Abstract:Aim To study the chemical constituents of the root of Angelica pubescens Maxim.f.biserrata Shan et Yuan in China National GAP Base of Chinese Materia Medica for Angelica pubescens Maxim.f.biserrata in Hubei of China.Methods The chemical components were isolated by solvent extraction and column chromatography,and their chemical structures were identified on the basis of physic-chemical constant and spectral data.Results Eight compounds were isolated and identified as angenomalin(Ⅰ),angelol A(Ⅱ),daucosterol(β-sitosteryl 3-O-β-D-glucopyranoside,Ⅲ),marmesinin(Ⅳ),adenosine(V),3-O-trans-coumaroylquinic acid(Ⅵ),3-O-trans-feruloylquinic acid(Ⅶ) and sucrose(Ⅷ).Conclusion Compounds Ⅰ,Ⅵ and Ⅶ were isolated from the titled plant for the first time.

Abstract:Angelicae Sinensis Radix,dried roots of Angelicae sinensis,is a well-known traditional Chinese medicine. Six compounds were isolated and purified by silica gel and preparative HPLC. According to physicochemical properties and spectral data,the compounds were identified as senkyunolide H-7-acetate( 1),o-phthalic acid( 2),diisobutyl phthalate( 3),p-hydroxyphenylethanol ferulate( 4),ferulic acid( 5) and coniferylferulate( 6). Abstract:A new natural coumarin, angepubebisin (1), and a new furan, angepubefurin (2), together with the five known compounds, umbelliferone, angelol B (3), ulopterol (4), peucedanol (5), and scopoletin, were isolated from the roots of Angelica pubescens Maxim. f. biserrata Shan et Yuan. The structures of angepubebisin (1) and known compounds were determined by spectroscopic methods, including IR, UV, EI-MS, HR-FTICR-MS, 1D-, and 2D-NMR spectral analyses, and angepubefurin (2) was determined by HR-FTICR-MS and X-ray diffraction analyses.

Abstract:The compounds were isolated and purified by HP20 macroporous adsorption resin, ODS, silica gel, and Sephadex LH-20 column chromatography, as well as semi-preparative HPLC chromatography from the 80% ethanol extract of the root of Angelica decursiva, and their structures were identified based on their physiochemical properties and spectroscopic data. Twelve compounds were structures were identified as (9R,10R)-9-acetoxy-8,8-dimethyl-9,10-dihydro-2H,8H-benzo[1,2-b：3,4-b']dipyran-2-one-10-yl ester (1), bakuchicin (2), (3', S,4'S)-disenecioyloxy-3',4'-dihydroseselin (3), (3'R,4'R)-3'-angeloyloxy-4'-senecioyloxy-3',4'-dihydroseselincalipteryxin (4), (+)-8,9-dihydro-8-(2-hydroxypropan-2-yl)-2-oxo-2H-furo[2,3h]chromen-9-yl-3-methylbut-2-enoate (5), libanoridin (6), selinidin (7), suberosin (8), crocatone (9), peujaponisinol B (10), peujaponisinol A (11), and ostenol (12), respectively. Compounds 1-5 were isolated from the plants of Angelica genus for the first time. Compounds 7-12 were isolated from A. decursiva for the first time.

Abstract:Objective To study the chemical constituents of Acanthopanax senticosus.Methods A.senticosus was extracted with 75% EtOH and separated with petroleum ether and EtOAc,successively.The EtOAc-soluble fraction was isolated by column chromatography and RP-preparative HPLC.Their structures were identified by spectroscopic method(MS,1H-NMR,and 13C-NMR).Results Nineteen compounds were isolated from the EtOAc fraction of A.senticosus and identified as quercetin(1),quercitrin(2),kaempferol(3),hyperoside(4),rutin(5),acacetin(6),daidzin(7),3′-methoxydaidzin(8),puerarin(9),3′-methoxypuerarin(10),4′-methoxypuerarin(11),syringaldehyde(12),syringic acid(13),glucosyringic acid(14),isofraxidin(15),2,3-di(3′,4′-methyl-endioxy-benzyl)-2-buten-4-olide(16),l-sesamin(17),methylpluviatolide(18),and 4′-hydroxy-2′-methoxycinnamaldehyde(19).Conclusion Compounds 6—11,14,16,18,and 19 are isolated from the plants in Acanthopanax(Deche et Planch.) Miq.for the first time.

Abstract:Objective To study the chemical constituents of the roots of Angelica dahurica, a well-known Chinese herbal medicine named Baizhi in Chinese. Methods Compounds were separated by various chromatographies, and the structures of the new compounds were elucidated based on the analysis of their spectroscopic and spectrometric data (1D, 2D NMR, HRESI MS, IR, and UV). The absolute configurations of the new compounds were determined by the calculated electronic circular dichroism and chemical derivatization. The inhibitory activities of all isolates against nitric oxide (NO) production were evaluated using lipopolysaccharide-activated RAW 264.7 macrophage cells. Results Seven new 3,4-dihydro-furanocoumarin derivatives (1a/1b, 2a/2b, 3a/3b, 4) together with a known furanocoumarin (5) were isolated from the roots of A. dahurica. The new compounds included three pairs of enantiomers, (4S, 2′′R)-angelicadin A (1a)/(4R, 2′′S)-angelicadin A (1b), (4S, 2′′S)-angelicadin A (2a)/(4R, 2′′R)-angelicadin A (2b), and (4S, 2′′S)-secoangelicadin A (3a)/(4R, 2′′R)-secoangelicadin A (3b), together with (4R, 2′′R)-secoangelicadin A methyl ester (4). The known xanthotoxol (5) inhibited the NO production with the half-maximal inhibitory concentration (IC50) value of (32.8±0.8) µmol/L, but all the new compounds showed no inhibitory activities at the concentration of 100 µmol/L. Conclusion This is the first report of the discovery of 3,4-dihydro-furanocoumarins from A. dahurica. The results are not only meaningful for the understanding of the chemical constituents of A. dahurica, but also enrich the reservoir of natural products.

Abstract:Seven new dimeric furanocoumarins, dahuribiethrins A-G (1-7), were isolated from the roots of Angelica dahurica. Their structures were determined by chemical derivatization and extensive spectroscopic techniques, including (1)H NMR, (13)C NMR, HSQC, (1)H-(1)H COSY, HMBC, and NOESY experiments. Compounds 2, 3, 4, and 5 exhibited significant inhibition of nitric oxide production in the lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells with IC50 values in the range of 8.8-9.8 μM.

Abstract:A new chromone, angeliticin B (1) together with nine known compounds, psoralene (2), isoimperatorin (3), (S)-(-)-2'-methoxypeucedanin hydrate (4), (S)-(-)-oxypeucedanin (5), xanthotoxin (6), isopimpinellin (7), 1'-O-β-D-glucopyranosyl-(2'S, 3'R)-3'-hydroxymarmesin (8), sec-O-glucosylhamaudol (9) and vanillin (10) were isolated from the methanol extract of Angelica polymorpha Maxim. The structures of these compounds were elucidated through a comprehensive analysis of standard spectral data (MS, IR, and NMR). Compound 1 exhibited antioxidant activity with IC50 = 198.57 μM in DPPH experiment and 31.71 μM in ABTS experiment. Compound 2, 6, 7 exhibited ABTS radical scavenging activity with IC50 ranging from 105.96 μM to 167.67 μM. Compound 3 demonstrated a synergistic induction effect on nigericin-activated NLRP3 inflammasome in THP-1 cell by LDH release method.
Rapid Separation and Identification of Furocoumarins in Angelica Dahurica by High-Performance Liquid Chromatography with Diode-Array Detection, Time-Of-Flight Mass Spectrometry and Quadrupole Ion Trap Mass Spectrometry

Abstract:High-performance liquid chromatography with diode-array detection (HPLC/DAD), time-of-flight mass spectrometry (HPLC/TOFMS) and quadrupole ion trap mass spectrometry (HPLC/QITMS) were used for separation, identification and structural analysis of furocoumarins in Angelica dahurica. Two furocoumarins (imperatorin and isoimperatorin) in Angelica dahurica extract were identified unambiguously by comparing their relative retention times, characteristic ultraviolet information and accurate mass measurement. A formula database of known furocoumarins in Angelica dahurica was established, against which the other 21 furocoumarins were identified effectively based on the accurate extract masses and formulae acquired by HPLC/TOFMS. In order to distinguish the isomers, multi-stage mass spectrometry (MSn, ion trap mass spectrometry) was used. General fragmentation behavior of the furocoumarins in the ion trap mass spectrometer was studied by the two furocoumarin standards, and their fragmentation rules in MS(n) spectra were summarized. These deduced fragmentation rules of furocoumarins were successfully implemented in distinguishing the three groups of isomers in Angelica dahurica by HPLC/QITMS. By using the three different analytical techniques, 23 furocoumarins in Angelica dahurica were tentatively identified within 30 min. Finally, HPLC/TOFMS fingerprints of Angelica dahurica were established by which it can be concluded that a rapid and effective method based on the three analytical techniques for identification of chemical components was established. This can provide help for further quality control of Angelica dahurica and pharmacology mechanism study of furocoumarins in Angelica dahurica.

Abstract:As a popular medicinal plant, Angelica dahurica has been attached to great importance in herbal medicine. Its main active compounds are furanocoumarins (FCs) including imperatorin and isoimperator which exhibit anti-oxidant, anti-tumor, and anti-virus activities. However, the enzymes catalyzing the crucial step of FCs' biosynthesis pathway remain unidentified. This study initially screened six prenyltransferases (PTs)-coding sequences through transcriptome and gene expression profile analysis. Combined with qRT-PCR and in vitro enzyme activity verification, two C-PTs ( Ad PT1, 2) which convert umbelliferone to 7-demethylsuberosin (DMS), and one O-PT ( Ad PT3) which transforms xanthotoxol to imperatorin were identified. Among these PTs, Ad PT2 also produces a trace amount of osthenol from umbelliferone. Homologous modeling, molecular docking, and site-directed mutagenesis analysis were also conducted and it is discovered that Asp301 and Asp305 in the two Asp-rich regions are vital for the activity of Ad PT1. The study is the first to conduct a frontier exploration of the biosynthesis pathways of FCs in A. dahurica and will establish a foundation for the molecular mechanism of further C-PTs and O-PTs evolutionary studies. CONCLUSION All the compounds were isolated from the plant for the first time.
