Abstract:Regioselectivity of lipases played a crucial role in structured lipids synthesis and oil modification.Because of differences between aqueous and non-aqueous systems,regioselectivity analyzed by the conventional hydrolysis method might not always match that in synthesis reactions.In this paper,acidolysis of lauric acid and camellia oil was developed to directly evaluate regioselectivity of lipases in solvent-free system.Fatty acid composition and positional distribution of reaction products were detected by gas chromatography.Through this protocol,Lipozyme RM IM,L02,L03 and L04 were identified as sn-1 (3) regioselectivity,L01 was partially selective,and Novozym 435 was nearly non-selective.Predictability of the model reaction had been verified by replacing substrates of lipases.According to the comparative analysis of results of the acidolysis method and hydrolysis one,regioselectivity of lipases under two situations was generally the same,except selectivity of Novozym 435 which was susceptible to solvent systems.Therefore the present method avoided limits of hydrolysis determination results in synthesis reactions.And moreover it lowered the requirement on purity of reactants,which means it can be used to screen numerous lipases at low cost.
Effect of Immobilization by Modified Ultrastable-Y Molecular Sieve on the Resolution of (R,s)-2-octanol in Nonaqueous Media by Lipase

Abstract:The lipase from Penicillium expansum PED-03 (PEL) was immobilized into a modified ultrastable-Y molecular sieve and the resolution of (R,S)-2-octanol was carried out in a bioreactor in nonaqueous media by the immobilized lipase to investigate the effect of immobilization on the catalytic property of PEL duing the enzymatic reaction. It was found that the conversion (c), enantiomeric excess (ee), and enantioselectivity (E) of the resolution catalyzed by the PEL immobilized on the modified ultrastable-Y molecular sieve were much higher than those of the reaction catalyzed by free PEL and PEL immobilized on other supports. Being immobilized on the modified Ultrastable-Y molecular sieve, the lipase exhibited obvious activity in a wider pH range and at a much higher temperature than the free PEL, and showed a marked enhancement of stability against thermal inactivation, by which the suitable pH value of the buffer used for immobilization could be "memorized". The conversion of the reaction catalyzed by PEL immobilized on the modified ultrastable-Y molecular sieve reached 97.68% of the theoretical value with excellent enantioselectivity (Average E of 8 batches >460) in nonaqueous media at "memorial" pH 9.5, 50 degrees C for 24 h, which showed a good application potential to the production of optically pure (R,S)-2-octanol.

Enzymatic Resolution of (R, S)-2-Octanol by Ultrastable-Y Molecular Sieve Immobilized-Lipase in Microaqueous Media

Abstract:The resolution of (R, S)-2-octanol was performed in microaqueous media by P. expansum PED-03 lipase (PEL) immobilized onto Ultrastable-Y modified molecular sieve. It was found that conversion(c), enantiomeric excess (e. e. ), and enantioselectivity (E) of the reaction catalyzed by PEL immobilized onto Ultrastable-Y modified molecular sieve were much higher, compared with free PEL and PEL immobilized by other carriers. Media type and water content played an important role in the resolution of (R,S)-2-octanol catalyzed by Ultrastable-Y modified molecular sieve immobilized-PEL, and the reaction eonversion(c) reached 97.68% of the theoretical value and the enantiomeric excess(e. e. ) reached 98.75% in n-hexane with 0.8% water at '' memorial '' pH=9.5 and 50 degrees C for 24 h. The modified Ultrastable-Y molecular sieve immobilized-PEL was a good biocatalyst with a fine enantio selectivity and stability, presenting a good promise in the enzymatic resolution of(R,S)-2-octanol.

Abstract:Six lipases were screened for their ability to catalyse the Knoevenagel condensation between benzaldehyde and methyl cyanoacetate. Lipase from porcine pancreas tolerated a variety of functional groups on the aromatic ring, produced the highest yields, and also catalysed transesterification of the product in the presence of an alcoholic cosolvent. We show that, in organic solvents, lipase from porcine pancreas has higher activity for this "promiscuous" reaction than for naturally occurring esterification catalysis.

Abstract:A broad exploitation of ester hydrolases from 7 typical bacteria was reported in this study. Thirty-two predicted esterases and hydrolases were cloned based on published genomic information. The catalytic activity of obtained clones was tested with p-nitrophenyl esters at various temperatures and pH values. The results indicated that eight enzymes presented with typical esterase activity on p-nitrophenyl butylate and caprylate. The result also showed that despite their great sequence difference, the eight enzymes shared similar properties (substrate specificity, optimal pH and temperature) with each other. Phylogenetic analysis revealed a close relationship between these eight enzymes and “true esterases”. As there was no information on enantioselectivity of these enzymes reported, the enantioselectivity of these enzymes to various chiral substrates was investigated for the first time. In comparison with commercial enzyme, Candida rugosa lipase (CRL), enzymes E12, E14, E18, E21 and E24 presented with equal or higher activity and enantioselectivity to the substrates. Furthermore, enzyme E14 (predicted carboxylesterase from Rhodobacter sphaeroides), E21 (S-formylglutathione hydrolase from Pseudomonas putida) and E24 (carboxylesterase from P. putida) presented with enantioselectivity in the resolution of methyl mandelate, 1-phenyethyl acetate and 2-octanol. These findings suggested that the novel ester hydrolases with high activity and enantioselectivity could be obtained from alpha/beta hydrolase family.

Abstract:Enzymatic esterification of resveratrol is crucial for its potential application in lipophilic foods and drugs. However, the poor activity of the free enzyme hinders the reaction. In this work, the highly efficient enzymatic synthesis of resveratrol ester derivatives was achieved by immobilized lipase on hydrophobic modified hollow mesoporous silicon spheres (HMSS-C8). We preliminarily explored the use of Candida sp. 99-125 lipase (CSL) for the acylation of resveratrol, with a regioselectivity toward 3-OH- over 4'-OH-acylation. HMSS-C8 provided ideal accommodation for CSL with a loading capacity of up to 652 mg/g. The catalytic efficiency of CSL@HMSS-C8 was 15 times higher than that of free CSL, and the conversion of resveratrol reached 98.7% within only 2 h, which is the fastest value recorded in the current literature. After 10 cycles, the conversion remained up to 86.3%. Benefiting from better lipid solubility, the relative oxidation stability index values of oil containing monoester derivatives were 43.1%-68.8% and 23.9%-33.2% higher than that of refined oil and oil containing resveratrol, respectively. This research provides a new pathway for efficient enzymatic synthesis of resveratrol ester derivatives and demonstrates the potential application of resveratrol monoester derivatives as a group of excellent lipid-soluble antioxidants.

Abstract:The esterification reactions catalyzed by lipases are among the most significant biochemical processes of industrial relevance. The lipases have the function of versatility by catalyzing a diversity of reactions with extreme ease, obtaining quality products with high yield. Therefore, enzyme-catalyzed esterification has gained increasing attention in many applications, due to the importance of derived products. More specifically, lipase-catalyzed esterification reactions have attracted interest in research over the past decade, due to the increased use of organic esters in the chemical and biotechnology industry. These esters can be obtained by three techniques: extraction from natural sources, chemical and enzymatic syntheses. Biotechnological processes have offered several advantages and are shown as a competitive alternative to chemical methods due to high catalytic efficiency, mild operating conditions, and selectivity of natural catalysts. These an industrial point of view, reactions catalyzed by enzymes are the most economical approach to achieve green products with no toxicity and no harm to human health. Thus, this review presents a descriptive evaluation of the trends and perspectives applied to enzymatic esterification, mainly for the synthesis of esters with different properties, such as aromatics, emulsifiers, and lubricants using the esterification process. An emphasis is given to essential factors, which affect the lipase-catalyzed esterification reaction. In which, the parameters are dependent on the lipase source, a form of the biocatalyst (free or immobilized), the polarity of the reaction medium, the molar ratio between alcohol and acid, among other variables, are also discussed.

Abstract:Triacylglycerides are naturally abundant and renewable feedstock for biofuels and chemicals. In this report, these seemingly stable compounds are shown to be reactive toward a variety of sulfonamides under Lewis acid catalysis. In these reactions, alkyl C(sp 3 )–O bonds are cleaved and C–N bonds constructed, providing functionalized value-added products directly from renewables. Mechanistic and scope study demonstrate that the origin of the reactivity could be the synergy of Lewis acid catalysis and neighboring group participation by the 2- or 3-acyloxy or acylamido group with respect to the reactive site. Since poly(ethylene terephthalate) (PET), a widely available consumer polyester, also contains 1,2-diol diester group as the repeating unit in the main chain, this chemistry can also be applied to efficient depolymerization of PET.

Abstract:Enzyme-catalyzed synthesis of fatty acid guaifenesin esters through transesterification was performed in acetone. Guaifenesin and vinyl fatty acid esters were used as substrates and the reactions were catalyzed by lipase from Mucor miehei (Lipozyme®). Guaifenesin was regioselectively acylated at the primary hydroxyl groups and guaifenesin derivatives with long chain acyl group were prepared in good yields. Various reaction conditions were examined in detail. The results showed that immobilized lipase from M. miehei had the highest activity and a maximum yield of 88% was obtained in acetone, which was accepted by the EEC for use in the manufacture of food products and pharmacology.

Abstract:Sugar esters have recently received increased attention from the viewpoint of renewable resources. Specifically vinyl esters from carbohydrates by enzyme-catalyzed regioselective transformations offer a new family of functional water-soluble monomers for preparation of sugar-based polymers. Here we reported enzyme-catalyzed regloselective synthesis of novel polymerizable raffinose vinyl ester. Transesterification of raffinose with divinyl succinate, divinyl adipate and divinyl sebacate in anhydrous pyridine at 50 degreesC for 6 d was catalyzed by an alkaline protease from Bacillus subtilis. Three raffinose vinyl esters with different acyl donor chain lengths were regioselectively synthesized at 55%, 48% and 30% yields, respectively. Characterization of the products by H-1 NMR, C-13 NMR, and H-1-H-1 correlation spectroscopy and H-1-C-13 heteronuclear multiple quantum coherence NMR revealed that the ester was substituted at C-1 position of beta-fructofuranose moiety in raffinose. Reaction conditions for the esterification were investigated. Activities of. seven commercial enzymes from different sources catalyzing synthesis of raffinose ester Were compared. Lipase from porcine pancreas(PPL) and alkaline protease from Bacillus subtilis had high activities. We also studied the effect of temperature and incubation time on raffinose esters synthesis catalyzed by alkaline protease from Bacillus subtilis and found the optimal temperature and reaction time were 50 degreesC and 6 d respectively.
Controlling Enantioselectivity of Esterase in Asymmetric Hydrolysis of Aryl Prochiral Diesters by Introducing Aromatic Interactions.

Abstract:Aromatic interactions specific to aryl radicals were introduced into two esterases, BioH from Escherichia coli and RspE from Rhodobacter sphaeroides to control their enantioselectivity in the asymmetric hydrolysis of prochiral aryl glutaric acid diesters. As a result, the enantiomeric excess (ee) of the S-product of dimethyl 3-phenylglutarate was increased from 25% (BioH wild type) to 96% (B_L83F/L86F) and from 13% (RspE wild type) to >99% (R_Y27R), respectively, while another variant of RspE R_M121F gave a reversed ee of 50% (R-product). Similar enhancement or reversion of enantioselectivity were also observed in the hydrolysis of three other prochiral aryl diesters (dimethyl 3-(4-flouro)-phenylglutarate, dimethyl 3-(4-cholo)-phenylglutarate and dimethyl 3-(3,4-dicholo)-phenylglutarate). Especially, the mutant R_Y27R was shown to be an excellent S-selective hydrolase for prochiral aryl diesters, with ee of all S-products >99%. In the mutants with altered enantioselectivity, the successful introduction of designed aromatic interactions was confirmed by molecular dynamics simulations and binding free energy analysis. These results demonstrate that aromatic interaction is one of the origins of enzyme enantioselectivity, the tuning of which leads to dramatic change in enantioselectivity. Besides, the successful engineering of the enantioselectivity in two different proteins toward four different substrates suggests that the introduction of aromatic interactions is a generally applicable strategy in the control of enantioselectivity toward aryl substrates.

Abstract:The chemoselective capabilities of porcine pancreatic lipase (PPL) in tetrahydrofuran andCandida rugosalipase (CRL) in diisopropyl ether have been investigated for selective acetylation and deacetylation of hydroxymethylated phenols and hydroxyaryl alkyl ketones and their peracetylated derivatives. Both PPL and CRL exhibited exclusive selectivity for the acetylation of alcoholic hydroxyl group over the phenolic hydroxyl group(s) of the hydroxymethylated phenols1–5and aryl alkyl ketones6–9, and for the deacetylation of ester group involving the phenolic hydroxyl group over the ester group involving alcoholic hydroxyl of the peracetates19–24. The preliminary results indicate that this strategy of chemoselective acetylation can also be used in the enantiomeric resolution of racemic ketones6–9. Single crystal X-ray diffraction studies have confirmed the structures of compounds4, 15, and17.

Study on the synthesis of pine sterol esters in solvent-free systems catalyzed by Candida rugosa lipase immobilized on hydrophobic macroporous resin

Abstract:Background: Pine sterol ester is a type of novel food source nutrient with great advantages in lowering blood cholesterol levels, inhibiting tumors, preventing prostate enlargement, and regulating immunity. Macroporous resins with large specific surface area, stable structures, and various functional groups (epoxy, amino, and octadecyl groups) have been selected for immobilization of Candida rugosa lipase (CRL) to improve its stability and efficiency in the synthesis of pine sterol esters. A solvent-free strategy using oleic acid (substrate) as an esterification reaction medium is an important alternative for avoiding the use of organic solvents. Results: The immobilization conditions of CRL immobilized on several types of commercial macroporous resins were optimized. Fortunately, by adsorption (hydrophobic interaction), a high immobilization efficiency of CRL was obtained using macroporous resins with hydrophobic octadecyl groups with an immobilization efficiency of 86.5%, enzyme loading of 138.5 mg g-1 and enzyme activity of 34.7 U g-1 . The results showed that a 95.1% yield could be obtained with a molar ratio of oleic acid to pine sterol of 5:1, an enzyme amount of 6.0 U g-1 (relative to pine sterol mass) at 50 °C for 48 h. Conclusion: The hydrophobic macroporous resin (ECR8806M) with a large specific surface area and abundant functional groups was used to achieve efficient immobilization of CRL. CRL@ECR8806M is an efficient catalyst for the synthesis of phytosterol esters and has the potential for further large-scale applications. Therefore, this simple, green, and low-cost strategy for lipase immobilization provides new possibilities for the high-efficiency production of pine sterol esters and other food source nutrients. © 2023 Society of Chemical Industry.

Abstract:Polysaccharides, as one of the most prominent natural biopolymers, display numerous biological activities and industrial applications. Nevertheless, some polysaccharides lack biological properties or display weak biological activities. Besides, practical usages of polysaccharides still remain challenging because of their poor solubility and inappropriate hydrophilic/hydrophobic balance. Hence, enzymatic or chemical modifications have been emerged to improve physicochemical and biological properties of polysaccharides by altering their structural and conformational features, and consequently to enlarge their industrial applications. Compared to chemical methods, enzymatic modifications have drawn more attention owing to decreasing the input energy and environmental pollution, as well as high specificity and selectivity. The present review describes different reactions, including hydrolysis, esterification, transesterification, polymer, and phenolics grafting, for the modification of cellulose, chitosan, starch and their derivatives by catalysis with free and immobilized enzymes. Beyond the reaction types and the enzymes, the properties and potential applications of the obtained products are also reviewed.

Abstract:The enantioselective hydrolysis of chiral esters using esterases and lipases gives access to key optically active intermediates en route to prostaglandins, coriolic acid, the anti-HIV agent carbovir and mevinic acid type hypocholestemic agents. The hydrolysis of meso-esters using hydrolases is a very efficient strategy in organic synthesis and has been used to prepare the carbocyclic nucleosides neplanocin and risteromycin. Acylases have been used to prepare (-)-carbovir and both enantiomers of a GABA-mimetic from 2-azabicyclo[2.2.1)hept-5-en-3-one. The employment of nitrilases and nitrile hydratases is gaining in popularity; for example, prochiral 2-benzoyloxypropane-1,3-dinitrile is hydrolysed to (S)-3-benzoyloxy-4-cyanobutanoic acid with exquisite selectivity. Lipases in organic solvents can effect esterification, transesterification and interesterification reactions and this popular methodology has been used to prepare key norcarbocyclic nucleotides and carbocyclic oxetanocin A in single enantiomer form. Yeast catalysed reductions of ketones afford optically active secondary alcohols, typically employed for the synthesis of pheromones, fragrances and chemotactic agents such as leukotriene-84. Instead of a whole-cell system such as yeast, partially purified dehydrogenases can be employed to synthesise (S)-secondary alcohols, for examplan intermediate to the antifungal agent brefeldin-A. Biohydroxylations are important reactions and are being applied to a wide range of substrates. The oxidation of benzene and derivatives to the corresponding cyclohexadiene diols are classic examples and have provided a route to analogues of cyclophellitol. Similarly, mono-oxygenase catalysed Baeyer-Villiger reactions are now well-documented and have furnished intermediates to carbocyclic-AZT, lipoic acid and azadirachtin. Sulfoxides of high optical purity have been prepared by yeast-catalysed oxidation, while enzymes in the transferase and lyase classes have been used to make carbohydrates and amino acids. In conclusion, the science of biotransformations opens up numerous synthetic routes to a wide variety of target molecules that are not easily accessible by other methods of synthetic organic chemistry.

Abstract:A27-YEAR-OLD WOMAN PRESENTED WITH BLISTERING LEsions on her hands, feet, and perioral area. The lesions had manifested more than 1 week before her visit. She said that her hair had become brittle and lighter in color and was shedding. She said that she had not used any medications before onset of the skin lesions. When the plantar bullae progressed to the point of interfering with her walking, she sought medical attention. The patient had a long history of restrictive eating patterns and nightly selfinduced vomiting. She also reported a recent history of diarrhea and intermittent amenorrhea. On physical examination, the patient was a thin woman with a body mass index of 17.5 (calculated as weight in kilograms divided by the square of height in meters; reference range for healthy adults, 18.5 to 24.9). Tense 4to 5-cm bullae were present on her hands, with erosions on her fingers, palms, soles (FIGURE 1A-C), perioral area, and oral mucous membranes. Multiple dusky vesicles with maceration were present in the vulvar area. Perifollicular hemorrhage was evident in the axillary and pretibial regions. The patient’s hair was thin, proximally hypopigmented, and was easily removed on gentle hair pull (Figure 1D). The patient was admitted to the hospital for further evaluation and treatment. Abstract:This paper describes highly selective transesterification reactions, catalyzed by an alkaline protease from Bacillus subtilis in pyridine, of several mono- and di-saccharides with divinyl dicarboxylates ranging from 4 to 10 carbon atoms. A series of polymerizable vinyl fatty acid sugar esters were obtained with good selectivity and high yields. Most products had high proportions of the alpha anomer. The influences of the enzymes, solvents, temperature, and acyl donor chain length on the reaction were studied. Vinyl sugar esters offer a new family of functional water-soluble monomers for preparation of sugar-containing polymers. (C) 2004 Elsevier Ltd. All rights reserved.
A Study on the Regioselective Acetylation of Flavonoid Aglycons Catalyzed by Immobilized Lipases

Abstract:This study aimed to explore the capacity of immobilized lipases on the acetylation of six aglycon flavonoids, namely myricetin, quercetin, luteolin, naringenin, fisetin and morin. For this purpose, lipase B from Candida antarctica (CaLB) and lipase from Thermomyces lanuginosus (TLL) were immobilized onto the surface of ZnOFe nanoparticles derived from an aqueous olive leaf extract. Various factors affecting the conversion of substrates and the formation of monoesterified and diesterified products, such as the amount of biocatalyst and the molar ratio of the substrates and reaction solvents were investigated. Both CaLB and TLL-ZnOFe achieved 100% conversion yield of naringenin to naringenin acetate after 72 h of reaction time, while TLL-ZnOFe achieved higher conversion yields of quercetin, morin and fisetin (73, 85 and 72% respectively). Notably, CaLB-ZnOFe displayed significantly lower conversion yields for morin compared with TLL-ZnOFe. Molecular docking analysis was used to elucidate this discrepancy, and it was revealed that the position of the hydroxyl groups of the B ring on morin introduced hindrances on the active site of CaLB. Finally, selected flavonoid esters showed significantly higher antimicrobial activity compared with the original compound. This work indicated that these lipase-based nanobiocatalysts can be successfully applied to produce lipophilic derivatives of aglycon flavonoids with improved antimicrobial activity.
