Abstract:We present a practical and convergent synthesis of glycoalkaloids solasonine 1 and its saponin derivative 2 , incorporating a {3- O -α- L -rhamnopyranosyl-(1→2)- O -[β- D -glucopyranosyl-(1→3)]-β- D -galactopyranoside} moiety. The key features of this strategy include the following: (1) AuCl 3 – t BuCN cooperative catalysis enabling 1,2- trans stereoselective glycosidation of 2-branched trisaccharide trichloroacetimidate donors with steroidal aglycons, in the absence of neighboring group participation; (2) "cyanide effect" mediated regioselective benzoylation of the 4- and 6-hydroxyl groups of galactopyranosyl disaccharide; and (3) an effective approach to prevent orthoester byproduct formation.

Abstract:Total synthesis of simonsol C has been achieved, focusing on the postdearomatization transformations. Our methodology integrates an efficient combination of dearomatization and Zn/AcOH reduction to introduce an allyl group, followed by oxo-Michael addition, to construct the 6/5/6 benzofuran skeleton. It offers a novel method for synthesizing allyl-containing quaternary carbon atoms in a straightforward manner.

Abstract:Solanesol, a main polyprenol occurred in tobacco, has gained the attention of biochemists because of its value as a source of isoprene units for the synthesis of metabolically active quinones and vitamin K analogs. This article deals with extraction and purifyication of solanesol. Ground tobacco leaf was extracted with petroleum ether in 50°C water bath under reflux for 2h, and then filtrated, the powder leaf was re-extracted successively with petroleum ether and the extracts were pooled and concentrated to get pasty residue. The residue was saponified and subsequently loaded onto silica gel column chromatography. The chemical was eluted using a binary solvent mixture of petroleum ether–acetone (90:10, v/v) and the eluent was collected by fraction size of 5ml. Tentative identification was carried out using TLC. The fractions mainly contained solanesol were pooled and dried by rotary evaporation. The yield of solenosol was 0.38% of the dry weight of tobacco leaf, and the purity was 83.04%.

Abstract:The method of solid-phase extraction (SPE) for the concentration and clean-up of tobacco extract samples during solanesol analysis was proposed in this work. A column (200 mm × 4 mm i.d.) packed with 0.10 g silica gel (with particle size of 70 μm, porosity of 0.5 and surface area of 400 m2 g−1) was used as SPE cartridge. Several extraction parameters, such as sample loading flow (0.3–7 mL min−1), sample volume (5–50 mL), the column adsorption capacity and elution were evaluated to provide a fast, quantitative and reproducible SPE method. A mean solanesol recovery was 97.5 ± 1.6% (mean ± sd) and mean intra- and inter-day reproducibility was higher than 97%. It can be used in the analysis of solanesol in tobacco extracts.

Abstract:Solanesol is a non-cyclic terpene alcohol composed of nine isoprene units that mainly accumulates in solanaceous plants. Solanesol plays an important role in the interactions between plants and environmental factors such as pathogen infections and moderate-to-high temperatures. Additionally, it is a key intermediate for the pharmaceutical synthesis of ubiquinone-based drugs such as coenzyme Q10 and vitamin K2, and anti-cancer agent synergizers such as N-solanesyl-N,N'-bis(3,4-dimethoxybenzyl) ethylenediamine (SDB). In plants, solanesol is formed by the 2-C-methyl-d-erythritol 4-phosphate (MEP) pathway within plastids. Solanesol's biosynthetic pathway involves the generation of C5 precursors, followed by the generation of direct precursors, and then the biosynthesis and modification of terpenoids; the first two stages of this pathway are well understood. Based on the current understanding of solanesol biosynthesis, we here review the key enzymes involved, including 1-deoxy-d-xylulose 5-phosphate synthase (DXS), 1-deoxy-d-xylulose 5-phosphate reductoisomerase (DXR), isopentenyl diphosphate isomerase (IPI), geranyl geranyl diphosphate synthase (GGPPS), and solanesyl diphosphate synthase (SPS), as well as their biological functions. Notably, studies on microbial heterologous expression and overexpression of key enzymatic genes in tobacco solanesol biosynthesis are of significant importance for medical uses of tobacco.

Abstract:A concise asymmetric synthesis of two naturally occurring seco-type cholestane alkaloids (-)-solanidine and (-)-tomatidenol from (-)-diosgenin with a linear reaction sequence of 12 steps and 13 steps, respectively, is reported. The synthetic strategy includes the highly controlled establishment of highly functionalized octahydroindolizine ((-)-solanidine) and 1-oxa-6-azaspiro[4.5]decane ((-)-tomatidenol) cores with five stereocenters, respectively, from (-)-diosgenin, featuring two stereoselective cascade transformations including a modified cascade ring-switching process of furostan-26-acid to open the E-ring of (-)-diosgenin and a cascade azide reduction/intramolecular reductive amination to close the E- and F-rings of (-)-solanidine and (-)-tomatidenol. This work should enable further explorations of chemical and biological spaces based on solanidine, tomatidenol and related natural products.

Abstract:Ultrasound irradiation (USI), by virtue of cavitational collapse, is able to conduct diverse organic reactions with outstanding outcomes in terms of reaction rates, yields, and selectivities. On another note, solvent-free synthesis has received considerable attention owing to the rising concern for the effect of solvents on the environment. USI combined with no solvent conditions leads to eco-friendly synthetic conditions together with considerable improved selectivity, cleaner reactions, higher yields and purities, and reduction of reaction times. This chapter includes material illustrating up-to-date developments on US-assisted solvent-free synthesis of heterocyclic compounds. To be more precise, the chapter consists of two parts. The first part deals with US-assisted solvent- and catalyst-free one-pot multicomponent synthesis of 1,4-diazabutadienes, pyrrolidines, α-aminophosphonates, formamidines, pyrazolones, 1,4-dihydropyridines, amines, formamides, and silyl ethers. The second part focuses on US-assisted solvent-free one-pot multicomponent synthesis of trans-chalcones, 3,4-dihydro-2(1H)-quinolinones, imidazoles, coumarins, 1H-1,2,3,4-tetrazoles, β-indolylketones, 1,5-benzodiazepines, α-oxycarbanilinophosphonates, dihydropyrimidinones, β-iodo vinylthiocyanates, α-hydroxyphosphonates, α-aminophosphonates, and 2-aminothiophenes.

Abstract:Stereoselective syntheses of terpenoids in a more efficient manner have been a long-term pursuit for synthetic chemists. Herein we describe the two-step, enantiospecific and protecting-group-free synthesis of (+)-schisanwilsonene A from a carotane compound, which was produced in <i>E. coli</i>. We also completed the first enantiomeric synthesis of (+)-tormesol in five steps.
