Abstract:A versatile reaction cascade leading to highly substituted 1,2,3,6-tetrahydropyridines has been developed. It comprises rhodium(I)-catalyzed C-H activation-alkyne coupling followed by electrocyclization and subsequent acid/borohydride-promoted reduction. This one-pot procedure affords the target compounds in up to 95% yield with >95% diastereomeric purity.

Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A metal- and hypervalent iodine reagent-free electrochemical oxidative dearomatization-induced [5+2] cycloaddition/pinacol rearrangement cascade reaction was described. The electrosynthetic method showed strong tolerance for vinylphenols, ethynylphenols, and allenylphenols, which thus enabled the rapid assembly of diversely functionalized bicyclo[3.2.1]octanes in 41-95% yields and up to >20:1 dr. This protocol could be scaled up to gram amounts and should find wide application in complex natural product synthesis.

Abstract:An alternative route has been developed for the construction of multisubstituted pyrrole derivatives from readily available, doubly activated cyclopropanes and anilines using an iron-mediated oxidation domino reaction (i.e., sequential ring-opening, cyclization, and dehydrogenation reactions). This reaction uses readily available reactants and is tolerant of a broad range of substrates, with the desired products being formed in good to excellent yields.

The formation of the target pyrrole products involves the formation of one new C-C and two new C-N bonds via the major functionalization of six C(sp3)-H bonds, presenting a highly novel and efficient synthetic protocol toward pyrrole scaffolds.

Abstract:Herein, we report a new process for the synthesis of highly functionalized pyridines based on a tandem Pummerer-type rearrangement, aza-Prins cyclization, and elimination-induced aromatization. This formal [5+1] cyclization provides pyridines in good yields with easily accessible starting materials. Abstract:A simple and efficient protocol for direct synthesis of multisubstituted 6-alkyl-4-chromenyl-1-aryl-3,4-dihydro-1 H -pyrrolo[3,4- b ]pyridine-2,5,7(6 H )-trione derivatives is presented by a cascade reaction of 3-formylchromones, Meldrum's acid and α -aminomaleimides as readily accessible starting materials. In this chemoselective, metal catalyst-free approach Michael addition/intramolecular cyclization and elimination of acetone and carbon dioxide occur consecutively. The significant advantages of this strategy are creating a C–N and a C–C bond, short reaction times, excellent yields (68–80%), and a metal-free catalyst.

Abstract:An efficient asymmetric Mannich/cyclization cascade strategy was established from 2-benzothiazolimines with N-acylpyrazoles to provide optical active benzothiazolopyrimidine derivatives using a copper-based complex. The mild cascade process constructed various structurally diverse products with broad scope of substrates together with excellent enantioselectivities (up to 99 % ee) and diastereoselectivities (up to 99:1 d.r.).

Abstract:A highly efficient catalyst-free cascade reaction of air-stable isochromenylium tetrafluoroborates and bifunctional styrenes containing a 1,3-diketone moiety has been developed, affording the corresponding single diastereomeric ocatahydrophenanthrene derivatives (21 examples, up to 86% yield). A cascade process of [4 + 2]-cyclization and subsequent intramolecular nucleophilic addition is proposed to generate the three new C-C bonds diastereoselectively in the reactions.

Abstract:A simple and efficient copper-catalyzed method has been developed for synthesis of bicyclic pyrimidinones containing six-, seven-, eight-membered rings under mild conditions. The protocol uses readily available 2-bromocycloalk-1-enecarboxylic acids, amidines, and guanidines as the starting materials, copper-catalyzed cascade couplings provide the corresponding bicyclic pyrimidinones without addition of any ligand or additive, and the method is of economical and practical advantages.

Abstract:An efficient one-pot synthesis of highly functionalized bicyclic pyrrole derivatives by a three-component domino reaction of heterocyclic ketene aminals (HKAs), arylglyoxal monohydrate, and indoles in ethanol medium catalyzed by acetic acid is described. In this procedure, three sigma bonds were formed simultaneously. The present synthesis features excellent regio-selectivity, easy purification as well as simple starting materials.

Abstract:A one-pot synthesis of substituted pyridines via a cascade reaction of 2-azido-24-dienoates with alpha-diazocarbonyl compounds and triphenylphosphine is reported. The process involves a Staudinger-Meyer reaction, a Wolff rearrangement, ail aza-Wittig reaction, and an electrocyclic ring- closure. The procedure is general and efficient. The substrates are readily available.

Abstract:An efficient pyridine-catalyzed chemoselective four-component cascade reaction of aromatic aldehydes, malononitrile/cyanoacetates, Morita-Baylis-Hillman (MBH) carbonates, and alcohols has been established. This one-pot reaction progressed in an unusual reaction with solvent participation via a Knoevenagel condensation/oxa-Michael addition/SN2' substitution sequence. This method allowed for facile access to an array of functionalized chain alkylbenzenes and dihydroquinolinones bearing one all-carbon quaternary center in moderate to excellent yields. It is worth noting that the configuration of the all-carbon quaternary center could be modulated by changing only the electron-withdrawing groups via a tandem reduction/cyclization reaction.

Abstract:A copper-catalyzed decarboxylative cascade cyclization of propargylic cyclic carbonates/carbamates with pyridinium 1,4-zwitterionic thiolates is developed. A range of fused polyheterocyclic compounds are obtained in moderate to good yields with excellent diastereoselectivities. Of particular note is that four new bonds (two C-C, one C-O/N, one C-S) and four new stereocenters could be efficiently embedded into the tetracyclic fused scaffolds in a single step.

Abstract:Pyrroles, furans, and thiophenes are important structural motifs in biologically active substances, pharmaceuticals and functional materials. In this paper, we disclosed an efficient synthetic strategy for the rapid construction of multisubstituted pyrroles, furans, and thiophenes via NXS mediated desulfonylative/dehydrogenative cyclization of vinylidenecyclopropanes (VDCPs). The advantages of this method include wide substrate range, high efficiency and synthetic usefulness of the heterocyclic products under metal-free and mild conditions. The derivatization of pyrrole products and the preparation of functional molecules successfully demonstrated the synthetic potential of the products as platform molecules. The reaction mechanism has been investigated on the basis of control experiments and DFT calculations.

Abstract:A novel K3PO4-catalyzed synthesis of polysubstituted pyrroles by a Michael addition/alkyne carbocyclization of activated alkynes and N-propargylamines has been developed. This transition-metal-free cascade process represents an environmental friendly and efficient way to construct polysubstituted pyrroles in good yields.
