Abstract:A series of alkyl 4-dialkylaminophenyl ketones were prepared and reduced asymmetrically by borane under the chiral oxazaborolidine catalysis. The results indicate that the ketones show a more obvious substituent effect on the enantioselectivity than the corresponding 4-alkyl/alkoxy/alkylthiophenyl ketones in, the asymmetric reduction because of the existence of a strong coordinate nitrogen atom with the boron atom in the catalyst and borane.

Abstract:The method of reduction of 2,2',4'-trichloroacetophenone with sodium borohydride to prepare 1-(2,4-dichlorophenyl)-2-chloroethanol was studied. Temperature effect on the reaction was investigated and the mechanism to form objective product and byproducts was discussed. Calcium chloride and lanthainium chloride etc. efficiently improved the reaction specificity. In the presence of amberlyst-732(H+) the reaction was smoothly carried out in aprotic solvent such as THF with high yield and specificity. The structure of the objective product was characterized by IR, H-1 NMR and C-13 NMR spectra.

Abstract:The reductive alkylation reaction of aldehyde/ketone and amine is an important reaction to synthesis alkylated amine.Two main methods to achieve the synthesis were introduced,which were catalytic hydrogenation and borohydride reduction.Described the main research progress and introduced the catalysts used in the two methods.Pointed out sulfidizing of catalyst metal was effective to achieve high selectivity.Introduced the commonly catalytic system used in borohydride chemical reduction method and discussed the effects of the reaction conditions such as steric hindrance of reactant,dehydrating agent,raw material ratio and solvent.Modifying the reaction conditions such as raw material ratio and solvent could improve the selectivity of target product.

Abstract:The chemoselective catalytic reduction of nitro scaffolds to value-added amine functions is desirable but challenging. We developed a combined KOtBu/BEt3 catalyst for deoxygenative reduction of nitroarenes, nitro heteroarenes and even notoriously challenging nitroalkanes using pinacolborane under mild reaction conditions. The novel transition-metal-free strategy is readily applicable to the preparation of commercially available pharmaceuticals.

Abstract:A rapid and mild protocol for the exhaustive deoxygenation of various aromatic ketones to corresponding alkanes was described, which was mediated by TiCl4 and using ammonia borane (AB) as the reductant. This reduction protocol applies to a wide range of substrates in moderate to excellent yields at room temperature. The gram-scale reactions and syntheses of some key building blocks for SGLT2 inhibitors demonstrated the practicability of this methodology. Preliminary mechanistic studies revealed that the ketone is first converted into an alcohol, which then undergoes a carbocation to give the alkane via hydrogenolysis.

Abstract:An efficient and practical preparation of homochiral linear secondary alcohols, 1-(4-alkylphenyl) and 1-(4-alkoxylphenyl) alcohols, via the asymmetric oxazaborolidine-catalyzed borane reduction of prochiral ketones is described. The phenomenon of the enantioselectivity of 1-(4-alkoxylphenyl) alcohols lower than that of 1-(4-alkylphenyl) alcohols was found and rationalized to the coordination of the oxygen atom in the alkoxy groups to the catalyst and borane. Based on the rationale, the enantioselectivity of 1-(4-alkoxylphenyl) alcohols in the asymmetric reduction was improved with increasing the amount of the catalyst.

Abstract:The effect of borane source on enantioselectivity in the enantiopure oxazaborolidine-catalyzed asymmetric borane reduction of ketones has been investigated by using (S)-3,1,2-oxazaborobicyclo[3.3.0]octane and (S)-7,3,1,2-thiaxazaborobicyclo[3.3.0]octane as catalysts. The results indicate that the enantioselective order of different borane sources is borane-dimethyl sulfide < borane-N,N-diethylaniline < borane-THF for the asymmetric reduction of a ketone under the same conditions. Abstract:Chemoselective reduction of aromatic nitro compounds ( ortho -nitrophenol derivatives) using Borane in THF (BH 3 -THF) at room temperature, without using an additional metal catalyst is reported. Interestingly, nitroarenes containing two or three groups, a selective reduction of the nitro group adjacent to the hydroxy group is observed which happens to be the first Chemoselective reduction protocol for nitro groups using BH 3 -THF. The presence of the phenolic hydroxyl group is found to be the key that aids in hydrogen transfer to the nitro group through a cyclic transition step.

Abstract:An unusual reduction of some aromatic aldehydes and ketones by sodium borohydride was discovered. In a THF/H2O or DMSO/H2O solvent system the aromatic aldehydes and ketones with acyloxy substituent on the ortho-position to the carbonyl group can be reduced to the corresponding alkyl phenols. This unusual reduction is: applicable also to the corresponding alcohols of all the above aldehydes and ketones. A putative mechanism was suggested. Tn addition to the above aldehydes, ketones and benzyl alcohols, certain 4-acyloxybenzyl esters(probably also the 2-substituted analogues) such as 4-benzoyloxybenzyl benzoate was, also found to be reduced to methylphenol by this unusual reduction.

Abstract:The enantioselective 1,2-reduction of α,β-unsaturated ketones was achieved using a NiH catalyst in the presence of pinacolborane. This mild process represents a general method to access a wide variety of structurally diverse α-chiral allylic alcohols in excellent yields and enantioselectivity, as well as very high levels of ambidoselectivity for 1,2- over 1,4-reduction. Furthermore, for reactions on a 10 mmol scale, catalyst loadings as low as 0.5 mol % could be employed to deliver product without any detrimental effect on the yield, enantio-, or ambidoselectivity.

Abstract:An operationally convenient Zn-catalyzed synthesis of alcohols by the reduction of aldehydes, ketones, and α,β-unsaturated aldehydes/ketones is reported. It is a rare example of using mild and sustainable HBpin as a reductant for catalytic reduction of carbonyl compounds in the absence of acid or base as hydrolysis reagent. The reaction is upscalable and proceeds in high selectivity without the formation of boronate ester by-products, and tolerates sensitive functionalities, such as iodo, bromo, chloro, fluoro, nitro, trifluoromethyl, aminomethyl, alkynyl, and amide. The Zn(OAc)2/HBpin combination has been also proved to be chemoselective for the C=N reduction of imine analogs.

Abstract:The asymmetric borane reduction of prochiral ketones with a series of easily constructed chiral C-3-symmetric tripodal tris(beta-hydroxyamide) ligands was investigated. The borane complex of chiral ligand 1,1',1"-(1,3,5-benzenetricarbonyl)-tris[(2S)-alpha,alpha-diphenl-2-pyrrolidinemethanol] (1h) was found to be an efficient catalyst in asymmetric borane reduction of prochiral ketones and excellent enantioselectivities were obtained with both electron-deficient and electron-rich prochiral ketones (up to 97% ee).
