Abstract:Although pH-switchable behaviors have been reported based on multilayer films modified electrodes, their pH-switchable biosensing is still difficult due to the existence of the electroactive mediator. In this study, we report the pH-dependable determination of hydrogen peroxide (H2O2) based on a four-bilayer film fabricated through layer by layer assembly between hemoglobin (Hb) and multiwall carbon nanotubes (MWCNTs). We observed that response of electroactive probe Fe(CN)6(3-) at the multilayer films was very sensitive and reversible to pH values of phosphate buffer solutions phosphate buffer solution with cyclic voltammetry. The reduction peak height of Fe(CN)6(3-) at the multilayer film could reach ∼221μA at pH 3.0 while 0μA at pH 9.0. The linear range for the detection of H2O2 at pH 3.0 was from 12.5μM to 10.4mM, which was much wider than that at pH 9.0. Our results demonstrated that the detection of H2O2 with the proposed modified electrode is dependent on pH values of phosphate buffer solution. Moreover, the component of multilayer films has impacts on the performance of biosensors with pH-switchable behaviors.
Disposable L-Lactate Biosensor Based on A Screen-Printed Carbon Electrode Enhanced by Graphene

Abstract:In this work, an amperometric L-lactate biosensor based on a graphene-modified screen-printed carbon electrode (SPCE) was constructed. First, the electrocatalytic performance of the SPCE modified with graphene by a one-step electrodeposition process (OerGO/SPCE) was investigated. The cyclic voltammogram of OerGO/SPCE, which showed a well-defined redox peak, had a smaller peak potential separation than that of SPCE, revealing the improvement in electron transfer speed brought about by modifying with graphene. Next, lactate oxidase and potassium ferricyanide were dropped on the OerGO/SPCE to construct a graphene-modified L-lactate biosensor (LOD/K3[Fe(CN)6]/OerGO/SPCE). The proposed biosensor, with a detection limit of 60 04, had a high sensitivity (42.42 mu A mM(-1) cm(-2)) when working at a low working potential (0.15V). The linear range was 0.5 mM-15mM, covering the detecting range of L-lactate in clinical applications. The L-lactate biosensor had a short response time (10s) and required only 10 mu l of the sample. This L-lactate sensor modified with electrodeposited graphene had a larger sensitivity than that based on the bare SPCE. Thus, our low-cost and disposable L-lactate biosensor enhanced by graphene can perform as an attractive electrochemical device that can be manufactured for point-of-care testing (POCT) devices and be employed in POCT applications.

Abstract:Potentiometric biosensors based on flexible arrayed silver paste electrode and copper-doped zinc oxide sensing film modified by iron-platinum nanoparticles (FePt NPs) are designed and manufactured to detect lactate in human. The sensing film is made of copper-doped zinc oxide (CZO) by a radio frequency (RF) sputtering system, and then modified by iron-platinum nanoparticles (FePt NPs). The surface morphology of copper-doped zinc oxide (CZO) is analyzed by scanning electron microscope (SEM). FePt NPs are analyzed by X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR). The average sensitivity, response time, and interference effect of the lactate biosensors are analyzed by voltage-time (V-T) measurement system. The electrochemical impedance is analyzed by electrochemical impedance spectroscopy (EIS). The average sensitivity and linearity over the concentration range 0.2–5 mM are 25.32 mV/mM and 0.977 mV/mM, respectively. The response time of the lactate biosensor is 16 s, with excellent selectivity.

Abstract:In this study, we developed an electrochemical microchannel biosensor platform to analyse lactate metabolism in cells. This biosensor platform was fabricated by photolithography, thin-film deposition and microfluidic technology. A kind of functional biomaterial was prepared by mixing lactate oxidase, single-walled carbon nanotubes and chitosan, and platinum as working and blank electrodes of the biosensor was modified by a thin Prussian blue layer. The lactate biosensor was obtained by dropping functional biomaterials on the electrode. The results demonstrated that the sensitivity of the electrochemical biosensor was up to 567 nA mM-1 mm-2 and the limit of detection was 4.5 μM (vs. Ag/AgCl as the counter/reference electrode). The biosensor used to quantitatively detect metabolic lactate concentrations in HepG2 cells cultured with cancer drugs showed high sensitivity, selectivity and stability, and has potential applications in organ-on-a-chip and tissue engineering technologies, which typically involve low concentrations of metabolites.

Abstract:A novel disposable lactate biosensor was reported, which can measure the lactate in whole blood and other bio-fluids rapidly, accurately, expediently and directly. The biosensor was based on two kinds of electron mediator, the ferricyanide and the modified carbon nanotubes (CNT). Lactate oxidase (LOD) was coated on the electrodes. The sensor was fabricated by screen-printed techniques, the sample was supplied through capillary action and the volume was controlled by micro cavity. The liner range is 1~15 mmol/l, precision reaches 0.2 mmol/l . Compared with the respondence of sensor without carbon nanotubes, the characteristic of sensor include that is improved obviously.

Abstract:We report on the formation of colloidal biocomposites resulting from the electrostatic self-assembly of negatively charged lactate oxidase (LOD) and oppositely charged nanoscaled cobalt phthalocyanine (NanoCoPc) colloid under enzyme-friendly conditions. The biocomposites were further used to fabricate lactate biosensors due to their excellent film-forming ability and strong adsorbability on the surface of glassy carbon electrode. The electrochemical assays of such films revealed a large capacity of NanoCoPc for the retention of LOD. Here, NanoCoPc colloid was not only used as carriers for immobilization of LOD, but also displayed intrinsic electrocatalytic activity for the oxidation of H2O2, a product of enzymatic reaction. A chitosan film containing MnO2 nanoparticles was further electrodeposited as an external layer, which could effectively eliminate the interference from ascorbic acid. Under optimal conditions, the biosensor showed a wide linear response to lactate in the range of 0.020–4.0mM, with high sensitivity (3.98μAcm−2mM−1), as well as good reproducibility and long-term stability. The biosensor has been used for the determination of lactate in real samples with an acceptable accuracy.

Abstract:Biosensors based on Organic Electrochemical Transistors (OECTs) are developed for the selective detection of glucose and lactate. The transistor architecture provides signal amplification (gain) with respect to the simple amperometric response. The biosensors are based on a poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) channel and the gate electrode is functionalised with glucose oxidase (GOx) or lactate oxidase (LOx) enzymes, which are immobilised within a Ni/Al Layered Double Hydroxide (LDH) through a one-step electrodeposition procedure. The here-designed OECT architecture allows minimising the required amount of enzyme during electrodeposition. The output signal of the biosensor is the drain current (Id), which decreases as the analyte concentration increases. In the optimised conditions, the biosensor responds to glucose in the range of 0.1–8.0 mM with a limit of detection (LOD) of 0.02 mM. Two regimes of proportionality are observed. For concentrations lower than 1.0 mM, a linear response is obtained with a mean gain of 360, whereas for concentrations higher than 1.0 mM, Id is proportional to the logarithm of glucose concentration, with a gain of 220. For lactate detection, the biosensor response is linear in the whole concentration range (0.05–8.0 mM). A LOD of 0.04 mM is reached, with a net gain equal to 400.

Abstract:An excellent biosensor with ZnO nanowires-gated AlGaAs/GaAs high electron mobility transistor (HEMT) was used to detect lactic acid. Due to the new structure, addition of the Si-doped GaAs cap layer, the HEMT biosensor could detect a wide range of lactic acid concentrations from 0.03 nM to 300 mM. The novel biosensor exhibiting good performance along with fast response, high sensitivity, wide detection range, and long-term stability, can be integrated with a commercially available transmitter to realize lactic acid detection.

Abstract:A LOx-based electrochemical biosensor for high-level lactate determination was developed. For the construction of the biosensor, chitosan and Nafion layers were integrated by using a spin coating procedure, leading to less porous surfaces in comparison with those recorded after a drop casting procedure. The analytical performance of the resulting biosensor for lactate determination was evaluated in batch and flow regime, displaying satisfactory results in both modes ranging from 0.5 to 20 mM concentration range for assessing the lactic acidosis. Finally, the lactate levels in raw serum samples were estimated using the biosensor developed and verified with a blood gas analyzer. Based on these results, the biosensor developed is promising for its use in healthcare environment, after its proper miniaturization. A pH probe based on common polyaniline-based electrochemical sensor was also developed to assist the biosensor for the lactic acidosis monitoring, leading to excellent results in stock solutions ranging from 6.0 to 8.0 mM and raw plasma samples. The results were confirmed by using two different approaches, blood gas analyzer and pH-meter. Consequently, the lactic acidosis monitoring could be achieved in continuous flow regime using both (bio)sensors.
Wearable Carbon Nanotube-Based Biosensors on Gloves for Lactate

Abstract:Developing a simple and direct approach for interfacing a sensor and a target analyte is of great interest for fields such as medical diagnosis, threat detection, food quality control, and environmental monitoring. Gloves provide a unique interface for sensing applications. Here, we show for the first time the development of wearable carbon nanotube (CNT)-based amperometric biosensors painted onto gloves as a new sensing platform, used here for the determination of lactate. Three sensor types were studied, configured as: two CNT electrodes; one CNT electrode, and an Ag/AgCl electrode, and two CNT electrodes and an Ag/AgCl electrode. The sensors are constructed by painting the electrodes using CNT or Ag/AgCl inks. By immobilizing lactate oxidase onto the CNT-based working electrodes, the sensors show sensitive detections of lactate. Comparison of sensor performance shows that a combination of CNT and Ag/AgCl is necessary for highly sensitive detection. We anticipate that these findings could open exciting avenues for fundamental studies of wearable bioelectronics, as well as practical applications in fields such as healthcare and defense.

Abstract:Lactate is an important organic molecule that is produced in excess during anaerobic metabolism when oxygen is absent in the human organism. The concentration of this substance in the body can be related to several medical conditions, such as hemorrhage, respiratory failure, and ischemia. Herein, we describe a graphene-based lactate biosensor to detect the concentrations of L-lactic acid in different fluids (buffer solution and plasma). The active surface (graphene) of the device was functionalized with lactate dehydrogenase enzyme using different substances (Nafion, chitosan, and glutaraldehyde) to guarantee stability and increase selectivity. The devices presented linear responses for the concentration ranges tested in the different fluids. An interference study was performed using ascorbic acid, uric acid, and glucose, and there was a minimum variation in the Dirac point voltage during detection of lactate in any of the samples. The stability of the devices was verified at up to 50 days while kept in a dry box at room temperature, and device operation was stable until 12 days. This study demonstrated graphene performance to monitor L-lactic acid production in human samples, indicating that this material can be implemented in more simple and low-cost devices, such as flexible sensors, for point-of-care applications.

Abstract:The development of new bioelectronic sensors could enable significant advances in clinical analysis, environmental monitoring, and defense. Cotton fabric is a wearable material, which can contact the skin closely. Here, we demonstrate for the first time the development of a wearable amperometric biosensor on a cotton fabric for the detection of lactate. The sensor is constructed by printing carbon graphite ink and Ag/AgCl ink on a cotton fabric as the working, reference, and counter electrode. Via the immobilization of lactate oxidase, the sensor shows a sensitive detection of lactate with a detection range of 0.05-1.5 mM and a rapid measuring time of around 5 min. We anticipate that these results could open exciting opportunities for fundamental studies and practical applications of wearable bioelectronics in areas ranging from healthcare to defense.

Abstract:Generally a glucose-sensitive enzyme field-effect transistor (ENFET) is based on local pH change in biomembranes resulted from the formation of gluconic acid. Here we proposed a glucose ENFET based on a new principle. The glucose ENFET was fabricated by coimmobilizing glucose oxidase (GOD) and MnO2 nanoparticles on the gate of an ion-sensitive field-effect transistor (ISFET). The proposed glucose biosensor shows a significant local pH increase in the sensitive membrane with the increase of glucose concentration. The driving force of the pH change in our sensor is essentially different from all the other glucose ENFETs, including those prepared by bulk MnO2. The special reaction ability of MnO2 nanoparticles with hydrogen peroxide might be the main cause of the pH change. In addition, the influence of buffer concentration, pH and ionic strength on the glucose ENFET is investigated in detail. It is found that the ionic strength has little effect on the performance of the ENFET. Under optimal conditions, the proposed ENFET exhibits a linear response with glucose in the range of 0.025–1.90mM, an extended dynamic upper limit of 3.5mM glucose, and considerable good reproducibility and stability.

Abstract:Lactate levels in humans reveal intensity and duration of exertion and provide a critical readout for the severity of life-threatening illnesses such as pediatric sepsis. Using the lactate oxidase enzyme (Lox) from Aerococcus viridians, we demonstrated its functionality for lactate electrochemical sensing in physiological fluids in a lab setting. The structure and dynamics of LOx were validated by crystallography, X-ray scattering, and hydroxyl radical protein footprinting. This provided a validated protein template for understanding and designing an enzyme-based electrochemical sensing elements. Using this template, LOx enzyme variants were generated and compared. Comparison of the variants demonstrates that one exhibits effective lactate sensing at significantly reduced operating voltages. Additionally, we demonstrate that the four hexahistidine-tags on each enzyme tetramer are sufficient for immobilization to create a durable, functional sensor, with no need for a covalent attachment, enabling self-immobilization and eliminating the need for additional immobilization steps. The functionality of the LOx enzyme variants was verified at physiological lactate concentrations in both human serum (0-4 mM) and artificial sweat (0-100 mM) using 3-electrode setups for analysis of the three variants in parallel. Accuracy of measurement in both artificial sweat and human serum were high. Employing a microfluidic flow cell, we successfully monitored varying lactate levels in physiological fluids continuously over a 2h period. Overall, this optimized LOx enzyme, which self-immobilizes onto gold sensing electrodes, facilitates efficient and reliable lactate detection and continuous monitoring at reduced operating voltages suitable for further development towards commercial use.

Abstract:In the wearable electrochemical biosensors, sensing signal duration is significantly dependent on the long-term stability of functional materials modified on the flexible substrate, the effect of pH changes of sweat on the sensing device and signal fluctuation caused by the bending of sensor. Here, we proposed a wearable biosensor based on the lactate-sensing membrane mainly constituted by Prussian blue (PB), reduced graphene oxide (rGO), Au nanoparticles and lactate oxidase (LOx). Based on the in-situ layer-by-layer spin-coating preparation method, the electrode surface was covered with an extensive and uniform PB/GO membrane with a high stability. After the electro-reduction of GO to rGO and the combination of urchin-like Au particles with sufficient tentacles to LOx, the sensing membrane showed the improved electron transport from the enzyme active center to the electrode. Therefore, the wearable biosensor achieved a high sensitivity of 40.6 μA mM−1 cm−2 in a range of 1–222 μM and a low sensitivity of 1.9 μA mM−1 cm−2 in a wide range of 0.222–25 mM, satisfying the requirement of the typical test. In addition, with the excellent running and mechanical stability, the lactate biosensor was successfully applied on volunteers' skin for real-time monitoring of perspiration in vivo. The results were comparable with ex vivo measurements achieved by a commercial lactate sensor. The wearable electrochemical biosensor provides a good candidate in the future for the evaluation of human sweat in sports and biomedical fields.

Abstract:L-Lactate is an important bioanalyte in the food industry, biotechnology, and human healthcare. In this work, we report the development of a new L-lactate electrochemical biosensor based on the use of multiwalled carbon nanotubes non-covalently functionalized with avidin (MWCNT-Av) deposited at glassy carbon electrodes (GCEs) as anchoring sites for the bioaffinity-based immobilization of a new recombinant biotinylated lactate oxidase (bLOx) produced in Escherichia coli through in vivo biotinylation. The specific binding of MWCNT-Av to bLOx was characterized by amperometry, surface plasmon resonance (SPR), and electrochemical impedance spectroscopy (EIS). The amperometric detection of L-lactate was performed at −0.100 V, with a linear range between 100 and 700 μM, a detection limit of 33 μM, and a quantification limit of 100 μM. The proposed biosensor (GCE/MWCNT-Av/bLOx) showed a reproducibility of 6.0% and it was successfully used for determining L-lactate in food and enriched serum samples.

Abstract:Emerging wearable devices with non-invasively biosensing technics have drawn considerable attention to continuously monitor several metabolites in body fluids, such as a tear, saliva, and sweat, to diagnose human health conditions. Most importantly, wearable sensors could offer unique possibilities for online, real-time and non-invasive monitoring of health compared to traditional invasive biosensors. Among lots of analytes, lactic acid concentration in the human body exhibits a high relationship with several diseases such as acute heart diseases, hypoxia, muscle fatigue, meningitis, and cystic fibrosis, and it could also cause muscle pain in athletes. To further boost up the reproducibility and reliability of wearable biosensors to detect lactate concentration levels from human sweat, Ni-based layered double hydroxide (LDH) with various secondary transition metals (Fe and Co) was proposed as electrocatalysts in an enzyme-free electrochemical lactate sensor. According to the mechanism of lactate oxidation on the transition metal-based electrocatalyst, secondary transition metal of Co could serve as the active site for lactate oxidation and facilitate the adsorption of OH - in the alkaline electrolyte. To further increasing the active surface area for enhancing the sensitivity of Ni-based LDH, ZIF-67 derived NiCo LDH was synthesized as the electrocatalyst for non-enzymatic lactate detection. Co-based ZIF-67 served as self-sacrificial templates to fabricate hierarchically structural NiCo LDH with uniform porosity and high electrochemically active surface area to achieve outstanding electrocatalytic performance for lactate sensing. After optimizing the particle size of ZIF-67 and transformation times, ZIF-67 derived NiCo LDH reached the ultrahigh sensitivity of 83.98 μA mM - 1 cm - 2 at an applied potential of 0.55 V (vs. Ag/AgCl KCl sat’d) in the concentration range from 2 to 26 mM. On the other hand, pioneering works in biosensors for lactate detection in sweat has been encountered major challenges such as noble material usage, immobile power supply, and complicated circuit connection to realize the compact sustainable sensing systems. To solve these restrictions, herein, the self-powered molecular imprinted polymers based triboelectric sensor (MIP-TES) was designed to offer a multifunctional noninvasive approach for specific and simultaneous lactate detection. Free-standing PVDF/graphene flexible electrode modified poly(3-aminophenyl boronic acid) imprinted lactate molecule demonstrated the change of the surface properties afterlactate adsorption. MIP-modified electrode revealed the selective lactate sensing over non molecular imprinted polymers (NIP) electrode through the superior and stable signal change with variation of lactate concentration in human sweat. Moreover, MIP modified lactate sensor was further introduced in the triboelectric nanogenerator system to harvest mechanical energy from contact and separation into electrical output. The more adsorbed lactate led to lower energy barriers and decreasing electrical potential when detecting higher lactate concentration. Self-power triboelectric lactate sensor could directly power the number of LED lights without an external energy supply. Eventually, it was validated the feasible application of wearable sensors on human skin. After introducing noninvasive enzyme-free biosensors and triboelectric sensors, an innovatively continuous non-invasive health monitoring platforms can be achieved for practical applications, especially in the areas of home medical examination and wearable personal biosensors.

Abstract:Room temperature Ionic liquids (RTILs) have evolved as a new type of solvent for biocatalysis, mainly due to their unique and tunable physical properties.[1] In addition, within the family of organic semiconductor-based sensors, organic electrochemical transistors (OECTs) have attracted particular interest.[2] Here, we present a simple and robust biosensor, based on a OECT, capable of measuring lactic acid using a gel-like polymeric materials that endow RTIL (ionogel)[3] as solid-state electrolyte both to immobilise the enzyme and to serve as a supporting electrolyte.[4] This represents the first step towards the achievement of a fast, flexible, miniaturised and cheap way of measuring lactate concentration in sweat.

Abstract:Organic electrochemical transistors (OECTs) have emerged as an advantageous choice for constructing biosensors due to their remarkable water compatibility, low operating voltage, and inherent amplification capability. However, the current research on bio-sensing based on OECTs predominantly employs p-type material PEDOT:PSS as the channel material. Nevertheless, the utilization of a single material and its depleting characteristics impose significant limitations on device miniaturization and integration applications. We present a high-sensitivity glucose sensor based on n-channel accumulation-type OECT through immobilization the glucose oxidase on Pt gate electrode with an blending approach. The glucose oxidase directly mixed with bovine serum albumin, chitosan and immolizied on the gate electode, demonstrating a stable and sensitive response to the glucose. The modified n-channel organic electrochemical transistor demonstrates a highly sensitive response to glucose across a concentration range from M to mM, with a quantified stable sensitivity over 2.69/mmol by normalizing the current change with respect to concentration. The device also exhibits selectivity towards glucose (compared with high concentrations of lactic acid, different concentration gradients of ascorbic acid, and uric acid), rendering it suitable for noninvasive glucose detection in body fluid like sweat, saliva etc. This flexible and ensitive electrochemical transistor sensor holds immense potential for the development of potable healthcare biosensing.
