Abstract:One new monacolin analog, monacolin V (1), together with two new monacolin‐like natural products, 6‐hydroxyl monacolin P (2) and 3‐keto monacolin S (3), were isolated from the ethyl acetate portion of red yeast rice ethanol extract. The complete assignments of 1H and 13C NMR data for three compounds were obtained by the aid of HSQC, HMBC, 1H‐1H COSY, and NOESY data. One new monacolin analog, monacolin V (1), together with two new monacolin‐like natural products, 6‐hydroxyl monacolin P (2) and 3‐keto monacolin S (3), were isolated from the ethyl acetate portion of red yeast rice ethanol extract. Their structures were identified by HRESIMS and NMR experiments, and the complete assignments of 1H and 13C NMR data for three compounds were obtained by the aid of HSQC, HMBC, 1H‐1H COSY, and NOESY data. This is the first time that the NMR data of compounds 2 and 3 have been fully assigned.

Abstract:Xuezhikang capsule (ethanol extract of red yeast rice) which produced by Beijing WBL Peking University Biotech Co., Ltd., is a traditional Chinese medication with 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase inhibiting activity. Clinical trials indicated that Xuezhikang with lipid-lowering action could reduce the risk of cardiac events and total mortality of Chinese coronary heart disease patients. To exactly explain the clinical features of Xuezhikang, we undertook a complete study of the chemical constituents of Xuezhikang. This study resulted in the isolation of a new monacolin analogue, named alpha, beta-dehydromonacolin L (1), along with two known compounds: monacolin L (2) and 3-(2, 6-dimethyl-1, 2, 4a, 5, 6, 7, 8, 8a-octahydronaphthalen-1-yl)propanoic acid (3). The chemical structures were determined by extensive 1D and 2D NMR and MS spectroscopic analysis.

Abstract:Detailed analyses were undertaken of the natural constituents of red yeast rice, a traditional Chinese medicine and food known for centuries to improve blood circulation. Preparation of red yeast rice following ancient methods by fermenting the fungal strain Monascus purpureus Went on moist and sterile rice indicated the presence of a group of metabolites belonging to the monacolin family of polyketides, together with fatty acids, and trace elements. The presence of these compounds may explain in part the cholesterol-lowering ability associated with this traditional Chinese food.

Abstract:Further study on a 95% EtOH extract of the red mold rice fermented with the mutant of the fungus Monascus sp. led to the isolation of one new natural pyranoid derivative, namely monascuslactone ( 1 ), together with three known compounds. The structure of the new compound was determined as (4 S * , 6 S *)-4-hydroxy-4,6-dimethyltetrahydro-2 H -pyran-2-one. The known compounds were identified as monascin ( 2 ), ankaflavin ( 3 ), and methyl N -methyl anthranylate ( 4 ). Their structures were elucidated by 1D and 2D NMR spectroscopy together with HR-ESI-MS analysis, and comparison of the spectroscopic data with those reported for structurally related compounds.

Abstract:Seven compounds were isolated from Monascus-fermented rice by column chromatography with silica gel and semiprep HPLC. Their structures were elucidated by extensive spectroscopic methods. All compounds displayed HMG-CoA reductase inhibitory potential, among them compound 7 exhibited strong inhibition with IC50 value comparable with lovastatin. In this study, two compounds (1 and 2) were obtained from natural source for the first time.

Abstract:A novel decalin derivative, trans-1-oxo-2,4-diacetylaminodecalin (1) with anti- Candida activity, had been isolated from Streptomyces chrestomyceticus strain ADP4. The structure of the compound was determined from the analysis of spectral data (LCMS/MS, UV, FTIR, 1D- and 2D-NMR). The anti- Candida activity of 1 was specific to Candida albicans and Candida auris . Further, it displayed inhibition of the early-stage biofilm of C. albicans . In-silico analysis of the compound revealed its drug likeness properties without any violations and PAINS alert when investigated for ADME properties Along with the overall bioavailability, compound 1 did not show any predicted bioaccumulation and mutagenicity in the analysis by TEST software. Non-cytotoxic property was further confirmed by in-vitro assay on the HepG2 cell line.

Abstract:Red yeast rice is consumed as a medicinal food to lower blood lipids. Besides, it is used to color food, make wine, etc. In this study, water-soluble oligosaccharides in red yeast rice were extracted by ultrasonic-assisted extraction method. The parameters to extract oligosaccharides from red yeast rice were optimized by the Box-Behnken design under the following optimal extraction conditions: extraction temperature, 60°C; extraction time, 97 min; and liquid/material ratio, 25 ml/g. The structure and the antioxidant activity of the new oligosaccharide were preliminarily investigated. Total carbohydrates extracted from red yeast rice with 80% ethanol-water solution (v/v) were first removed from pigments using D101 macroporous adsorption resin. The total sugar contents were then purified by DE52 resins and Sephadex G-25 resins to obtain red yeast rice oligosaccharides, coded as RYRO1. Structural characterization experiments indicated that RYRO1 is an oligosaccharide with a weight average molecular weight of 874 Da and a theoretical degree of polymerization of 4.86. RYRO1 is composed of mannose, glucosamine, glucose, and galactose with a molar ratio of 0.248:0.019:1:0.026. The ABTS, DPPH, and hydroxyl free radical scavenging assays showed antioxidant nature of RYRO1.

Abstract:Covering: up to December 2023 Decalin-containing tetramic acid derivatives, especially 3-decalinoyltetramic acids (3-DTAs), are commonly found as fungal secondary metabolites. Numerous biological activities of this class of compounds, such as antibiotic, antiviral, antifungal, antiplasmodial, and antiprotozoal properties, have been the subject of ongoing research. For this reason, these molecules have attracted a lot of interest from the scientific community and various efforts including semi-synthesis, co-culturing with bacteria and biosynthetic gene sequencing have been made to obtain more derivatives. In this review, 3-DTAs are classified into four major groups based on the absolute configuration of the bicyclic decalin ring. Their biosynthetic pathways, various biological activities, and structure–activity relationship are then introduced.

Abstract:Red yeast rice (RYRP) has been utilized for coloring food, brewing wine, and preserving meat, which is also used as a folk medicine for centuries. In this study, a water‐soluble nonstarch polysaccharide from RYRP was extracted by using ultrasonic‐assisted extraction method. By using the Box–Behnken design to optimize the parameters for extracting the RYRP, the maximum extraction yield (3.37 ± 0.78%) was obtained under the optimal extraction conditions as follows: ratio of water to raw material (40 mL/g), extraction temperature (62 °C), extraction time (75 Min), and ultrasonic power (200 W). Moreover, monosaccharide composition analysis showed that RYRP was consisted of mannose, glucosamine, glucose, and galactose with a molar ratio of 0.152:0.015:1:0.149. The molecular weight distribution analysis showed that the average molecular weight of the RYRP fraction was about 3.49 × 103 Da. Furthermore, RYRP exhibited significant antioxidant activities in vitro and the gastrointestinal‐protective effect in vivo using gastrointestinal disorders model mice. RYRP could be explored as a potential source in the pharmaceutical and functional food industries.

Abstract:Nitricquinomycins A-C (1 similar to 3), three uncommon naphthopyrrolediones, together with one new compound (4), were isolated from the modified rice solid fermentation of the marine-derived Streptomyces sp. ZS-A45, an actinobacterium isolated from the marine sediments collected from Zhoushan island. Their structures were elucidated on the basis of MS, NMR spectroscopic and quantum chemical calculations of the electronic circular dichroism (ECD) spectra. Compounds 3 and 4 exhibited significant cytotoxicity against A2780 cell lines with IC50 values of 4.77 and 2.92 mu M, respectively; on the other hand, compound 3 exhibited moderate antibacterial activities against Escherichia coli, Staphylococcus aureus and Candida albicans with MIC values of 20.0 40.0 and 40.0 mu M, respectively. Abstract:Twelve compounds were isolated from Liquidambaris Resina by silica gel column chromatography and thin layer chromatography. Their structures were identified on the basis of spectral data, electron capture detector data, and physicochemical properties as(2'R, 3'R)-2',3'-dihydroxy-hydrocinnamyl-(E)-cinnamate(1),(E)-cinnamyl-(E)-cinnamate(2), cinnamic acid(3), 28-norlup-20(29)-en-3-one-17β-hydroperoxide(4), erythrodiol(5), 13β,28-epoxy-30-hydroxyolean-1-en-3-one(6),(3β)-olean-12-ene-3,23-diol(7), 2α,3α-dihydroxy-olean-12-en-28-oic acid(8), 28-hydroxyolean-12-en-3-one(9), 3-epi-oleanolic acid(10), 3-oxo-oleanolic acid(11), and hederagenin(12). Compound 1 was a new cinnamic acid ester derivative and compounds 2-4,6-8, and 12 were isolated from Liquidambaris Resina for the first time. Compounds 4, 5, 10, and 12 exerted inhibitory effects on the proliferation of human umbilical vein endothelial cells(HUVEC) with the IC_(50) values of(17.43±2.17),(35.32±0.61),(27.50±0.80), and(46.30±0.30) μmol·L~(-1), respectively.

Abstract:A new flavonoid glycoside, diosmetin 7-O-beta-D-xylopyranosyl(1-6)-beta-D-glucopyranoside, named raddeanalin (1), was isolated from the ethanolic extract of the leaves of Salix raddeana Laksh. together with three known compounds, kaempferol 3-O-glucoside (2), quercetin 3-O-glucoside (3) and quercetin 3-O-rutinoside (4). The structure of new compound was established by the spectral data and chemical properties.

Abstract:A new sesquiterpenoid, 1β,5α-guaiane-4β,10α-diol-6-one (1), was isolated from 70% EtOH extract of the rhizomes of Acorus calamus. The structure was determined on spectroscopic methods, especially 2D NMR techniques. The absolute configuration of 1 was confirmed by TDDFT quantum chemical calculation of its ECD spectrum. Compound 1 showed promising anti-diabetic activity on a insulin-mediated glucose consumption model of HepG2 cells.

Abstract:Red yeast rice (RYR) is produced by the fermentation of rice ( Oryza sativa ) using Monascus sp., and has a long history as a food colorant and a folk medicine in China. RYR is well-known for lowering blood cholesterol levels due to its bioactive component, monacolin K (lovastatin). In addition to antihypercholesterolemic effect, RYR showed evidence to be effective for the control of diabetes, hypertension, obesity, as well as for the prevention of Alzheimer's disease and osteoporosis. This review is focused on the study of the bioactive and pharmacological effects of RYR.

Abstract:A new dicoumarin, named as dimeresculetin (1), together with another dicoumarin, euphorbetin (2) and esculetin (3) were isolated from the ethyl acetate extract of the dried whole plants of Viola yedoensis Makino. The structure of 1 was elucidated as 7-hydroxy-6-[(6,7-dihydroxy-2-oxo-2H-1-benzopyran-5-yl)oxy]-2H-1-benzopyran-2-one on the basis of extensive NMR, as well as the other spectral analysis. Compounds 1-3 exhibited anticoagulant activities with respect to activated partial thromboplastin time (APTT), prothrombin time (PT) and thrombin time (TT).

Abstract:A new cladosporol derivative xylophilum A ( 1 ), together with 10 known compounds ( 2 – 11 ), were isolated from the rice fermentation of the fungus Cladosporium xylophilum . Their structures were established by extensive spectroscopic methods and comparison of their NMR data with literatures. The antimicrobial activity of compound 1 against 11 kinds of pathogenic microbial was evaluated, but no significant activity was found (MIC >100 μg/ml).

Abstract:An improved synthesis of (2S, 4S)- and (2S, 4R)-2-amino-4-methyldecanoic acids was accomplished using a glutamate derivative as starting material and Evans' asymmetric alkylation as the decisive step. The NMR data of the two diastereomers were measured and compared with those of the natural product.
