In-situ laser-induced fluorescence system for nanoliter sample detection

Abstract:Aiming at the key problem of detecting extra small volume sample in bioscience research,an in-situ laser-induced fluorescence system was constructed for nanoliter sample detection.In this research,we used a capillary as detection cell,a diode-pumped solid state laser as light source,and a high resolution spectrometer as detector for constructing an in-situ laser-induced fluorescence system.In addition,the fluorescent photons emitted from a sample in the capillary were directly collected in the common end of bifurcated fiber where the excitation lights came from.An experimental design for rhodamine B detection has been performed for system validation.The results show that the concentrations have a good linearity with the fluorescence intensity in the range of 93pg/mL-15ng/mL.The correlation coefficient is 0.9982 with a detection limit down to 28pg/mL.The newly built detection system satisfies the requests of biomass detection practice,and possesses advantages of low noise,low reagent consumption,high sensitivity and good precision.
