Abstract:An efficient and practical preparation of homochiral linear secondary alcohols, 1-(4-alkylphenyl) and 1-(4-alkoxylphenyl) alcohols, via the asymmetric oxazaborolidine-catalyzed borane reduction of prochiral ketones is described. The phenomenon of the enantioselectivity of 1-(4-alkoxylphenyl) alcohols lower than that of 1-(4-alkylphenyl) alcohols was found and rationalized to the coordination of the oxygen atom in the alkoxy groups to the catalyst and borane. Based on the rationale, the enantioselectivity of 1-(4-alkoxylphenyl) alcohols in the asymmetric reduction was improved with increasing the amount of the catalyst.

Abstract:The first catalytic enantioselective 1,2-reduction of enones with 0.45 mol equiv potassium borohydride solution catalyzed by a chiral N,N'-dioxide-Sc(III) complex catalyst was accomplished under mild reaction conditions. A number of optically active allylic alcohols were obtained in good to excellent enantioselectivities (up to 95% ee) with nearly quantitative yields.

Abstract:Highly optically pure homochiral 1-(4-alkylthiophenyl) alcohols were prepared efficiently and practically via the oxazaborolidine-catalyzed asymmetric borane reduction of prochiral ketones in toluene at 25 degrees C. The coordination of the sulfur atom in the ketones to the boron atom in the catalyst and borane can be inhibited under these reduction conditions.

Abstract:Veratryl alcohol was synthesized through the reduction of sodium borohydride from veratraldehyde with isopropanol as solvent, followed by solvent extraction and distillation under reduced pressure. Better reaction conditions are: n(sodium borohydride):n(veratraldehyde)=0.32:1, temperature 20 ℃, under which the reduction efficiency is improved and the consumption of sodium borohydride is reduced. The product is highly pure (GC>98%) with an overall yield of 85.2% after rectification. Structure of the product was confirmed by GC-MS.

Abstract:Asymmetric catalytic activity of the chiral spiroborate esters 1-9 with a O3BN framework (see Fig. 1) toward borane reduction of prochiral ketones was examined. In the presence of 0.1 equiv. of a chiral spiroborate ester, prochiral ketones were reduced by 0.6 equiv. of borane in THF to give (R)-secondary alcohols in up to 92% ee and 98% isolated yields (Scheme 7). The stereoselectivity of the reductions depends on the constituents of the chiral spiroborate ester (Table 2) and the structure of the prochiral ketones (Table 1). The configuration of the products is independent of the chirality of the diol-derived parts of the catalysts. A mechanism for the catalytic behavior of the chiral spiroborate esters (R,S)-2 and (S,S)-2 during the reduction is also suggested.

Abstract:A series of alkyl 4-dialkylaminophenyl ketones were prepared and reduced asymmetrically by borane under the chiral oxazaborolidine catalysis. The results indicate that the ketones show a more obvious substituent effect on the enantioselectivity than the corresponding 4-alkyl/alkoxy/alkylthiophenyl ketones in, the asymmetric reduction because of the existence of a strong coordinate nitrogen atom with the boron atom in the catalyst and borane.

Abstract:The asymmetric borane reduction of prochiral ketones with a series of easily constructed chiral C-3-symmetric tripodal tris(beta-hydroxyamide) ligands was investigated. The borane complex of chiral ligand 1,1',1"-(1,3,5-benzenetricarbonyl)-tris[(2S)-alpha,alpha-diphenl-2-pyrrolidinemethanol] (1h) was found to be an efficient catalyst in asymmetric borane reduction of prochiral ketones and excellent enantioselectivities were obtained with both electron-deficient and electron-rich prochiral ketones (up to 97% ee).

Abstract:The secondary reduction in the direct and oxazaborolidine-catalyzed asymmetric borane reduction of ketones was investigated by the use of GC/MS tracing titration and control experiments. The results indicate that the secondary reduction affects the enantioselectivity only in noncoordinated solvents at low temperature and not under the usual catalytic reduction conditions because the intermediate alkoxyborane is unstable and quickly converts to borane and dialkoxyborane. The function of an alcohol additive in the asymmetric borane reduction of ketones is to consume excess borane in the reduction system thus inhibiting noncatalytic reduction, which leads to increased enantioselectivity in the catalytic reduction.

Abstract:Fluorinated motifs are frequently encountered in drugs and agrochemicals. Incorporating fluorine-containing motifs in drug candidates for lead optimization in pharmaceutical research and development has emerged as a powerful tool. The construction of molecules that feature a trifluoromethyl (CF3−) group on a stereogenic carbon has accumulated broad research efforts. Unlike its well-explored, biologically active methyl counterpart, asymmetric construction of β-trifluoromethylated alcohols bearing adjacent stereocenters still remains elusive. Through retrosynthetic analysis, we posited that followed by sequential reduction of carbonyl, the initial construction of chiral α-trifluoromethylated ketones could render the desired product in a facile, one-pot fashion. Herein, we developed the first example of nickel-catalyzed asymmtric reductive cross-coupling trifluoroalkylation of acyl chlorides for enantioselective synthesis of diverse α-trifluoromethylated ketones. The one-pot reduction of these α-trifluoromethylated ketones furnished corresponding alcohols bearing β-CF3-substituted stereogenic carbons with excellent diastereoselectivity and complete enantioselective retention. High yields/enantioselectivity, mild conditions, and good functional group compatibility are shown in the system. Abstract:Chiral tertiary alcohols are an important structural motif, however, the general and efficient methodologies for their synthesis are less reported. Herein, we report a Ni(ІІ)-catalyzed asymmetric alkenylation and arylation of aryl ketones with organoborons under air via a 1,5-metalate shift strategy to obtain chiral tertiary allylic alcohols and diaryl alcohols. The reaction demonstrates good functional group tolerance and delivers chiral tertiary alcohols with good to excellent results. Furthermore, this method can be applied to the late-stage modification of drugs and the efficient synthesis of natural products. Notably, the reaction proceeds through an outer-sphere mechanism. The Ni(II) complex functions both as a Lewis acid to activate the ketone and create a chiral environment, and as coordination bridge linking the ketone and the organoboron-derived "ate" complex, facilitating the 1,5-metalate shift without forming a C-Ni bond. This approach contrasts with traditional transition metal-catalyzed nucleophilic addition reactions that involve carbon-metal bond formation.

Abstract:Optically active aryl alcohols are a kind of important chiral building blocks for the synthesis of chiral drugs. In recent years, the preparations of enantiopure aryl alcohols have become one of the focus points in organic chemistry. Compared with conventional chemical processes, biocatalysis is more attractive due to its high enantioselectivity, mild and safe reaction conditions and less environmental hazards. Asymmetric bioreduction of aryl ketones is the most effective method for the synthesis of enantiopure aryl alcohols. This paper mainly reviews the recent progress in the preparation of chiral aryl alcohols by asymmetric bioreduction catalyzed by microbial whole-cells, enzymes, genetically engineered bacteria or yeasts, as well as immobilized cells. The effects of substrate co-solvents such as organic solvents, surfactants and ionic liquids on asymmetric bioreduction are further summarized. The prospect of the preparation of enantiopure aryl alcohols by asymmetric bioreduction is also discussed.
