Abstract:The synthesis of primary amines via the reductive amination of alcohols involves a hydrogen-borrowing or hydrogen-transfer mechanism, which consists of three main steps: alcohol hydroxyl dehydrogenation, carbonyl imidization, and imine hydrogenation. Heterogeneous catalysts are widely used for this reaction because of their high performance and amenability to separation and reuse. However, the efficiency of reductive amination is limited by the dehydrogenation step, which is severely affected by the competitive adsorption of NH3. We hope to improve the efficiency of reductive amination by increasing dehydrogenation efficiency. Therefore, in this overview, we introduce the research progress of alcohol reductive amination reaction catalyzed by heterogeneous metal catalysts, focusing on methods of enhancing dehydrogenation efficiency by screening the metal component and the acidity/alkalinity of the support. Finally, we propose some new strategies for the preparation of catalysts from the perspective of overcoming the competitive adsorption of NH3 and speculate on the design and synthesis of novel catalysts with high performance in the future.

Abstract:Recently, the asymmetric reduction of imines catalyzed by Lewis-basic organocatalyst has been received much attention as the cheap hydrogen source and the simple post treatment. Based on the different functional groups of organocatalysts, this paper introduced formamide, pyridine amide, sulfonamide, supported and other Lewis basic organocatalysts. The structural characteristics, catalytic activity and mechanism of the Lewis-basic organocatalysts were summarized.

Abstract:The reductive alkylation reaction of aldehyde/ketone and amine is an important reaction to synthesis alkylated amine.Two main methods to achieve the synthesis were introduced,which were catalytic hydrogenation and borohydride reduction.Described the main research progress and introduced the catalysts used in the two methods.Pointed out sulfidizing of catalyst metal was effective to achieve high selectivity.Introduced the commonly catalytic system used in borohydride chemical reduction method and discussed the effects of the reaction conditions such as steric hindrance of reactant,dehydrating agent,raw material ratio and solvent.Modifying the reaction conditions such as raw material ratio and solvent could improve the selectivity of target product.

Abstract:A new method for reduction of N-nitroso aza-aliphatic cyclic compounds employing zinc in pressurized CO2–H2O medium has been developed. H2O and NH4Cl were used as hydrogen donors, and reduction was performed under environmentally benign conditions. The presented approach allowed to obtain the respective N-amino products selectively and in excellent yields (up to 97%).

Abstract:Herein, we report the first example of efficient reductive amination of ketones/aldehydes with amines using BH3N(C2H5)3 as a catalyst and a reductant under mild conditions, affording various tertiary and secondary amines in excellent yields. A mechanistic study indicates that BH3N(C2H5)3 plays a dual function role of promoting imine and iminium formation and serving as a reductant in reductive amination.

Abstract:Zinc dust in glacial acetic acid (Zn/HOAc) reagent system is an important traditional reductive reagent in organic chemistry, which still plays a unique role in a wide range of functional group transformations in organic synthesis. We now present a systematic outline of the applications of this reduction system in practical organic synthesis. This article mainly surveys the hydrogenative reductions of CC, C C, CO, CN, CS, N–N, NN, NO, N–O, O–O, S–S, reductive cleavages of C−halo, C–N, C–O, C–S, and some interesting reductive rearrangement reactions mediated by zinc in acetic acid, respectively. The relevant plausible reaction mechanisms under Zn/HOAc conditions were also discussed briefly.

Abstract:A photoredox-catalyzed sequential decarboxylative/defluorinative aminoalkylation of CF3-alkenes with N-arylglycines is described. This metal-free and redox-neutral protocol provided efficient access to the monofluoroalkenyl-1,5-diamines in good yields with excellent functional group compatibility. Mechanistic studies revealed that the reaction proceeds via a radical pathway with the gem-difluoroalkenyl amine as an intermediate.

Abstract:An expeditious, practical, and mild method for the reduction of amides to amines is reported. The procedure is based on the activation of amides with titanium tetrachloride followed by reduction with lithium aluminum hydride. The reducing system can be applied to the reduction of tertiary and secondary amides giving the corresponding amines in good yields. The protocol was also extended to the reduction of amides of Nα-protected amino acid and dipeptides. The corresponding 1,2 diamines and diaminoalcohols were produced in high yields and with retention of configuration at the chiral centers.

Abstract:The direct oxidative N-acylation reaction of primary amides with aryl/alpha,beta-unsaturated aldehydes was achieved in the presence of azolium salt C3 and an inorganic base using 3,3',5,5'-tetra-tert-butyldipherioquinone as the oxidant, thus providing an efficient approach for the synthesis of three types of imide compounds including N-sulfonylcarboxamides, N-sulfinylcarboxamides, and dicarboxyimides in good yield.

Abstract:A step-wise reductive method that converts nonracemic γ-chiral α-ketoamides into amide derivatives was developed under mild conditions. The method involves a highly efficient three-step transformation including NaBH4-reduction, bromination and Pd/C hydrogenation reactions. As a consequence, a variety of nonracemic γ-diaryl amides products were obtained in high overall yields with excellent enantiomeric specificity. The utility of the method is demonstrated through the concise formal synthesis of (+)-sertraline in good overall yield.

Abstract:LiAlH4 and NaBH4 were found to mediate the conversion of 2-(pyrimidyl-2-ylsulfanyl)-N-arylbenzamides and 2-(triazinyl-2-ylsulfanyl)-N-arylbenzamides into pyrimidyl and triazinyl amines Under notably mild conditions via a novel reductive rearrangement mechanism. These reactions invent a new route to prepare amines, which are a kind of important biologically active compounds and provide the first insight into a novel hydride-promoted reductive rearrangement of amides. Abstract:Asymmetric reductive amination of poorly nucleophilic sulfonamides was realized in the presence of nickel catalysts and titanium alkoxide. A wide range of ketones, including enolizable ketones and some biaryl ones, were converted into sulfonamides in excellent enantiomeric excess. The cyclization of sulfamates and intermolecular reductive amination of a diarylphosphinamide were also successful. Formic acid was used as a safe and economic surrogate of high-pressure hydrogen gas.

Abstract:AbstractTransformation of biomass into valuable nitrogen‐containing compounds is highly desired, yet limited success has been achieved. Here we report an efficient catalyst system, partially reduced Ru/ZrO2, which could catalyze the reductive amination of a variety of biomass‐derived aldehydes/ketones in aqueous ammonia. With this approach, a spectrum of renewable primary amines was produced in good to excellent yields. Moreover, we have demonstrated a two‐step approach for production of ethanolamine, a large‐market nitrogen‐containing chemical, from lignocellulose in an overall yield of 10 %. Extensive characterizations showed that Ru/ZrO2‐containing multivalence Ru association species worked as a bifunctional catalyst, with RuO2 as acidic promoter to facilitate the activation of carbonyl groups and Ru as active sites for the subsequent imine hydrogenation.

Abstract:An efficient and general protocol for copper-catalyzed N-arylation of aliphatic amines and amino acids has been developed using aryl iodides under mild conditions (coupling temperature at 25-35 degrees C). For the N-(o-nitrophenyl) amino acid derivatives, subsequent reduction of the nitro group in the presence of tin(II) chloride resulted in 3,4-dihydroquinoxalin-2(1H)-one derivatives in good yields.
