Abstract:A new easy-to-run route for synthesizing some novel pendant-armed benzene-containing cyclen derivatives was proposed.The titled compounds were synthesized by the reaction of 1,4,7,10-tetraazacyclododecane(cyclen)and the electrophilic reagent with potassium carbonate as the proton trapper and were characterized by MS and 1HNMR.The method features a lot of advantages,such as simplicity in operation,mild reaction conditions,high yield and environmental friendliness with the use of potassium carbonate instead of triethylamine and(N,N)-diisopropylethylamine.

Abstract:An efficient tandem process was developed to synthesize diversified benzo[a]xanthones from 2-methyl-3-(1-alkynyl)chromones with electron-deficient chromone-fused dienes. This unusual reaction, involving multiple steps and not requiring the use of transition metal catalysts or an inert atmosphere, results in the formation of three new C-C bonds and one C-O bond.

Abstract:Seven-membered polycyclic architectures, widely present in natural products and molecular drugs, are challenging synthetic targets. However, methods for synthesizing fused medium-sized bicyclo[<i>m</i>.<i>n</i>.0] ring systems, including the benzo-cycloheptane systems, are still urgent. Herein we describe a base-induced ring expansion as a general strategy to construct a wide range of fused seven-membered ring systems. The application of this method was demonstrated by the efficient total syntheses of two sesquiterpenoids, plecarpenene and plecarpenone, both bearing a fused bicyclo[5.3.0]decane skeleton.

Abstract:A copper-catalyzed decarboxylative cascade cyclization of propargylic cyclic carbonates/carbamates with pyridinium 1,4-zwitterionic thiolates is developed. A range of fused polyheterocyclic compounds are obtained in moderate to good yields with excellent diastereoselectivities. Of particular note is that four new bonds (two C-C, one C-O/N, one C-S) and four new stereocenters could be efficiently embedded into the tetracyclic fused scaffolds in a single step.

Abstract:The construction of fused [5.5.0] bicyclic heterocycles with precise regio-, stereo-, and enantioselective control remains a significant challenge in asymmetric catalysis. In this work, we introduce a novel gold-catalyzed asymmetric [8+4] cycloaddition of 1-(1-alkynyl)cyclopropyl ketones with simple tropones, yielding highly functionalized cyclohepta[b]furo[3,4-d]oxepine derivatives with excellent diastereo- and enantioselectivity (38 examples, all >20:1 dr, up to 95% ee). Additionally, an efficient kinetic resolution (KR) process is achieved (s factor up to 104). The gram-scale synthesis and subsequent synthetic transformations of the cycloadducts further demonstrate the synthetic potential of this method. Furthermore, the cycloadduct can undergo a [1,5]-H shift under acid catalysis, adding another dimension to the structural diversity.

Abstract:A facile, versatile and stereoselective synthesis of bicyclic polyhydroxylated alkaloids as castanospermine analogues is described. The synthetic route started from methyl pyranosides. The key steps involved a high-yielding expeditious one-pot tandem reaction from alkenes to N-substituted delta-lactams. The delta-lactams were stereoselectively vinylated to give the dienes, which were followed by the ring-closing metathesis to produce the cyclized products. The functional group transformations of the resulting bicyclic compounds furnished diverse polyhydroxylated alkaloids in good yields.

Abstract:We successfully developed an unprecedented visible light-promoted synthesis route to fused tetracyclic skeletons with rich sp 3 carbons through stepwise radical cycloadditions and further cyclization of the final radical intermediates with adjacent aromatic rings. This convenient and sustainable protocol features mild reaction conditions, excellent H 2 O and air tolerance, good functional group compatibility and wide substrate adaptability. Moreover, based on direct excitation, radical-trapping, radical clock, and EPR experiments, a possible stepwise formal [3 + 2]/[4 or 5 + 2] cycloaddition process was proposed.

Abstract:A series of 1,3-cyclodiketone- and tetrahydroepoxyisoindole-fused β-enamino dicarbonyl heterocycles were synthesized via a 1,4-diazabicyclo[2.2.2]octane-catalyzed, CH3NO2-mediated three-component reaction of 1,3-cyclodiketone, furfural, and allylamine in toluene. The target compounds were generated via the formation of β-enamino diketone as a key intermediate, followed by intramolecular Diels-Alder cycloaddition. The prepared molecules bearing a quinoline-2,4-dione moiety could be further brominated with N-bromosuccinimide and diastereoselectively reduced by NaBH4 to afford pyrano[3,2-c]quinolin-5-one-derived heterocycles with six vicinal stereogenic centers.

Abstract:In this article we report a new strategy to build fused/spiro/bridged carbocyclic systems with a novel 6-C synthon from readily available diallyl diacetates through the sequential Pd-catalyzed double allyl alkylation and Diels-Alder annulation. Further exploration on the application of this strategy can construct useful complex scaffolds.

Abstract:An unprecedented, caged 2H-benzo-dioxo-pentacycloundecane (BDPC) framework was serendipitously obtained in a single transformation via a triple-dearomative photocycloaddition of chromone esters with furans. This caged structure was generated during the course of an effort to access a tricyclic, oxygen-bridged intermediate enroute to the dihydroxanthone natural product nidulalin A. Reaction scope and limitations were thoroughly investigated, revealing the ability to access a multitude of synthetically challenging caged scaffolds in a two-step sequence. Photophysical studies provided key mechanistic insights on the process for formation of the novel caged scaffold.

Abstract:A novel three-component reaction of 2-oxo-2H-chromene-3-carbaldehydes with isocyanides and anilines was developed for one-pot assembly of biologically intriguing chromeno[4,3-b]pyrrol-4(1H)-ones, which can also be transferred into diverse polycyclic fused scaffolds through further synthetic manipulations. The described method tolerates a broad substrate scope and proceeds in moderate to good yield via a sequential multicomponent reaction and intramolecular Michael cyclization.

Abstract:Herein, an atom-economic method for the synthesis of fused-ring pyrrolizine derivatives by a cycloaddition reaction of easily accessible N -substituted pyrrole-2-carboxaldehydes with N -substituted maleimides in the presence of di- tert -butyl peroxide has been successfully developed. A total of 23 compounds were obtained by using this method, with a maximum yield of 72%, providing a practical and efficient method for the synthesis of tricyclic pyrrolizine frameworks. Abstract:An efficient protocol for the direct transformation of chrornan-4-ones to tricyclic fused pyridines with the skeleton of cassiarins, a family of alkaloids with antimalarial activity, was developed. Also, a general strategy for modular construction of polyheterocydes with diverse natural product-like skeletons was developed by using ketone-alkyne bifunctional substrates. These reactions involved a one-pot cascade oximation of ketones, rhodium-catalyzed C-H activation, and intermolecular/intramolecular alkyne annulations under mild conditions with high atom, step, and redox economy.
