Abstract:Three new dimeric furanocoumarins, dahuribiethrins H-J (1-3), and a new ester coumarin, dahurinol A (4), were isolated from the roots of Angelica dahurica. Their structures were elucidated on the basis of extensive spectroscopic data including UV, IR, HRESIMS, 1D and 2D NMR. Compounds 2 and 3 exhibited inhibition of nitric oxide production in the lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells with IC50 values of 8.7 ± 0.6 and 27.3 ± 0.9 μM, respectively.

Abstract:Seven new dimeric furanocoumarins, dahuribiethrins A-G (1-7), were isolated from the roots of Angelica dahurica. Their structures were determined by chemical derivatization and extensive spectroscopic techniques, including (1)H NMR, (13)C NMR, HSQC, (1)H-(1)H COSY, HMBC, and NOESY experiments. Compounds 2, 3, 4, and 5 exhibited significant inhibition of nitric oxide production in the lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells with IC50 values in the range of 8.8-9.8 μM.

Abstract:Objective To study the chemical constituents of the roots of Angelica dahurica, a well-known Chinese herbal medicine named Baizhi in Chinese. Methods Compounds were separated by various chromatographies, and the structures of the new compounds were elucidated based on the analysis of their spectroscopic and spectrometric data (1D, 2D NMR, HRESI MS, IR, and UV). The absolute configurations of the new compounds were determined by the calculated electronic circular dichroism and chemical derivatization. The inhibitory activities of all isolates against nitric oxide (NO) production were evaluated using lipopolysaccharide-activated RAW 264.7 macrophage cells. Results Seven new 3,4-dihydro-furanocoumarin derivatives (1a/1b, 2a/2b, 3a/3b, 4) together with a known furanocoumarin (5) were isolated from the roots of A. dahurica. The new compounds included three pairs of enantiomers, (4S, 2′′R)-angelicadin A (1a)/(4R, 2′′S)-angelicadin A (1b), (4S, 2′′S)-angelicadin A (2a)/(4R, 2′′R)-angelicadin A (2b), and (4S, 2′′S)-secoangelicadin A (3a)/(4R, 2′′R)-secoangelicadin A (3b), together with (4R, 2′′R)-secoangelicadin A methyl ester (4). The known xanthotoxol (5) inhibited the NO production with the half-maximal inhibitory concentration (IC50) value of (32.8±0.8) µmol/L, but all the new compounds showed no inhibitory activities at the concentration of 100 µmol/L. Conclusion This is the first report of the discovery of 3,4-dihydro-furanocoumarins from A. dahurica. The results are not only meaningful for the understanding of the chemical constituents of A. dahurica, but also enrich the reservoir of natural products.

Methods: The chemical constituents were isolated and purified by AB-8 macroporous adsorption resin and Sephadex LH-20 column as well as semi-preparative reversed phase HPLC. The chemical structures were identified by spectral data. Results: Ten compounds were isolated and identified as xanthoarnol-3'-O-beta-D-glucopyranoside (1), angedahuricoside A (2), angedahuricoside B (3), isofraxidin-7-O-beta-D-glucopyranoside (4), fraxidin-8-O-beta-D-glucopyranoside (5), (-)-marmesinin (6), (2'S,3'R)-3'-hydroxymarmesinin (7), hyuganoside V (8), daucosterol (9), and sucrose (10). Conclusion: Compounds 1-3 are new ones and compounds 4-6 and 8 are obtained from title plant for the first time. Abstract:In this study, a new chromone, six coumarins, and a phenolic compound were isolated from a MeOH extract of an Angelica dahurica stems. Through the use of various spectral techniques, the structures of the isolated compounds were determined to be scopoletin ( 1 ), xanthyletin ( 2 ), xanthotoxin ( 3 ), hopeyhopin ( 4 ), thamnosmonin ( 5 ), 6-[(1S, 2R)-2,3-dihydroxy-1-methoxy-3-methylbutyl]-7-methoxycoumarin ( 6 ), decursidate ( 7 ), and new compound 8 , named dahuricalol. Among the isolatled and identified compounds, (S)-5-hydroxy-8-(hydroxymethyl)-2,2-dimethyl-6-oxo-3,4-dihydro-2H,6H-pyrano[3,2-g]chromen-3-yl (Z)-2-(hydroxymethyl)but-2-enoate ( 8 ) was isolated from plant for the first time.

Abstract:OBJECTIVE:To study the coumarin glucosides of Angelica dahurica.METHODS:Fresh roots and rhizomes of Angelica dahurica were extracted with ethanol at room temperature. Repeated column chromatography and preparative HPLC were used to isolate and purify the compounds. Their structures were elucidated on the basis of chemical evidence and spectral analysis.RESULTS:Twenty-six coumarin glucosides were isolated from Angelica dahurica, while here we reported 7 of them: sec.-O-beta-D-Galactopyranosyl-(R)-byakangelicin (I);8-O-beta-D-Galactopyranosyl-xanthotoxol (II); 7-O-beta-D-Apiofuranosyl-(1-->6)-beta-D-Glucopyranosyl-peucedanol (III); (R)-peucedanol-7-O-beta-D-Glucopyranoside (IV); sec.-O-beta-D-Glucopyranosyl-(R)-Oxypeucedaninhydrate (V); 7-O-beta-D-Galactopyranosyl-Sco-poletin( VI); Aesculin (VII).CONCLUSION:Compound V is a new compound, Compound VII is isolated from Umbellifera for the first time, compounds III, IV are isolated from Angelica for the first time,while compound I, II and VI are isolated from this plant for the first time.

Abstract:Two new 6-alkylcoumarins, angelitriol(1) and angelol J (2), were isolated from the EtOAc-soluble fraction of the roots of Angelica pubescens Maxim f. biserrata Shan et Yuan. Their structures were elucidated as 6-[(1R,2S)-1,2,3-trihydroxy-3-methylbutyl]-7-methoxycoumarin (1) and 6-[(1R,2R)-1-ethoxy-2,3-dihydroxy-3-methylbutyl]-7-methoxycoumarin (2), respectively, on the basis of spectral evidence. Both of them showed strong inhibitory effects on human platelet aggregation.

Abstract:As a popular medicinal plant, Angelica dahurica has been attached to great importance in herbal medicine. Its main active compounds are furanocoumarins (FCs) including imperatorin and isoimperator which exhibit anti-oxidant, anti-tumor, and anti-virus activities. However, the enzymes catalyzing the crucial step of FCs' biosynthesis pathway remain unidentified. This study initially screened six prenyltransferases (PTs)-coding sequences through transcriptome and gene expression profile analysis. Combined with qRT-PCR and in vitro enzyme activity verification, two C-PTs ( Ad PT1, 2) which convert umbelliferone to 7-demethylsuberosin (DMS), and one O-PT ( Ad PT3) which transforms xanthotoxol to imperatorin were identified. Among these PTs, Ad PT2 also produces a trace amount of osthenol from umbelliferone. Homologous modeling, molecular docking, and site-directed mutagenesis analysis were also conducted and it is discovered that Asp301 and Asp305 in the two Asp-rich regions are vital for the activity of Ad PT1. The study is the first to conduct a frontier exploration of the biosynthesis pathways of FCs in A. dahurica and will establish a foundation for the molecular mechanism of further C-PTs and O-PTs evolutionary studies. Abstract:Six new pyrrole 2-carbaldehyde derived alkaloids, dahurines A-F (1-6), along with five known ones (7-11) and butyl 2-pyrrolidone-5-carboxylate (12) were isolated from the roots of Angelica dahurica. Their structures were determined by extensive spectroscopic and spectrometric data (1D and 2D NMR, IR, and HRESIMS) and calculated electronic circular dichroism (ECD) methods. Although compounds 7 and 8 have been chemically synthesized, they were obtained from natural materials for the first time. Compounds 2, 3, 4, 10, and 11 exhibited acetylcholinesterase inhibitory activity with IC50 values in the range of 47.5-52.5 μM.
