Effect of Electroporation Conditions on Cell Viability of Haematococcus Pluvialis

Abstract:Haematococcus pluvialis is one of the important species of microalgae with great economic value.However,no report on the successful transformation of foreign genes by electroporation has been made to date due to the lack of effective genetic transformation methods.The effect of electroporation conditions on cell viability of Haematococcus pluvialis was systematically investigated in this paper.The results showed that the viability of cells during the early log phase was obviously higher than that during the mid-log phase or late log phase by electroporation under the same conditions.Comparing with square wave pulses and exponential decay wave pulses,a decline of cell viability was found with the increase of electrical field strength or pulse length.A cell viability of approximate 50% was achieved by using square wave pulses with 600 V/cm and 2 ms,or by using exponential decay wave pulses with either 500 V/cm,5.3 ms or 600 V/cm,2 ms.Under the same electrical field strength and pulse length,the cell viability by exponential decay wave pulses was higher than that by square wave pulses.These results provided useful data and practical guide for establishing a successful method for transformation of Haematococcus pluvialis with high efficiency by electroporation.
