THE COMPARATIVE STUDY ON THE EFFECT OF DIFFERENT DIETARY FAT ON LIPID METABOLISM IN HYPERLIPIDEMIC RATS

Abstract:Objective To investigate the effects and mechanism of different dietary fat on lipid metabolism in hyperlipidemic rats. Method Twenty eight male OLETF rats were randomly divided into 4 groups and fed with modified AIN76 diets which included 8% corn oil.Each diet was added with 2% palm oil,2% borage seed oil,2% perilla oil or 2% fish oil,respectively.After 4 w,serum triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),nonesterified fatty acid(NEFA),glucose levels,and hepatic lipid concentrations(TG,TC and phospholipids) were measured.To determine the mechanism of different dietary fat on lipid metabolism in hyperlipidemic rats,the mRNA expression of key enzymes related to lipids metabolism,such as fatty acid synthase(FAS),malic enzyme(ME),glucose-6-phosphate dehydrogenase(G6PDH) and carnitine palmitoyl transferase(CPT) were detected.Results The serum NEFA and TG were remarkably decreased in borage seed oil group compared with the palm oil group(P0.05).The serum NEFA and TG levels were dramatically decreased by perilla oil feeding(P0.05).The concentrations of serum NEFA,TC,TG,HDL-C and glucose(P0.05) were significantly decreased in fish oil group.Moreover,the hepatic TG and TC concentrations were significantly decreased(P0.05) in borage seed oil group and perilla oil group.The concentration of serum TC was similar between fish oil group and control group,but TG level was remarkably decreased in fish oil group.On the other hand,the mRNA expression of G6PDH,ME,FAS genes were significantly decreased,while the mRNA expression of CPT-1 and CPT-2 genes increased remarkably in borage seed oil group,perilla oil group and fish oil group.Conclusion Borage seed oil,perilla oil and fish oil attenuated the serum and hepatic lipids accumulation,mainly via reduction of hepatic fatty acid synthesis and up-regulation of fatty acid catabolism.
