Abstract:Chemical investigation of a co-culture of two endophytic fungi Aspergillus fumigatus D and Fusarium oxysporum R1 from two traditional medicinal plants, Edgeworthia chrysantha Lindl. and Rumex madaio Makino, led to isolation of a new amide 1 and six known compounds, including neovasinin (2), neovasifuranone B (3), N-(2-phenylethyl)acetamide (4), α-linolenic acid (5), α-elaeostearic acid (6), palmitoleic acid (7). On the basis of extensive spectroscopic analysis including 1D and 2D NMR, HR-ESI-MS and optical rotation measurement as well as comparison of literature data, chemical structure of 1 was unambiguous elucidated as (S, E)-methyl-2-(2,4-dimethylhex-2-enamido)acetate. Bioassay results indicated that none of these compounds exhibited strong inhibitory effect on three human pathogens Escherichia coli, Staphyloccocus aureus and Candida albicans with MIC values of ≥25 μM.
Identification and characterization of a novel decalin derivative with anti-Candida activity from Streptomyces chrestomyceticus strain ADP4

Abstract:A novel decalin derivative, trans-1-oxo-2,4-diacetylaminodecalin (1) with anti- Candida activity, had been isolated from Streptomyces chrestomyceticus strain ADP4. The structure of the compound was determined from the analysis of spectral data (LCMS/MS, UV, FTIR, 1D- and 2D-NMR). The anti- Candida activity of 1 was specific to Candida albicans and Candida auris . Further, it displayed inhibition of the early-stage biofilm of C. albicans . In-silico analysis of the compound revealed its drug likeness properties without any violations and PAINS alert when investigated for ADME properties Along with the overall bioavailability, compound 1 did not show any predicted bioaccumulation and mutagenicity in the analysis by TEST software. Non-cytotoxic property was further confirmed by in-vitro assay on the HepG2 cell line.

Abstract:Marine derived rare actinomycetes is emerging as one of the new sources for various natural products for further drug discovery. Dimeric indole alkaloids represent a group of structurally diverse natural products and N-N linkage is a special dimerization mode. Here, we report the isolation of 1,1'-([1,1'-biindole]-3,3'-diyl) bis (ethane-1,2-diol), a new tryptophan-derived indole alkaloid from the marine sponge-derived Kocuria sp. S42. The structure was established based on extensive spectroscopic analyses, including nuclear magnetic resonance (NMR) and high-resolution electrospray ionization mass (HR-ESI-MS) spectrometry. Abstract:Four new bisamides 1-4, and two new rhamnosides (5, 6), along with four known compounds 7-10, were isolated from a scale culture of the mangrove-derived actinomycete Streptomyces sp. SZ-A15. All structures were determined through analysis of the UV, IR, HRESIMS, 1D and 2D NMR spectra as well as by comparison with literature data. BRD4 inhibition of all isolated compounds was evaluated. As for the ability to inhibit protein BRD4, compound 9 exhibited moderate activity with the value of 78.4 +/- 2.2% at 10 mu M. [GRAPHICS] .

Abstract:An endophytic fungus was isolated from the root of the medicinal plant Moringa oleifera Lam. Based on analyzing the rDNA sequence, the fungus was identified as Nigrospora sp. This is the first report of the isolation of endophytic Nigrospora from M. oleifera . By bioassay-guided fractionation, four antifungal secondary metabolites were isolated from liquid cultures of the fungus Nigrospora sp. LLGLM003, and their chemical structures were determined to be griseofulvin ( 1 ), dechlorogriseofulvin ( 2 ), 8-dihydroramulosin ( 3 ) and mellein ( 4 ) on the basis of spectroscopic analyses. Compound 2 , 3 and 4 were isolated from Nigrospora sp. for the first time. In vitro antifungal assay showed that griseofulvin displayed clear inhibition of the growth of 8 plant pathogenic fungi. Dechlorogriseofulvin and mellein exhibited only weak antifungal activities, whereas 8-dihydroramulosin displayed no antifungal activities.

Abstract:A new depsidone derivative botryorhodine I (1), along with eight known compounds (2-9) were obtained from solid rice cultures of the fungal strain, Lasiodiplodia theobromae M4.2-2 isolated from a mangrove sediment sample. The structures of the isolated compounds were elucidated on the basis of 1 D and 2 D NMR analysis as well as by HRESIMS. All compounds were evaluated for their cytotoxic potential against the mouse lymphoma cell line L5178Y as well as for their antibacterial activities against a panel of Gram-positive and Gram-negative bacterial strains. Compound 3 revealed potent cytotoxic activity with an IC50 of 7.3 µM whereas compound 7 showed selective anti-bacterial activity against different S. aureus and E. faecium bacterial strains with MIC value of 25 µg/ml.

Abstract:Four new polyketide decalin derivatives, penicisteck acids A–D (1–4), and three new nitrogenous compounds (7–9), together with eight previously reported compounds, were isolated from the mangrove endophytic fungus Penicillium steckii SCSIO 41025. Their structures and absolute configurations were characterized by analysis of comprehensive spectroscopic data combined with ECD calculations and modified Marfey's method. Structurally, compounds 1–3 were highly oxygenated decalin derivatives harboring an unusual propanoic acid unit at C‐1. Compound 8 was identified as a rare natural alkaloid‐N‐oxide confirmed by single crystal X‐ray diffraction analysis. A plausible biogenetic pathway of compounds 1–6 is proposed. Compounds 13 and 15 showed significant anti‐bacterial activity against Staphylococcus aureus with the MIC values of 3.9 and 7.8 μg/mL, respectively. Compounds 10 and 12 exhibited potent α‐glucosidase inhibitory activity with the IC50 values of 64.1 and 191.4 μM, respectively.This article is protected by copyright. All rights reserved.

Abstract:A new alkaloid, 1,2-dihydrophenopyrrozin (1), along with five known compounds (2-6) was isolated from an axenic culture of the endophytic fungus, Bionectria sp., obtained from seeds of the tropical plant Raphia taedigera. Co-cultivation of this fungus either with Bacillus subtilis or with Streptomyces lividans resulted in the production of two new o-aminobenzoic acid derivatives, bionectriamines A and B (7 and 8) as well as of two additional known compounds (9 and 10). None of the latter compounds (7-10) were detected in axenic cultures of the fungus or of the bacteria indicating activation of silent biogenetic gene clusters through co-cultivation with bacteria. The structures of the new compounds were unambiguously determined based on detailed NMR and MS spectroscopic analysis and by comparison with the literature. The crystal structure of agathic acid (6) is reported here for the first time. Penicolinate A (4) exhibited potent cytotoxic activity against the human ovarian cancer cell line A2780 with an IC50 value of 4.1μM.
Nmr Structural Elucidation of 3,4-Seco-olean-11,13-dien-4,15 Alpha,22 Beta,24-Tetraol-3-Oic Acid with Modulation of Water Peak

Abstract:The structure of a new compound, isolated from the endophyte of the semi-mangrove plant Hibiscus tilianceus, was elucidated with nuclear magnetic resonance spectroscopy. In the primary H-1 NMR test, it was found that some signal lines were submerged under the "water peak", and this would interfere with the analysis of NMR spectra. In this work, the variational rule between the water-peak chemical shift and the water content was investigated for solvent DMSO-d(6). Using the results, the water-peak chemical shift was modulated with certain amount of extra water introduced into the practical sample tests. As a result, the interference of water peak was overcome, and a group of suitable spectra including H-1 NMR, C-13 NMR, COSY, HMQC, HMBC, etc., were obtained. The structural elucidation of the new compound was achieved, and it was confirmed that this compound belonged to a triterpenoid of oleanane, and identified as 3,4-seco-olean-11,13-dien-4,15 alpha,22 beta,24-tetraol-3-oic acid.

Abstract:Two new bicyclic sesquiterpenes,Δ 9 -2, 5, 11-trihydroxyl- β -cis-bergamotene ( 3 ) and Nigrohydroin A ( 4 ), together with ten known compounds ( 1, 2 and 5 - 12 ) were obtained from endophytic fungus Nigrospora sp. E121. The structures were elucidated on the basis of their 1D and 2D NMR spectra and mass spectrometric data. The possible biosynthetic pathway of compounds 1 , 2 , 3 and 4 in Nigrospora sp. E121were reported according to literature. The phytotoxic assay results indicated that the acetyl fragment in α-acetylorcinol may contribute to the phytotoxic activity of this compound.

Abstract:Chemical investigation of the ethyl acetate extract of Corynespora cassiicola, isolated from leaf tissues of the Chinese mangrove medicinal plant Laguncularia racemosa, yielded four new secondary metabolites, including three decalactones, xestodecalactones DF (13) as well as corynesidone C (4), in addition to four known compounds. The structures of the new compounds were determined on the basis of one- and two-dimensional NMR spectroscopy as well as by high-resolution mass spectrometry. Absolute configurations of the optically active compounds 13 were determined by TDDFT ECD calculations of their solution conformers, proving that they belong to the (11S) series of xestodecalactones, opposite to the (11R) configuration of the known xestodecalactones AC. All compounds were tested against a panel of human protein kinases. Among the isolated compounds, two inhibited several kinases such as IGF1-R and VEGF-R2 with IC50 values mostly in the low micromolar range.

Abstract:A new altersolanol derivative, 4-dehydroxyaltersolanol A (9), along with two known sesquiterpenoids, (S)-7′-hydroxyabscisic acid (7) and (S)-abscisic acid (8) were obtained from the endophytic fungus, Nigrospora oryzae, isolated from leaves of Combretum dolichopetalum. The host plant yielded six known compounds including ellagic acid (1), 3, 3′, 4-tri-O-methylellagic acid (2), arjunolic acid (3), 4′-dihydrophaseic acid (4), echinulin (5) and arestrictin B (6). Close structural similarities with regard to compounds 4, 7 and 8 were observed between the metabolites from the host plant and those of the endophytic fungus. Furthermore compounds 5 and 6 are related to alkaloids isolated from N. oryzae previously thus stressing the notion that some of the isolated plant metabolites may actually be of fungal origin. The structures of the isolated compounds were established by spectroscopic methods including 1D, 2D NMR, MS, and by comparison with the literature. 4-Dehydroxyaltersolanol A (9) and 3, 3′, 4-tri-O-methylellagic acid (2) showed cytotoxicity against L5178Y mouse lymphoma cells with IC50 values of 9.4 and 29.0μM, respectively.

Abstract:A new drimane sesquiterpene 11-methoxyl-danilol ( 1 ) was obtained from endophytic fungus Nigrospora chinensis of Gannan navel orange pulp. Its structure was established to possess a natural rarely-occurring tricyclic acetal fused ring system by means of spectroscopic data analyses. Meanwhile, five known compounds danilol ( 2 ), redoxcitrinin ( 3 ), euphorbol ( 4 ), ergosta-7,24(24')-dien-3 β -ol ( 5 ), and ergosta-4,6,8(14),22-tetraen-3-one ( 6 ) were also co-isolated in this fungus. The results of antibacterial and cytotoxic activity screenings showed that compound 5 displayed antibacterial activities against Staphylococcus aureus and MRSA (methicillin-resistant S. aureus ) with MIC value of 50 μg/mL. Abstract:Abstract In the search for new bioactive compounds from extremophilic actinomycetes, a new marine actinomycete strain, Nocardiopsis lucentensis sp. ASMR2 has been isolated and taxonomically identified from marine plants collected in the Red Sea at Hurghada coasts. A large-scale fermentation of the strain on modified rice solid medium was performed, followed by work-up and purification of the obtained extract using a series of chromatographic purifications, delivering the novel butenolide system 3′-hydroxy- N -(2-oxo-2,5-dihydrofuran-4-yl)propionamide ( 1a ) along with the naturally new 4-methoxy-2 H -isoquinolin-1-one ( 2 ). Furthermore, eight known bioactive compounds are also reported, namely, indole-3-carboxylic acid, indole-3-acetic acid, indole-3-acetic acid methyl ester, furan-2,5-dimethanol, tyrosol, glycerol linoleate, cyclo-(Tyr, Pro), and adenosine. The chemical structures of the new compounds ( 1a , 2 ) were confirmed by extensive one- and two-dimensional (1D and 2D) nuclear magnetic resonance (NMR) spectroscopy, electron ionization high resolution (EI-HR) mass spectrometry, and by comparison with literature data. The antimicrobial activity of the strain extract, as well as of compounds 1a and 2 , were studied using a panel of pathogenic microorganisms. The in vitro cytotoxicity of the bacterial extract and compounds 1a and 2 were studied against the human cervix carcinoma cell line (KB-3-1) and its multidrug-resistant subclone (KB-V1).

Abstract:Investigation of secondary metabolites from the mangrove endophytic fungus Talaromyces sp. SAF14 led to the isolation of two new polyketides, methyl ( R )-3-(6,8-dihydroxy-7-methoxy-1-oxoisochroman-3-yl)propanoate ( 1 ), ( R )-3-(5,8- dihydroxy-1-oxoisochroman-3-yl)propanoic acid ( 2 ), together with four known alkaloids ( 3 – 6 ). The planar structures of new compounds were elucidated by comprehensive analysis of HR-ESI-MS and NMR data. The absolute configurations were determined by comparison of the calculated ECD spectrum with the measured one. All the isolated compounds were tested for cytotoxic activities against three human cancer cell lines. The known beauvericin ( 3 ) exhibited strong cytotoxic activity against A549, MCF-7, and KB cell lines with IC 50 values of 5.36 ± 2.49, 1.96 ± 1.09 and 4.46 ± 0.68 μM, respectively.

Abstract:Three pairs of enantiomers (1–3)—the new 12R-aloesol (1a) and two new fatty acids (2 and 3)—and one new natural product (4) together three known compounds (5–7) were isolated from a coral-reef-derived Streptomyces sp. SCSIO 66814. Their structures were determined through extensive spectroscopic analysis, chiral analysis, and single-crystal X-ray diffraction data. Compounds 2 and 3 were presumed to be intermediates for further generating homononactic acid (5) and nonactic acid, and the latter two molecules were able to act as precursors to form macrotetrolides with remarkable biological activity. The isolation of related precursors, compounds 2–5, provided more evidence to support the proposal of a plausible biosynthetic pathway for nonactic acid and its homologs. Additionally, (+)-1 exhibited a weak activity against DPPH radicals.

Abstract:Four new p-aminoacetophenonic acids, named (2E)-11-(4'-aminophenyl)-5,9-dihydroxy-4,6,8-trimethyl-11-oxo-undec-2-enoic acid (1), 9-(4'-aminophenyl)-3,7-dihydroxy-2,4,6-trimethyl-9-oxo-nonoic acid (2), (2E)-11-(4'-aminophenyl)-5,9-O-cyclo-4,6,8-trimethyl-11-oxo-undec-2-enoic acid (3) and 9-(4'-aminophenyl)-3,7-O-cyclo-2,4,6trimethyl-9-oxo-nonoic acid (4), were isolated from an endophyte Streptomyces sp. (strain HK10552) of the mangrove plant Aegiceras corniculatum. The structures of 1-4 were elucidated by using spectroscopic analyses. The relative stereoconfigurations of compounds 3 and 4 were determined by NOESY experiments. In the bioassay test, 1-4 showed no cytotoxicity against the Hela cell lines. Compound 4 also showed no inhibitory bioactivity on HCV protease and SecA ATPase and wasn't active against VSVG/HIV-luc pseudotyping virus.

Abstract:Two new cyclic depsipeptides, W493 C (1) and D (2), along with two known derivatives W493 A (3) and B (4) were obtained from the endophytic fungus Fusarium sp. isolated from the Mangrove plant Ceriops tagal. The structures of the new compounds were determined on the basis of one- and two dimensional NMR and high-resolution mass spectroscopic data. The absolute configurations of the amino acid residues of 1 and 2 were confirmed by application of Marfey's method. W493 A (3) and B (4) exhibited moderate activity against the fungus Cladosporium cladosporiodes and weak antitumor activity against the human ovarian cancer cell line A2780.
