Abstract:Skin-interfaced electrochemical sensing devices are widely developed for biochemical sensing at molecular levels. However, the sensing electrodes fabricated with photolithography or printing technique are hard to achieve high stretchability without special designs like serpentine shape. Also, most of them require wired connections with external electrochemical workstations for data acquisition or on-board batteries to power the circuits, which largely restrain the flexibility, simplification, and miniaturization of the devices. Here, a battery-free, wireless, and epidermal electrochemical system is presented for in situ biochemical sensing. The sensing component of the system is a stretchable electrode array all-printed with soft polymer and conductive silver nanowires. The electrodes show stable electrical properties within strain range of 0-50%, without serpentine designs. The electronic component of the system is a fully integrated flexible circuit board with near field communication module, which enables wireless energy harvesting and data transmission with smartphones. The system demonstrates good performance in real-time on-body sweat analysis for simultaneous quantitative detections of glucose, hydrogen, sodium, and potassium. This battery-free and wireless epidermal electrochemical system provides a simplified, miniaturized, and flexible solution for a wide range of biochemical platforms, including wearable and implanted bioelectronics.

Abstract:In the wearable electrochemical biosensors, sensing signal duration is significantly dependent on the long-term stability of functional materials modified on the flexible substrate, the effect of pH changes of sweat on the sensing device and signal fluctuation caused by the bending of sensor. Here, we proposed a wearable biosensor based on the lactate-sensing membrane mainly constituted by Prussian blue (PB), reduced graphene oxide (rGO), Au nanoparticles and lactate oxidase (LOx). Based on the in-situ layer-by-layer spin-coating preparation method, the electrode surface was covered with an extensive and uniform PB/GO membrane with a high stability. After the electro-reduction of GO to rGO and the combination of urchin-like Au particles with sufficient tentacles to LOx, the sensing membrane showed the improved electron transport from the enzyme active center to the electrode. Therefore, the wearable biosensor achieved a high sensitivity of 40.6 μA mM−1 cm−2 in a range of 1–222 μM and a low sensitivity of 1.9 μA mM−1 cm−2 in a wide range of 0.222–25 mM, satisfying the requirement of the typical test. In addition, with the excellent running and mechanical stability, the lactate biosensor was successfully applied on volunteers' skin for real-time monitoring of perspiration in vivo. The results were comparable with ex vivo measurements achieved by a commercial lactate sensor. The wearable electrochemical biosensor provides a good candidate in the future for the evaluation of human sweat in sports and biomedical fields.

Lactate Biosensing for Reliable On-Body Sweat Analysis

Abstract:Wearable lactate sensors for sweat analysis are highly appealing for both the sports and healthcare fields. Electrochemical biosensing is the approach most widely used for lactate determination, and this technology generally demonstrates a linear range of response far below the expected lactate levels in sweat together with a high influence of pH and temperature. In this work, we present a novel analytical strategy based on the restriction of the lactate flux that reaches the enzyme lactate oxidase, which is immobilized in the biosensor core. This is accomplished by means of an outer plasticized polymeric layer containing the quaternary salt tetradodecylammonium tetrakis(4-chlorophenyl) borate (traditionally known as ETH500). Also, this layer prevents the enzyme from being in direct contact with the sample, and hence, any influence with the pH and temperature is dramatically reduced. An expanded limit of detection in the millimolar range (from 1 to 50 mM) is demonstrated with this new biosensor, in addition to an acceptable response time; appropriate repeatability, reproducibility, and reversibility (variations lower than 5% for the sensitivity); good resiliency; excellent selectivity; low drift; negligible influence of the flow rate; and extraordinary correlation (Pearson coefficient of 0.97) with a standardized method for lactate detection such as ion chromatography (through analysis of 22 sweat samples collected from 6 different subjects performing cycling or running). The developed lactate biosensor is suitable for on-body sweat lactate monitoring via a microfluidic epidermal patch additionally containing pH and temperature sensors. This applicability was demonstrated in three different body locations (forehead, thigh, and back) in a total of five on-body tests while cycling, achieving appropriate performance and validation. Moreover, the epidermal patch for lactate sensing is convenient for the analysis of sweat stimulated by iontophoresis in the subjects' arm, which is of great potential toward healthcare applications.This article has not yet been cited by other publications.

Abstract:Wearable non-invasive biosensors for the continuous monitoring of metabolites in sweat can detect a few analytes at sufficiently high concentrations, typically during vigorous exercise so as to generate sufficient quantity of the biofluid. Here we report the design and performance of a wearable electrochemical biosensor for the continuous analysis, in sweat during physical exercise and at rest, of trace levels of multiple metabolites and nutrients, including all essential amino acids and vitamins. The biosensor consists of graphene electrodes that can be repeatedly regenerated in situ, functionalized with metabolite-specific antibody-like molecularly imprinted polymers and redox-active reporter nanoparticles, and integrated with modules for iontophoresis-based sweat induction, microfluidic sweat sampling, signal processing and calibration, and wireless communication. In volunteers, the biosensor enabled the real-time monitoring of the intake of amino acids and their levels during physical exercise, as well as the assessment of the risk of metabolic syndrome (by correlating amino acid levels in serum and sweat). The monitoring of metabolites for the early identification of abnormal health conditions could facilitate applications in precision nutrition.

Abstract:A wearable sweat sensor, which could continuously monitor biomolecules related to the human physiological state, is emerging as a promising piece of health surveillance equipment. However, current sensors cannot simultaneously achieve a detection performance that equates to that of traditional sensors and satisfactory mechanical strength. Herein, a wearable sweat sensor with excellent detection performance and mechanical stability is designed and fabricated. Based on the integration of laser-induced graphene electrodes and a screen printing technique, this wearable sweat sensor could realize both the separate and simultaneous detection of uric acid (UA), tyrosine (Tyr), and ascorbic acid (AA) with high sensitivity. Good UA sensing performance in artificial sweat could be maintained even after 20 000 bends. In addition, the sensor can operate well in the wearing state or in a complex bovine whole blood sample. For the detection of human sweat, the changes in UA concentration after a purine-rich meal are continuously monitored and the results are in accordance with the corresponding serum UA detection results tested with a commercial serum UA meter. These results suggest its application potential in health monitoring for both gout patients and healthy humans.

Abstract:Emerging wearable devices with non-invasively biosensing technics have drawn considerable attention to continuously monitor several metabolites in body fluids, such as a tear, saliva, and sweat, to diagnose human health conditions. Most importantly, wearable sensors could offer unique possibilities for online, real-time and non-invasive monitoring of health compared to traditional invasive biosensors. Among lots of analytes, lactic acid concentration in the human body exhibits a high relationship with several diseases such as acute heart diseases, hypoxia, muscle fatigue, meningitis, and cystic fibrosis, and it could also cause muscle pain in athletes. To further boost up the reproducibility and reliability of wearable biosensors to detect lactate concentration levels from human sweat, Ni-based layered double hydroxide (LDH) with various secondary transition metals (Fe and Co) was proposed as electrocatalysts in an enzyme-free electrochemical lactate sensor. According to the mechanism of lactate oxidation on the transition metal-based electrocatalyst, secondary transition metal of Co could serve as the active site for lactate oxidation and facilitate the adsorption of OH - in the alkaline electrolyte. To further increasing the active surface area for enhancing the sensitivity of Ni-based LDH, ZIF-67 derived NiCo LDH was synthesized as the electrocatalyst for non-enzymatic lactate detection. Co-based ZIF-67 served as self-sacrificial templates to fabricate hierarchically structural NiCo LDH with uniform porosity and high electrochemically active surface area to achieve outstanding electrocatalytic performance for lactate sensing. After optimizing the particle size of ZIF-67 and transformation times, ZIF-67 derived NiCo LDH reached the ultrahigh sensitivity of 83.98 μA mM - 1 cm - 2 at an applied potential of 0.55 V (vs. Ag/AgCl KCl sat’d) in the concentration range from 2 to 26 mM. On the other hand, pioneering works in biosensors for lactate detection in sweat has been encountered major challenges such as noble material usage, immobile power supply, and complicated circuit connection to realize the compact sustainable sensing systems. To solve these restrictions, herein, the self-powered molecular imprinted polymers based triboelectric sensor (MIP-TES) was designed to offer a multifunctional noninvasive approach for specific and simultaneous lactate detection. Free-standing PVDF/graphene flexible electrode modified poly(3-aminophenyl boronic acid) imprinted lactate molecule demonstrated the change of the surface properties afterlactate adsorption. MIP-modified electrode revealed the selective lactate sensing over non molecular imprinted polymers (NIP) electrode through the superior and stable signal change with variation of lactate concentration in human sweat. Moreover, MIP modified lactate sensor was further introduced in the triboelectric nanogenerator system to harvest mechanical energy from contact and separation into electrical output. The more adsorbed lactate led to lower energy barriers and decreasing electrical potential when detecting higher lactate concentration. Self-power triboelectric lactate sensor could directly power the number of LED lights without an external energy supply. Eventually, it was validated the feasible application of wearable sensors on human skin. After introducing noninvasive enzyme-free biosensors and triboelectric sensors, an innovatively continuous non-invasive health monitoring platforms can be achieved for practical applications, especially in the areas of home medical examination and wearable personal biosensors.

Abstract:Flexible electrochemical biosensors used for sweat analysis have aroused great interests in the field of intelligent instruments. Due to the complexity of body fluids, development of ultrasensitive and miniaturized sensors for analysis of sweat is always challengeable. In this research, we fabricate a sensitive and flexible sensor for real-time monitoring of glucose and lactate in sweat at micromolar level and millimolar level by using gold nanopine needles (AuNNs) as the signal amplification strategy. A flexible chip with gold electrode arrays was fabricated in a standard semi-conductor laboratory. AuNNs were deposited on the working electrode by electrochemical deposition method. Enzyme immobilization was carried out on the electrode surface via a cross-linker poly(ethylene glycol) diglycidylether (PEGDE), which is an important component of redox hydrogels capable of retaining the activity of the enzyme to a greater extent than other linkers like glutaraldehyde. The catalytic behavior of AuNNs under different conditions was investigated. The as-fabricated electrochemical sweat biosensor was found to achieve low detection limit down to 7 μmol L−1 for glucose, 54 μmol L−1 for lactate, respectively. The sensor also exhibited great selectivity, reproducibility and stability. In addition, human sweat samples were analyzed by the sensor, indicating its potential applications in determination of glucose and lactate as a wearable skin sensor.

Abstract:ABSTRACT Introduction: Attention is given to developing electrochemical sensors for the rapid and real-time measurement of lactate levels. The synthesis of electrochemical sensors is based on an electrode modified with a nanocomposite. Objective: Analyze an electrochemical sensor's feasibility for sports monitoring sweat in lactate. The Au@CNTs were the main focus of this study. Methods: The Au@CNTs composite was synthesized on the GCE surface and tested under pre-established protocols as a sensor. Results: The shape and structure of the modified electrodes were analyzed using SEM. The results showed that the Au@CNTs nanoparticles in the Au@CNTs nanocomposite were evenly distributed throughout the porous CNTs network. The performance of the developed sensor was measured using cyclic voltammetry and amperometry. The electrochemical biosensor responded linearly to lactate over phosphate buffer solution with a low detection limit and sensitivity. Conclusion: The experiment of this sensor evaluated lactate concentrations in real sweat samples that were exceptionally close to the injection amount, enabling it as an effective biosensor for the detection of lactate in sweat samples. Abstract:Real-time and continuous monitoring of lactate levels in sweat has been used as an indicator of physiological information to evaluate exercise outcomes and sports performance. We developed an optimal enzyme-based biosensor to detect the concentrations of lactate in different fluids (i.e., a buffer solution and human sweat). The surface of the screen-printed carbon electrode (SPCE) was first treated with oxygen plasma and then surface-modified by lactate dehydrogenase (LDH). The optimal sensing surface of the LDH-modified SPCE was identified by Fourier transform infrared spectroscopy and electron spectroscopy for chemical analysis. After connecting the LDH-modified SPCE to a benchtop E4980A precision LCR meter, our results showed that the measured response was dependent on the lactate concentration. The recorded data exhibited a broad dynamic range of 0.1–100 mM (R 2 = 0.95) and a limit of detection of 0.1 mM, which was unachievable without the incorporation of redox species. A state-of-the-art electrochemical impedance spectroscopy (EIS) chip was developed to integrate the LDH-modified SPCE for a portable bioelectronic platform in the detection of lactate in human sweat. We believe the optimal sensing surface can improve the sensitivity of lactate sensing in a portable bioelectronic EIS platform for early diagnosis or real-time monitoring during different physical activities.

Abstract:Integrating sweat biosensors into daily wear has the potential to effectively monitor personal lactate levels, offering the advantages of convenience, precision, and multi-point detection. However, existing wearable fabric sweat biosensors have limitations in terms of long-term repeatable detection and washability due to the difficulty of building a stable layer of recognition materials on the highly curved fiber surface. Here, a long-term repeatable and washable electrochemical fabric biosensor is reported based on a fiber electrode with multi-scaled aligned channels for efficient and reliable lactate detection. Through the functionalization of a molecularly imprinted polymer with redox-sensitive nano reporters, the fabric biosensor exhibits impressive in situ long-term repeatable detection for more than 400 times, washability, and stability under deformations. The multi-scaled channels contribute to a desired electric field distribution and thus a reliable interface between the polymer and the fiber. Employing industrial sewing technology, a 20 m (L) x 0.5 m (W) sensing fabric is further produced. Ultimately, combined with a wireless communication element, the fabric biosensing system can immediately track lactate levels. By functionalizing a molecularly imprinted polymer with redox-sensitive nano reporters on a fiber electrode featuring multi-scaled channels, a fabric biosensor is developed with remarkable in situ long-term repeatable detection, capable of detecting lactate over 400 times, while maintaining washability and stability under deformations. Abstract:Herein we report a wearable sweat sensor of a Janus fabric based on surface enhanced Raman scattering (SERS) technology, mainly detecting the two important metabolites glucose and lactate. Janus fabric is composed of electrospinning PU on a piece of medical gauze (cotton), working as the unidirectional moisture transport component (R = 1305%) to collect and transfer sweat efficiently. SERS tags with different structures act as the probe to recognize and detect the glucose and lactate in high sensitivity. Core-shell structured gold nanorods with DTNB inside (AuNRs@DTNB@Au) are used to detect lactate, while gold nanorods with MPBA (AuNRs@MPBA) are used to detect glucose. Through the characteristic SERS information, two calibration functions were established for the concentration determination of glucose and lactate. The concentrations of glucose and lactate in sweat of a 23 years volunteer during three-stage interval running are tested to be 95.5, 53.2, 30.5 μM and 4.9, 13.9, 10.8 mM, indicating the glucose (energy) consumption during exercise and the rapid accumulation of lactate at the early stage accompanied by the subsequent relief. As expected, this sensing system is able to provide a novel strategy for effective acquisition and rapid detection of essential biomarkers in sweat.

Abstract:Sweat could be a carrier of informative biomarkers for health status identification;therefore,wearable sweat sensors have attracted significant attention for research.An external power source is an important component of wearable sensors,however,the current power supplies,i.e.,batteries,limit further shrinking down the size of these devices and thus limit their application areas and scenarios.Herein,we report a stretchable self-powered biosensor with epidermal electronic format that enables the in situ detec-tion of lactate and glucose concentration in sweat.Enzymatic biofuel cells serve as self-powered sensing modules allowing the sweat sensor to exhibit a determination coefficient (R2) of 0.98 with a sensitivity of 2.48 mV/mM for lactate detection,and R2 of 0.96 with a sensitivity of 0.11 mV/μM for glucose detection.The microfluidic channels developed in an ultra-thin soft flexible polydimethylsiloxane layer not only enable the effective collection of sweat,but also provide excellent mechanical properties with stable performance output even under 30％ stretching.The presented soft sweat sensors can be integrated at nearly any location of the body for the continuous monitoring of lactate and glucose changes during normal daily activities such as exercise.Our results provide a promising approach to develop next-generation sweat sensors for real-time and in situ sweat analysis.

Abstract:Despite improvements in diagnosis and therapy, morbidity and mortality have remained high and appropriate treatment of patients still is a major challenge. Most cases are diagnosed with locally advanced disease, and multimodal treatment consists of concomitant chemoradiotherapy or surgery followed by riskadapted radio(chemo)therapy. Although it is conceivable that distinct patient subgroups might benefit from treatment de-escalation (e.g., HPV-related HNSCC) or targeted therapy, effective personalized treatment options are not yet in clinical use for HNSCC. The aim of this issue of Oncology Research and Treatment is to highlight new insights into the biology of HNSCC, to discuss their possible implications for established treatment modalities, to point to future therapeutic developments, and to call attention to open questions which should be addressed by clinical trials. Jou and Hess [3] focused on recent data derived from integrative genomics analysis and multi-scale modeling approaches regarding the biological and clinical diversity of HNSCC. Wagner et al. [4] gave an overview of the newly recognized tumor entity ‘HPV-related HNSCC’. Semrau [5] discussed modern treatment technologies and new developments of radiation oncology. In their original article, Wuerdemann et al. [6] compared the outcome of patient cohorts with HPV-related and -unrelated oropharyngeal cancers treated with primary surgery. Schuler at al. [7] outlined novel treatment options for HNSCC, especially in the field of immunotherapy.

The possibility of developing a wearable amperometric lactate biosensor using a screen-printed carbon electrode (SPCE) impregnated by Ag nanoparticles (Ag NPs) for sweat determination in sports monitoring is questioned. Objective: To develop a sensor with GCE substrate coated on Ag NPs/SPCE. Methods: FESEM and XRD analysis were used for the morphological and structural characterizations of Ag and SPCE NPs, respectively. Results: FESEM, EDS, and XRD revealed that Ag NPs were uniformly dispersed in SPCE. The electrochemical biosensor responded linearly to lactate in phosphate buffer solutions, with detection and sensitivity limits of 1.2 μM and 14.2 mAcm-2 mM-1, respectively. Conclusion: The results suggest that Ag NPs/SPCE can be used to continuously monitor lactate levels in sweat as a practical and reliable biosensor for use. Abstract:Emerging wearable devices with non-invasively biosensing technics have drawn considerable attention to continuously monitor several metabolites in body fluids, such as a tear, saliva, and sweat, to diagnose human health conditions. To further boost up the reproducibility and reliability of wearable biosensors to detect lactate concentration levels from human sweat, in this study, ZIF-67 derived NiCo LDH was synthesized as the electrocatalyst for non-enzymatic lactate detection. Co-based ZIF-67 served as self-sacrificial templates to fabricate hierarchically structural NiCo LDH with uniform porosity and high electrochemically active surface area to achieve outstanding electrocatalytic performance for lactate sensing. After optimizing the particle size of ZIF-67 and transformation times, ZIF-67 derived NiCo LDH reached the ultrahigh sensitivity of 83.98 μA mM−1 cm−2 at an applied potential of 0.55 V (vs. Ag/AgCl KCl sat’d) in the concentration range from 2 to 26 mM. Moreover, the lactate concentration difference in the human sweat can be verified under different exercise conditions, namely anaerobic exercise and aerobic exercise, using ZIF-67 derived NiCo LDH modified electrode. Overall, this research provides a potential electrocatalyst for utilizing enzyme-free electrochemical lactate sensors with reliable and stable performance to introduce non-invasive human sweat monitoring on wearable bioelectronics.

Abstract:Lactate levels in humans reveal intensity and duration of exertion and provide a critical readout for the severity of life-threatening illnesses such as pediatric sepsis. Using the lactate oxidase enzyme (Lox) from Aerococcus viridians, we demonstrated its functionality for lactate electrochemical sensing in physiological fluids in a lab setting. The structure and dynamics of LOx were validated by crystallography, X-ray scattering, and hydroxyl radical protein footprinting. This provided a validated protein template for understanding and designing an enzyme-based electrochemical sensing elements. Using this template, LOx enzyme variants were generated and compared. Comparison of the variants demonstrates that one exhibits effective lactate sensing at significantly reduced operating voltages. Additionally, we demonstrate that the four hexahistidine-tags on each enzyme tetramer are sufficient for immobilization to create a durable, functional sensor, with no need for a covalent attachment, enabling self-immobilization and eliminating the need for additional immobilization steps. The functionality of the LOx enzyme variants was verified at physiological lactate concentrations in both human serum (0-4 mM) and artificial sweat (0-100 mM) using 3-electrode setups for analysis of the three variants in parallel. Accuracy of measurement in both artificial sweat and human serum were high. Employing a microfluidic flow cell, we successfully monitored varying lactate levels in physiological fluids continuously over a 2h period. Overall, this optimized LOx enzyme, which self-immobilizes onto gold sensing electrodes, facilitates efficient and reliable lactate detection and continuous monitoring at reduced operating voltages suitable for further development towards commercial use.

Abstract:Recent progress in wearable sweat sensors had overcome numerous limitations of conventional sensors and provided methods of collecting molecular-level insight into the dynamics of human bodies. Lactate, a major metabolite in human sweat, can serve as the critical limiting factor for continuing physical activity. However, pioneering works in biosensors for lactate detection in sweat has been encountered major challenges such as noble material usage, immobile power supply, and complicated circuit connection to realize the compact sustainable sensing systems. To solve these restrictions, herein, the self-powered molecular imprinted polymers-based triboelectric sensor (MIP-TES) was designed to offer a multifunctional noninvasive approach for specific and simultaneous lactate detection. Free-standing PVDF/graphene flexible electrode modified poly(3-aminophenyl boronic acid) imprinted lactate molecule demonstrated the change of the surface properties after lactate adsorption. MIP-modified electrode revealed the selective lactate sensing over non molecular imprinted polymers (NIP) electrode through the superior and stable signal change with variation of lactate concentration in human sweat. Moreover, MIP modified lactate sensor was further introduced in the triboelectric nanogenerator system to harvest mechanical energy from contact and separation into electrical output. The more adsorbed lactate led to lower energy barriers and decreasing electrical potential when detecting higher lactate concentration. Self-power triboelectric lactate sensor could directly power the number of LED lights without an external energy supply. Eventually, it was validated the feasible application of wearable sensors on human skin. The proposed self-powered lactate has created a fascinating horizon in portability, compact, and the real-time analysis of people’s perspiration in the future.

Abstract:In the field of wearable sensing, small and precise sensors can greatly reduce the burden on the wearer and improve the sense of experience, which is the future direction of sensing development. Herein, we introduce an ultra-small wearable biosensor system that integrates an MS02 chip for real-time and highly accurate sweat detection. The whole system mainly includes flexible electrodes and a printed circle board (PCB). The size of the PCB is only 1.5 cm × 0.8 cm, which greatly minimizes the size of the sweat system and improves wearing comfort. Notably, this miniaturized system is comparable to a commercial electrochemical workstation, ensuring the reliability and accuracy of real-time analysis. The core processing MS02 chip, with a dimension of 1.2 mm × 1.1 mm, is used to perform electrochemical signal processing. By performing electrochemical characterization and measurements of the ultra-small wearable biosensor system, on-body monitoring of four biomarkers (glucose, lactate, Na+, and K+) in sweat of human volunteers has been successfully achieved. With the help of this electrochemical sensor system, mass of biochemical data from perspiration can be acquired to better understand the body's response to daily activities, which will facilitate the early prediction of abnormal physiological changes in the future.
