Abstract:Curdlan is a bacterial, water-insoluble, linear homopolysaccharide that has been widely used in the food industry. In this study, genome information of strain CGMCC 11546, a UV-induced high-yield mutant of the model curdlan-producing strain Agrobacterium sp. ATCC 31749, was used to investigate the molecular mechanism of curdlan biosynthesis. The maximum curdlan yield of 47.97 ± 0.57 g/L was obtained from strain CGMCC 11546 by using optimal media containing 60 g/L sucrose, 6 g/L yeast, 2 g/L KH2PO4, 0.4 g/L MgSO4·7H2O, 2 g/L CaCO3, 0.1 g/L FeSO4·7H2O, 0.04 g/L MnSO4, and 0.02 g/L ZnCl2 at 30 °C and 280 rpm after 96 h of fermentation. The gel strength of curdlan was improved by 41 % by knocking out the β-1,3-glucanase genes exoK and exsH of strain CGMCC 11546. Furthermore, the application of curdlan from the ΔexoK-exsH strain in noodles significantly improved the eating quality of both raw and cooked noodles.

Abstract:Curdlan polysaccharide was isolated from the fermentation broth of Alcaligenes faecelis var.C125 by acid and alkali and the rude sample CD-1 was gained. The protein and pigment were removed from CD-1 by Sevage method and cellulose column chromatography, respectively. The pure sample CD-2 was separated by Sephadex G-100 column chromatography. The CD-2 was a homogeneous pure polysaccharide confirmed by UV-spectrometer and Sephadex G-200 chromatography, and its molecular weight determined by Sephtrometer-200 column chromatography was 65300D. CD-2 was composed of only glucose, which was proved by paper chromatography analysis after complete hydrolysis of the pure sample by sulfuric acid . And the chemical structure of CD-2 was indicated as a kind of linear glucan linked by β-(1,3) linkage by infrared spectroscopy and ()~(13)C-NMR.

Abstract:Curdlan is an exopolysaccharide, which is composed of glucose linked with β-(1,3)-glycosidic bond and is produced by bacteria, such as Alcaligenes spp., Agrobacterium spp., Paenibacillus spp., Rhizobium spp., Saccharomyces cerevisiae, Candida spp., and fungal sources like Aureobasidium pullulan, Poria cocos , etc. Curdlan has been utilized in the food and pharmaceutical industries for its prebiotic, viscosifying, and water-holding properties for decades. Recently, the usefulness of curdlan has been further explored by the pharmaceutical industry for its potential therapeutic applications. Curdlan has exhibited immunoregulatory and antitumor activity in preclinical settings. It was observed that curdlan can prevent the proliferation of malarial merozoites in vivo ; therefore, it may be considered as a promising therapy for the treatment of end-stage malaria. In addition, curdlan has demonstrated potent antiviral effects against human immunodeficiency virus (HIV) and Aedes aegypti virus. It has been suggested that the virucidal properties of curdlans should be extended further for other deadly viruses, such as severe acute respiratory syndrome (SARS), Middle East respiratory syndrome (MERS), and the current severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2/COVID-19). The prebiotic property of curdlan would confer beneficial effects on the host by promoting the growth of healthy microbiota in the gut and consequently help to reduce gastrointestinal disorders. Therefore, curdlan can be employed in the manufacture of prebiotics for the management of various gastrointestinal dysbiosis problems. Studies on the mechanism of action of curdlan-induced suppression in microbial and tumor cells at the cellular and molecular levels would not only enhance our understanding regarding the therapeutic effectiveness of curdlan but also help in the discovery of new drugs and dietary supplements. The primary focus of this review is to highlight the therapeutic interventions of curdlan as an anticancer, anti-malaria, antiviral, and antibacterial agent in humans. In addition, our review provides the latest information about the chemistry and biosynthesis of curdlan and its applications for making novel dairy products, functional foods, and nutraceuticals and also details about the recent patents of curdlan and its derivatives.

Abstract:Polysaccharides are polymers of natural origin that are formed by microorganisms, animals and plants. Polysaccharides play an important part in the progress of living creatures because these are important particles for various life activities. These biologically derived polysaccharides are interesting because of their important characteristics like biodegradability, non-toxicity availability, biocompatibility, stability and low-cost production. In the recent years, biomaterials of polysaccharide origin have become innovative research goal in the arena of material science and engineering since polysaccharides establish a significant constituent of life matter and shows outstanding properties, which are the representative features of biomaterials. Polysaccharide biomaterials have been broadly proposed as scaffold materials in tissue engineering applications as gene carriers and for drug delivery systems. Curdlan is a class of natural polymer with exceptional pharmacological characteristics and has the capability to form single and triple helical structures with various biological important bioactive molecules for anticancer and antimicrobial applications. Curdlan is favorable for increasing the solubility in comparison to other polysaccharides therefore provides the possibilities for processing this polysaccharide with bioactive molecules into biomacromolecules of desired functions. Curdlan conjugated with various anticancer and antimicrobial agents have been explored, either as a delivery carrier for the drug or as a biomaterial with the drug. This review summarized almost all studies of curdlan with anticancer, antimicrobial applications, the analysis of different curdlan formulations and the influence of bioactive molecules on their anticancer and antimicrobial properties.

Abstract:Curdlan is an edible microbial polysaccharide and can be used in food, biomedical and biomaterial fields. To reduce the cost of curdlan production, this study investigated the suitability of cassava starch hydrolysates as carbon source for curdlan production. Cassava starch was hydrolyzed into maltose syrup using β-amylase and pullulanase at various enzyme dosages, temperature, time and addition order of two enzymes. The maltose yield of 53.17% was achieved at starch loading 30% by simultaneous addition β-amylase 210 U/g starch and pullulanase 3 U/g starch at 60 °C for 9 h. Cassava starch hydrolysates were used as carbon source for curdlan production by Agrobacterium sp. DH-2. The curdlan production reached 28.4 g/L with the yield of 0.79 g/g consumed sugar and molecular weight of 1.26 × 106 Da at 96 h with cassava starch hydrolysate at 90 g/L initial sugar concentration. Curdlan produced from cassava starch hydrolysates was characterized using FT-IR spectra and thermo gravimetric analysis. This work indicated that cassava starch was a potential renewable feedstock for curdlan production.

Abstract:In this study, a curdlan-producing bacterium was isolated from Cow pea soil and identified as Priestia megaterium based on 16 S rRNA sequencing. To identify the most suitable carbon and nitrogen sources for curdlan production, submerged fermentation studies with different sources was carried out. To enhance the curdlan yield, optimization by one-factor-at-a-time approach was conducted. The optimal fermentation media consisted of 15% (w/v) sucrose, 0.1% (w/v) urea, 0.1% (w/v) KH 2 PO 4 , 0.04% (w/v) MgSO 4 ·7H 2 O, trace elements, initial pH of 7.0 with 10% (v/v) inoculum size and agitation speed of 180 rpm. Kinetics of growth, curdlan yield, sucrose and ammonia depletion were studied for a period of 168 h. Maximum curdlan yield (0.31 g/L) was achieved at 96 h of fermentation. At this point, the fermentation media had an optical density of 9.68, biomass concentration of 4.26 mg/mL, and viable count of 2.4 × 10 4 CFU/mL. Additionally, the maximum percentage consumption of sucrose and ammonia over 168 h of fermentation were 75 and 62.5%, respectively. Finally, the identity of biopolymer curdlan was validated through characterization techniques such as Fourier Transform Infrared Spectroscopy (FTIR), Scanning Electron Microscopy (SEM) and Thermogravimetric analysis (TGA). Some characteristic features of curdlan such as the β-1,3-linkage was depicted by the absorption band at 890 cm −1 in FTIR, flaky granules with irregularities as seen in SEM, and thermal degradation between 235 and 350 °C by TGA. To the best of our knowledge, this is the first report on curdlan production from Priestia megaterium.

Abstract:This review examines the production of the biopolymer curdlan, synthesized by Agrobacterium species (sp.), on processing coproducts and plant lignocellulosic hydrolysates. Curdlan is a β-(1→3)-D-glucan that has various food, non-food and biomedical applications. A number of carbon sources support bacterial curdlan production upon depletion of nitrogen in the culture medium. The influence of culture medium pH is critical to the synthesis of curdlan. The biosynthesis of the β-(1→3)-D-glucan is likely controlled by a regulatory protein that controls the genes involved in the bacterial production of curdlan. Curdlan overproducer mutant strains have been isolated from Agrobacterium sp. ATCC 31749 and ATCC 31750 by chemical mutagenesis and different selection procedures. Several processing coproducts of crops have been utilized to support the production of curdlan. Of the processing coproducts investigated, cassava starch waste hydrolysate as a carbon source or wheat bran as a nitrogen source supported the highest curdlan production by ATCC 31749 grown at 30 °C. To a lesser extent, plant biomass hydrolysates have been explored as possible substrates for curdlan production by ATCC 31749. Prairie cordgrass hydrolysates have been shown to support curdlan production by ATCC 31749 although a curdlan overproducer mutant strain, derived from ATCC 31749, was shown to support nearly double the level of ATCC 31749 curdlan production under the same growth conditions.

Abstract:A key goal in current research on industrial curdlan production is the expansion of carbon sources for fermentation. In this study, a recombinant bacterial strain, sp-AmyAXCC, capable of fermenting and synthesizing curdlan using dextrin as a carbon source, was produced via heterologous expression of IPTG-inducible α-amylase from Xanthomonas campestris NRRL B-1459 in Agrobacterium tumefaciens ATCC31749. External expression of the enzyme was confirmed by western blotting, and the expression levels of exogenous proteins during the fermentation process were monitored. Additionally, the properties of the curdlan product were characterized using attenuated total reflectance-Fourier transform infrared spectroscopy and X-ray diffraction. The recombinant strain produced curdlan at a titer of 30.40 ± 0.14 g/L, gel strength of 703.5 ± 34.2 g/cm2, and a molecular weight of 3.58 × 106 Da, which is 33% greater than the molecular weight of native curdlan (2.69 × 106 Da). In the batch fermentation of sp-AmyAXCC with 12% dextrin as a carbon source, the titer of curdlan was 66.7 g/L with a yield of 0.56 g/g, and a productivity rate of 0.62 g/L/h at 108 h. The results of this study expand the substrate spectrum for Agrobacterium fermentation in curdlan production and provides guidance for further industrialization of curdlan production.

Abstract:Curdlan's application is constrained by high gelation concentration, poor water solubility, and incompatibility with other polysaccharides. To address these limitations, this study investigated the effects of different concentrations (0.05–0.3%) of non-ionic polysaccharides (pullulan (PL), locust bean gum (LBG), guar gum (GG), and konjac gum (KGM)) on the heat-induced gelling properties of curdlan. PL with no branch showed 0.3% enhanced gel hardness. LBG with a small amount of galactose residue and KGM with an acetyl group had similar effects on hardness, while GG with a large amount of galactose residue slightly weakened the mixed gel. The rheological results showed that PL had little effect on curdlan, and LBG and KGM had a positive effect on curdlan unfolding, but 0.3% GG was significantly antagonistic to curdlan. The above results implied that non-ionic polysaccharides without side chains interacted weakly with the curdlan and hardly changed the properties of curdlan. Curdlan unfolding and stable suspension were favored if the structure contained galactose or acetyl side chains that interacted with curdlan through hydrogen bonding. These results suggested an effective way to modify curdlan by strengthening the interaction of curdlan with others and weakening the hydrogen bonding of curdlan to broaden its application in food colloids.

Abstract:In this study, the upstream and downstream production processes of curdlan from Priestia megaterium were optimized to enhance its yield. Additionally, a novel extraction method was developed for curdlan recovery. Optimization studies were conducted using Central composite design (CCD). Curdlan yield improved from 0.15 g/L (unoptimized) to 0.46 g/L (3-fold increase) when fermentation was carried out in CCD-optimized media of (w/v) sucrose 20%, urea 0.1%, KH 2 PO 4 0.02%, agitation speed 250 rpm. To further enhance curdlan yield during extraction, ultrasonication was incorporated as a novel step into the conventional method of acid/alkali-assisted curdlan recovery. A two-step optimization was chosen for extraction, namely, one-factor-at-a-time (OFAT) and CCD, wherein the optimized extraction parameters were determined to be 25 s sonication, 1 N NaOH, and 2 h solubilization time. The curdlan yield improved by 1.5-fold (0.70 g/L) post optimization, in comparison with unoptimized conventional extraction step. Finally, the biopolymer was validated through characterization by nuclear magnetic resonance (NMR) which showed characteristic curdlan spectra in the 13 C and 1 H NMR studies. To the best of our knowledge, this study represents the first documented report on curdlan extraction using this novel method of ultrasonication.

Effect of curdlan on the gel properties and interactions of whey protein isolate gels

Abstract:This study investigated the influence of curdlan on the gel properties of whey protein isolate (WPI). Results demonstrated that curdlan significantly improved the water-holding capacity, gel strength and rheological properties of the WPI gels. Moreover, it promoted the unfolding of the molecular structures of WPI, which was manifested by the transition from α-helix to β-sheet, an increase in free sulfhydryl content and a decrease in surface hydrophobicity. Furthermore, 4 % curdlan promoted the formation of WPI with uniform and compact elastic gel network structures, primarily attributed to disulphide bonds, hydrogen bonds and hydrophobic interactions. However, when the addition of curdlan exceeds 4 %, excessive entanglement of curdlan chains and steric hindrance effects hinder the unfolding and folding of protein structures, weaken their interaction, result in a loose network structure and affect the gel properties. In conclusion, this study demonstrates that curdlan can effectively improve the gelling properties of WPI, suggesting its potential application in low-calorie gel-based dairy products.
Curdlan-polyphenol complexes prepared by pH-driven effectively enhanced their physicochemical stability, antioxidant and prebiotic activities

Abstract:In this study, the curdlan-polyphenol complexes were constructed by a pH-driven method. The interaction between curdlan and various hydrophobic polyphenols (curcumin, quercetin, and chlorogenic acid) was investigated. Curdlan could self-assemble into particles for loading polyphenols through hydrogen bonding and hydrophobic interactions. The three polyphenols were embedded in curdlan in an amorphous state. The curdlan-curcumin complex showed the lowest viscoelasticity but exhibited the highest curcumin loading ability (34.04 ± 1.73 mg/g). However, the curdlan-chlorogenic acid complex emerged the opposite trend, indicating that the loading capacity was associated with the hydrophobicity of polyphenols. The antioxidant activity of curdlan significantly increased after combining with polyphenols, which could be maintained during in vitro simulated gastrointestinal digestion. In particular, the curdlan-quercetin complex exhibited the highest antioxidant activity and short-chain fatty acid concentration, which could influence gut microbiota composition by promoting the proliferation of Prevotella and inhibiting the growth of Escherichia_Shigella. In conclusion, the curdlan-polyphenol complexes prepared by an alcohol-free pH-driven method could effectively enhance the gastrointestinal stability of polyphenols as well as increase the antioxidant and prebiotic activities of curdlan, which could be applied as a functional ingredient to improve gut health.
Interleukin-8 Regulation of the Ras/Raf/Mitogen-activated Protein Kinase Pathway in Human Neutrophils (*)

Abstract:Interleukin-8 (IL-8), the prototypic member of the CXC subfamily of chemokines, induces in neutrophils chemotaxis, the respiratory burst, granule release, and increased cell adhesion. The IL-8 receptor is a seven-transmembrane spanning receptor coupled to specific heterotrimeric G proteins including G and G. IL-8 stimulation of its receptor on neutrophils activates Ras GTP loading and the mitogen-activated protein kinase (MAPK) pathway including Raf-1 and B-Raf. The properties of IL-8 stimulation of the MAPK pathway differ from those observed for chemoattractants such as C5a. Even though Ras GTP loading is similar for IL-8 and C5a, the maximal activation of Raf-1 and B-Raf is approximately 2-fold and 3-7-fold, respectively, less for IL-8 than that observed for C5a. Raf-1 activation is rapid but transient, returning to near basal levels by 10 min. B-Raf activation is slower in onset and does not return to basal levels for nearly 30 min. IL-8 activation of MAPK follows a time course suggesting an involvement of both Raf-1 and B-Raf. Surprisingly, wortmannin, at low concentrations, inhibits Raf-1, B-Raf, and MAPK activation in response to IL-8 and C5a demonstrating a role for phosphatidylinositol 3-kinase in the activation of Raf kinases in G protein-coupled receptor systems in human neutrophils. Furthermore, wortmannin inhibits IL-8 stimulated granule release and neutrophil adherence. These findings demonstrate the control of Raf kinases, the MAPK pathway and specific neutrophil functions by phosphatidylinositol 3-kinase enzymes.
Production of Curdlan by Agrobacterium sp. DH-2 Using Sugarcane Molasses-Based Medium

Abstract:Curdlan is an extracellular polysaccharide produced by microorganisms with various applications in the food and biomedical fields. In order to reduce the cost of producing curdlan, the production of curdlan by Agrobacterium sp. DH-2 using sugarcane molasses as a carbon source was investigated. Pretreatment of sugarcane molasses reduced the metal content and resulted in an increase in curdlan production compared to untreated molasses. The composition of the medium based on pretreated molasses was optimized for curdlan production. The addition of (NH4)2HPO4 was found to be effective in increasing curdlan production, while the addition of corn steep liquor, CaCO3, KH2PO4, and MgSO4 did not have a significant effect. An orthogonal design was used to optimize the amount of (NH4)2HPO4 added and the sugar concentration of pretreated molasses. Maximum curdlan production of 13.0 g/L was obtained in the optimized medium, which contained a sugar concentration of 60 g/L of pretreated molasses and 1.0 g/L of (NH4)2HPO4. The curdlan produced in the molasses-based medium had an average molecular weight of 0.49 × 106 Da. FT-IR spectra and TGA thermal analysis showed that there were no significant differences between the curdlan produced and commercial curdlan. These results suggested that pretreated sugarcane molasses supplemented with (NH4)2HPO4 as a nitrogen resource could be used for economical production of curdlan.

Actual and Simulated Injury of Creontiades signatus (Heteroptera: Miridae) Feeding on Cotton Bolls

Abstract:The actual feeding injury of Creontiades signatus Distant (Heteroptera: Miridae) was compared with a simulated technique during 2005, 2006 and 2008 by injecting varying dilutions of pectinase into cotton bolls at different boll sizes (ages) and into 2 or 4 locules to determine if such a technique could be used to reduce the time and labor involved with conducting economic injury level studies in the field. The most accurate simulation occurred in 2008 by injecting 1 μL of 10% pectinase into all 4 locules of a cotton boll. This improved the relationships of injury score to seed cotton, seed, and lint weights. The youngest boll age class of ≥ 2 cm diam. (2 d of age) was not significantly more damaged than the medium age ≥ 2.5 cm (8 d of age) bolls, and both sustained significantly more injury than the large boll classification of ≥ 3 cm (12 d of age). However, small bolls were at least 3 times more likely to abscise than medium-sized bolls, and large bolls did not abscise regardless of treatment. Some damage was observed for large bolls from the injected and actual feeding compared with the controls, but the lint and seed weights were not significantly different for any of the treatments including the controls. Our study characterizes the feeding injury caused by C. signatus and describes a simulated technique that may be used to further economic injury studies.

Abstract:High-moisture extrusion is a promising thermomechanical technology extensively employed in manufacturing fibrous meat analogues from plant-based proteins, garnering considerable research attention. However, polysaccharide-based extrusion has been rarely explored. The present study investigates the effects of varying extruder barrel temperatures (130 °C–200 °C) on the texture and structure of curdlan extrudates, and highlights the formation mechanism. Results showed that the single chain of curdlan aggregates to form triple-helix chains upon extrusion, consequently enhancing the crystallinity, particularly at 170 °C. The hardness, chewiness, and mechanical properties improved with increasing barrel temperature. Moreover, barrel temperatures affected the macrostructure, the extrudates maintained intact morphologies except at 160 °C due to the melting of curdlan gel as confirmed by the differential scanning calorimetry thermogram. Microstructural analysis revealed that curdlan extrudates transited through three phases: original gel (130 °C, 140 °C, and 150 °C), transition state (160 °C), and regenerated gel (170 °C, 180 °C, 190 °C, and 200 °C). The steady state of regenerated gel (170 °C) exhibited higher crystallinity and smaller fractal dimension, resulting in a more compact and crosslinked gel network. This study elucidates the structure transition of curdlan gel at extremely high temperatures, offering valuable technical insights for developing theories and methods with respect to polysaccharide-based extrusion that may find applications in food-related fields.

Abstract:This study delves into the effects of curdlan integration and thermal sterilization on the rheological properties, structure, and quality attributes of concentrated rice starch gel. Acting as a heat-set polysaccharide, curdlan established a dual-network gel structure with rice starch gel, displaying strong interactions with rice starch, as confirmed by confocal laser scanning microscopy and Fourier-transform infrared spectroscopy. The addition of curdlan expedited the gel formation of rice starch, yielding a denser gel structure. Consequently, this enhanced G', solid-like behavior, textural properties, and cooking quality while reducing frequency-dependence. Given the cooling-induced gelation behavior of pure rice starch, thermal treatment disrupted inter-chain hydrogen bonding, compromising the structural integrity of the gel. This disruption manifested in a softer texture and diminished mechanical properties and cooking quality. Notably, this decline in mechanical properties and cooking quality of rice starch gel was markedly ameliorated with the incorporation of curdlan, particularly at a content of ≥1.0 %. Compared with pure RS, 1.0 % CD inclusion showed a reduction in cooking breakage rate by 30.69 % and an increase in hardness by 38.04 %. This work provides valuable insights for the advancement of fresh starch gel-based foods that exhibit exceptional quality and an extended shelf life.

This somewhat surprising fact may be panly due to the relative youth or brain injury rehabilitation as a srecialry. In 1980 there were fewer than 12 centers in the United States that specialized in brain injury rehabilitation, but this field has grown rapidly over the last 20 years with, more recently, some reduction in services. This paper attempts to account for some of these changes and describes factors that have led to the increased scrutiny of brain injury rehabilitation in the managed care environment. An overview of the recent history of brain injmy rehabilitation is prOVided, and areas of panicular interest to occupational therapists are emrhasized. By focusing on what we have \earned aboul persons with brain injury, we will be in a better rosition to advocate for the provision of ;,Jdequate <Inel cost-effective services. Abstract:The cbm6e gene from Saccharophagus degradans 2-40 T was cloned and expressed in Escherichia coli. CBM6E contains a glycoside hydrolase family 128 (GH128) catalytic module and a C-terminal carbohydrate-binding module (CBM) grouped into CBM family 6. The purified recombinant CBM6E displayed high substrate specificity toward curdlan as an endo-β-1,3-glucanase and had maximal activity at pH 6.0 and 35 ℃. The hydrolytic products against curdlan were predominantly laminaritriose (L3) and laminaritetraose (L4) along with a lower amount of laminaripentaose (L5) and laminarihexaose (L6). The CBM6 module selectively enhanced the enzyme activity against curdlan and displayed strict binding specificity to curdlan, no matter in its powder or high-set gel forms. This study laid a foundation for enzymatic degradation of curdlan to produce high-value β-1,3-glucooligosaccharides at moderate temperatures and provided a novel CBM tag for enzyme immobilization on curdlan.
