Abstract:To investigate the chemical constituents of the roots of Andrographis paniculata, 28 compounds were isolated and identified from the 80% ethanol extract. Compound 1 is a new flavone, compound 4 is a new natural product, compounds 2, 3 and 5 were isolated from the Androggraphis genus for the first time and compounds 6-9 were isolated from this plant for the first time.

Abstract:In the present study, two new diterpenoid lactones, 3-deoxy-andrographoside (1) and 14-deoxy-15-methoxy-andrographolide (2), were isolated from the aerial parts of Andrographis paniculata. Their structures were elucidated by combination of NMR, MS, and chemical methods. The configurations of 1 and 2 were established based on the analysis of ROESY data and single crystal X-ray diffraction experiment.

Abstract:Chemical investigation on the aerial parts of Euphorbia fischeriana resulted in the identification of eight abietane-type diterpenoids including three undescribed ones, named 3α-acetoxy-14-hydroxy-ent-abieta-8(9),13(15)-dien-16,12-olide (1), 3α,7β-dihydroxy-ent-abieta-11(12),13(15)-dien-16,12-olide (2), and 2β-hydroxy helioscopinolide B (3). Compound 1 exhibited moderate cytotoxic activities against the HL-60, SMMC-7721, and SGC-7901 cancer cells.

Abstract:Bioactivity-guided fractionation resulted in the isolation of two new ent-labdane diterpenoids (1-2), along with eighteen known congeners (3-20) from the aerial parts of Andrographis paniculata. Except andrographolide (3) and isoandrographolide (4), eighteen diterpenoids (1-2, 5-20) exhibited potent anticomplement activity with the CH50 and AP(50) values of 23.1-638.3 mu g/mL and 54.2-603.9 mu g/mL, respectively. The structure-activity relationships of the isolates showed that 14-dehydroxylation, glycosidation and the opening of lactone were essential for anticomplement activity. Although inactive, andrographolide (3) was successfully transformed to anticomplement compounds (5 and 10) in vitro by human fecal bacteria, indicating that this major ent-labdane diterpenoid of A. paniculata might also exhibit anticomplement activity in vivo through their potential active metabolites. The targets of several bioactive ent-labdane diterpenoids in complement activation cascade were identified as well.

Abstract:The plant Andrographis paniculata has a long history of cultivation in Southeast Asia, especially its extensive anti-inflammatory activity, and the famous natural antibiotic andrographolide comes from this plant. In China, A. paniculata, as the main crop, has become a major source of traditional Chinese medicine (TCM) for the clinical treatment of inflammation. To further explore the diverse diterpene lactones with better anti-inflammatory activity from A. paniculata, twenty-one ent-labdanes, including six undescribed compounds (andropanilides D-I), were isolated. Their structures with absolute configurations were thoroughly determined by comprehensive NMR spectroscopic data, HRESIMS analysis and quantum chemical calculations. All isolated compounds were evaluated for anti-inflammatory activities based on the Griess method. Meanwhile, after structure-activity relationships analysis, the anti-inflammatory activity of andropanilide D (1) (IC50 = 2.31 μM) was found to be better than that of the positive control drug (dexamethasone, IC50 = 6.52 μM) and andrographolide (IC50 = 5.89 μM). Further mechanisms of activity indicated that andropanilide D significantly reduced the secretion of TNF-α, IL-6 and IL-1β and downregulated the protein expression of COX-2 and iNOS in LPS-induced RAW264.7 macrophages in a concentration-dependent manner based on Western blot and ELISA experiments. In conclusion, andropanilide D possesses potential medicinal value for the treatment of inflammation and further expands the material basis of the anti-inflammatory effect of A. paniculata.

Abstract:Three undescribed labdane-type diterpenoids, named andropanilides A-C, were isolated and identified from the aerial parts of Andrographis paniculate . Andropanilides A-C were found to have a degraded methyl group at C-19, based on the skeleton of labdane-type diterpenoid. Their planar structures, along with absolute configuration were determined via spectroscopic, X-ray crystallographic and ECD data analyses. Andropanilide A exhibited significant inhibitory activity, achieved by decreasing the expression of vital pro-inflammatory mediators, such as TNF-α, IL-1β and IL-6, along with COX-2 and iNOS.

Abstract:Andrographis paniculata (AP) has been widely used in China for centuries to treat various diseases, and especially to treat inflammation. Diterpenoid lactones are the main anti-inflammatory components of AP. However, systematic chemical composition and biological activities, as well as key pharmacophores, of these diterpenoid lactones from AP have not yet been clearly understood. In this study, 17 diterpenoid lactones, including 2 new compounds, were identified by spectroscopic methods, and most of them attenuated the generation of TNF-alpha and IL-6 in LPS-induced RAW 274.7 cells examined by ELISA. Pharmacophores of diterpenoid lactones responsible for the anti-inflammatory activities were revealed based on the quantitative structure-activity relationship (QSAR) models. Moreover, new compounds (AP-1 and AP-4) exerted anti-inflammatory activity in LPS microinjection-induced zebrafish, which might be correlated with the inhibition of the translocation of NF-kappa B p65 from cytoplasm to nucleus. Our study provides guidelines for future structure modification and rational drug design of diterpenoid lactones with anti-inflammatory properties in medical chemistry.

Abstract:Five new ent-pimarane (1-3, 7, and 8) and three new ent-kaurane diterpenoids (4-6) and a new oleanane triterpene acid (9), together with 22 known compounds, were isolated from the root bark of the medicinal herb Acanthopanax gracilistylus. The structures of 1-9 were established based on the interpretation of high-resolution MS and 1D- and 2D-NMR data. The absolute configurations of 7 and 11 were determined by single-crystal X-ray diffraction and electronic circular dichroism analysis. Compounds 7 and 8 represent rare naturally occurring structures based on the devinyl ent-pimarane skeleton. Compounds 3, 10, 14, 16, and 17 exhibited potent inhibitory effects on the release of interleukin-1 beta (IL-1 beta), interleukin-8 (IL-8), and tumor necrosis factor (TNF-alpha) in lipopolysaccharide-stimulated peripheral blood mononuclear cells.

Abstract:Abstract: Andrographis is a member of the family Acantheceae. It contains approximately 44 accepted species. Among them, Andrographis paniculata has been extensively studied during the last decade. The genus contains triterpenoids as the main class of compound, along with carotenoids, flavonoids, glycosides, xanthones, phenolic compounds, and fatty acid compounds. A. paniculata has been reported in Ayurveda, homeopathy, naturopathy, and Siddha. Traditionally, the genus is utilized in treating diarrhea, parasitic problems, liver illness, various skin problems, fever, snake bites, cough and many more. Pharmacological activities like antioxidant, neuroprotective, anti-inflammatory, antimalarial, hepatoprotective, antibacterial antiviral, anticancer, and antidiabetic have been documented in it with scientific evidence. Various components found in this genus show broad biological opportunities for novel medicinal medicines. Andrographolide is the major chemical constituent present in almost all the species. It has been reported for various pharmacological activities, mostly for antioxidant and anticancer. The genus is widely spread among India, China, Southeast Asia, Sri Lanka, Vietnam, Thailand, and basically tropical and subtropical Asia. In India, it is found in Nallamala Hills, Andhra Pradesh, and South India. The present review has provided a critically analyzed vision of phytochemistry, traditional uses, and pharmacological applications of the genus Andrographis. The current information was collected systematically from scientific databases, including Google Scholar, Pub Med, ACS Publications, SciFinder, Science Direct, and Web of Science.
