Immobilization of Enzymes on Polymers Modified by Ultrasonic Irradiation

Abstract:Polystyrene (PS) was copolymerized with methyl methacrylate (MMA) and/or glycidyl methacrylate (GMA) through ultrasonic irradiation. Block copolymers PS-b-PMMA, PS-b-PGMA and PS-b-P(MMA-co-GMA) were obtained; their compositions were determined by FTIR and 1H-NMR. The variation of MMA content in PS-b-PMMA with irradiation time was examined. Copolymers PS-b-PGMA and PS-b-P(MMA-co-GMA) were used as supports for immobilizing trypsin and catalase. It was found that PS-b-P(MMA-co-GMA) had a lower capacity for enzyme binding because of the presence of MMA units. The trypsin bound on PS-b-P(MMA-co-GMA) exhibited an activity about 40% of that of free trypsin; if it was bound on PS-b-PGMA, it lost all of its activity. Catalase on both copolymers retained roughly 25% of its activity.
