Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:Phosphachroman-2,4-dione and 4-alkoxyphosphacoumarin derivatives, phosphonic analogues of chromones and coumarins with potential biological activities, were synthesized in good yields through sequential base-catalyzed intramolecular cyclization of O-alkyl-O′-(2′-methoxycarbonyl phenyl)-(substituted) benzyl phosphonates and alkylation. The synthesis sets the stage for an examination of the biological activities.

Abstract:A highly efficient and convenient procedure to access a series of coumarin-containing phthalides and derivatives is developed. The novel bis-heterocyclic products are obtained via reactions between 4-hydroxycoumarins (or 4-hydroxy-2-quinolones) with different 2-formylbenzoic acids in water as an environmentally friendly solvent. 4-Aryl substituted derivatives of 3-(4-hydroxy)coumarinylphthalide are obtained in excellent yields by initial conversion into the corresponding triflate and subsequent palladium-catalyzed Suzuki–Miyaura coupling with arylboronic acids.

Abstract:The cyclization of various nucleophiles, such as 1,3-dicarbonyl compounds, 1,3-bis(silyloxy)-1,3-butadienes, hydrazones, hydroxylamine, amidines, thioglycolic esters, heterocyclic enamines and others, with 4-chlorocoumarins, containing an electron-withdrawing group at position 3, allows for a convenient synthesis of a great variety of benzo[c]coumarins and related fused coumarins. Suzuki–Miyaura reactions of coumarin-derived bis(triflates) result in formation of arylated coumarins with excellent regioselectivity that is controlled by electronic and steric features of the substrate. The combination of Suzuki–Miyaura with lactonization reactions of carba- and heterocyclic substrates allows for the synthesis of benzo[c]coumarins and of other fused coumarins. Domino Michael–retro-Michael–lactonization reactions of 1,3-bis(silyloxy)-1,3-butadienes with chromones provide a convenient access to hydroxylated benzo[c]coumarins. Abstract:A concise and efficient cross-coupling synthetic strategy has been developed to construct 2-aryl-4H-thiochromen-4-one derivatives from 2-sulfinyl-thiochromones and arylboronic acids. This reaction proceeds via a catalyst system of Lewis acid and palladium(II) combined with XPhos as an optimal ligand in moderate to good yields. Besides, this flexible methodology provides a wide scope for the synthesis of different functionally substituted thiochromone scaffolds and can be further exploited to construct diverse thioflavone libraries for pharmaceutical research.

Abstract:An efficient approach was developed for synthesis of 3-tosyloxy-4-hydroxycoumarins under mild conditions by using Koser's reagents. The reaction tolerated various functional groups, and the products served as useful aromatic building blocks. Additionally, a plausible mechanism via iodonium ylide was proposed, and the oral anticoagulant Warfarin was synthesized in good yield.

Abstract:An expedient and practical route for realizing a novel, substrate-controlled diastereoselective switch strategy has been successfully developed. The [4+1] annulation of thioamides with sulfur ylides proceeded smoothly via a Michael addition triggered sequential process, and trans-dihydrothieno[3,4-c]coumarins were obtained because of steric hindrance effects between the benzoyl group and the unit of 2H-chromen-2-one as shown in the Newman projection. Nucleophilic substitution was observed as the key step when the sulfur ylides were replaced with acetophenones in the [4+1] annulation, and a double pi,pi-stacking donor-acceptor interaction stabilized the transition state that led to the cis-dihydrothieno[3,4-c]coumarins. In addition, a facile chemoselective reaction was also investigated, providing a series of thieno[3,4-c]coumarins.

Abstract:Abstract A series of 2-(hetero)aryl-4-phosphorylated quinazolines was successfully synthesized in moderate to excellent yields via a palladium-catalyzed C–O/P–H cross-coupling reaction of 4-(tosyloxy)quinazolines with H-phosphonates and phosphine oxides. This efficient and green methodology provides an alternative straightforward protocol for the introduction of phosphorus groups to quinazoline compounds at the C4 position via C–O activation.

Abstract:Decarboxylative C(sp2)-heteroatom cross-coupling reactions hold extraordinary potential for the sustainable preparation of biologically active scaffolds. Herein, we report a copper sulfate/1,10-phenathroline catalytic system for the decarboxylative intramolecular C(sp2)-O, C(sp2)-S, and C(sp2)-N coupling reactions leading to the construction of a series of benzo[b]furans, benzo[b]thiophenes, and indole derivatives from the corresponding coumarins, thiocoumarins, or quinolones, respectively. Our mechanistic study based on benzo[b]furan formation suggests a three-step process of the transformations, which consists of (i) base-mediated hydrolytic ring opening of coumarin, (ii) copper-oxygen co-initiated radical decarboxylation, and (iii) copper-catalyzed C-heteroatom cross coupling. Application of this method in the total synthesis of egonol, a bioactive natural product, was demonstrated successfully, with an overall yield of 51.7%.
