The neural substrate during early stage processing of emotional picture stimuli in major depression:a low frequency magnetoencephalography study

Abstract:Objective: To explore the neural substrate of major depression during early stage (0 ~ 100 ms,100 ~ 200 ms,200 ~ 300 ms) processing of emotional picture stimuli using low frequency Magnetoencephalography(MEG) . Method: Eight depressive patients and twelve right-handed healthy individuals were silently viewing positive,neutral,negative emotional pictures from International Affective Picture System(IAPS) . Data was collected using MEG and analyzed using SPM8b software. Two sample t-test was used to find the difference of brain activation,voxels with a P value < 0. 01(uncorrected) and cluster size≥10 voxels were determined significant difference. Results: Compared to controls,during positive picture presenting,depressive patients showed increased activity in left inferior frontal gyrus,right subcallosal gyrus,right medial frontal gyrus,right parahippocampal gyrus in 100 ~ 200 ms. While neutral picture presenting,depressive patients exhibited increased brain activity in right lentiform nucleus,right insula,right superior temporal gyrus,left medial frontal gyrus in 0 ~ 100 ms; increased activation in right insual,right lentiform nucleus,right claustrum,left inferior and superior frontal gyrus in 100 ~ 200 ms; increased activation in right insula,right lentiform nucleus,right claustrum in 200 ~ 300 ms. During negative picture presenting,depressive subjects showed increased activity in right superior temporal gyrus,right insula,right caudate head,right middle and inferior frontal gyrus in 0 ~ 100 ms; increased activity in right middle temporal gyrus,right caudate body in 100 ~ 200 ms,increased activity in right inferior frontal gyrus in 200 ~ 300 ms. Also we find coincident activity in the parietal cortex such as precuneus, posterior cingulate gyrus. Conclusion: Parietal cortex such as precuneus which has function to modulate attention is impaired in depressive patients,and it can't suppress the pathway from visual cortex to frontal cortex during emotional processing,more emotional information arrived at ventral frontal cortex,insula and so on,this hypothesis maybe pathophysiology of major depressive disorder.
