Establishment and Optimization of SSR-PCR Reaction System in Flax

Abstract:The factors which affected the SSR-PCR reaction results were studied in the experiment,the optimum SSR reaction system of flax was established,it was showed as follows:total volume of reaction was 20 μL,the optimum concentrations of Mg2+ and dNTPs were 1.9 mmol·L-1 and 0.55 mmol·L-1 respectively,moreover,1.5 U Taq DNA polymerase,50 ng·μL-1 template DNA 100ng,25 ng·μL-1 primer 75 ng.To evaluate the SSR system,10 different flax germplasm were used and tested by 1.5 % agarose gel,the PCR results showed that this SSR reaction system was steady and reproducible.
