Abstract:Regioselectivity of lipases played a crucial role in structured lipids synthesis and oil modification.Because of differences between aqueous and non-aqueous systems,regioselectivity analyzed by the conventional hydrolysis method might not always match that in synthesis reactions.In this paper,acidolysis of lauric acid and camellia oil was developed to directly evaluate regioselectivity of lipases in solvent-free system.Fatty acid composition and positional distribution of reaction products were detected by gas chromatography.Through this protocol,Lipozyme RM IM,L02,L03 and L04 were identified as sn-1 (3) regioselectivity,L01 was partially selective,and Novozym 435 was nearly non-selective.Predictability of the model reaction had been verified by replacing substrates of lipases.According to the comparative analysis of results of the acidolysis method and hydrolysis one,regioselectivity of lipases under two situations was generally the same,except selectivity of Novozym 435 which was susceptible to solvent systems.Therefore the present method avoided limits of hydrolysis determination results in synthesis reactions.And moreover it lowered the requirement on purity of reactants,which means it can be used to screen numerous lipases at low cost.

Abstract:Our previous work has indicated that enzymatic resolution of glycidyl butyrate are strongly affected by many factors, including concentration of substrate, amount of lipase, the temperature, pH, shaking speed and reaction time. In this study, Plackett-Burman design was undertaken to evaluate the effects of the six factors. By regression analysis, concentration of substrate, amount of lipase and the temperature were found to be important for enzymatic resolution of glycidyl butyrate. In the second phase of the optimization process, a response surfacemethodology (RSM) was used to optimize the above critical factors, and to find out the optimal concentration levels and the relationships between these factors. By solving the quadratic regression model equation using appropriate statistic methods, the optimal parameter of the variables were determined as: 0.499 mol/L glycidyl butyrate, 30.23 mg/g lipase and 29.68 degrees C. In the optimum condition, the value of enantiomeric excess(ee%) was 93.28%. Compared to 84.65% which was the maximum ee% under the non-optimized condition, this study has a significant advancement. The experimental data under various conditions have validated the theoretical values.

Abstract:Diglycerol obtained from biomass glycerol has been valorized into selective products by acetalization reaction in the presence of ZnCl2. A synthesis method of monoacetals via catalyst-transfer is carried out encompassing two phase-separated reagents, diglycerol with n-octanal. Mechanistic analysis has revealed that the formation of unstable emulsion promotes the reaction, and the selectivity is affected by the catalyst-transfer direction. By measuring the Zn content in the two phases, we demonstrated the transfer direction of ZnCl2 and the monoacetal were opposite, resulting in separation of the catalyst and the substrate. And the formation of diacetal was retarded. The products monoacetals exhibit superior surfactant properties, especially in terms of foam stability and alkali tolerance.

Abstract:We investigated the influence of solvent on the positional selectivity of Novozym 435 which was the immobilized Candida antarctica lipase B (CALB) during the esterification of oleic acid with glycerol for 1,3-diolein preparation previously. Herein, molecular modeling was used to elucidate the underlying mechanism of the solvent effect on the positional selectivity of the enzyme. The results showed that the binding energy of sn-1 hydroxyl of glycerol molecular with CALB became higher, and the binding energy of sn-2 hydroxyl of glycerol molecular with CALB became lower along with the increase of the solvent log P. It was demonstrated that, increasing log P of the solvent, the enzyme selectivity to sn-1 hydroxyl of glycerol molecular grew weaker, and the selectivity to sn-2 hydroxyl of glycerol molecular grew stronger.
Synthesis of flavor and fragrance esters using Candida antarctica lipase

Abstract:Candida antarctica lipase fraction B (CAL-B) showed substrate specificity in the synthesis of esters in hexane involving reactions of short-chain acids having linear (acetic and butyric acids) and branched chain (isovaleric acid) structures, an unsaturated (tiglic acid) fatty acid, and phenylacetic acid with n-butanol and geraniol. The variation in the conversion to the esters was ca. 10%. Similar results were observed in a study of the alcohol specificity of the enzyme for esterification of acetic and butyric acids with four alcohols: n-butyl, isopentyl, 2-phenylethyl, and geraniol. Enantioselectivity of CAL-B in hexane with a range of chiral alpha-substituted or beta-substituted carboxylic acids and n-butyl alcohol was analyzed. The results show that CAL-B can be employed as a robust biocatalyst in esterification reactions due to the high conversions obtained in the synthesis of short-chain flavor esters in an organic solvent, although this enzyme exhibited modest enantioselectivity with chiral short-chain carboxylic acids.
Optimization of the Lipase-Catalyzed Selective Amidation of Phenylglycinol

Abstract:Ceramides and their analogs have a regulatory effect on inflammatory cytokines expression. It was found that a kind of ceramides analog synthesized from phenylglycinol could inhibit the production of cytokine TNF-α. However, two active hydrogen groups are present in the phenylglycinol molecule. It is difficult to control the process without hydroxyl group protection to dominantly produce amide in the traditional chemical synthesis. A selective catalytic the amidation route of phenylglycinol by lipases was investigated in this research. The results indicated that the commercial immobilized lipase Novozym 435 has the best regio-selectivity on the amide group. Based on the experimental results and in silico simulation, it was found that the mechanism of specific N-acyl selectivity of lipase was not only from intramolecular migration and proton shuttle mechanism, but also from the special structure of active site of enzyme. The optimal reaction yield of aromatic amide compound in a solvent-free system with lipase loading of 15 wt% (to the weight of total substrate) reached 89.41 ± 2.8% with very few of byproducts detected (0.21 ± 0.1% ester and 0.64 ± 0.2% diacetylated compound). Compare to other reported works, this work have the advantages such as low enzyme loading, solvent free, and high N-acylation selectivity. Meanwhile, this Novozym 435 lipase based synthesis method has an excellent regio-selectivity on most kinds of amino alcohol compounds. Compared to the chemical method, the enzymatic synthesis exhibited high regio-selectivity, and conversion rates. The method could be a promising alternative strategy for the synthesis of aromatic alkanolamides.

Abstract:To valorize oleaginous Schizochytrium sp. biomass, a novel sequential process was addressed and developed to simultaneously produce high value DHA-enriched glycerides and low value bulky biodiesels. In this process, Schizochytrium sp. biomass was first treated by alkaline protease; then the resulting biomass was directly subjected to enzymatic ethanolysis catalyzed by liquid enzymes from C. antarctica (CALA), T. lanuginosus (TL), R. miehei (RM) and A. oryzae (PLA). For all four liquid lipases, the alkaline protease treatment on Schizochytrium sp. biomass significantly enhanced total lipid recovery, biodiesel yield and yield of DHA-enriched glycerides. Under optimized conditions, CALA and TL achieved the highest biodiesel yields 76.28-76.66 wt% based on microalgae TFAs while PLA had the lowest (55.25 wt%). Moreover, CALA attained the highest DHA (77.93 wt%) and LC PUFAs (93.08 wt%) contents in glyceride fraction, suggesting a superior performance to concentrate DHA and LC PUFAs in glycerides. C-13 NMR analysis illustrated that non-regiospecificity and unfavorability towards LC PUFAs of CALA made it possible to transesterify saturated fatty acids located in all 3 positions of microalgal oil; thus LC PUFAs were highly enriched in glycerides. Overall, the sequential process using enzymes and ethanol as green media was successfully established for the co-production of high-value microalgal bioproducts rich in LC PUFAs and sustainable biodiesels in a comprehensive manner.
[Rifampicin in the treatment of Staphylococcus arthritis].

Abstract:Rifampicin is used for treating 20 Staphylococcus septic arthritis, in combination with another antibiotic. The former is chosen for its striking bone diffusion. The average duration of the whole antibiotic treatment is 3 months. Rifampicin is usually prescribed, in daily doses of 1 200 mg for 60 days, after one month of parenteral antibiotherapy. The bactericidal activity is controlled regularly by measuring serum inhibitory titre. For 2 patients, the administration of rifampicin is suspended after 12 and 25 days respectively, due to digestive disorders. Results are evaluated at the end of a period averaging 18 months. Articular infection has been cured in all patients. Taking these results into account, we are able to emphasize that the inclusion of rifampicin in the antibiotic scheme is of great value in treating bone and articular infections.

Abstract:Glycerol esterification produces a variety of valuable chemicals and has been a subject of great interest in recent years. Lipase-catalysed synthesis of acylglycerols reactions have many potential applications, including in the production of structured lipids, biofuels and pharmaceuticals. It offers several benefits over traditional chemical methods, including higher selectivity, milder reaction conditions and increased sustainability. In this systematic review, we summarize 52 primary research of lipase catalysed synthesis of acylglycerol published from 2012 to 2022. Different types of reactions employed in the synthesis of acylglycerols using various lipases are thoroughly discussed. The use of lipase in as biocatalyst in the synthesis of acylglycerols is safe for consumption and sustainable as it can reduce the utilization of toxic and hazardous organic solvents and chemicals that cause harm to human health and the environment. This review serves as a reference for the research and development of acylglycerols using biocatalysts and reactors. It could be an interest to the food and pharmaceutical industries or individuals who are interested to explore this field. Several limitations associated with the overall research on production of acylglycerols are presented in the conclusion section.

Abstract:The esterification of some natural antioxidants such as cinnamic acid derivatives and ascorbic acid in non‐aqueous media, catalyzed by immobilized lipases from Candida antarctica and Rhizomucor miehei, was investigated. The alcohol chain length affected the rate of esterification of cinnamic acids by both lipases. Higher reaction rates were observed when the esterification was carried out with medium‐ or long‐chain alcohols. The rate also depended on aromatic acid structure. The reactivity of the carboxylic function of the cinnamic acids was affected by electron‐donating substituents in the aromatic ring. Higher yields were observed for the esterification of p‐hydroxyphenylacetic acid (97%) catalyzed by C. antarctica lipase and for the esterification of cinnamic acid (59%) catalyzed by R. miehei lipase. Candida antarctica lipase was more suitable for producing ascorbic acid fatty esters, catalyzing with a relatively high yield (up to 65% within 24 h) the regioselective esterification of ascorbic acid with various fatty acids in 2‐methyl‐2‐propanol. The reaction rate and yield depended on the fatty acid chain length and on the molar ratio of reactants. All ascorbic acid fatty esters produced by this procedure exhibited a significant antioxidant activity in a micellar substrate composed of linoleic acid.

Abstract:On the basis of our previous finding that humulone, a bitter acid from beer hop extract, was a potent inhibitor of bone resorption and inhibited the catalytic activity of cyclooxygenase-2 (COX-2) and more potently the transcription of the COX-2 gene, we examined the effect of humulone on angiogenesis, using chick embryo chorioallantoic membranes (CAMs) and vascular endothelial and tumor cells. Humulone significantly prevented in vivo angiogenesis in CAM in a dose-dependent manner with an ED(50) of 1.5 microg/CAM. Humulone also inhibited in vitro tube formation of vascular endothelial cells. Moreover, it suppressed the proliferation of endothelial cells and the production of vascular endothelial growth factor (VEGF), an angiogenic growth factor, in endothelial and tumor cells. Thus, humulone is a potent angiogenic inhibitor, and may be a novel powerful tool for the therapy of various angiogenic diseases involving solid tumor growth and metastasis.
Optimization of solvent-free enzymatic esterification in eutectic substrate reaction mixture

Abstract:The Candida rugosa lipase catalyzed esterification of (-)-menthol and lauric acid (LA) was studied in a eutectic mixture formed by both substrates((-)-menthol:LA 3:1, mol/mol). No additional reaction solvent was necessary, since the (-)-menthol:LA deep eutectic solvent (DES) acts as combined reaction medium and substrate pool. Therefore, the esterification is conducted under solvent-free conditions. The thermodynamic water activity (aw) was identified as a key parameter affecting the esterification performance in the (-)-menthol:LA DES. A response surface methodology was applied to optimize the esterification conditions in terms aw, amount of C. rugosa lipase (mCRL) and reaction temperature. Under the optimized reaction conditions (aw = 0.55; mCRL =60 mg; T =45 °C), a conversion of 95 ± 1% LA was achieved (one day), the final (-)-menthyl lauric acid ester concentration reached 1.36 ± 0.04 M (2.25 days). The experimental product formation rate agreed very well with the model prediction.

Abstract:The effect of reaction media, cosolvent and additives on candida antarctica lipase B(Novozym 435)-catalyzed enantioselective esterification of ketoprofen was systematically exp-lored. Novozym 435 showed high catalytic activity and enantioselectivity in cyclohexane. Cosolvent and additives have profound effects on Novozym 435-catalyzed enantioselective esterification of ketoprofen: polar cosolvent benzene and toluene can improve enzymatic enantioselectivity; a small amount of 18-Crown-6 could increase both the esterification rate and the enantioselectivity. Dimethylsulfoxide (DMF) however, could not improve the enanti-oselective esterification within the scope studied.

Abstract:The preparation of ester enantiomers (acetates, butanoates, hexanoates and octanoates) of the secondary alcohols 2-pentanol, 2-heptanol and 2-nonanol via lipase-catalyzed kinetic resolutions was investigated. Conversion rates and stereochemical courses of esterification and hydrolysis reactions catalyzed by commercially available enzyme preparations were followed for the homologous series of these passion fruit-typical 2-alkyl esters by capillary gas chromatography using heptakis(2,3-di-O-methyl-6-O-tert-butyldimethylsilyl)-beta-cyclodextrin as chiral stationary phase. An efficient method was developed to prepare the ester enantiomers via lipase-catalyzed esterifications: optically pure (R)-2-alkyl esters (ee > 99.9%) were obtained by esterification of the racemic alcohols with enantioselective Candida antarctica lipase B (immobilized) as catalyst. The subsequent esterification of the unreacted alcohols using lipase from Candida cylindracea yielded the optically enriched (S)-esters (ee > 81.4%). The separation of the products via liquid solid chromatography using a mixture of silica gel and aluminum oxide (basic) resulted in high chemical purities and yields (> 40 mol %).
Parameters Influencing Lipase-Catalyzed Glycolipid Synthesis by (Trans-)Esterification Reaction

Abstract:Glycolipids are biodegradable, non-toxic surfactants with a wide range of applications. Enzymatic esterification or transesterification facilitated in reaction media of low water activity is a reaction strategy for the production of tailor-made glycolipids as a high structural diversity can be achieved. Organic solvents, ionic liquids, and deep eutectic solvents have been applied as reaction media. However, several challenges need to be addressed for efficient (trans-)esterification reactions, especially for the lipophilization of polar substrates. Therefore, crucial parameters in (trans-)esterification reactions in conventional and non-conventional media are discussed and compared in this review with a special focus on glycolipid synthesis.

Abstract:1,3-Diacylglycerol preparation has roused increasing attention in recent years as the 1,3-diacylglycerol-rich oils can suppress the deposition of visceral fat and prevent the body weight increasing. Lipozyme TL IM-mediated esterification of oleic acid with monoolein was effective for 1,3-diacylglycerol production. During the esterification process, the solvent shows obvious influence on the diolein synthesis as well as the 1,3-diolein production. This work investigated the related kinetics and mechanism of the solvent effect on the esterification and Lipozyme TL IM performance. The results indicated that both the esterification rate constant and the acyl migration rate constant positively correlated with the logP of the solvent, while the site specificity of lipase has negative correlation with solvent logP. The acylation toward the 2-position of 1-monoolein was more sensitive to the solvent logP compared to the 1-position of glycerides. Molecular dynamics simulation revealed that solvents with different logP influenced the structure of Lipozyme TL IM including RMSD, hydrogen bond, and radial distribution function to a large extent, which subsequently led to the catalytic activity and selectivity variation of the lipase.

Abstract:Chiral tertiary alcohols are important organic compounds in science as well as in industry. However, their preparation in enantiomerically pure form is still a challenge due to their complex structure and steric hindrances compared with primary and secondary alcohols, so kinetic resolution could be an attractive approach. Lipase A from Candida antarctica (CAL‐A) has been shown to catalyze the enantioselective esterification of various tertiary alcohols with excellent enantioselectivity but low activity. Here we report a mutagenesis study by rational design to improve CAL‐A activity against tertiary alcohols. Single mutants of CAL‐A were selected, expressed, immobilized and screened for esterification of the tertiary alcohol 1,2,3,4‐tetrahydronaphthalene‐1‐ol. A double mutant V278S+S429G showed a 1.5‐fold higher reaction rate than that of the wild type CAL‐A, while maintaining excellent enantioselectivity.

Efficient water removal in lipase-catalyzed esterifications using a low-boiling-point azeotrope

Abstract:High conversions in lipase-catalyzed syntheses of esters from free acyl donors and an alcohol requires efficient removal of water preferentially at temperatures compatible to enzyme activity. Using a lipase B from Candida antarctica (CAL-B)-mediated synthesis of sugar fatty-acid esters, we show that a mixture of ethyl methylketone (EMK) and hexane (best ratio: 4:1, vo/vo) allows efficient removal of water generated during esterification. Azeotropic distillation of the solvent mixture (composition: 26% EMK, 55% hexane, 19% water) takes place at 59 degrees C, which closely matches the optimum temperature reported for CAL-B. Water is then removed from the azeotrope by membrane vapor permeation. In case of glucose stearate, 93% yield was achieved after 48 h using an equimolar ratio of glucose and stearic acid. CAL-B could be reused for seven reaction cycles, with 86% residual activity after 14 d total reaction time at 59 degrees C. A decrease in fatty-acid chain length as well as increasing temperatures (75 degrees C) resulted in lower conversions. In addition, immobilization of CAL-B on a magnetic polypropylene carrier (EP 100) facilitated separation of the biocatalyst.
Performance enhancement of chemically modified lipase in the organic phase guided by molecular simulation

Abstract:Lipase is one of the most important biocatalysts because of its stereoselectivity towards chiral substrates and reasonable adaptiveness of non-conventional reaction media,including hydrophobic organic solvents. Increasing the selectivity of lipase in such non-conventional environments has been a continuous hotspot. However,current researches on improving lipase selectivity usually cause critical decrease of catalytic activity. In this work,a novel computation- directed approach to enhance lipase enantioselectivity in organic media without losing activity was introduced. Stereoselective acylation of chiral secondary alcohols catalyzed byPcL(Pseudomonas cepacia lipase) inn-hexane was taken as the model reaction. According to the results from molecular dynamics simulation,when the Tyr29 inside thePcL catalytic cavity was modified by N-acetylimidazole (NAI),the enantioselectivity of the lipase was significantly increased. The simulation result was agreed with the experimental data. MALDI-TOF-MS confirmed the Tyr29 modified position,and kinetic resolution of chiral secondary alcohols by NAI-modifiedPcL displayed an increase of enantioselectivity ratio from 12.6 to 48.1 and higher activity.

Abstract:Hypothesis A critical challenge in the enzymatic conversion of acylglycerols is the limited exposure of the enzyme dissolved in the aqueous solution to the hydrophobic substrate in the oil phase. Positioning the enzyme in a microenvironment with balanced hydrophobicity and hydrophilicity in Pickering emulsion will facilitate the acylglycerol-catalyzing reactions at the interface between the oil and liquid phases. Experiments In this work, to overcome the challenge of biphasic catalysis, we report a method to immobilize enzymes in polyethylene glycol (PEG)-based hydrogel microparticles (HMPs) at the interface between the oil and water phases in Pickering emulsion to promote the enzymatic conversion of acylglycerols. Findings 3¬Ýwt% of HMPs can stabilize the oil-in-water Pickering emulsion for at least 14¬Ýdays and increase the viscosity of emulsions. Lipase-HMP conjugates showed significantly higher hydrolytic activity in Pickering emulsion; HMP-immobilized lipase SMG1 showed an activity about three times that of free lipase SMG1. Co-immobilization of a lipase and a fatty acid photodecarboxylase from Chlorella variabilis (CvFAP) in Pickering emulsion enables light-driven cascade conversion of triacylglycerols to hydrocarbons, transforming waste oil to renewable biofuels in a green and sustainable approach. HMPs stabilize the Pickering emulsion and promote interfacial biocatalysis in converting acylglycerols to renewable biofuels.
