Abstract:Atomic force microscopy (AFM) has many unique features compared with other conventional microscopies, such as high magnification with high resolution, minimal sample preparation, acquiring 2D and 3D images at the same time, observing ongoing processes directly, the possibility of manipulating macromolecules, etc. As a nanotechnology tool, AFM has been used to investigate the nanostructure of materials in many fields. This mini-review focuses mainly on its latest application to characterise the macromolecular nanostructure and surface topography of agricultural products. First the fundamentals of AFM are briefly explained. Then the macromolecular nanostructure information on agricultural products from AFM images is introduced by exploring the structure-function relationship in three aspects: agricultural product processing, agricultural product ripening and storage, and genetic and environmental factors. The surface topography characterisation of agricultural products using AFM is also discussed. The results reveal that AFM could be a powerful nanotechnology tool to acquire a deeper understanding of the mechanisms of structure and quality variations of agricultural products, which could be instructive in improving processing and storage technologies, and AFM is also helpful to reveal the essential nature of a product at nanoscale. Abstract:Ultrafiltration had wide applications in the separation, concentration, and purification of polysaccharides of the following field: Chinese herb polysaccharides extracted from Ganoderma japonicum, Rheum tanguticum, LWDH decoction, Astragalus membranceus, Ganoderma sinense, Redix ginseng; marine active polysaccharides, such as shark cartilage mocopolyaccharides, Cristaria plicata polysaccharides, Porphyra haitanensis polysaccharides, fucoidan, carrageenan; ferment polysaccharides extracted from rhizomorph (Armillariella mellea), from PS-9415 fermentation broth, from mycelia(Cordyceps sinensis); and edible plant polysaccharides extracted from tea, lentinan, flammulina velutipes, aloe, Lyceum chinensa. Ultrafiltraion had a high recovery rate, not so easy to damage the bioactivity of polysaccharides, low energy consuming, and was suitable to industrial production of polysaccharides.

Abstract:With the deepening of polysaccharide research, whether the real polysaccharide structure can be clearly observed directly affects researchers' understanding of polysaccharide structure and function. Therefore, the establishment of a suitable method to study polysaccharide structure is urgent. Konjac glucomannan (KGM), agar, and curdlan were selected in this paper to investigate the effects of drying temperature, plasticizer type and dosage on the molecular structure of polysaccharides in the preparation of polysaccharide samples for AFM testing. The experimental results showed that the molecular morphology changes of different polysaccharides varied at different drying temperatures. Adding a certain dose of plasticizer could relieve the shrinkage of the hydrophilic polysaccharide molecular chain during the drying process. Compared with Tween 20 and Tween 80, glycerol showed a superior effect on relieving molecular chain shrinkage with the same addition amount. Finally, a new polysaccharide sample preparation method for AFM was proposed, and the feasibility of the method was confirmed, which provided a new idea for better use of AFM to observe the morphology of polysaccharides.

Abstract:Polysaccharide materials and biomaterials gain the focus of intense research owing to their great versatility in chemical structures and modification possibilities, as well as their biocompatibility, degradability, and sustainability features. This review focuses on the recent advances in the application of SANS on polysaccharide systems covering a broad range of materials such as nanoparticulate assemblies, hydrogels, nanocomposites, and plant-originating nanostructured systems. It motivates the use of SANS in its full potential by demonstrating the features of contrast variation and contrast matching methods and by reporting the methodologies for data analysis and interpretation. As these soft matter systems may be organized in multiple length scales depending on the interactions and chemical bonds between their components, SANS offers exceptional and unique opportunities for advanced characterization and optimization of new nanostructured polysaccharide materials.

Abstract:High-speed atomic force microscopy (HS-AFM) is a technique that enables real-time imaging of nanoscale phenomena in solution. It was originally developed to visualize biomolecules, whose dynamics in solution significantly affect the manifestation of their functions, and has contributed to the understanding of molecular mechanisms based on the observation of single-molecule dynamics of proteins. In recent years, its application has broadened to include not only biomolecules, but also the structural dynamics of supramolecular assemblies that associate and dissociate in solution, as well as the evaluation of synthetic molecules such as polymer gels that swell in solution. In this paper, we review some of our recent studies on the application of HS-AFM to supramolecular polymers and hydrogel particles.

Abstract:Natural polysaccharides exhibit a wide range of biological activities, which are closely related to their structural characteristics, including their molecular weight distribution, size, monosaccharide composition, glycosidic bond types and spatial conformation, etc. Size exclusion chromatography (SEC) and asymmetrical flow field-flow fractionation (AF4), as two potent separation techniques, both harbor potential for continuous development and enhancement. This manuscript reviewed the fundamental principles and separation applications of SEC and AF4. The structural information and spatial conformation of polysaccharides can be obtained using SEC or AF4 coupled with multiple detectors. In addition, this manuscript elaborates in detail on the shear degradation of samples such as polysaccharides separated by SEC. In addition, the abnormal elution that occurs during the application of the two methods is also discussed. Both SEC and AF4 possess considerable potential for ongoing development and refinement, thereby offering increased possibilities and opportunities for polysaccharide separation and characterization.

Abstract:Scanning probe microscopy (SPM) allows the spatial imaging, measurement, and manipulation of nano and atomic scale surfaces in real space. In the last two decades, numerous advanced and functional SPM methods, particularly atomic force microscopy (AFM), have been developed and applied in various research fields, from mapping sample morphology to measuring physical properties. Herein, we review the recent progress in functional AFM methods and their applications in studies of two-dimensional (2D) materials, particularly their interfacial physical properties on the substrates. This review can inspire more exciting application works using advanced AFM modes in the 2D and functional materials fields.

Abstract:In the past decades,AFM have great advances in high resolution molecular imaging and single molecule detection applications.AFM single molecular force spectroscopy(SMFS)are available to explore polymer structural changes,molecular identification of bio-signal transmission or protein folding process and unfolding process,the evidence of the relation between structural changes and functions is provided.Some progresses are mentioned such as using AFM in functionalization decoration,in locating using SMFS surface protein and polysaccharide and their elasticity.Prospects are also provided concerning future work and difficulties in AFM application.

Abstract:Over the past 30 years, atomic force microscopy (AFM) has played an important role in elucidating the structure and properties of polymer surfaces. AFM-based techniques have enabled the quantitative determination of the physicochemical properties of polymer surfaces with high spatial resolution and under a wide variety of conditions. Coupled with the improvements in spatial and temporal resolution, multiparametric and multifunctional characterization has revealed the delicate interplay between structure, dynamics, and properties at the surfaces of complex systems. Here we summarize some of the significant advances that have been made in synthetic polymeric materials, most in the past 10 years, where AFM has been crucial, and we provide our perspective on where AFM will be insightful in future and instrumental in advancing emerging areas.

Abstract:AFM-based single-molecule force spectroscopy was employed to measure the nanomechanical properties of 1,4-linked polysaccharides. Single-molecule force spectroscopy clearly reflected the difference in the mechanical properties of alpha-(1,4)- and beta-(1,4)-linked polysaccharides. A force-induced chair-twist boat conformational transition in alpha-(1,4)-linked polysaccharides was discovered. This chair-twist boat conformational transition is a nanomechanical fingerprint of alpha-(1,4)-linked polysaccharides. (C) 1999 Elsevier Science B.V. All rights reserved.

Abstract:Atomic force microscopy (AFM) is a useful tool in the imaging and investigation of surfaces and surface interactions. The use of the technique in microbiology has grown over the years, with publications using AFM to investigate various properties of microbes such as surface structures, membrane arrangements and even visualisation of individual proteins and cell division. AFM has also shown utility when it comes to measuring biological interactions, in particular receptor–ligand interactions, where the AFM is able to measure the strength of individual interactions and map the location of interactions to the surface location on the cell. Combining the use of AFM with binding assays and immunoassays provides the ability to profile and analyse the surface of microbes such as bacteria and viruses. Furthermore, utilising carbohydrate binding monoclonal antibodies and lectins provides the opportunity to better understand the glycosylation patterns of these cells and particles giving unique insights into how microbes react with the environment and binding agents. This chapter discusses the methods used in investigating microbes and the carbohydrates decorating their surfaces with AFM and immunoassays.

Abstract:This review surveys recent advances in the material functionalization of structural polysaccharides such as cellulose and chitin. This functionalization includes improving their mechanical properties, subtly regulating optical performance, developing cytocompatibility, etc. , and has been accomplished through nano-architectonic methods such as the orientation control of segments and molecules and the intimate nanoscopic mixing of different components, each on a scale larger than a single molecule. It is hoped that this fundamental data will lead to further functionalization of biomass-based materials, thereby expanding their bioscientiﬁc perspective.

Abstract:Atomic force microscopy (AFM), as a type of scanning probe microscopy (SPM), possesses formidable capabilities for nanoscale imaging and force spectroscopy. Due to its advantages such as high resolution, nondestructive detection, minimal environmental restrictions, strong versatility, and real-time in situ analysis, AFM has become an indispensable tool in surface science and materials research, finding extensive applications in the study of the membrane separation and fouling processes. The tremendous advantages of AFM in characterization applications stem from its diverse tip functionalization techniques. This review encompasses the preparation of AFM probe tips and the modification techniques of special tips, including carbon nanotube (CNT) probes, metal nanowire probes, colloidal probes, and single-cell/molecule probes. Furthermore, it highlights the applications and advancements of AFM and probe modification techniques in membrane technology research. With the continuous development of tip modification techniques, the analytical capabilities of AFM will be further expanded, promising broader prospects for its application in the study of membrane fouling mechanisms and the development of antifouling membrane materials.

Abstract:Fruits and vegetables are essential horticultural crops for humans. The quality of fruits and vegetables is critical in determining their nutritional value and edibility, which are decisive to their commercial value. Besides, it is also important to understand the changes in key substances involved in the preservation and processing of fruits and vegetables. Atomic force microscopy (AFM), a powerful technique for investigating biological surfaces, has been widely used to characterize the quality of fruits and vegetables and the substances involved in their preservation and processing from the perspective of nanoscale structure and mechanics. This review summarizes the applications of AFM to investigate the texture, appearance, and nutrients of fruits and vegetables based on structural imaging and force measurements. Additionally, the review highlights the application of AFM in characterizing the morphological and mechanical properties of nanomaterials involved in preserving and processing fruits and vegetables, including films and coatings for preservation, bioactive compounds for processing purposes, nanofiltration membrane for concentration, and nanoencapsulation for delivery of bioactive compounds. Furthermore, the strengths and weaknesses of AFM for characterizing the quality of fruits and vegetables and the substances involved in their preservation and processing are examined, followed by a discussion on the prospects of AFM in this field.

Abstract:Polysaccharides are a class of bio-macromolecules with highly complex structures that are widely found in living organisms such as microorganisms, plants and animals. Polysaccharides serve not only as structural components and energy sources of cells, but also as important signaling molecules which are involved in many key biological processes. Studies on polysaccharide-mediated biological processes require access to structurally defined molecules, which approach the size and complexity of those found in nature, but naturally-occurring polysaccharides usually exist in microheterogeneous forms, making it difficult or even impossible to isolate pure polysaccharides from natural sources in most cases. Chemical synthesis represents a reliable solution to this problem, which can provide polysaccharide samples with defined chemical structures for functional studies and even a library of analogs of natural glycans for structure-activity relationship investigations. But unlike oligonucleotides and peptides, which can already be obtained by automated synthesizers in a very short of time, the chemical synthesis of glycans remains a great challenge for synthetic chemists. The major challenge for glycan synthesis lies in the need to handle both stereo-and regio-chemistry in the construction of each glycosyl linkage, and the extensive protecting-group manipulations as well as much intermediate separation make it a tedious and time-consuming process. Over the past decades, carbohydrate chemists have developed many glycosylation reactions. A series of strategies for glycan assembly have been also established. The advances in both synthetic methods and strategies have significantly increased the synthetic efficiency of carbohydrate molecules, and many great accomplishments in the field of polysaccharide synthesis have been witnessed in recent decades. Some representative methods and strategies, and their successful applications in the chemical synthesis of complex polysaccharides are summarized in this review.

Abstract:Colloids, polymers, gels, and biological materials are widely used for numerous technological applications. The design, fabrication, and understanding of the physico-chemical properties of such (bio)materials, however, represent a challenge for scientists and technologists. This review is a concise update of the latest achievements in surface and bulk analytical techniques applied to biomaterials and soft matter systems. Specific emphasis is devoted to their structural, mechanical, surface, and chemical properties. The review introduces the basics of atomic force microscopy (AFM) and discusses its combination with optical microscopies and spectroscopic techniques. In particular, we report the AFM combination with fluorescence, confocal microscopy, stimulated emission depletion, Raman, and infrared spectroscopy. The review highlights the impact and the potential applications of the combination of such experimental techniques on current problems and questions related to material and polymer science, cell biology, biochemical engineering, and protein chemistry. This review is an accessible introduction to AFM for the newcomer and a source of new experimental information for the experienced researcher.
