New supports for enzyme immobilization based on the copolymers of poly(vinylene carbonate) and α-(2-aminoethylene amino)-ω-(2-aminoethylene amino)–poly(ethylene oxide)

Abstract:Poly(ethylene oxide) end-capped with amine groups (aa-PEO) was prepared in this study. It then was dissolved with poly(vinylene carbonate) (PVCA) in DMF. The solution was suspended in paraffin oil with stirring and consequently the beaded hydrophilic supports containing reactive cyclic carbonate groups for enzyme immobilization were produced. In the crosslinking reaction above, aa-PEO was served as both a crosslinking agent and hydrophilic modifier for PVCA. Evaluation of the properties of the supports by attachment of trypsin shows that specific activity of immobilized trypsin and amount of enzymes coupled to supports were related to the proportion of aa-PEO and PVCA, reaction time, enzyme concentration and pH value of enzyme solution, etc. The amount of trypsin coupled and specific activity reached their maximum at PVCA/aa-PEO=1/2. The optimal pH for trypsin immobilized increased compared with the optimal pH for native trypsin. Km for both immobilized and native trypsin was 32.5 and 15.2 mmol/l, respectively.
