Expression of survivin and vascular endothelial growth factor and their relationship in bladder cancer and ASODN targeting survivin inducing apoptosis of bladder cancer cell line in vitro

Abstract:OBJECTIVE:To investigate the correlation between the expression of survivin and VEGF in bladder cancer and the effect of ASODN targeting survivin on apoptosis and proliferation of bladder cancer cells. METHODS:The expressions of survivin and VEGF were detected by immunohischemistry and Western-blot in 41 specimens of bladder cancer tissues and non-cancerous adjacent bladder tissues. After being transfected with ASODN, the cell morphological changes were observed under a microscope. Survivin protein expression was detected by the immunohistochemical method and flow cytometry.AI,PI and IR were also examined.RESULTS:In 41 cases, 29(67.4%) cases of bladder cancer expressed survivin, and 25(61.0%) cases were positive or slight positive for VEGF. Of there was no survivin expression in non-cancerous adjacent bladder tissues.The expression of survivin in bladder cancer had association with metastasis (P=0.002).Survivin in bladder cancer had association with VEGF (P=0.001). The morphological changes of cells were observed in transfected groups.Survivin in ASODN groups significantly decreased.AI in ASODN groups was significantly higher (P=0.012). There was no significance in PI and AI rats among the different control groups (P>0.05).CONCLUSION:Survivin may play an important role in formation and metastasis in bladder cancer.It probably promotes growth and metastasis of bladder cancer by up-regulating the expression of VEGF.Survivin may decreased in T24 cells after ASODN transfection. ASODN can induce T24 cells apoptosis and inhibit the proliferation.
