The Effect of Human Urinary Kallidinogenase on the Cerebrovascular Reserve Capacity in Cerebral Ischemic Rabbits

Abstract:Objective To study the cerebrovascular reserve capacity effected by human urinary kallidinogenase(hK)in the local cerebral ischemic rabbits.Methods Total 17 healthy New Zealand rabbits were randomly divided into the control group,the placebo group and the hK group.hK or physiological saline was injected in the ischemia-reperfused rabbit models which were occluded with a thread in the middle cerebral artery(MCAO).The neurological function deficiency evaluation was performed followed by the positron emission computerized tomography(PET)/computer tomography(CT)examination to evaluate local glucose metabolic and blood fluid volume of the brain at 3,7 and 14 days respectively.The rabbits were executed and the brains were taken out by staining with Ulex agglutinin to measure cerebral vessels at 14 days.Results(1)Both the placebo group and the hK group had neurological function deficiency after MCAO.The score was 3.90±0.31,3.32±0.23,2.84±0.28 in placebo group and 3.17±0.32,2.62±0.21,1.09±0.17 in hK group at 3,7,14 days respectively.The improvement was more obvious in the hK group.And the difference between two groups was significant(3d,7d,P<0.05,14d,P<0.01).(2)The metabolic level was symmetrical in the control group.In the placebo group,the metabolic level in ischemic area was much lower than the normal area,30% at 3 days,and about 51% at 14 days.But the metabolic level was obviously increased after treatment with hK.At 3 days,the ratio was about 41% and was about 91% at 14 days,which was significantly different compared with the placebo group.(3)At the 14 days,the minor vessels number in the hK group was 14.12±1.32 which was much more than that in the control group(3.04±0.82)and placebo group(5.45±0.67)(P<0.01).Conclusion hK can improve the neurological function deficiency and increase the vascular reserve capacity by regenerating cerebral vessels.
