alpha-1 antitrypsin is the best genetic proof. Four single nucleotide polymorphisms (SNPs) of beta2-adrenergic receptor (beta(2)AR) result in single amino acid substitution. Two loci had been extensively studied and found that they could change the function of beta(2)AR. Two SNPs consist of substitutions of glycine for arginine at amino acid position 16, glutamic acid for glutamine at position 27. Many studies proved that polymorphisms at position 16 and 27 altered the lung function of COPD patients or the patient's susceptibility to the development of COPD. However, there was no exclusive conclusion. Therefore, a meta analysis was done to investigate the effect of polymorphisms in the beta 2-adrenergic receptor (ADRB2) gene on the risk of COPD and lung function. Methods Comprehensive searches of MEDLINE, Embase, Ovid, HighWire, Cochrane Library, and Chinese databases (CBMdisc, VIP, CNKI, and Wanfang data) from January 1980 to September 2011 were performed, using the keywords: COPD OR chronic obstructive pulmonary disease AND adrenoreceptor OR adrenergic receptor AND polymorphism OR mutation OR variation. Case-control research or cross sectional studies in which diagnosis of COPD met the Global Initiative for Chronic Obstructive Lung Disease (GOLD) guidelines; all the studies reported the ADRB2 genotype at position 16 or 27. Outcomes measured were genotype frequency and forced expiratory volume in the first second (FEV1%) in both the case and control. Results Twelve case-control studies and eight cross-sectional studies were included. Compared to the control (n=1225), neither Gly/Gly (n=527) nor Arg/Arg (n=422) homozygotes at position 16 demonstrated increased susceptibility to COPD, with odds ratios (ORs) of 0.95 (95% CI (0.68, 1.31), z=0.33, P=0.740) and 0.82 (95% CI (0.52, 1.28), z=0.88, P=0.381), respectively. Similar results were obtained for position 27, with ORs of 0.97 (95% CI (0.77, 1.23), z=0.21, P=0.833) for Glu/Glu homozygotes (n=357) and 0.82 (95% CI (0.53, 1.29), z=0.85, P=0.393) for Gln/Gln homozygotes (n=704) (control=1183). In patients with COPD, Arg/Arg homozygotes (n=41) had a similar FEV1% compared with Gly/Gly homozygotes (n=102) (standardized mean difference (SMD)=0.88, 95% CI (-0.85, 2.62), z=1.00, P=0.319). The genotype distribution was different between Caucasian and Asian populations (all P <0.05 except the genotype Arg/Gly) for both position 16 and 27. Conclusions Polymorphisms of ADRB2 at positions 16 and 27 did not change the risk of COPD nor affect lung function or disease severity. Abstract:We sought to investigate the association of single nucleotide polymorphisms (SNPs) of the genes involved in βAR signaling with the response of patients to βAR blockers. A total of 2403 hospitalized patients with chronic heart failure (HF) were enrolled in a multicenter observational study as the first cohort and followed up for a mean period of 20 months. Genes for β1AR, β2AR, and the major cardiac G-protein-coupled receptor kinases (GRKs) GRK2 and GRK5 were analyzed to identify SNPs, and patients were stratified according to genotypes. A second independent cohort enrolling 919 patients with chronic HF was applied to validate the observed associations. The signaling properties of the key identified SNPs were assessed in vitro. Our data showed that HF patients harboring the Gly16 allele in the gene for β2AR ( ADRB2 ) had an increased risk of the composite end point relative to patients who were homozygous for Arg16. Notably, these patients showed a beneficial response to βAR-blocker treatment in a G allele-dose-dependent manner, whereas Arg16 homozygotes had no response to βAR-blocker therapy. This Arg16Gly genotype-dependent heterogeneity in clinical outcomes of HF was successfully validated in the second independent population. Besides, the in vitro experiments revealed that G allele carriers were defective in β2AR-coupled inhibitory adenylate cyclase g (G i ) protein signaling.
The Effect of Common Polymorphisms of the β<sub>2</sub>-Adrenergic Receptor on Agonist-Mediated Vascular Desensitization

Abstract:Background: With continuous exposure to beta (2)-adrenergic agonists, vascular tissue becomes desensitized to agonist-mediated vasodilatation. We studied the effects of two common polymorphisms of the beta (2)-adrenergic receptor, one at codon 16 and one at codon 27, on agonist-mediated vasodilatation and desensitization in the vascular bed. Methods: We studied 26 healthy subjects who were selected to represent three genotypes: 7 were homozygous for the alleles encoding Arg16 and GIn27, 8 were homozygous for the alleles encoding Gly16 and Gln27, and 11 were homozygous for the alleles encoding Gly16 and Glu27. Vascular responses were assessed by measuring changes in the diameter of a dorsal hand vein. A dose-response curve of the effect of the beta (2)-adrenergic-receptor agonist isoproterenol was constructed (dose range, 4 to 480 ng per minute). Desensitization was then induced by a 2-hour continuous infusion of isoproterenol, and venodilatation was measured 30, 60, 90, and 120 minutes after the start of the infusion. Results: Subjects who were homozygous for Arg16 had almost complete desensitization; venodilatation in response to isoproterenol in this group decreased from a mean (+/-SE) of 44+/-11 percent to 8+/-4 percent (P=0.006). In contrast, subjects who were homozygous for Gly16 did not have significant desensitization, irrespective of the amino acid encoded by codon 27. Subjects who were homozygous for Glu27 had higher maximal venodilatation in response to isoproterenol than those who were homozygous for Gln27 (86+/-13 percent vs. 54+/-8 percent, P=0.03). Conclusions: The Arg16 polymorphism of the beta (2)-adrenergic receptor is associated with enhanced agonist-mediated desensitization in the vasculature, and the Glu27 polymorphism is associated with increased agonist-mediated responsiveness. Therefore, polymorphisms of the beta (2)-adrenergic receptor are potentially important determinants of the vascular response to stress. (N Engl J Med 2001;345:1030-5.)
